Effects of Arctium lappa Linne on the DNFB-induced allergic dermatitis (우방자(牛蒡子)의 아토피 피부염에 대한 효과)

  • Objectives : Atopic dermatitis is a chronic inflammatory skin disease which is characterized by severe pruritis, erythema, edema, effusion and scabs. The aim of this study was to understand effects of Arctium lappa Linne, which is well known for its efficacy for various skin diseases, on atopic dermatitis Methods : We conducted this experiment using the DNFB-induced NC/Nga mice. After two weeks of applying DNFB to NC/Nga mice, severe symptoms of atopic dermatitis occurred. We divided the mice into three different groups: a control group which was given no treatment at all, a group treated with dexamethasone (1mg/kg), and another group treated with Arctium lappa Linne (300mg/kg). After one week of treatment, results were recorded according to their improvement on skin, itching behavior, IL-4, and INF-$\gamma$ measurement, which is a significant criterion for diagnosing atopic dermatitis. Results : Itching behavior showed significant improvement in the Arctium lappa Linne group. However Arctium lappa Linne failed to reduce IL-4 and INF-$\gamma$ count. Also there was no satisfying improvement on AD-like skin lesions on the rostral back of the NC/Nga mice treated with Arctium lappa Linne. Conclusions : Arctium lappa Linne showed improvement in itching behavior in NC/Nga mice induced by DNFB. Arctium lappa Linne showed neither significant improvement on skin lesions nor in IL-4 and INF-$\gamma$ measurement.

Effect of β-glucan on Epithelial Inflammation Induced by Atopic Dermatitis through Endocannabinoid System Activity (β-glucan의 Endocannabinoid system 활성을 통한 아토피피부염 유발 상피 염증 억제 효과)

  • Objectives This study was conducted to confirm the inhibitory effect of β-glucan on epithelial inflammation induced by atopic dermatitis through Endocannabinoid system (ECS) activity. Methods Six-week-old NC/Nga mice were divided into a control group (Ctrl), atopic dermatitis elicitation group (ADE), and a β-glucan-treated group (β-glucan treatment after atopy dermatitis elicitation, β-GT). After 3 weeks, CB1, CB2, and GPR55 were observed to confirm the regulation of ECS activity, and filaggrin in the stratum corneum and Kallikrein-related peptidase (KLK) 7 in the stratum corneum and protease activated receptor (PAR)-2 were observed to confirm the inhibition of the inflammation, Phosphorylated extracellular signal-related kinase (p-ERK), Phosphorylated mammalian target of rapamycin (p-mTOR), and E-Cadherin were observed to confirm microenvironmental regulation. Results β-GT was significantly increased in CB1, CB2, and GPR55 positive reactions compared to that of the ADE. In positive reaction of the filaggrin in the stratum corneum, β-GT was significantly increased than that of the ADE. For KLK7 positive and PAR2 positive, β-GT was significantly reduced compared to the ADE. The p-ERK-positive and p-mTOR-positive reactions were significantly reduced in β-GT than in ADE. E-cadherin positive reaction was significantly increased in β-GT than in ADE (All p < 0.01). Conclusions It was confirmed that β-glucan has the effect of inhibiting the epithelium induced by atopic dermatitis through the ECS activity.

Yangkyuksanhwa-tang effected to Atopic Dermatitis (양격산화탕(凉膈散火湯)이 Atopy 피부염(皮膚炎)에 미치는 영향(影響))

  • 1. Objectives Yangkyuksanhwa-tang is used mush in pruritus and dermatopathy of Soyangin. It is suggested this prescription is effective on atopy dermatitis. 2. Methods For observation of Yangkyuksanhwa-tang effected to atopic dermatitis, extract of Yangkyuksanhwa-tang has been dispensed to the stratum corneum of epithelium in dermatome of murine after making damage to its defense mechanism against fat and causing atopic dermatitis artificially. After that, the change in outer dermatome and minute mechanism of epidermis, the change of eosinophil, the change in distribution of soybean agglutinin, the change in distribution of fat and ceramide in stratum corneum, the change in inflammation in dermatome, the change of cell accrementition and apoptosis, and the effect on anaphylaxis and Staphylococcus aureus was observed. 3. Results After administration of Yangkyuksanhwa-tang, severe skin damage such as eczema and psoriasis, that was observed in the case of atopy dermatitis, was decreased and the increase of eosinophil in serum was suppressed. Lipid lamella was recovered, so epidermal demage was relieved. The distribution of HSP70 in the outer skin was decreased. Yangkyuksanhwa-tang suppressed activation of $NF-_{\kappa}B$ p50, induced CD11/18b not to be generated, and suppressed inflammatory response of skin. Anaphylaxis and groth of Staphylococcus aureus was suppressed. 4. Conclusions Yangkyuksanhwa-tang decreased skin damage of atopy dermatitis. It has antibiosis about Staphylococcus aureus, it can be medicinal substances on atopy dermatitis. In addition, it is possible that it can be medicinal substances on regional skin allergy.

A Case Report of Atopic Dematitis treated by Osuyu-tang (Wuzhuyu-tang) based on Shanghanlun provisions (『상한론(傷寒論)』 육경(六經) 진단체계에 근거하여 오수유탕(吳茱萸湯)을 투여한 양명병(陽明病) 아토피 피부염 1례)

  • Objective : This case report aims to show the effect of Osuyu-tang on an atopic dermatitis. The patient was diagnosed with Yangmyung-byung based on Shanghanlun provisions. Methods : According to Diagnostic system based on Shanghanlun provisions, the patient was diagnosed with Yanamyung-byung and has been unusual dietary habits. So she took Osuyu-tang decoction for 2 weeks. We evaluated the results of treatment by SCORAD Index and before and after treatment photos. Results : After taking Osuyu-tang decoction for 2weeks, the patient's SCORAD INDEX and symptoms was decreased from 49.5(severe) to 3.5(mile). Also, the patient's chronic diagestive disorder improved. Conclusions : This case report showed an effectiveness of using Osuyu-tang according to Diagnostic system based on Shanghanlun provisions on atopic dermatitis.

