Direct Triazine Herbicide Detection Using a Self-Assembled Photosynthetic Reaction Center from Purple Bacterium

  • In this study, a direct detection system for triazine derivative herbicides was developed using the photosynthetic reaction center (RC) from the purple bacterium, Rhodobacter sphaeroides, and surface plasmon resonance (SPR) apparatus. The histidine-tagged RCs were immobilized on an SPR gold chip using nickel-nitrilotriacetic acid groups as a binder for one of the triazine herbicide, atrazine. The SPR responses were proportional to the sample concentrations of atrazine in the range 0.1-1 $\mu\textrm{g}$/mL. The sensitivity of the direct detection of atrazine using the RC-assembled sensor chip was higher than that using the antibody-immobilized chip. The other types of herbicides, DCMU or MCPP, were not detected with such high sensitivity. The results indicated the high binding selectivity of the RC complex.

Enzymatic Properties of Atrazine Chlorohydrolase Entrapped in Biomimetic Silica

  • Purified atrazine chlorohydrolase (AtzA) was entrapped in the nanoparticles of biomimetically synthesized silica at the ambient condition within 20 min. Entrapped AtzA in biomimetic silica was less affected by pH change and showed higher thermostability than free enzymes. The entrapped AtzA was also more tolerant against proteolysis, with 80% of the initial activity remaining and retained 82% of the initial activity even after four cycles of usage. These results suggest that entrapment of AtzA in biomimetic silica could be utilized under diverse environmental conditions with the active catalytic performance sustained.

  • Characteristics of sorption and desorption in soils affect chemical fate, soil-remediation time, and selection of remediation technology. The sorption and desorption behavior of atrazine and naphthalene on soils was studied. Six soils collected at Gwangju area were used as sorbents and the organic matter contents ranged from 1.28 to 5.21%. Sorption and desorption experiments were conducted and sorption distribution coefficients(Kd) of atrazine and naphthalene were nearly linear$(R^2=0.93{\sim}0.97)$. Desorption parameters were evaluated using three site desorption model included equilibrium, nonequilibrium and nondesorption sites. Non-desorbable site fraction for atrazine was evaluated, but for naphthalene it was not enumerated during the experimental period. Through the series dilution desorption experiments, non-desorpbable sites were observed for both chemicals.

Cyanobacteria를 이용한 광합성 전자전달저해제의 생합리적 스크리닝 (Biorational Screening System Using Cyanobacteria(Anacystis nidulans $R_2$) for Searching the Photosynthetic Electron Transport Inhibitors)

  • 광합성 전자전달저해제의 탐색을 위한 biorational biorational screening 방법으로 광합성 박테리아의 일종인 cyanobacteria의 표준형과 $D_1$단백질의 구성 아미노산이 변환된 변이주 Di-1, G-264, Di-22, TRY-5를 사용하여 기존 약제의 혼합효과와 몇가지 합성화합물의 Hill반응 저해활성을 측정 조사하였고, 또한 시금치로 부터 분리한 chloroplast와 cyanobacteria로 부터 분리한 thylakoid membrane에 대한 기존 제초제의 반응성을 조사하였다. 1. 광합성억제형 제초제에 대한 반응성 비교에서 cyanobacteria로 부터 분리한 thylakoid membrane이 시금치로 부터 분리한 chloroplast보다 약제에 대하여 민감한 반응을 나타내어 광합성 전자전달저해제의 탐색을 위한 재료로 보다 적합하였다. 2. Diuron에 대하여 wild type보다 Di-22와 D-5 변이주는 각각 약 2,000배와 800배의 저항성을 나타내었고 atrazine에 대하여는 G-264 변이주만이 약 1,000배의 저항성을 나타내었으며, 기타 네 약제에 대하여는 뚜렷한 저항성을 나타내는 변이주가 없었다. 3. D1 단백절의 264번 위치의 serine이 다른 아미노산으로 치환된 Di-1과 D-22 및 G-264 변이주에 대한 diuron과 atrazine의 혼합처리 효과를 검토한 결과 상호 경합적으로 작용하였으며 그 정도는 G-264 변이주에 대해서 가장 뚜렷하게 나타났다. 그러나 diurone과 atrazine의 혼합처리에 의한 thermoluminescence band의 변화는 인정할 수 없었다. 4. 변이주 Di-1과 D-22는 dinoseb에 대해서 저항성을 나타내지 않았으나. dinoseb과 diuron의 혼합처리시 diuron에 대한 저항성이 크게 약화되었다.

