Studies on the Toxicity of $\delta$-endotoxin of Bacillus thuringiensis to the Several Tissues of Hyphantria cunea Drury (미국흰불나방(Hyphantria cunea Drury)에 대한 Bacillus thuringiensis 내독소단백질의 독성효과에 관한 연구)

  • Ultrastructural changes of tissues caused by Bacillus thuringiensis var. kurstaki $\delta$-endotoxin intoxication of Hyphantria cunea were observed by transmission electron microscopy. Bt $\delta$-endotoxin crystals induced the disruption of microvilli, vacuolation of cytoplasm, changes in the cisternae of the endoplasmic reticulum, disappearance of basal striations, loss of ribosomes, and changes in the configurations of mitochondria in the columnar cell of midgut. The fat body cells also showed spherical endoplasmic reticulum and distorted mitochondria, and then the cells were destroyed.

  • The toxic effect of Bacillus thuringiensis var, kurstaki $\delta$-endotoxin was determined in the midgut, fat body and Malpighian lobules from larvae of Hyphantria cunea using the au-toradiography and Western blot methods. Bt $\delta$-endotoxin crystals inhibited protein synthesis and elongation factor-2 activity of all tissues tested.

Biological Control of Fusarium Wilt by Antagonistic Microorganism in Greenhouse Grown Cucumber Plants (오이의 온실재배에서 발생하는 위조병의 미생물학적 제어)

  • This study was carried out to clarify the effects of antagonistic microorganism, Bacillus sp. JC181 isolated from the greenhouse soil grown cucumber plants on the growth inhibition of plant pathogen, fusarium wilt (Fusarium oxysporum) occurred in cucumber plants in greenhouse. Antagonistic bacterial strains were isolated and were investigated into the antifungal activity of the antagonistic microorganism against fusarium wilt. Screened fourteen bacterial strains which strongly inhibited F. oxysporum were isolated from thc greenhouse soil grown cucumber plants, and the best antagonistic bacterial strain designated as JC181, was finally selected. Antagonistic bacterial strain JC181 was identified to be the genus Bacillus sp. based on the morphological and biochemical characterization. Bacillus sp. JC181 showed 58.2% of antifungal activity against the plant pathogen growth of F. oxysporum. By the bacterialization of culture broth and heated filtrates of culture broth, Bacterial strain, Bacillus sp. JC181. showed 91.2% and 260% of antifungal activity against F. oxysporum, respectivrly.

Prediction of Lytic Segments from Bacillus thuringiensis var israelensis 130 kDa and 72 kDa Proteins

  • The amino acid sequences of 130 kDa and 72 kDa proteins responsible for the larvicidal activity of Bacillus thuringiensis var israelensis (Bti) were analyzed by hydrophobic moment plots. A search for highly amphiphilic $\alpha$-helices was made in these proteins using the helical hydrophobic moment as a criterion of amphiphilicity The protein segments of the largest hydrophobic moments were analyzed. In the present communication we report the surface seeking helices in 130 kDa and 72 kDa proteins of Bacillus thuringiensis var israelensis. It is assumed that the surface seeking segments may participate in one of the membrane-related functions of Bacillus thuringiensis.

Production Condition and Utilities of Extracellular Biopolymer from Bacillus licheniformis (Bacillus licheniformis가 생산하는 Extracellular Biopolymer의 생산조건 및 이용특성)

  • A bacterium that produce biopolymer was isolated from Gochujang, one of Korean traditional fermented foods, and identified as Bacillus licheniformis. The production of biopolymer was highest and 34mg/250ml, when the baterium was cultivated in condition of sucrose 6.0%, Yeast extract 0.1%, peptone 0.l%, NaCl 3.0%, and pH 6.0. The 1% solution of this biopolymer was able to form a translucent and glossy film. And the solution of biopolymer was found to precipitate Kaolin solution and also stabilize the suspension of lactic acid bacteria within the test range of 0.25-1.00%.   • Ten(10) strains of aerobic bacteria and two(2) strains of microaerobic bacteria were isolated out of soysauce which was brewed by the conventional process. The following bacteria were identified by studying their morphorogies and physiological characters. Bacillus pumillus-R-2; Bacillus subtilis var., aterimus-S-1; Bacillus licheriformis var-S-2; Bacillus subtilis-T-1; Sarcina maxima-T-2; Pediococcus acidi lactici-Z-2; Bacillus citreus var, . soyaB-Z-5. T-2 and Z-5 of the isolated bacteria were found good in growth even in the 24%-salted density, and Z-5 was more vigorous than T-2 though stinking. S-1 produced black-brown pigment from the medium containing various kinds of carbohydrate and the medium of soysauce which are available to S-1.

