석회유황합제의 다중처리에 따른 '후지' 및 '홍로' 사과의 적과 효과 (Multiple Applications of Lime Sulfur for Fruit Thinning of 'Fuji' and 'Hongro' Apple Trees)

  • 본 연구는 2년간 '홍로'/M.9 및 '후지'/M.9 품종의 적과를 위한 석회유황합제 및 어유와의 혼합처리 효과를 조사하기 위해 진행하였다. '후지' 1년 차에 있어, 만개 시 석회유황합제 1% 또는 2% 처리는 대조구에 비해 정과 및 액과의 유과수를 감소시켰다. 하지만 2차년도에는 석회유황합제 1% 또는 2% 단용처리에서는 정과 유과수의 차이를 나타내지 않았다. 다만 석회유황합제 1%의 3회 처리에서는 1차년도와 2차년도 모두 유의적인 적과효과를 나타내었다. 석회유황합제 1%의 다중처리는 '홍로'에서 가장 효율적인 적과효과를 나타내었다. 1차년도의 경우, 석회유황합제 1%의 2회 또는 3회 살포가 효과적인 적과효과를 보였으며, 2차년도에서는 석회유황합제 1% 1회처리를 제외하고는 모두 유의적인 적과효과를 나타내었다. 첨가된 어유의 경우 대부분의 경우 석회유황합제의 적과효과를 강화하지 않았다. 본 시험에 사용된 모든 처리구는 과실의 동녹 형성에 악영향을 미치지 않았으며, 또한 '후지' 및 '홍로'의 과실 중량, 경도, 가용성고형물 및 적정산도와 같은 과실품질에 영향을 미치지 않았다.

사과의 결과지 길이와 꽃눈 위치에 따른 개화 및 과실 특성 (Characteristics of Flowering and Fruit According to Bearing Branch length and Flower Bud Position in Apples)

  • 국내 육성 품종인 3년생 '홍로'와 도입 품종인 4년생 '갤럭시갈라' 사과를 대상으로 정아 고사 시 장과지 및 액아의 활용 가치를 알아보고자 결과지 길이 및 꽃눈 위치 따른 개화 및 과실 특성을 조사하였다. '홍로'와 '갤럭시갈라' 모두 개화시, 만개기 및 낙화기는 20 cm 이상 결과지에서 5 cm 미만과 $10{\sim}15cm$ 결과지보다 다소 늦었다. 화총당 꽃수도 때 정도 적었다. 과중, 종자수, 당산비 등 대부분의 과실 특성은 5 cm 미만과 $10{\sim}15cm$ 20 cm 이상 결과지보다 높았지만, 당도와 산 함량은 반대의 경향이었다. 20 cm 이상 결과지의 정아와 액아 간 대부분의 과실 특성은 액아에서 좋은 경향을 나타내었지만, '홍로' 사과의 종자수, '갤럭시갈라'의 Hunter a값을 제외하고는 유의한 차이를 나타내지 않았다. 그리고 두 품종 모두 과중과 종자 간 높은 상관성을 나타내었다. 따라서 정아 고사 시 장과지로 대체 활용할 수 있고 '갤럭시갈라'의 경우 액화아의 이용도 가능하였다.

개량머루(Vitis sp.)의 생육기에 따른 과실 품질 및 형태학적 관찰 (Evaluation of Fruit Quality and the Anatomical Characteristics of Gaeryangmeoru (Vitis sp.) Grapes Depending on the Growth Stage)

  • 개량머루의 시기별 생육 특성을 구명하기 위하여 액아 발달, 과실 품질, 과실 조직의 해부학적 발달과정을 관찰하였다. 액아는 신초 발생부터 낙엽기까지 가지에서 마디 위치에 관계없이 괴사가 전혀 발생하지 않아 모두 충실하였다. 성숙기 과실 특성은 과립중 1.2g, 과립수 98개 및 과방중 128.8g이었고, 당도 $16.9^{\circ}Brix$, 적정산도 1.57%이었다. 당의 조성은 과당과 포도당이었고 성숙기의 과당과 포도당의 비는 1:1에 도달하였다. 사과산이 유기산 중에서 가장 많았으며 시기별 적정 산도의 변화가 일치하였다. 당과 산은 변색 1주전부터 급격하게 변화하였다. 배주의 퇴화는 수정 전후에 모두 발견되었고 성숙기의 과피는 1층의 epidermis와 7~10층의 hypodermis로 구성되었다.

