  • A survey of the free amino acids and free sugars in tomato, watermelon, muskmelon, peach and plum was made by means of amino acid autoanalyzer and thin layer chromatography. The results of the survey are summarized as follows. 1) Fifteen amino acids found in fruit were aspartic acid, glutamic acid, alanine, serine, asparagine, lysine, valine, glycine, methionine, histidine, threonine, leucine, isoleucine, proline and arginine, and an unknown was found. 2) Ten kinds of amino acids were detected in tomato, peach and plum, thirteen amine acids in watermelon and muskmelon (edible part), and eleven amino acids in muskmelon (rind). 3) In general, these fruits contained similar amounts of these thirteen amino acids, and although they were not outstandingly high in any one acid they did contain a nutritionally well-balanced mixture. 4) Amino acids found in the greatest amount in fruit were following: glutamic acid and aspartic acid in tomato, asparagine and lysine in watermelon, alanine, serine and aspartic acid in musk-melon, and aspartic acid and serine in peach and plum. 5) Glucose, fructose, sucrose and maltose were detected in all fruit. The contents of glucose and fructose were high, and those of sucrose and maltose were low.

  • The change of food components in seaweeds, masangi (Capsosiphon fulvecense), gashiparae (Enteromorpha prolifera) and cheonggak (Codium fragile) was investigated at various harvest times. The crude protein was $34.1\~43.8\%$ for masangi and $22.5\~35.7\%$ for gashiparae, respectively. The content of glutamic acid, aspartic acid and leucine among total amino acid was high. The major free amino acids were proline and alanine for masangi, asparagine and glutamic acid for gashipare, and hdyrolxyproline, glutamic acid and alanine for cheonggak. In gashipare, the content of aspargine was greatly decreased, while one of sarcosine was increased in March. The ratio of polyene was $63.91\~74.04\%$ for masangi, $62.87\~68.23\%$ for gashipare, and $40.26\~44.61\%$ for cheonggak. The levels of Ca, K, Mg and Fe were high. In masangi and gashiparae, the chlorophyll a and b was greatly decreased in March.

  • The contents of free amino acids in deembryod brown rice of two varieties were investigated by amino acid autoanalizer in relation to specific gravity grade. The analytical methods of free amino acid were also discussed. 1) The lower the specific gravity of the unhulled rice the higher the content of total free amino acids in the deembryod brown rice, and the similar trend appears to hold on each amino acids. 2) Main free amino acids were serine+asparagine, glutamic acid, aspartic acid, alanine and valine, and maximum values of them were 7.3, 5.1, 4.0, 3.4, 0.9mg/100g rice, respectively. They consist about 85% of total free amino acids in most cases. 3) The contents of soluble nitrogen and free amino acids appear to be lower in high protein variety (IR 667) than in low protein variety (Jinhung). The percentage of free amino acid nitrogen to soluble nitrogen, however, appears to be higher in high protein variety (IR 667). 4) Alanine was much lower than aspartic acid in IR 667 having Indica blood while alanine appears to be higher than aspartic acid in Jinhung (Japonica rice) suggesting varietal difference in amino acid metabolism. 5) Threonine peak was overlaped with glutamine, and serine was with asparagine in this study.

  • An actinomycete, AS-568, which produced an inhibitory substance against succinate dehydrogenase of Ehrlich ascites carcinoma and Sarcoma-180 cells of mouse, was isolated. The inhibitory activity was determined by SDI (Succinate Dehydrogenase Inhibition) method. The active substance was specific against carcinoma cells compared to normal cells in mouse; liver, kidney and brain. The inhibitory ratio was about 50% after one hr treatment at 37$^{\circ}C$ in vitro. Maximal productivity of active substance was recognized by 5 days culture in glucose-asparagine. The active component in cultural liquid was stable in neutral pH range and heat treatment reasonably, add it was recovered from precipitate by ammonium sulfate or non-dialyrable fraction in cellophane membrane as showing the behavior of high molecular substance.