Estimating dehalogenation reactivity of nanoscale zero-valent iron by simple colorimetric assay by way of 4-chlorophenol reduction

  • A number of different nanoscale zero-valent iron (nZVI) materials have been prepared and compared depending on the desired properties for the particular application, but different physicochemical properties of this prepared nZVI make it difficult to universally compare and standardize them to the same scale. In this study, we aimed to demonstrate a simple microplate-based colorimetric assay using 4-chlorophenol as an indicator with respect to the remediation of real treatment targets, such as trichloroethylene (TCE), 1,1,1-trichloroethane (TCA), and atrazine. Effect of nickel contents on 4-chlorophenol reduction was successfully investigated by the miniaturized colorimetric assay. In the same manner, the effect of nickel contents on dehalogenation of TCE, TCA, and atrazine was investigated and the pseudo-first-order kinetic constants were compared with the results for 4-chlorophenol. The similar pattern could be observed between 4-chlorophenol reduction obtained by colorimetric assay and TCE, TCA, atrazine reduction obtained by a traditional chromatographic method. The reaction kinetics does not match perfectly, but the degree of reaction can be estimated. Therefore, the colorimetric assay can be a useful and simple screening tool to determine nZVI reactivity toward halogenated organics before it is applied to a particular remediation site.

지표수질 모의를 위한 NAPRA WWW 시스템의 적용 (Application of NAPRA WWW for Modeling Surface Water Quality)

  • National Agricultural Pesticide Risk Analysis (NAPRA) WWW 시스템 (http://pasture.ecn.purdue.edu/${\sim}napra$)은 각기 다른 영농방법이 지표수질, 유사, 그리고 지하수질에 미치는 영향을 평가하기 위하여 개발되었다. 이 NAPRA WWW 시스템은 Total Maximum Daily Loads와 같은 수질 요건을 만족시킬 수 있는 최적영농 방법이 무엇인지 찾는데, 그리고 수질측면에서 취약한 지역을 찾아내는데 매우 효율적인 시스템이다. 이 NAPRA WWW 시스템을 이용하여 미국 인디애나주의 수계에 대해서, NAPRA 모의 Nitrogen과 Atrazine 결과를 실측치와 비교하였다. 18개 수계에 대해서 NAPRA 예측 질소값과 실측 질소값을 비교한 결과 $R^2$ 값은 0.51이고, 6개 수계에 대해서 NAPRA 예측 Atrazine값과 실측값을 비교한 결과 $R^2$ 값은 0.87이었다. 이 연구에서 보여지는 바와 같이 NAPRA WWW 시스템은 수계내에서 질소와 Atrazine에 따른 오염지역을 찾아내는데 효율적으로 사용될 수 있는 시스템이다.

신규(新規) 합성화합물들이 cyanobacteria의 광합성전자전달계에 미치는 영향 (The Effect of Newly Synthesized Compounds on the Photosynthetic Electron Transport of Cyanobacteria (Anacystis nidulans $R_2$))

  • 광합성 전자전달저해제의 탐색을 위한 biorational screening 방법으로 광합성 박테리아의 일종인 cyanobacteria의 표준형(wild type)과 $D_1$ 단백질의 구성 아미노산이 변환된 변이주 Di-1, G-264. Di-22 등 4종을 사용하여 신규합성 화합물에 대한 광합성 전자전달저해활성을 조사하여 구조-활성간의 특성을 검토하였고, 이들의 thermoluminescence band를 이용하여 결합부위를 비교하였다. 1. Phenol 화합물(E-series)들은 cyanobacteria를 이용한 광합성 전자전달 저해활성의 양상을 볼 때 diuron보다는 dinoseb과 비슷하였다. 2. 합성한 Phenol화합물에서 나타난 구조활성간의 특정으로는 치환기의 phenyl ring에 C1이 치환되지 않은 경우 Hill반응 저해활성이 나타나지 않았고, 화합물에 있는 두개의 ring을 연결하는 bridge의 길이가 길어질수록 Hill반응 저해활성은 약하게 나타났다. 3. Triazine 화합물(T-series)의 경우 T-27, T-28화합물은 대조약제 atrazine보다 각각 10, 30배 강한 저해활성을 나타내었고, 변이주 G-264에 대 한 저항성 비율이 atrazine보다 더욱 크게 나타나 이들 화합물들의 결합부위가 $D_1$ 단백질의 264번 위치와 깊은 관계가 있을 것으로 생각되었다. 4. Triazine 화합물에 있어서 치환기 -C-NH-는 전자전달계에 중요한 작용기로 생각되었다. 5. Thermoluninescence band를 이용하여 결합부위를 비교할 수는 있었지만, 명료하게 구분되지 않았다.

