The Influence of the Nucleotide Sequences of Random Shine-Dalgarno and Spacer Region on Bovine Growth Hormone Gene Expression

  • To investigate the effects of the nucleotide sequences in Shine-Dalgarno (SD) and the spacer region (SD-ATG) on bovine growth hormone (bGH) gene expression, the expression vectors under the control of the T7 promoter (pT7-7 vector) were constructed using bGH derivatives (bGH1 & bGH14) which have different 5'-coding regions and were induced in E. coli BL21 (DE3). Oligonucleotides containing random SD sequences and a spacer region were chemically synthesized and the distance between the SD region and the initiation codon were fixed to nine bases in length. The oligonucleotides were annealed and fused to the bGH1 and bGH14 cDNA, respectively. When the bGH gene was induced with IPTG in E. coli BL21(DE3), some clones containing only bGH14 cDNA produced considerable levels of bGH in the range of $6.9\%\;to\;8.5\%$ of total cell proteins by SDS-PAGE and Western blot. Otherwise, the bGH was not detected in any clones with bGH1 cDNA. Accordingly, the nucleotide sequences of SD and the spacer region affect on bGH expression indicates that the sequences sufficiently destabilize the mRNA secondary structure of the bGH14 gene. When the free energy was calculated from the transcription initiation site to the +51 nucleotide of bGH cDNA using a program of nucleic acid folding and hybridization prediction, the constructs with values below -26.3 kcal/mole (toward minus direction) were not expressed. The constructs with the original sequence of bGH cDNA also did not show any expression, regardless of the free energy values. Thus, the disruption of the mRNA secondary structure may be a major factor regulating bGH expression in the translation initiation process. Accordingly, the first stem-loop among two secondary structures present in the 5'-end region of the bGH gene should be disrupted for the effective expression of bGH.

Expression of Bovine Growth Hormone Gene in a Baculovirus, Hyphantria cunea Nuclear Polyhedrosis Virus

  • Bovine growth hormone (bGH) gene was expressed in an insect Spodoptera frugiperda cell line using a Baculovirus, Hyphantria cunea nuclear polyhedrosis virus (HcNPV). The bGH gene in pbGH plasmid was sequenced and amplified by PCR technique with two primers containing NcoI sites. The bGH gene consisted of 654 bp (217 amino acid residues), the 5'-untranslated region of the cloned bGH cDNA contains 56 bp, and the 3'-untranslated region contains 145 bp and two pallindromic regions. The amplified bGH gene DNA fragment (654 bp) was inserted into the NcoI site of the pHcEVII vector, which was named pHcbGH. The pHcbGH transfer vector DNA and the wild type HcNPV DNA were cotransfected into S. frugiperda cells to construct a recombinant virus. Eight recombinant viruses were selected and named HcbGH. One clone, HcbGH-4-1 showed largest plaque size, therefore the recombinant virus was further studied. The multiplication pattern of the recombinant HcbGH-4-1 was similar to that of the wild type HcNPV. The bGH gene DNA in the HcbGH-4-1 recombinant was confirmed by Southern blot hybridization. The amount of the bGH (217 amino acid residues, 21 kDa) produced in S. frugiperda cells infected with the HcbGH-4-1 recombinant was approximately 5.5 ng per ml ($10^6$ cells) by radioimmunoassay.

  • Improved hypophysectomy technique and the efficacy of bGH in hypophysectomized female rats were studied as a basis for the function of endocrine gland and hormones. 1. With improved sublaryngeal a, pp.oach by hypophysectomy, 34 rats out of 40 were successfully hypophysectomized(more than 80% of success). The surgical time for hypophysectomy per rat was taken about 3.0$\pm$1.0 min. 2. For the effect of hypophysectomy and bGH on body weight gain among the 5 gruops of A, B, C, D, and E, the weight gains for A, B, C, D, and E, were 93g, 9.2g, 10.7g, 25.1g and 36.2g, respectively, which implied that hypophysectomy had a major effect on body weight. And also, the effect of bGH was paralleled with the level of bGH. There were significant difference between control and hypophysectomized groups, and between the bGH treated and bGH non-treated groups after hypophysectomy(p<0.01). 3. For the effect of hypophysectomy and bGH on the length of femur, tibia, humerus, and anterbrachi, longest was for A, shortest for B, and longer in order of C, D, and E. The A group was significantly longer than B, C, D, and E groups. The B group was shorter than C, D, and E groups(p<0.01). 4. For the effect of hypophysectomy and bGH on the weight of femur, tibia, humerus, and antebrachi, heaviest was for A, lightest for B, and heavier in order of C, D, and E. The A group was significantly heavier than B, C, D, and E groups. The B group was lighter than C, D, and E groups(p<0.01). 5. In changes of body length, and weights of liver and heart, A group was most drastically increased. Among the hypophysectomized groups, the change was increased in order of B, C, D, and E. Especially, the change of A group was significantly greater than B, C, D and E gruops. However, no significant differences were found among B, C, D, and E groups. 6. The hypophysectomy in rat had a significant effect on body growth. The effect of hormone was remarkably different depending upon the level of hormone. It was found that 300$\mu\textrm{g}$ of bGH/kg of body wt/day was slightly lower for normal growth.