In vitro shoot initiation of Artocarpus heterophyllus Lam. (Jak Fruit) Effect of the explant type and the season of explant collection

  • A method for rapid propagation of mature Jack fruit was developed. Four types of explants (mature embryos, apical meristems of young seedlings, apices from mature plants and nodal segments) were used. It has been found 88% of young apical meristems produced shoots in Campbell and Durzan (CD) medium compared to 60% in Murashige and Skoog (MS) medium. Only 1/3 of them produced multiple shoots. Shoot initiation from nodal segments was very rare. Mature apices produced callus. Although removal of the sheathing cover around mature buds enhanced the shoot initiation but success rate was low in growth regulator free medium. Embryos respond to the CD medium but not to the MS medium. Embryos from seeds soaked in water for 24 hours produced shoots after 8 weeks of incubation and the success rate was 70% while embryos from dry seeds only produced roots. There was no significant effect of cold storage (refrigeration) for 7 days on shoot initiation from mature embryos (65%) but the ability for shoot induction declines with storage time (55% after 21 days of cold storage). Mature axillary buds were established in Modified Campbell and Durzan (CD) medium supplemented with 0.5mg/1 and IBA. There was a significant difference in the growth performance of shoots according to the period of the year in which explants were collected. Highest (60%) was observed in November-January period. It was only 30% when the explants were collected in February-April or May-July and decreased to 20% in August-October. The shoots produced in November-January showed a higher vigor than those produced in other months. Since Jak fruit show seasonal changes in fruit bearing and shedding of leaves, it can be suggested that the difference in growth performances of tissues cultured in artificial culture media would have been affected by endogenous rhythms.

Photoreversibility of Fruiting and Growth in Oriental Melon (Cucumis melo L.)

  • BACKGROUND: Photoreversibility, a reversion of the inductive effect of a brief red light pulse by a subsequent far red light pulse, is a property of photo responses regulated by the plant photoreceptor phytochrome B. Plants use photoreceptors to sense photo signal and to adapt and modify their morphological and physiological properties. Phytochrome recognizes red light and far red light and plays an important role in regulating plant growth and development. METHODS AND RESULTS: The reversal responses of growth and fruiting characteristics were investigated to increase the yield of oriental melon (Cucumis Melo L. var. Kumsargakieuncheon) by means of controlling light quality in a plastic house. Red (R:660nm) and far red (FR:730nm) lights were subsequently irradiated on the whole stems and leaves of the oriental melon plant during growing periods, using red and far red LEDs as light sources, from 9:00 PM daily for 15 minutes. The intensities of R and FR light were 0.322-0.430 μmol m-2s-1 and 0.250-0.366 μmol m-2s-1, respectively. Compared to R light irradiation, combination of R and FR light irradiation increased the length of internode, number of axillary stems, number of female flowers, and fruit number of oriental melons. The results of treatment with R were similar to R-FR-R light irradiation in terms of length of internode, number of axillary stems, number of female flowers, and number of fruits. When FR treatment was considered, R-FR and R-FR-R-FR light irradiation had similarities in responses. These reversal responses revealed that oriental melon showed a photoreversibility of growth characteristics, flowering, and fruiting. CONCLUSION: These results suggested the possibility of phytochrome regulation of female flower formation and fruiting in oriental melon. The fruit weight of the oriental melon was the heaviest with the R light irradiation, while the number of fruits was the highest with the FR light. With the FR light irradiation, the fruit weight was not significantly higher compared to that of the control. Meanwhile, the yield of oriental melon fruits increased by 28-36% according to the intensities of the FR light due to the increases of the number of fruits.