A case of eczema herpeticum with atopic dermatitis treated by Oriental Medicine (아토피 환자에 병발한 포진상 습진 치험 1례)

  • Eczema herpeticum is a widespread Herpes simplex virus(HSV) infection, which usually develops in patients with atopic dermatitis, because prutitis in patients with atopic dermatits leads them to scratch their bodies, it cause the dissemination of Herpes simplex virus. This study carried out to observe the progress of Eczema herpeticum and find effective remedy. The pastient was taken Herb-medicine with acupunture, cupping theraphy, aroma theraphy, and etc. After the treatment the grade of VAS was decressed and clinical symptoms were gradually disappeared. Those results suggest that herbal treatment was effective to Eczema herpeticum.

The Inhibitory Effect of Gooseberry on DNCB-induced Atopic Dermatitis in vivo and in vitro

  • Generally, berry fruits have various pharmacological activities such as anti-inflammation, anti-oxidation and anti-cancer effects. The effects of gooseberry, a berry fruits, on atopic dermatitis (AD) have not been widely examined. The aim of this present study is to investigate whether gooseberry modulates AD. We examined the pharmacological effects of gooseberry on 2, 4-dinitrochlorobenzene (DNCB)-induced AD symptoms in mice. To determine the anti-atopic mechanism of gooseberry, we investigated its effects on the production of inflammatory cytokines and activation of nuclear factor-${\kappa}B$ in PMA + ionophore -stimulated human mast cells (HMC-1). The results demonstrated that gooseberry attenuated AD clinical symptoms such as erythema, edema and dryness as well as histamine and IgE serum levels in DNCB-induced AD model mice. Additionally, gooseberry suppressed the expression of inflammatory cytokines and activation of nuclear factor-${\kappa}B$ in stimulated HMC-1. These findings demonstrate that gooseberry is potential agent for treating AD and allergic inflammation.

Oldenlandia diffusa Ameliorates on Atopic Dermatitis in Mice

  • Oldenlandia diffusa (OD), a member of the Rubiaceae family, has been used for treating inflammatory and infectious diseases. However, the anti-atopic effects of OD are not completely understood yet. Thus, the aim of the present study was to elucidate beneficial effects of OD on atopic dermatitis (AD) in vivo. We examined the pharmacological effects of OD on compound 48/80- or histamine-induced scratching behaviors and 2, 4-dinitrochlrobenzene (DNCB)-induced AD-like skin lesions in mice. Additionally, we evaluated regulatory effects of OD on the expression of interleukin (IL)-6 and tumor necrosis factor (TNF)-α in DNCB-induced AD-like skin lesions. Results showed that OD inhibited the scratching behavior and AD symptoms. It also decreased serum levels of IgE and histamine in mice. Moreover, OD significantly ameliorated the expression of IL-6 and TNF-α in AD-like skin lesion. These finding suggest experimental evidence of that OD's potential for treating AD.

  • Atopic dermatitis (AD) is an allergic disorder characterized by skin inflammation. It is well known that the activation of various inflammatory cells and the generation of inflammatory molecules are closely linked to the development of AD. There is accumulating evidence demonstrating the beneficial effects of herbal extracts (HEs) on the regulation of inflammatory response in both in vitro and in vivo studies of AD. This review summarizes the anti-atopic effects of HEs and its associated underlying mechanisms, with a brief introduction of in vitro and in vivo experiment models of AD based on previous and recent studies. Thus, this review confirms the utility of HEs for AD therapy.

A Study of Atopic Dermatitis Treated with Ceramic Based Infra-red Therapy (아토피성 피부염의 광선치료(GI-2000) 효과)

  • Objectives : Ceramic based intra-red Therapy has been effective on dermatitis and immune system. The purpose of this study was to know the clinical effect of ceramic based infra-red therapy on Atopic Dermatitis(AD). Methods: For 3 months from Dec. 2003 to Feb. 2004, we treated ceramic based intra-red therapy to 21 outpatients with AD who visited Hana Oriental hospital. Ceramic based infra-red therapy using GI-2000 system was lighted to undressed outpatients for 15 minutes, three times a week. Results and Conclusions: Ceramic based intra-red therapy showed useful effect on AD, especially itching sign.

Application of oral cyclosporine in a dog with atopic dermatitis (개의 아토피성 피부염에 대한 cyclosporine의 경구 요법)

  • A 7.1 kg, seven-year old, castrated male, Shih-Tzu with severe pruritus, chronic otitis externa and Malassezia infection was referred to Veterinary Medical Teaching Hospital of Chungnam National University. In local animal hospital, steroid therapy was used to treat uncontrollable pruritus, but the clinical signs were recurrent when steroid therapy was discontinued. On physical examination, generalized alopecia, erythema, papules, severe crust and diffuse lichenification were presented. Tape strip test of skin lesions revealed cocci and Malassezia infections. Based on the result of history, clinical signs and examination described above, canine atopic dermatitis with secondary superficial pyoderma and Malassezia dermatitis was diagnosed. Oral challenge with cyclosporine and antibiotics had good results in clinical signs. Clinical sign scores were evaluated by investigator with CADESI at 2weeks, 4weeks, 6weeks, 8weeks and 10weeks after cyclosporine administration. And in the result of comparing of allergen-specific IgE value, the level of allergen-specific IgE to general causative allergen after 10 weeks of cyclosporine therapy was higher than that before cyclosporine therapy.