  • To investigate the fidelity of transgene transmission and expression, we produced transgenic mice carrying bovine $\beta$-casein/bovine grwoth hormone(bGH) fusion gene and examined transmission efficiency and expression level of the transgene in the founders and their progeny. The transgene was composed of 1.8 kb bovine $\beta$-casein promoter and 2.1 kb bGH gene. Ten transgenic mice were produced. Milk and mammary gland were collected from eight transgenic lines at 10-day lactation and a, pp.ied to Western and Northern blot analyses. The bGH expression was detected in four of them. The concentrations of bGH in milk were highly variable from 4$\mu\textrm{g}$/ml to 600$\mu\textrm{g}$/ml depending on the lines. The bGH mRNA level in mammary gland was closely correlated with the bGH concentration in milk in each transgenic line. These results indicated that bGH transgene expression was a, pp.opriately regulated in the mammary gland and secreted into milk in transgenic mice. By using two transgenic lines(#2, #7) secreting a considerable amoung of bGH into their milk, the inferitance and maintenance of transgenic phenotype were assessed in successive four generations. The mean transmission frequencies of transgene in lines #2 and #7 were 34% and 40%, respectively. The bGH concentration in milk were 80, 240, 120, 60$\mu\textrm{g}$/ml in each G0(generation 0), G2, G3, G4 generation of line #2 and 600, 1600, 860, 900$\mu\textrm{g}$/ml in each G1. G2, G3, G4 generation of line #7. These results demonstrated that bovine $\beta$-casein/bGH gene was stably transmitted from generation to generation in a Menelian fashion in trasgenic mice and consistenly expressed in their milk throughout the generations, although there was a little variation in the transmission frequency and expression level of the transgene between generations.

Cellvibrio sp. KY-GH-1의 아가로오스 당화 관련 엑소형 GH50A β-아가레이즈와 GH117A α-NABH의 특성 및 NA2와 L-AHG 양산에의 적용 가능성 (Characterization of Exolytic GH50A β-Agarase and GH117A α-NABH Involved in Agarose Saccharification of Cellvibrio sp. KY-GH-1 and Possible Application to Mass Production of NA2 and L-AHG)

