Azinphos-methyl이 랫트 태아에 미치는 기형학적 연구 (Teratological Evaluation of Azinphos-Methyl in Rat Fetuses)

  • azinphos-methyl을 S.D.랫트에 투여하여 기형이 유발되는지의 여부와 태아의 기관형성 및 자궁내에서 태아의 발달에 미치는 영향을 알아보기 위하여 무처치의 negative control군, 수도물을 경구투여한 sham control군, 이미 기형효과가 있는 것으로 알려진 aspirin투여의 positive control군, 그리고 azinphosmethyl 0.094 mg/kg, 0.4 mg/kg 및 1.5mg/kg 투여군으로 나누어 각군 30마리씩으로, 임신 6~15일 사이에 경구투여한 결과는 다음과 같다. 즉, 모체의 체중증가율은 임신 7~14일, 즉 약물투여기에 aspirin 투여군과 azinphos-methyl 1.5mg/kg 투여군에서 현저한 감소를 보였다(p<0.01). 모체의 장기무게는 azinphos-methyl 1.5mg/kg 투여군에서 간장의 절대무게비가 유의성(p<0.05) 있는 감소를 보였으며, 신장의 절대 및 상대무게비는 aspirin군에서 (p<0.05, p<0.01), 또 난소의 절대 및 상대무게비는 aspirin 투여군(p<0.01)과 azinphos-methyl 전처치군(p<0.05)에서 유의성 있게 증가하였다. 모체 간장의 단백질량은 aspirin과 azinphos-methyl 1.5mg/kg 투여군에서 현저히 감소되었다(p<0.01). 모체의 배아와 태자에 대한 관찰 결과 azinphos-methyl 1.5mg/kg 투여군에서 암수의 비율에 있어 현저하게 증가(p<0.01) 했으며, 체중은 aspirin과 azinphos-methyl 1.5mg/kg 투여군에서 고도의 유의성(p<0.01) 있게 감소하였고, azinphos-methyl 0.4 mg/kg 투여군에서도 현저하게 감소하였다(p<0.05). 그리고 미숙태자와 흡수태자는 aspirin군에서, 죽은 태자수는 azinphos-methyl 1.5mg/kg 투여군에서 유의성 있게 증가하였다(p<0.05, p<0.01). 태자의 장기에서 나타난 기형은 aspirin과 azinphos-methyl 1.5mg/kg 투여군 공히 횡경막에서 횡격막 천공, 두부에서 무안구증, olfactory bulb의 확장, 수두뇌, 그리고 3뇌실과 측뇌실의 결손, 심장에서 좌심실벽 위축, 심첨확장 등이 유의성(P<0.01, P<0.05) 있게 관찰되었으며, 특히 횡경막, 심장 및 안구등에 높은 기형발생율을 보였다. 또 태자의 골화지연은 두개골에서 후두골, 접형골, 구개, 흉골에서 4번째 흉골편, 검상돌기, 척추에서 경추, 흉추, 미추, 전${\cdot}$후지골, 중족골에서 관찰되었다. 한편, 자연 분만시킨 태자의 사망율은 azinphos-methyl 1.5mg/kg 투여군에서 negative control군에 비해 고도의 유의성(p<0.01) 있게 높음을 알 수 있었으며, 체중도 역시 현저하게(p<0.01) 감소하였고, 발육지표중 pivoting, 체모형성, 청각능력, 시각능력, 사지근육 발달정도 및 고환하강시기 등에서 고도의 유의성(p<0.01)이 관찰되었다.

  • Platelet anti-aggregating activities were tested with analogs of protocatechuic acid and gallic acid. Six of them which showed comparable inhibitory effects with aspirin against collagen induced platelet aggregation were selected and their anti-thrombotic effects were evaluated in the mouse thrombosis model and compared with those of aspirin and paeonol. At the dose of 50 mg/kg, p.o., ethyl gailate(13) treated group showed higher % of recovery within 6 min of thrombotic challenge and lower mortality within 5 min than aspirin treated group.

Lately it has been established by double-blind and randomized clinical studies that regular aspirin intake reduces the incidence of mild strokes and heart attackes.

  • Specific molecular recognition of cyclodextrin and aspirin was determined. A stable 1:1 inclusion complex was established in solution. The distinct structure of inclusion complex was elucidated by FT-IR, FAB-MS, UV, 1H NMR, and 13C NMR spectroscopy. Based on the 1H NMR data, a time-averaged conformation of $\alpha$-cyclodextrin exhibited significant catalytic activity toward the hydrolysis of aspirin in alkaline solution.

