Responses of Edwardsiella tarda to innate immunity of olive flounder, Paralichthys olivaceus (넙치, Paralichthys olivaceus의 비특이적 방어기작에 대한 Edwardsiella tarda 균의 반응)

  • This paper aims to compare difference with the in an ability of their resistance and survival against in a non-specific defence mechanism of the olive flounder, between the virulent and the avirulent E. tarda strains. The tested E. tarda strains, we divided into the virulent and the avirulent strain groups on the basis of a value of 50% lethal dose (LD50) for the olive flounder weighed 10.3 g in average. The strains of LD50 101.6~104.2 cfu/fish were grouped as virulent strains, such as KE-1, KE-3, KE-5 and FSW910410. The group of avirulent strains as LD50 exceeded 108.7 cfu/fish were included the strains, SU100 and AL92448. A test was conducted to understand the survival ability of each strain in the mucus of the skin and the intestine of olive flounders. The results showed KE-1, KE-3, KE-5 and FSW910410 were highly to survive between 6 hours and 24 hours in intestine. The survival ability in the bile of olive flounder the number of avirulent strains declined during incubation but the virulent strain showed the number of alive bacteria having sustained or increased. In the test for the survival of bacteria in fresh sera of olive flounder, the virulent strains also had tendency to multiply. Concerning the tested bacteria internalization into the head kidney macrophages and the intracellurar replication in the macrophages of olive flounder. The virulent strains exhibited strong internalization, followed high rate replication. According to the results, virulent strains of E. tarda revealed more ability to resist and survive in the face of humoral and cellular defence factors than avirulent strains.

  • Control effect of an avirulent bacterlocin-producing strain(ABPS) Y6l-1 of Pseudomonas solanacearum on bacterial wilt was 58.8 % when the bacterial suspension had poured onto the rhizosphere soil of tobacco cultivar NC82 on one day before transplanting to the field and of hilling time. Until eight weeks after inoculation, survival of the strain on rhizoplane and in stem of the plants inoculated was better than that of other four strains tested. It suggested that survival of the ABPS in and on the plants should be supported for the sufficient protection.

  • An understanding of the nature of the immune response to asexual erythrocytic stages of malaria parasites will facilitate vaccine development by identifying which responses the vaccine should preferentially induce. The present study examined and compared the immune responses of NIH mice in either single or mixed infections with avirulent (DK) or virulent (DS) strains of Plasmodium chabaudi adami using the ELISA test for detecting and measurement of cytokines and antibody production. In both single and mixed infections, the study showed that both cell- and antibody-mediated responses were activated. In all experiments, an early relatively high level of IFN-$\gamma$ and IgG2a during the acute phase of the infection, and later elevation of IL-4 and IgG1, suggested that there was a sequential Th1/Th2 response. However, in the avirulent DK strain infection a stronger Th1 response was observed compared to the virulent DS strain-infection or in mixed infections. In the virulent DS infection, there was a stronger Th2 response compared to that in the DK and mixed infections. The faster proliferation rate of the virulent DS strain compared to the DK strain was also evident.

  • The global pattern of growth-dependent gene expression in Streptococcus pneumoniae strains was evaluated using a high-density DNA microarray. Total RNAs obtained from an avirulent S. pneumoniae strain R6 and a virulent strain AMC96-6 were used to compare the expression patterns at seven time points (2.5, 3.5, 4.5, 5.5, 6.0, 6.5, and 8.0 h). The expression profile of strain R6 changed between log and stationary growth (the Log-Stat switch). There were clear differences between the growth-dependent gene expression profiles of the virulent and avirulent pneumococcal strains in 367 of 1,112 genes. Transcripts of genes associated with bacterial competence and capsular polysaccharide formation, as well as clpP and cbpA, were higher in the virulent strain. Our data suggest that late log or early stationary phase may be the most virulent phase of S. pneumoniae.

  • To study the determinants which are involved in the loss of pathogenicity in wilt-inducing Pseudomoms solamcewum, several physlologica I functions were compared in a virulent P. solanacearum strain and an avirulent, spontaneously derived mutant strain. The polyacrylamide gel electrophoresis showed the distinction between two strains in the patterns and the relative intensity of proteins produced intracellularly or extracellularly. Enzyme assays showed that the level of polygalacturonase activity in the culture filtrate of the avirulent mutant was markedly reduced, while carboxymethylcellulase(rondoglucanase) activity in both strains were nearly negligible. These results suggest that the loss of pathogenicity in mutant strain is attributed in part to the reduced production of polygalacturonase. In audit ion, comparative analyses by agarose gel electrophoresis of DNA molecules isolated from both strains show that the pathogenicity genes of p. solanaceerum are not located on plasmid but are on chromosome.

