Matrix Solid-Phase Dispersion (MSPD) Isolation and Liquid Chromatographic Determination of Residual Furazolidone in Eggs (MSPD 전처리법과 HPLC를 이용한 Furazolidone의 계란내 잔류분석)

  • A liquid chromatographic method, using matrix solid-phase dispersion (MSPD) is developed for the extraction of residual furazolidone in chicken eggs. Blank or fortified egg samples (0.5 g) were blended with Octadecylsilyl (Bulk $C_{18}$, 40${\mu}{\textrm}{m}$, 18%. load, endcapped. 2 g) derivatized silica. After homogenization, $C_{18}$/egg and Na$_2$S $O_4$matrix were transferred to a column made of 10 ml glass syringe and filter paper and compressed 4.0∼4.5 ml volume. The column was washed with 8 ml of hexane and dried under $N_2$ gas. Furazolidone was eluted with acetonitrile (8 ml) under gravity. The eluate containing furazolidone was free from interfering compounds when analyzed by HPLC with UV detection (365 nm, photodiode array). Calibration curves were linear (r = 0.99985) and inter- (1.47%) and intra-assay (5.29%) variabilities for the concentration range examined (7.8∼497 ng/g of eggs, 20 ${mu}ell$ injection volume) were indicative of an acceptable methodology for the analysis of furazolidone. Average recovery of furazolidone added to egg was 96.2%. The limit of detection for the proposed method was 1 ng/g for furazolidone. The method using MSPD is proposed as an alternative assay to the classical method which involves the use of large volumes of a harmful solvent and requires a long tedious separation and clean-up processes prior to its determination.

Physicochemical Properties of the Exopolysaccharides Produced by Marine Bacterium Zoogloea sp. KCCM10036

  • The physicochemical properties of the exopolysaccharide (EPS) produced by marine bacterium Zoogloea sp. KCCM10036 were investigated. Two types of isolated EPSs were shown to have average relative molecular masses $(M_r)\;of\;4.07{\times}10^6$ of CBP (cell-bound polysaccharide) and $3.43{\times}10^6$ of WSP (water-soluble polysaccharide), respectively. When the CBP was utilized as an emulsifier, it stabilized the emulsifier, for up to 148 h. Compared with other commercially available hydrocolloids such as xanthan gum, the Tween series, and Triton, the CBP showed much better emulsifying capability on a water-in-oil system. Phase separation occurred in the Tween series after 24 h, whereas the emulsion was better stabilized by the CBP. The CBP thus has potential as an emulsifying agent in commercial emulsions. The flocculating activity was also greatest at 0.01% (w/v) and decreased at higher concentrations than the optimized concentration of the WSP and CBP. The results also showed that both types of expolysaccharides from Zoogloea sp. had excellent flocculating activity.

Application of tube-type ceramic microfiltration membrane for post-treatment of effluent from biological wastewater treatment process using phase separation

  • A tube-type ceramic membrane for microfiltration was developed, and the membrane module comprised of three membranes was also applied to biological carbon and nitrogen removal processes for post-treatment. Manufacturing the microfiltration membrane was successful with the structure and boundary of the coated and support layers within the membrane module clearly observable. Total kjeldahl nitrogen removal from effluent was additionally achieved through the elimination of solids containing organic nitrogen by use of the ceramic membrane module. Removal of suspended solids and colloidal substances were noticeably improved after membrane filtration, and the filtration function of the ceramic membrane could also easily be recovered by physical cleaning. By using the ceramic membrane module, the system showed average removals of organics, nitrogen, and solids up to 98%, 80% and 99.9%, respectively. Thus, this microfiltration system appears to be an alternative and flexible option for existing biological nutrient removal processes suffering from poor settling performance due to the use of a clarifier.

Study on the flow characteristics and heat transfer enhancement on flat plate in potential core region of 2-dimensional air jet (포텐셜 코어내에 설치된 충돌평판상의 열전달증진 및 유동특성에 관한 연구)

  • A heat exchanging system employing the impinging air jet is still widely used In the various fields due to its inherent merits that include the easiness in engineering applications and the high heat and/or mass transfer characteristics. The purpose of this study is to investigate the enhancement of heat transfer and flow characteristics by placing a turbulence promoters in front of heat exchanging surface. In this study, a series of circular rods are placed at the upstream of a flat plate heat exchanger that is located at potential core region(H/W=2) of a two-dimensional impinging air jet. Heat transfer enhancement is achieved by inserting turbulence promoter that results in the flow acceleration and disturbance of boundary layer. The average Nusselt number of the flat plate with the turbulence promoters is found to be around 1.42 times higher than that of the flat plate without the turbulence promoters. Based on the results of flow visualization with a smoke wire, it is confirmed that the heat transfer enhancement is caused by the flow separation and disturbance of boundary layer by inserting the turbulence promoter.