  • Aspirin이 위점막(胃粘膜)에 손상(損傷)을 일으키는 기전(機轉)을 해명(解明)하고 침해(侵害)받는 세포(細胞)의 종류(種類) 및 형태(形態)를 구명(究明)하고자 rat에 체중(體重) 1kg당(當) 60mg을 경구(經口) 및 피하(皮下)로 투여(投與)한 후(後) 경시적(經時的)으로 임상소견(臨床所見)을 관찰(觀察)함과 동시(同時)에 위점막세포(胃粘膜細胞)의 형태적변화(形態的變化)를 광학(光學) 및 전자현미경적(電子顯微鏡的)으로 관찰(觀察)한 결과(結果)를 요약(要約)하면 다음과 같다. aspirin의 경구(經口) 및 피하투여(皮下投與)에 의(衣)하여 특별(特別)한 임상소견(臨床所見)이나 위점막(胃粘膜)의 육안적(肉眼的) 및 광학현미경적(光學顯微鏡的) 소견(所見)은 인정(認定)되지 않았으나 미세형태학적(微細形態學的)으로는 투여경로(投與經路)에 관계(關係)없이 명백(明白)한 변화(變化)를 인정(認定)할 수 있었다. 즉(卽) 위점막세포중(胃粘膜細胞中) 가장 심하게 손상(損傷)받는 세포(細胞)는 벽세포(壁細胞)와 주세포(主細胞)로서 벽세포(壁細胞)에서는 SER과 intracellular canaliculi의 증식(增殖)이, 그리고 주세포(主細胞)에서는 RER의 단열(斷裂), 확장(擴張) 및 효소원과립(酵素原顆粒)의 수적감소(數的減少)와 구조적이상(構造的異常)과 같은 가역적(可逆的) 변성변화(變性變化)가 인정(認定)되었으며 점액세포(粘液細胞), 점액경세포(粘液頸細胞), 기저과립세포(基底顆粒細胞)는 비교적(比較的) 경(經)한 변화(變化)를 보였다. 경구투여군(經口投與群)보다는 경미(輕微)하지만 피하주입군(皮下注入群)에서도 경구투여군(經口投與群)에 상응(相應)하는 변화(變化)를 나타낸 점(点)은 aspirin에 의한 위점막(胃粘膜) 손상(損傷)이 aspirin의 직접적영향외(直接的影響外)에 혈중(血中) aspirin 농도(濃度)에 의(依)한 간접적손상(間接的損傷)도 가미(加味)된다는 것을 시사(示唆)하는 형태적소견(形態的所見)이라고 생각(生覺)되었다.

백서에서 Aspirin과 Prostaglandin E1이 미세혈관 문합의 개존에 미치는 효과 (The Effect of Aspirin and Prostaglandin E1 on the Patency of Microvascular Anastomosis in Rat)

  • In the field of microsurgery, the vascular obstruction of the microvascular anastomosis by thrombus is one of the most important complication. The purpose of this study is to compare the effect between aspirin and prostaglandin $E_1$($PGE_1$) which act as the peripheral vasodilatation and platelet disaggregation. We have used total 48 white male rats and divided them into three gruoups(A, B and C group). Each group consists of 16 rats respectively. A group is as control, B group is medicated with aspirin(3.0mg/kg/day) and C group with $PGE_1(1.2{\mu}g/kg/day)$. The gross and histopathologic findings at anastomosed site were observed on 3, 5, 10 and 15 days after vascular anastomosis and the results were obtained as the followings. 1. The microvascular patency rate is 81.2% in control group, 93.8% in aspirin group and 100% in $PGE_1$ group. 2. On the histologic examination, the formation of mural thrombus is decreased both in the aspirin and $PGE_1$ group as comparing with the control group and also the hypertrophy of the intima forming from media is less formed in $PGE_1$ group than aspirin group and the degree of thickeness is also less. 3. The fibrosis of media is less observed in $PGE_1$ group than aspirin group. According to the above results, the application of $PGE_1$ to the microsurgery is considered to be effective on the prevention of the thrombus formation and on providing high patency rate.