  • Effect of an avirulent bacteriocin-producing strain(ABPS) of Pseudomonas solanacearum adapted to low temperature on the control of tobacco bacterial wilt was examined under the natural field conditions. The ABPS of p. solanacearum were succeeding-cultured at gradually low temperature, 3$0^{\circ}C$ to 13$^{\circ}C$. The isolates adapted to low temperature grew faster than the wild type either in artificial media or on the tobacco rhizoplane. The control effect of one of the isolates on bacterial wilt was higher than that of the wild type when the bacterial suspension had been poured onto the tobacco rhizosphere soil on 1 day before and 15 days after transplanting to the field. It was suggested that ABPS of p. solanaceamm adapted to the low temperature, might be more effective biological control agent than the wild type.

Generation of transposon insertion mutants from type A Pasteurella multocida

  • The transposon TnphoA was used to generate avirulent mutants from a type A Pasteurella multocida. A suicide vector plasmid pRT733 carrying TnphoA, having the kanamycin resistant gene and harbored in Escherichia coli K-12 strain SM10(${\lambda}pir$), was mated with streptomycin resistant P. multocida P-1059 strain as recipient. This resulted in the generation of two TnphoA insertion mutants (transconjugants, tc95-a and tc95-b) which were resistant both to kanamycin ($Km^{R}$) and streptomycin ($Sm^{R}$), secreted alkaline phosphatase, and were avirulent to turkeys. Southern blot hybridization using two probes derived from internal fragments of TnphoA, confirmed the insertion of TnphoA into 12.9kb or 13.7kb DNA fragment from the EcoRV digested genomic fragments of transconjugants. The two transconjugants, tc95-a and tc95-b, were distinguishable from their parent strains by differences in ribotypes, and outer membrane protein profiles. TnphoA insertion in both transconjugants also resulted in constitutive expression of a 33Kd iron regulated outer membrane protein (IROMP). The gene encoding $Sm^{R}$ was also located within the same 12.9kb EcoRV genomic fragment from both transconjugants. Furthermore, our finding that the recipient P. multocida P-1059 $Sm^{R}$ strain and both transconjugants were avirulent to turkeys suggest that the either 12.9kb or 13.7kb genomic DNA contains the virulence gene and speculate that the presence of $Sm^{R}$ gene or TnphoA insertion may be responsible for regulating and inactivating the gene(s) encoding virulence in P. multocida.

  • Agrobacterium tumefaciens KU12 contains pTiKU12 (240kb) of the octopine type Ti plamsid and pTi12 (45 kb) of the cryptic plasmid. To make the avirulent A. tumefaciens, the octopine type Ti plasmid, pTiKU12, was cured with elevated temperature (37${\circ}C$) and ethidium bromide (EtBr), respectively. Also the cryptic plasmid, pTi12, was cured by the introduction of recombinant plasmid, pYWXP, made by pTi12 replication origin and pUC19. pYWXP was cured by elevated temperature (37${\circ}C$) and EtBr simultaneously.

Virulence characters of Yersinia enterocolitica isolated in Korea (한국에서 분리한 Yersinia enterocolitica의 병원성에 관한 연구)

  • In order to investigate the virulence of Y. enterocolitica isolated in Korea, all necessary experiments were done including several virulence determinating tests-autoagglutination test, calcium-dependency test, HeLa cell invasion test, Sereny test, crystal violet binding test, and electrophoresis for plasmid pattern. The obtained results are as follows: The virulent strains of Y. enterocolitica revealed positive reactions on autoagglutination test, cacium-dependency test, and drystal violet binding test, while the avirulent strains did not. A positive reaction was observed only at $37^{\circ}C$ implying that the expression of virulence is temperature-dependent. In Sereny test, the standard reference virulent strain (serotype 0:8) showed positive reactions while the virylent experimental strains (serotype 1:3, 0:9) revealed negative results, which indicates that the virulence of Y. enterocolitica in experimental animals varied according to their serotypes. Most of the virulent strains contained 36-38Mdal plasmids, but the avirulent strains did not. In addition, it was noted that autoagglutination, calcium-dependency, and crystal violet binding were related to the presence of plasmids.

Identification of N,N',N"-triacetylfusarinine C as a key metabolite for root rot disease virulence in American ginseng

  • Background: It is estimated that 20-30% of ginseng crops in Canada are lost to root rot each harvest. This disease is commonly caused by fungal infection with Ilyonectria, previously known as Cylindrocarpon. Previous reports have linked the virulence of fungal disease to the production of siderophores, a class of small-molecule iron chelators. However, these siderophores have not been identified in Ilyonectria. Methods: High-resolution LC-MS/MS was used to screen Ilyonectria and Cylindrocarpon strain extracts for secondary metabolite production. These strains were also tested for their ability to cause root rot in American ginseng and categorized as virulent or avirulent. The differences in detected metabolites between the virulent and avirulent strains were compared with a focus on siderophores. Results: For the first time, a siderophore N,N',N"-triacetylfusarinine C (TAFC) has been identified in Ilyonectria, and it appears to be linked to disease virulence. Siderophore production was suppressed as the concentration of iron increased, which is in agreement with previous reports. Conclusion: The identification of the siderophore produced by Ilyonectria gives us further insight into the root rot disease that heavily affects ginseng crop yields. This research identifies a molecular pathway previously unknown for ginseng root rot and could lead to new disease treatment options.