  • 본 연구에서는 밤차로부터 용출한 차액의 단당류, 아미노산 및 카페인을 시판중인 현미녹차와 결명자차의 차액과 비교분석함을써 밤차의 기능성 건강음료로 활용하기 위한 기초자료를 규명하였으며 그 결과는 다음과 같다. 1. 현미녹차의 차액에는glucose와 galactose약 20:1 정도의 비율로 검출되었으며 용출시간이나 용출온도에 따른 당류의 변화는 적다. 2. 밤차의 차액에는 glucose, galactose, mannose등이 약 100:1:10의 비율로 존재하여 용출시간이나 용출온도에 의한 변화는 적다. 특히 mannoxe는 천연에서 유리상태로존재하기 어려운 희귀한 유리당이다. 3. 결명자차 차액에서는 당류를 확인할 수 없을 정도로 낮은 농도를 나타냈으며 결명자차의 amino산 함량은 현미녹착나 밤속껍질차의 아미노산 함량에 의해 1.6~1/7에 지나지 않았다.4. 현미녹차의 차액 중 아미노산은 glutamic acid, aspartic acid, serine, methionine, alanine 등의 순서로 검출되었으며, 용출온도와 용출시간에 비례하여 농도가 증가하였고, 아미노산의 용출은 대부분 첫회에 이루어지고 있다는 것을 알 수 있었다. 5. 밤차의 차액 중에 아미노산은 alanine, asparagine, aspartic acid, proline, valine 등의 순으로 많이 함유되어 있고, alanine, asparagine, proline등은 현미녹차에 비하여 함유량이 많으며 glutamic acid, serine, methionine, glutamine 등은 상대적으로 소량 함유되어 있었다. 용출온도와 용출시간에 비례하여 농도가 증가하는 경향을 나타냈으며 아미노산의 요출은 대부분 첫회에 이루어지고 있다는 것을 알수 있었다. 6. 현미녹차는 카페인을 126~162 mg/L 정도로 다량 함유하고 있으나 밤차나 결명자차는 함유하지 않았다.

  • Free amino acids at five different developemntal stages (Gastrulation-Hatching -out stage) of Hynobius leechi BOULENGER were analyzed qualitatively by the use of paper paitition chromatography. It was found that the number of free amino acids increased as the development proceeded. The free amino acids identified at each stages are as follows : Gastrulation stage : Alaninie, Aspartic acid, Glutamin acid, Histidine, Methionine. Neural plate formation stage : Alanine , Aspartic acid, Glutamic acid, Glycine, Histidine, MEthionine, Phenylalanine, Proline, Serine, Trypotophan. Middle tail-bud stage : Alanine, Arginine, Asparagine,Aspartic acid, Citrulline, Glutamic acid, Glycine, Histidie,Hydroxyproline, Proline, Leucine, Methionine, Ornithine, Phenylalanine, Serine, Threonine, Tryptophan. Late tail-bud stage : Alanine, Arginine, Asparagine, Aspartic acid, Citrulline, Glutamic acid. Glycine, Histidine, Hydroxyproline, Leucine, Methionine, Ornithine, Phenylalanine, Proline, Serine, Threonine, Tryptophan, Valine. Hatching -out stage : the same with the late tail-bud stage. It seems probable that the metabolic systems of amino acids before and after the middle tail-bud stage are quite different from each other. Before the middle tail=-bud stage, the reaction system of amino acids is thought not to be completed while after that stage the system has been completed , because in the former period of the development , the number of freeamino acids increased rapidly with the development , and after that stage, there is practically no change in the number of free amino acids.

  • In the preparation of a hydroxyapatite [HAp]/gelatin [GEL] nanocomposite, the GEL matrix in aqueous solution of $H_3PO_4$ was modified by the introduction of aspartic acid [Asp], asparagine [Asn], and glycine [Gly]. The addition of Asp, Asn and Gly greatly affected the slurry formation of HAp/GEL nanocomposite and the resulting dry body showed variations in toughness with the addition of the different amino acids. The introduction of Asn into HAp/GEL nanocomposite was effective for producing the organic-inorganic interaction between HAp and GEL, and caused the increase of toughness. The formation reaction of the modified HAP/GEL nanocomposites was investigated by using XRD and FT-IR. The organic-organic interaction between the GEL matrix and the additives of Asp, Asn and Gly was confirmed from FT-IR analysis, and the organic-inorganic interaction between HAp nanocrystallites and the modified GEL matrix was also discussed, using FT-IR spectra patterns. Nanocrystallites of HAp were covalently bound with the GEL macromolecules and differently influenced by the modification species of Asp, Asn, and Gly.