  • Beneficial bacteria, which have been used for medical purpose and for medicines for treating intestinal disorders, include strains of Bifidobacterium sp., Lactobacillus sp., Enterococcus sp., Clostridium butyricum, Lactobacillus sporogenes, Bacillus subtilis, Bacillus polyfermenticus and the like. Bacillus polyfermenticuss SCD with is commonly called as Bispan strain has been appropriately used for the treatment of long-term intestinal disorders, since the live strains in the form of active endospores can successfully reach the target intestine. In this study, the identification and characterization of Bispan strain was done using SEM observation, API 50CHB kits, isoprenoid quinone analysis, and fatty acid analysis. These results suggest that Bispan strain is very similar to Bacillus subtilis.

  • The complete nucleotide sequence of alkalophilic Bacillus sp. K-17 $\beta$-xylosidase gene and its flanking regions were established. A 1263-bp of an open reading frame for $\beta$-xylosidase was observed. The molecular weight (50, 521 dalton), deduced from the nucleotide sequence of $\beta$-xylosidase gene, agreed with the result obtained by SDS-polyacrylamide gel electrophoresis of the purified enzyme (51, 000 dalton). The Shine-Dalgarno sequence, 5'-GAGGAGG-3', was found 8 bp upstream of the initiation codon ATG. The -10 sequence (TAAAAT) in the promoter region for $\beta$-xylosidase gene was similar to the consensus sequence for Bacillus subtilis RNA polymerase, whereas the -35 sequence (TCGATCA) different from all the known -35 regions in the promoter for Bacillus subtilis RNA polymerase.

  • An extracellular fibrinolytic-enzyme-producing bacterium was isolated from Doen-Jang, a Korean traditional fermented flood, and identified as Bacillus sp. DJ based on its morphology and cellular fatty acid composition. The total extracellular fibrinase (EF) from Bacillus sp. DJ was analyzed using three fibrin zymographic techniques, SDS-fibrin zymography (SDS-FZ), isoelectrofocucing-fibrin zymographs(IEF-FZ), and a two-dimensional SDS-fibrin zymographic analysis (2D SDS-FZ). As a result, the EP map of Bacillus sp. DJ was established. The results suggest that the 2D SDS-FZ method will be a useful tool for the proteomic approach for many other bacterial pretenses.

  • Biocontrol of plant pathogens provides an alternative means of reducing the incidence of plant diseases without the negative aspects of chemical pesticides. Nowdays, as the resistant fungi about the chemical fungicides have revealed and the concern of environment has increased, the biological control of phytopathogenic fungi by the antagonistic microorganisms is very much indispensable. For the selection of strong antagonistic bacterium for biological control agent of rice leafblast and cucumber gray mold rot, the antifungal strain KL-3 strain was selected among 120 strains isolated from the rhizosphere soils. And the strain was identified to be a species of Bacillus subtilis or closely related strain. In several biochemical and in vitro antibiosis tests, antifungal substances of Bacillus sp. KL-3 were presumed heat stable, micromolecular antibiotic substances. In vivo test and vinyl house field test, the antifungal substances of Bacillus sp. KL-3 represented excellent biocontrol ability aganist Alternaria mali, Phyricularia oryzae, and Alternaria kikuchiana as well as broad spectrum of other fungi. In particular, Bacillus sp. KL-3 strain showed more predominant activity than some chemical fungicides against fungi shown to resist chemcal fungicides.