청폐사간탕(淸肺瀉肝湯)의 혈소판 응집억제 작용에 대한 in vitro 연구 (Anti-platelet Aggregation Effect of Cheongpyesagan-tang In Vitro)

  • Objective : The study was designed to test the anti-platelet effect of the extract Cheongpyesagan-tang and compare it with aspirin in vitro. Methods : The extract from Cheongpyesagan-tang was made by the pharmacy department of Kyung Hee Oriental Medical Hospital. The extract was investigated for inhibition against the collagen induced aggregation of human platelet suspensions on aggregometry. Aspirin and aspirin-Cheongpyesagan-tang were investigated together. Results : 1. In collagen induced human platelet aggregation test, the extract from Cheongpyesagan-tang significantly inhibited in concentration 30mg/ml (p<0.05), 40mg/ml, 50mg/ml (p<0.001) and the effect depended on concentration over 20mg/ml. 2. Aspirin and aspirin-Cheongpyesagan-tang inhibited collagen induced human platelet aggregation significantly (p<0.001). Aspirin-extract of Cheongpyesagan-tang inhibition rate was higher than aspirin only (p<0.05). Conclusions : The extract of Cheongpyesagan-tang has anti-platelet aggregation and synergic effect with aspirin on human platelet in vitro.

The Combined Effects of Ginkgo Biloba Extracts and Aspirin on Viability of SK-N-MC, Neuroblastoma Cell Line in Hypoxia and Reperfusion Condition

  • Objective: The purpose of this study is to investigate the combined effects of ginkgo biloba extract, ginkgolide A and B and aspirin on SK-N-MC, human neuroblastoma cell viability and mRNA expression of growth associated protein43 (GAP43), Microtubule-associated protein 2 (MAP2), B-cell lymphoma2 (Bcl2) and protein53 (p53) gene in hypoxia and reperfusion condition. Methods: SK-N-MC cells were cultured with Dulbecco's Modified Eagle's Medium (DMEM) media in $37^{\circ}C$, 5% $CO_2$ incubator. The cells were cultured for 8 hours in non-glucose media and hypoxic condition and for 12 hours in normal media and $O_2$ concentration. Cell survival rate was measured with Cell Counting Kit-8 (CCK-8) reagent assay. Reverse transcriptase polymerase chain reaction (RT-PCR) was used to estimate mRNA levels of GAP43, MAP2, Bcl2, and p53 genes. Results: The ginkgolide A and B increased viable cell number decreased in hypoxic and reperfused condition. The co-treatment of ginkgolide B with aspirin also increased the number of viable cells, however, there was no additive effect. Although there was no increase of mRNA expression of GAP43, MAP2, and Bcl2 in SK-N-MC cells with individual treatment of ginkgolide A, B or aspirin in hypoxic and reperfused condition, the co-treatment of ginkgolide A or B with aspirin significantly increased GAP43 and Bcl2 mRNA levels. In MAP2, only the co-treatment of ginkgolide A and aspirin showed increasing effect. The mRNA expression of p53 had no change in all treating conditions. Conclusion: This study suggests that the combined treatments of Ginkgo biloba extracts and aspirin increase the regeneration of neuroblastoma cells injured by hypoxia and reperfusion.

Anti-Platelet Drug Resistance in the Prediction of Thromboembolic Complications after Neurointervention

  • Objective : The aim of this study was to analyze the correlation between thromboembolic complications and anti platelet drugs before and after neurointervention. Methods : Blood samples and radiographic data of patients who received a neurointervention (coil embolization, stent placement or both) were collected prospectively. Rapid platelet function assay-aspirin (RPFA-ASA) was used to calculate aspirin resistance in aspirin reaction units (ARU). For clopidogrel resistance, a P2Y12 assay was used to analyze the percentage of platelet inhibition. ARU > 550 and platelet inhibition < 40% were defined as aspirin and clopidogrel resistance, respectively. Results : Both aspirin and clopidogrel oral pills were administered in fifty-three patients before and after neurointerventional procedures. The mean resistance values of all patients were 484 ARU and < 39%. Ten (17.0%) of 53 patients showed resistance to aspirin with an average of 597 ARU, and 33 (62.3%) of 53 patients showed resistance to clopidogrel with an average of < 26%. Ten patients demonstrated resistance to both drugs, 5 of which suffered a thromboembolic complication after neurointervention (mean values : 640 ARU and platelet inhibition < 23%). Diabetic patients and patients with hypercholesterolemia displayed mean aspirin resistances of 513.7 and 501.8 ARU, and mean clopidogrel resistances of < 33.8% and < 40.7%, respectively. Conclusion : Identifying individuals with poor platelet inhibition using standard regimens is of great clinical importance and may help prevent cerebral ischemic events in the future. Neurointerventional research should focus on ideal doses, timing, choices, safety, and reliable measurements of anti platelet drug therapy, as well as confirming the clinical relevance of aggregometry in cerebrovascular patients.

  • Molecular interaction between ${\alpha}$-cyclodextrin and aspirin was studied by UV, $^2H$-NMR and $^2H$-NMR spectroscopy analyses for solution complex and by FT-IR analyses for solid complex. The inclusion structure provides a basic understanding of the aspirin and ${\alpha}$-cyclodextrin interaction.