  • 최근, 본 연구진은 담수 환경 유래 한천 분해 세균인 Cellvibrio sp KY-GH-1 (KCTC13629BP)의 전체 유전체 염기 서열을 분석하여 아가로오스를 L-AHG 및 D-갈락토오스로 가수분해시키는 아가레이즈들을 암호화하는 유전 정보를 탐색하였다. 그 결과, KY-GH-1 균주는 유전체 상의 약 77 kb 길이의 아가레이즈 유전자 클러스터 내에 9개의 β-아가레이즈 유전자들과 2개의 α-네오아가로비오스 가수분해효소(α-NABH) 유전자들을 지닌 것으로 나타났다. 이러한 유전자 정보를 바탕으로 KY-GH-1 균주가 한천을 탄소원으로 자화하기 위해 단량체인 L-AHG와 D-갈락토오스로 분해시키는 공정은, 엔도형 GH16 β-아가레이즈, 엔도형 GH86 β-아가레이즈 등에 의해 개시되어 NA4, NA6, NA8 등을 생성시킨 후, 이들에 대해 엑소형 GH50 β-아가레이즈가 추가로 작용하여 NA2를 생성시키고, 이어서 GH117 α-NABH가 작용하여 생성된 NA2를 단량체 L-AHG와 D-갈락토오스로 분해함으로써 종결되는 것으로 예측되었다. 대장균 발현 시스템과 PET-30a 벡터를 함께 사용하여, KY-GH-1 균주 유래의 GH50 패밀리 β-아가레이즈 유전자들(GH50A, GH50B, GH50C)과 GH117 패밀리 α-NABH 유전자들(GH117A α-NABH, GH117B α- NABH)을 발현시켜 His-태그 재조합 효소단백질들로 확보하여, 이들 효소단백질을 이용하여 효소 활성을 비교 분석한 결과, 재조합 GH50A β-아가레이즈가 세 개의 GH50A 패밀리 β-아가레이즈 동위효소들 중에서 가장 높은 엑소형 β-아가레이즈 활성을 나타내며, 또한 재조합 GH117A α-NABH가 NA2를 L-AHG와 D-갈락토오스로 강력하게 가수분해할 수 있으나 재조합 GH117B α-NABH는 NA2 가수분해 활성이 없음을 확인하였다. 연이어 GH50A β-아가레이즈 및 GH117A α-NABH의 효소 특성을 추가로 조사하였다. 아울러 이들 각 효소가 나타내는, 아가로오스를 분해하여 NA2를 생성시키는 효율성과 NA2를 분해하여 L-AHG 및 D-갈락토오스를 생성시키는 효율성을 평가하였다. 본 총설에서는, L-AHG 및 D-갈락토오스의 양산을 위한 아가로오스의 효소적 가수분해에 성공적으로 활용될 수 있을 것으로 기대되는, 담수 유래 한천 분해 세균 Cellvibrio sp. KY-GH-1 유래의 재조합 GH50A β-아가레이즈 및 GH117A α-NABH의 장점들에 대해 기술한다.

  • In order to search for the effects of Shine-Dalgarno (SD) sequence and nucleotide sequence of spacer region (SD-ATG) on bGH expression, oligonucleotides containing random SD sequences and a spacer region were chemically synthesized. The distance between SD region and initiation codon (ATG) was fixed to 9 nucleotides in length. The expression vectors have been constructed using pT7-1 vector containing a T7 promoter. Positive clones were screened with colony hybridization and named pT7A or pT7B plasmid series. The selected clones were confirmed by DNA sequencing and finally, 19 clones having various SD combinations were obtained. When bovine growth hormone was induced by IPTG in E. coli BL21(DE3), all cells harboring these plasmids produced a detectable level of bGH in western blot analysis. However, various SD sequences did not affect on bGH expression, indicating that the sequences of SD and the spacer region did not sufficiently destabilize mRNA secondary structure of bGH gene. Therefore, these results indicate that the disruption of mRNA secondary structure might be a major factor for regulating bGH expression in the translational initiation process.

The Effect of Growth Hormone on mRNA Expression of the GABAB1 Receptor Subunit and GH/IGF Axis Genes in a Mouse Model of Prader-Willi Syndrome

  • Purpose: Growth hormone (GH) therapy substantially improves several cognitive functions in PWS. However, the molecular mechanisms underlying the beneficial effects of GH on cognition remain unclear in PWS. In this study, we investigated the effects of recombinant human GH on the gene expression of GABAB receptor subunits and GH/insulin-like growth factor (IGF) axis genes in the brain regions of PWS-mimicking mice (Snord116del). Methods: Snord116del mice were injected subcutaneously with 1.0 mg/kg GH or saline, once daily for 7 days. The collected brain tissues were analyzed for mRNA content using quantitative PCR (qPCR) in the cerebellum, hippocampus, and cerebral cortex. Results: GH increased the mRNA expression level of the $GABA_{B1}$ receptor subunit ($GABA_{BR1}$) and IGF-1R in the cerebellum. Furthermore, a significant positive correlation was found between the level of $GABA_{BR1}$ mRNA and the expression of the IGF-1R transcript. GH also induced an increase in the mRNA expression of IGF-2 and IGF-2R in the cerebellum. Conclusion: These data indicate that GH may provide beneficial effects on cognitive function through its influences on the expression of $GABA_{BR1}$ and GH/IGF-1 axis genes in PWS patients.

