This study was conducted to investigate physicochemical and microbiological properties during composting process when pig manure was composted with some bulking agents(Saw dust, Rice hull, Crushed wood). The pig manure consisted of 2.95% total nitrogen, 4.55% $P_2O_5$, 2.07% $K_2O$, 81.2% organic matter and 14.0 C/N, dry base. The inorganic content of bulking agents were similar one another, and C/N ratio was Saw dust 392. Rice hull 91.5, Crushed wood 266. The temperature of Saw dust composting slowly increased at initial stage, whereas one of Rice hull and Crushed wood rapidly increased and stabilized similarly to outdoor temperature after about 2 month. The pH of compost increased during initial 1 month after decreasing, but thereafter decreased rapidly. The C/N ratio rapidly decreased at initial stage, and it slowly decreased after 1 month. Total nitrogen somewhat increased according to composting process. Ammonium-nitrogen increased until 60th day, and thereafter it was rapidly inclined to decrease. The population of aerobic bacteria, Bacillus spp., actinomycetes and fungi increased up to > $10^{10}cfu\;g^{-1}$ fresh weight, > $10^7$, > $10^8$ and > $10^6$, respectively in Rice hull and Crushed wood composting, and more than ones in Saw dust composting, and specially actinomycetes. The persuasive Bacillus spp. isolated from composting process were B. lentimorbus, B. licheniformis, B. pumilus, B. megaterium, etc.
1) Many bacterial strains identified as Bacillus megeterium, Bacillus subtilis and Bacillus licheniformis were aboundantly found in summer jokal kimchi, but the most dominant strains in summer kimchi were Lactobacillus plantarum and Loctobacllus buchneri. 2) Bacillus groups found in kimchi were sensitive in a low concentration of AF-2, but groups of lactic acid bacteria were resistant to a high concentration of AF-2. 3) Allowable concentration of AF-2 in Korean kimchi is less than 10 p.p.m. 4) AF-2 was not suitable for the juicy kimchi as a preservative because the color of juicy kimchi was somewhat changed into orange red when 10 p.p.m. of AF-2 was added. 5) High concentration of AF-2 leads the hetero-fermentation of kimchi bacteria to the homofermentation. 6) Microflora of kimchi was influenced even in the concentration of 10 p.p.m. but it was impossible to check the acidification of kimchi in summer with 50 p.p.m. concentration of AF-2. 7) About 25% of AF-2 was consumed in kimchi fermentation for day at $23^{\circ}-25^{\circ}C$.
Asian dust storms originating in the arid desert of China and Mongolia usually occur from late winter through spring, and more than one million tons of dust per year is transported to the Korean Peninsula by the prevalent westerly winds. We supposed that these dust particles could include bioaerosols and act as carriers of microorganisms. In order to clarify the dynamics of microorganisms moving with these particles, the concentration and composition of microorganisms associated with settled particles were compared between samples collected during Asian dust events and those under non-dust periods. From February to April 2008, settled dust particles were collected at one location in Ulsan using rainfall meter of 200 mm diameter. During this period, there was one Asian dust event in Ulsan. The bacterial concentrations were higher in samples collected during Asian dust event than those under non-dust period, whereas fungal concentrations were rather similar regardless of the Asian dust event. We analyzed 16S rRNA gene sequences of 45 bacterial isolates obtained from the settled particle samples. These isolates belonged to either genus Bacillus or genus Streptococcus and were tentatively identified as B. amyloliquefaciens, B. aryabhattai, B. atrophaeus, B. licheniformis, B. megaterium, B. methylotrophicus, B. pumilus, B. sonorensis, B. subtlis, B. vallismortis, S. epidermidis, and S. succinus. In cases of fungal isolates, genera such as Mucor, Alternaria, Cladosporium, and Aspergillus were tentatively identified from samples collected at both Asian dust and non-Asian dust periods. It appears that endospore-forming bacteria such as Bacillus sp. rather than fungal spores are more likely to be associated with Asian dust particles.
For the synthesis of various pharmaceuticals, chiral alcohols are useful intermediates. Among them, (R)-ethyl-4-chloro-3-hydroxybutanoate ((R)-ECHB) is an important building block for the synthesis of L-carnitine. (R)-ECHB is produced from ethyl-4-chloro-3-oxobutanoate (ECOB) by a reductase-mediated, enantioselective reduction reaction. The Saccharomyces cerevisiae YOL151W reductase that is expressed in Escherichia coli cells exhibited an enantioselective reduction reaction toward ECOB. By virtue of the C-terminal His-tag, the YOL151W reductase was purified from the cell-free extract using Ni2+-NTA column chromatography and immobilized onto Ni2+-magnetic microparticles. The physical properties of the immobilized reductase (Imm-Red) were measured using electron microscopy, a magnetic property measurement system, and a zeta potential system; the average size of the particles was approximately 1 μm and the saturated magnetic value was 31.76 emu/g. A neodymium magnet was used to recover the immobilized enzyme within 2 min. The Imm-Red showed an optimum temperature at 45℃ and an optimum pH at 6.0. In addition, Bacillus megaterium glucose dehydrogenase (GDH) was produced in the E. coli cells and was used in the coupling reaction to regenerate the NADPH cofactor. The reduction/oxidation coupling reaction composed of the Imm-Red and GDH converted 20 mM ECOB exclusively into (R)-ECHB with an e.e.p value of 98%.