Effect of Carbohydrates on in vitro Shoot Growth of Various Prunus Species

  • Carbohydrate sources are one of important factors associated with macro- and micro nutrients and phytohormones in vitro culture medium for shoot growth. The optimal carbohydrates for eight species of the genus Prunus which are economically important fruit crop was evaluated at the initiation and elongation stages. All carbohydrate seemed utilized for the bud break and leaf growth at the early stage of culture. However, shoot elongation and fresh weight of species tested were superior in the medium containing 90 mM of fructose or glucose rather than sucrose. There was no difference between glucose and fructose. Adventitious shoots from the axillary buds were induced in most species but no significant differences were observed except for two species (P. salicina ‘Shiro’ and P. tomentosa). These result demonstrated that glucose and fructose were suitable carbohydrate sources for diverse Prunus species than sucrose, although the response to the carbohydrates in the medium were slightly different in the species.

Efficient Cryopreservation of in vitro Grown Shoot Tips of Pear (Pyrus spp.) by Droplet-vitrification

  • In this study, cryopreservation by droplet-vitrification was applied to pear (Pyrus spp.) germplasm. We focused on the development and assessment of various strategies for the selection of suitable tissue, osmoprotection, and dehydration. We also evaluated post-thaw recovery of cryopreserved explants by droplet-vitrification. Preferentially, we tested the effects of preculture and loading treatments to determine which tissues were more suitable, either the apical shoot tips or the axillary buds. Apical shoot tips showed the better regrowth rate than in vitro axillary buds. The most effective techniques for cryopreservation were as follows. Shoots from in vitro seedlings which had been cultured for about 5-6 weeks were cold-hardened at 4℃ for one week, excised shoot tips were precultured on liquid MS medium including 0.3 M sucrose for 31 hours and 0.7 M sucrose for 17 hours, osmoprotected in loading solution (LS) for 40 min, and then cryoprotected in dehydration solution (PVS3) for 90 min. In addition, we found that regrowth rates of explants on regrowth medium after exposure to liquid nitrogen (LN) were higher than those on MS medium. Results indicated that the highest regrowth percentage was 95.6% for 'Bartlett' cultivar and 68.9% for 'BaeYun No.3' cultivar. Consequently, apical shoot tips of two pear cultivars, 'Bartlett' (P. communis) and 'BaeYun No.3' (P. pyrifolia), were successfully cryopreserved by droplet-vitrification. Results of this study show that the enhanced droplet-vitrification method described in the present study could be used as an effective means for long-term storage of pear genetic resources.

한국산(韓國産) 옻나무속(屬) 과실(果實)과 종자(種子) 및 화서(花序)의 분류학적(分類學的) 재고(再考) (Reconsideration of Rhus (Anacardiaceae) with Respect to Fruit, Seed and Inflorescence)