  • Over an additive abelian group G of order g and for a given positive integer $\lambda$, a generalized Hadamard matrix GH(g, $\lambda$) is defined as a gλ$\times$gλ matrix[h(i, j)], where 1 $\leq i \leqg\lambda and 1 \leqj \leqg\lambda$, such that every element of G appears exactly $\lambd$atimes in the list h($i_1, 1) -h(i_2, 1), h(i_1, 2)-h(i_2, 2), …, h(i_1, g\lambda) -h(i_2, g\lambda), for any i_1\neqi_2$. In this paper, we propose a new method of expanding a GH(g^m, \lambda_1) = B = [B_{ij}] over G^m$ by replacing each of its m-tuple B_{ij} with B_{ij} + GH(g, $\lambda_2) where m = g\lambda_2. We may use g^m/\lambda_1 (not necessarily all distinct) GH(g, \lambda_2$)s for the substitution and the resulting matrix is defined over the group of order g.

Interaction of Bovine Growth Hormone with Buffalo Adipose Tissue and Identification of Signaling Molecules in Its Action

  • Results on localization of growth hormone receptor (GHR), interaction of growth hormone (GH) with receptor in buffalo adipose tissue and identification of activated signaling molecules in the action of GH are presented. Bovine GH (bGH) was labeled with fluorescein or biotin. Fluorescein-labelled bGH was used for localization of GHRs in buffalo adipocytes. The receptors were present on the cell surface. The affinity of binding of GH to its receptor was determined by designing an experiment in which buffalo adipose tissue explants, biotinylated GH and streptavidin-peroxidase conjugate were employed. The affinity constant was calculated to be $2{\times}10^8M^{-1}$. The receptor density on adipose tissue was found to be 1 femto mole per mg of tissue. Signalling molecules generated in the action of GH were tentatively identified by employing Western blot and enhanced chemiluminescence techniques using anti-phosphotyrosine antibody. Based on molecular weights of proteins reactive to anti-phosphotyrosine antibody, three signaling molecules viz. insulin receptor substrate, Janus activated kinase (Jak) and mitogen activated protein were tentatively identified. These signaling molecules appeared in a time (incubation time of explants with growth hormone) dependent way. The activation of Jak2 was confirmed by employing anti-Jak2 antibody in a Western blot. The activation of Jak2 occurred during 5 min incubation of buffalo adipose tissue explants with GH and incubation for an additional period, viz. 30 min. or 60 min., resulted in a drastic reduction in activation. The results suggest that Jak2 activation is an early event in the action of GH in buffalo adipose tissue.

Effects of 3,5,3'-triiodo-L-thyronine ($T^3$) on Growth of GR-transgenic Coho Salmon, Oncorhynchus Kitsutch

  • GH-transgenic coho salmon (Oncorhynchus kitsutch) juveniles in tGH*T$_3$and tGH*PTU were fed with the diets containing 1 ug/g fish of 3,5,3'-triiodo-L-thyronine (T$_3$) and 30 ug/g fish of 6-n-propyl-2- thiouracil (PTU), respectively, to assess the effect of these drugs on the change of physiological activity, growth and survival rate in comparison with normal transgenic (tGH*C) and nontransgenic coho salmon (Wild) for 90 days. Although the daily food intakes of all transgenic (tGH)-groups were higher than Wild, the amount was reduced by exogenous PTU supply. The fred efficiencies of tGH-groups were lower than Wild, but the efficiency was reduced both by T$_3$and PTU. The survival rate of tGH-group was significantly higher than that of Wild, but there was no significant difference among tGH-groups. Although the growth of tGH-coho salmon was faster than Wild. the growth rate of transgenic salmon was increased by exogenous T$_3$, but was reduced by PTU Plasma TT$_4$levels of tGH-groups was approximately 2-fold higher relative to Wild, but there were no difference of plasma TT$_4$levels among tGH-groups. plasma TT$_3$level or tGH-coho salmon was increased by exogenous T$_3$administration, but was reduced by exogenous PTU. In addition, although plasma GH levels of all tGH-groups were higher than that of Wild, the GH level in plasma of transgenic coho salmon was increased by exogenous T$_3$and reduced by exogenous PTU. In the meantime, the transgenic fishes also displayed head, jaw and opercular abnormalities typical of the offsets of this gene construct in coho salmon, indicating that some imbalance in growth processes has been induced. However, the abnormalities of transgenic coho salmon was reduced following exogenous PTU administration.