human tyrosinase (hTyr) catalyzes first and the rate limiting step in the synthesis of polymerized pigment, melanin which determines skin, hair and eye colors. Mutation of hTyr often brings about decrease of melanin production and further albinism. Meanwhile, a number of cosmetic companies providing skincare products for woman in Asia-Pacific region have tried to develop inhibitors to bright skin color for several decades. In this study, we built a 3D structure by comparative modeling technique based on the crystal structure of tyrosinase from bacillus megaterium as a template to serve structural information of hTyr. According to our model and sequence analysis of type 3 copper protein family proteins, two copper atoms of active site located deep inside are coordinated with six strictly conserved histidine residues coming from four-helix-bundle. In addition, protein-substrate and protein-inhibitor complex were modeled with the guide of van der waals surface generated by in house software. The enzyme expressed from strain L-49 was 2.01 times higher than that of original strain. Strain L-49 can grow on culture plate with $50{\mu}g/mL$ ampicillin. The synthesis of $\alpha$-amylase was greatly suppressed when strain L-49 was grown on monosaccharide such as glucose and polysaccharide at the same time cell concentration was low. Amylase production was enhanced when the bacterium was grown on starch and dextrin. Among different nitrogen sources tried, yeast extract was found to be the best followed by panpeptone, peptone, meat extract, bean meal, and corn steep liquor. The average rate of enzyme production was enhanced for 3~4 times in fermentation time from 24h to 44h. The sugar uptake rate has also increased. Low oxygen supply rate enhanced the rate of strain propagation but depressed the enzyme production. Hence it is benefit to obtain high enzyme activity that agitation speed maintained not lower than 400r/min and aeration rate maintained greater than 1:1vvm.
A maltopentaose-producing amylase (G5-amylase) from Bacillus megaterium KSM B-404 was applied to retard bread retrogradation. Retrogradation rates were determined by differential scanning calorimetry. Gel permeation chromatography determined changes in maltooligosaccharide composition and the molecular weight profiles of carbohydrate tractions. The baking process produced maltopentaose and maltotriose by the hydrolysis of starch molecules into small units. Amylose and amylopectin degradation as well as maltooligosaccharides produced by the enzyme were likely responsible for retarding starch retrogradation. Overall, addition of G5-amylase reduced the starch retrogradation rate, and was as effective as Novamyl(R), a commercial enzyme.
Amongst the various antimicrobial peptides, lysozyme plays a central role in initiating and maintaining the antibacterial defense response of insect. Here we propose the biosynthesis and refolding of recombinant lysozyme in Escherichia coli expressed in inclusion body form. The Agrius lysozyme gene was amplified using gene specific primers and then ligated into the pGEX-4T-1 vector, which contained the glutathione S-transferase (GST) gene as a fusion partner. A recombinant lysozyme was expressed in E. coli Rosetta cells using a pGEX-4T-1 expression vector, and the fusion protein was induced by ioporpyl-${\beta}$-D-thiogalactopyranoside (IPTG). The recombinant protein produced as an inclusion body was resolubilized in solubilization buffer, and the resultant solution was dialyzed in refolding buffer. After thrombin cleavage, the recombinant lysozyme was purified by ion exchange chromatography and reverse phase chromatography. The recombinant lysozyme was subjected to sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) analysis and immunoreactivity against the anti-Agrius lysozyme was observed by western blot analysis of this protein. The recombinant lysozyme displayed antibacterial activity against Bacillus megaterium and Micrococcus luteus, which was confirmed by the inhibition zone assay.
콩의 배당체 isoflavone류를 생리 활성형 aglycone으로 전환율을 증진시켜 콩 발효식품의 기능성을 증진시키고자, 식용버섯 균사체와 청국장 발효균 B. megateriw SMY-212를 이용하여 단독과 2단계 배양한 발효대두의 메탄올 추출물에 대한 항산화능과 인체 전립선암과 자궁암 세포주의 성장억제 효과를 조사하였다. 2단 발효대두 추출물의 수소공여능 효과는 1,000 $\mu$g/mL 농도에서 17.8∼26.9%로서 단독 발효대두보다 30∼40% 높았으며, 그 중에 동충하초와 SMY-212를 배양한 것이 가장 높았다. Linoleic acid에 대한 항산화 효과는 단독 발효대두는 동충하초가 가장 높았으며, 2단계 배양한 발효대두는 팽이균사체와 SMY-212가 오히려 높게 나타났다. 인체 전립선암 세포주(DU145)와 자궁암 세포주(Hela)에 대한 성장억제 효과는 단독배양에 서 모두 팽이버섯 균사체를 이용한 발효대두가 가장 높았으며, 2단계 배양에서 DU145에서는 팽이버섯과 아가리쿠스 균사체 배양물에 SMY-212를 이용한 발효대두가 높았고, HeLa에서는 팽이버섯, 동충하초, 새 송이버섯 균사체의 추출물 순으로 높았다. 결론적으로 삶은 콩에 버섯 균사체의 단독배양보다는 버섯균사체와 SMY-212를 2단 배양한 발효대두 추출물이 암세포주 성장억제 효과가 높게 나타났다.
The inoculation of some microorganisms into a microcosm containing soil from a barren lakeside area at Lake Paro in Kangwon-do enhanced plant growth significantly. The direct and viable counts of soil bacteria and soil microbial activities measured by electron transport system assay and fluorescein diacetate hydrolysis assay were higher in inoculated soil. The plant growth promoting effect of this inoculation may be caused by phytohormone production and the solubilization of insoluble phosphates by the inoculated bacteria. Three inoculated strains of Pseudomonas fluorescens produced several plant growth promoting phytohormones, including indole-3-acetic acid (auxin), which was confirmed by thin layer chromatography and GC/MS. P. fluorescens strain B16 and M45 produced 502.4 and 206.1 mg/l of soluble phosphate from Ca3(PO4)2 and hydroxyapatite, respectively. Bacillus megaterium showed similar solubilization rates of insoluble phosphates to those of Pseudomonas spp. We believe that this plant growth promoting capability may be used for the rapid revegetation of barren or disturbed land.