담배의 바이러스 병엽과 건전엽에 있어서의 유이아미노산에 관한 정량적 연구(예보)

  • A comparative study of free amino acid content in healthy and virus diseased tobacco leaves was carried out by author throughout the gorwing season from June to November of 1963. The methods of qualitative analysis of free amino acids applied in this experiment is followed by Moore and Stein. 1,2 Free amino acids determined in this experiment are shown in Fig. Ⅰ, Ⅱ and Table Ⅰ. As the figure and the table are shown, four more amino acids such as a spartic acid, glutamic acid, tyrosine and phenylalanine are detected in the healthy leaves; these four additional amino acids in the healthy leaves are conspicuous. More quantities of asparagine and alanine are detected in the diseased leaves than the healthy leaves and more quantities of tryptophan is detected in the healthy leaves. It is presumed that such amino acids as tyrosine and phenyllanine are decreased by the incooperation of free amino acid to TMV protein in the process of the process of the leaf protein metabolism which is caused by TMV-RNA trapping action in the diseased leaf protoplasm. It is thought that the decrease of asparagine and the increase of asparic acid in the healthy leaves are the results of in incooperaton of NH2, produced by the protein dissimilation in the diseased leaves, to aspartic acid; it's reaction is caused by the respiration of the diseased leaves accelerated by TMV attack. It is presumed, consequently, that the check of the diseased tobacco leave growth is influenced by the reduction of such amino acids as tryptophane and glutamic acid, which reduction may be due to the abnormal protein metabolism and the action of certain enzyme caused by TMV attack on host protoplast.

Isolation and Identification of Xylose fermenting Yeast (Xylose 발효효모의 분리 및 성질)

  • Ethanol productivity of a xylose fermenting yeast (Candida sp. X-6-4l) isolated from soil was investigated in laboratory scale using Erlenmeyer flask and mini-jar tormentor. The optimal conditions of xylose fermentation in flask experiment were pH 4, asparagine as nitrogen source, xylose 20g/$\ell$, and in these condition, ethanol yield was about 80% to theoretical yield. Using mini-jar fermentor containing 5% total sugar with 2.5% xylose and 2.5% glucose, we obtained 2.3%(v/ v) ethanol and the corresponding efficiency was 72.3% of total sugar. In this case, the consumming speed of sugar under aerobic condition was faster than that of anaerobic condition, and glucose was used previously to xylose. The optimum concentration of xylose for ethanol fermentation in mini-jar fer-mentor scale was 5%, and the efficiency was 69% of total sugar(Alc.2.2% v/v).

Remobilization of Nitrogen Reserves and Analysis of Xylem Exudate during Regrowth of Alfalfa (Medicago sativa) (알팔파의 예취 후 재생시 저장질소의 재이동과 목부 삼출액의 분석)

  • Experiments with non-nodulated alfalfa (Medicago sativa L. cv, Europe) plants grown in hydroponic cuiture, were carried out to estimate the remobiJization of nitrogen (N) reserves and to investigate the transported forms of reduced soluble-N in xylem during regrowth following shoot removal. Endogenous N remobilization were estimated by $^{15}N$ labelling and amino acids in xylem sap were analysed. The, $^{15}N$ contents of ~egr9wing leaves and stems increased as a result of remobilization of N reserves mainly from root ,system, Regrowing leaves were a stronger sink than regrowing stems, with about tWo-thirds of remobilized 15N being recoved in leaves. Endogenous N in lateral roots accounted for about 46% of the total N reserves used for regrowth, while tap roots accounted for 23%, About 72% of total endogenous N remobilized to regrowing shoot, occurred during the first 10 days of regrowth, The outflow of reduced soluble-N (mainly amino acids) was greater than that of protein-N, while the latter was the largest storage pool in tap root and lateral roots. It is suggested that amino acids-N was the most readily avaiable form of N reserves. Asparagine, which repre5ented about 75% of amino acids-N in xylem sap, was the main transported form of reduced N. Its relative contents, during the first 10 days of regrowth, decreased from 75% to 59%. This decline was accompanied by compensatory increase in the relative contents of asparatate and glutamine.