Stabilization for Heavy Metal Contamination Soils which Uses the Coal Mine Drainage Sludge (폐 석탄광산 배수처리 시 발생되는 슬러지를 이용한 오염토양 중금속 안정화)

  • In this study, to stabilize the heavy metal in the contaminated soils, the column leaching test based on rainfall and pH value was performed by using coal mine drainage sludge(CMDS): which was generated during electrical purification of abandoned coal mine wastewater. Four types of testing column were used in this study. That were the CMDS and the heavy metal contaminated soils well mixed in 0 wt%, 1 wt%, 3 wt% and 3 wt% layered column. According to the investigation, when the influent pH was $5.5{\sim}6.2$, there were no heavy metal elution at all conditions, and when the influent pH was $3{\sim}3.3$, the order of Cu, Zn, Pb, Cr elution concentration was 3 wt% M(mixed)<3 wt% S(separation)<1 wt% M<0 wt% and the average elution concentration was quite low, the value was 0.005 mg/L. Therefore, CMDS can used as new stabilizer of the heavy metal in the contaminated soils.

  • The $Cu_2O/TiO_2$ inverse opal heterojunction arrays were developed by electrochemical deposition of $Cu_2O$ nanoparticles on $TiO_2$ inverse opal arrays. The $Cu_2O$ nanoparticles completely filled the inner pores of $TiO_2$ inverse opal film (prepared by liquid phase deposition with an average thickness of 400 nm) and covered the entire area; exhibiting high crystalline properties of anatase and cubic phase from $TiO_2$ and $Cu_2O$, respectively. From asymmetric current-voltage profile, it was noticeable that a heterojunction was well formed for charge transport from $Cu_2O$ to $TiO_2$ film resulting from the enhanced charge separation yield. In addition, increased photocurrent of 0.19 $mA/cm^2$ (versus 0.08 $mA/cm^2$ under dark condition) was obtained at -0.35 V from the heterojunction structure in the 0.5M $Na_2SO_4$ solution.

  • This study is to investigate the heat transfer characteristics the for a round turbulent jet impinging on the flat plate with and without rib. Liquid crystal/transient method was used to determine the Nusselt number distributions along the surface. The temperature on the surface was measured using liquid crystal and a digital color image processing system. The experiments were made fur the jet Reynolds number (Re) 23,000, the dimensionless nozzle-to-surface distance (L/d) from 2 to 10, and the rib type [height ($d_1$) 2mm, pitch (p) from 12 to 36mm]. It was found that for $L/d{\ge}6$ the average Nusselt numbers on the flat plate with rib type C ($p/d_1=16$) are higher than those without rib, mainly due to an increase in the turbulent intensity caused by flow separation, recirculation and reattachment on the wall surface.

  • An experimental study was carried out to investigate near-wake characteristics of an elliptic airfoil oscillating in pitch. The airfoil was sinusoidally pitched about the half chord point between $-5^{\circ}C$ and $+25^{\circ}C$ angles of attack at the freestream velocities of 3.4 and 23.1 m/s The corresponding Reynolds numbers based on the chord length were $3.3{\times}10^4$ and $2.2{\times}10^5$, respectively. A hot-wire anemometer was used to measure the near-wake flow variable at the reduced frequency of 0.1. Ensemble-averaged velocity and turbulence intensity profile were presented to examine the near-wake characteristics depending on the Reynolds number. The axial velocity deficit in the near-wake region tend to decrease with the increase in the Reynolds number a found in many stationary airfoil test . Turbulence intensity in the near-wake region have a tendency to decrease with the increase in the Reynolds number during the pitch-up motion, whereas it shows different feature during the pitch-down motion either the laminar boundary layer or turbulent boundary layer separation.

  • The average heat transfer coefficient of 2-D impinging jet has been augmented as much as 60% on the wall with large-scale wavy roughness. The mechanism of this heat transfer augmentation is studied with emphasis on two primary flow structures in the impinging flow region by using either the surface floating method or the smoke-wire technique. They are the stream-wise vortex-like structure, which is characteristic to the impining jet, and the spanwise vortiecs associated with the flow separation around the roughness. The combined effect of these structures can effectively augment the heat transfer particularly in the downstream region where the teat transfer usually deteriorates consicerably.

  • An experimental investigation has been conducted to clarify roughness effects of geothermal water scale deposited onto a heating surface upon its forced convection heat transfer characteristics. Examined was a circular cylinder, on which particles of silica scale having five different sizes are uniformly distributed. The Reynolds number was varied from 13000 through 50000. Local and mean heat transfer characteristics were measured as functions of particle size and Reynolds number. Subsequently the mean fouling resistance was estimated from those results, and its characteristics are clarified. It was found that the heat transfer of cylinders greatly varies with the fouling of geothermal water scale, especially its scale height. Further, the local and average Nusselt numbers strongly depend upon the cylinder spacing and the Reynolds number.