  • 우리나라에 자생(自生)하고 있는 옻나무속(屬) 6종(種)과 외래종(外來種) 2종(種)에 대(對)한 화서(花序)의 형태(形態)와 과실(果實)과 종자(種子)의 형태(形態) 및 해부학적(解剖學的) 형질(形質)들을 광학현미경(光學顯微鏡)과 주사전자현미경(走査電磁顯微鏡)을 통해 관찰(觀察)한 결과(結果), 화서(花序)의 형태(形態)는 미국털옻나무가 정생(頂生)하고 직립(直立)하는 밀추화서(密錐花序)인 반면 붉나무는 밀추화서(密錐花序)와 유사한 원추화서(圓錐花序)였으며, 다른 수종(樹種)들은 액생(腋生)하고 드리워진 원추화서(圓錐花序)였다. 과실(果實)과 종자(種子)의 해부학적(解剖學的) 연구결과(硏究結果) 붉나무와 미국털옻나무는 외과피(外果皮)와 중과피(中果皮)가 완전(完全)히 분리(分離)되어 있지 않은 반면, 다른 수종(樹種)들은 모두 내외피(內果皮)가 3개(個)의 lignified cell layer와 crystal layer로 이루져 있었다. 또한 종피(種皮)의 외부(外部) 미세구조(微細構造)도 종(種)에 따라 고유(固有)한 특징(特徵)을 지니고 있어 종간(種間)의 식별(識別)과 분류(分類)가 가능(可能)하였다. 과실(果實)과 종자(種子)의 형태(形態) 및 해부학적(解剖學的) 형질(形質)에 의한 유집분석(類集分析) 결과(結果) 개옻나무-붉나무-덩굴옻나무류집군(類集群)과 옻나무-산검양옻나무-검양옻나무류집군(類集群)으로 분류(分類)되었다. 이들중 17개(個)의 기본 형질(形質)에 대(對)한 주성분(主成分) 분석결과(分析結果) Eigenvalue 1.0 이상(以上)이 3개(個)의 주성분군(主成分群)으로 분리(分離)되었으며, 제(第)2 주성분(主成分)까지의 누적기여율(累積寄與率)은 89.47%로 높은 설명력(說明力)을 보였고, 종자(種子)의 길이, 과실(果實)의 길이, 종자(種子)의 폭(幅), 종자(種子)의 무게, 과실(果實)의 폭순(幅順)으로 높은 기여도(寄與度)를 보였다. 화서(花序)와 종실(種實)의 외부(外部) 형태적(形態的), 해부학적(解剖學的) 특징(特徵)을 종합(綜合)하여 종간(種間)의 검색표(檢索表)를 제시하였다.

조기낙엽에 따른 참다래(골드러쉬) 무착과 유목 액아의 발아와 착화 (Regrowth of Buds and Flower Bud Formation in Kiwifruit as Affected by Early Defoliation)

  • 본 연구는 태풍에 의한 참다래 (골드러쉬) 조기 낙엽이 무착과 유목의 액아 발아와 착화에 미치는 영향을 분석하기 위해서 7, 8, 9, 및 10월에 인위적인 적엽처리를 하였다. 먼저 당해연도 액아의 재발아는 7월 적엽처리에서는 무처리의 23.2%와 마찬가지로 높은 재발아율을 나타냈다. 또한 8월의 경우 무처리의 재발아율은 5.8%로 낮았지만 적엽처리구는 17.23%의 높은 재발아율을 나타냈다. 하지만, 9월 적엽처리는 무처리의4.2%와 비슷한 낮은 재발아율을 나타냈으며, 10월 적엽처리에서는 재발아가 전혀 되지 않아 액아가 완전히 휴면에 진입한 것으로 판단되었다. 적엽처리에 의한 익년 착화율은 적엽시기와 적엽률에 따라 큰 영향을 받았다. 적엽 시기 중 7, 8, 9월에 75% 및 100% 적엽처리구는 무처리에 비해 유의성 있는 착화율 감소를 보인 반면, 10월 적엽처리는 적엽률에 관계없이 착화율에는 영향을 미치지 않는 것으로 나타났다. 그리고 착화 신초 한 개에 맺히는 꽃봉오리 수의 경우 75% 이상 적엽이 될 경우 무처리에 비해 절반 이하로 감소 되었다. 이와 같은 결과는 7월과 8월의 경우 적엽에 의해 꽃눈의 원기형성이 진행되어야할 액아의 재발아율이 높아 원기 형성이 감소했기 때문이며, 9월과 10월은 액아의 꽃눈 원기 형성은 어느 정도 이루어졌으나 광합성 산물 생산을 위한 잎의 부족 또는 부재로 저장양분 축적이 부족하여 이듬해 봄 꽃눈의 형태적 분화가 감소되었기 때문인 것으로 판단된다.