Simultaneous Analysis of 13 Pesticides in Groundwater and Evaluation of its Persistent Characteristics

  • For this study, groundwater samples for 3 years from 2011 through 2013 were collected at 106 groundwater monitoring site in Korea. These groundwater samples were analyzed for 13 pesticides such as cabofuran, pentachlorobenzene, hexachlorobenzene, simazine, atrazine, lindane (gamma-HCH), alachlor, heptachlor, chlordane (total), endosulfan (1, 2), dieldrin, endrin, 4,4-DDT. The objectives of this study were to determine the detection frequency and their concentrations of 13 pesticides and evaluate the health risk level considering ingestion, inhalation, and skin contact using concentrations of 13 pesticides in groundwater samples. An analysis was used for the simultaneous determination for 13 pesticides using GC-MS. GC-MS was performed on HP-5ms, using helium ($1ml\;min^{-1}$) as carrier gas. The average recoveries of the pesticides were from 92.8% to 120.8%. The limits of detection (LODs) were between $0.004{\mu}g\;L^{-1}$ and $0.118{\mu}g\;L^{-1}$ and the limits of quantification (LOQs) were between $0.012{\mu}g\;L^{-1}$ and $0.354{\mu}g\;L^{-1}$. 106 groundwater wells were selected. 54 wells were from well to monitor background groundwater quality and 52 wells were from well to monitor groundwater quality in industrial or contamination source area. Eight pesticides including pentachlorobenzene, lindane (Gamma-HCH), heptachlor, chlordane (total), Endosulfan (1, 2), dieldrin, endrin, and 4,4-DDT were not detected in groundwater samples. The detection frequency for hexachlorobenzene, alachlor, carbofuran and simazine was 23.4%, 11.4%, 7.3%, and 1.0%, respectively. Atrazine was detected once in 2011. The average concentrations were $0.00423{\mu}g\;L^{-1}$ for carbofuran, $0.000243{\mu}g\;L^{-1}$ for alachlor, $0.00015{\mu}g\;L^{-1}$ for simazine, and $0.00001{\mu}g\;L^{-1}$ for hexachlorobenzene. The detection frequency of hexachlorobenzene was high, but the average concentration was low. In the contaminated groundwater, the detection frequency for hexachlorobenzene, alachlor, carbofuran, simazine and atrazine was 26.1%, 21.3%, 7.1%, 1.9% and 0.3%, respectively. In the uncontaminated groundwater, detection frequency for hexachlorobenzene, carbofuran and alachlor were 20.2%, 7.5%, and 1.9% respectively. Simazine and atrazine were not detected at uncontaminated groundwater wells. According to the purpose of groundwater use, atrazine was detected for agricultural groundwater use. Hexachlorobenzene showed high detection frequency at agricultural groundwater use area where the animal feeding area and golf course area were located. Alachlor showed more than 50% detection frequency at cropping area, pollution concern river area, and golf course area. Atrazine was detected in agricultural water use area. By land use, the maximum detection frequency of alachlor was found near an orchard. For human risk assessment, the cancer risk for the 5 pesticides was between $10^{-7}$ and $10^{-10}$, while the non-cancer risk (HQ value) was between $10^{-4}$ and $10^{-6}$. For conclusion, these monitoring study needs to continue because of the possibility of groundwater contamination based on various purpose of groundwater use.

CE-AD기반의 Microfluidic chip을 이용한 Simazine과 Atrazine 그리고 Ametryn Herbicides의 검출 (Detection of Simazine, Atrazine and Ametryn Herbicides on a Microfluidic Chip Based on CE-AD)

  • A simple and rapid capillary electrophoresis method was developed for the quantitative analysis of common triazine herbicides. Cyclic voltammetry was employed to clarify the detection voltage which showed characteristic irreversible cathodic peaks. For the analysis, the mixture of triazine herbicides was applied in a microfluidic chip to determine the CE-separated peaks. Soil sample extracts were analyzed directly after drying and redissolution with the supporting electrolyte but without other pretreatment. The results were comparable to those obtained by HPLC with UV detection. Therefore, this method can be used in the rapid determination of pesticide/herbicide residues.
