The Nutritional Composition of Bamboo Shoots and the Effects of its Fiber on Intestinal Microorganisms (죽순의 영양성분 및 죽순의 식이섬유가 장내미생물에 미치는 영향)

  • This study evaluated the composition two popular species of edible bamboo shoots in Korea (Phyllostachyspubescens and Sinoarundinarianigra) and the effect of their abundant dietary fiber on intestinal microorganisms in healthy young women. The ranges of total moisture, crude protein, crude lipid, crude ash, and dietary fiber content were 87.190.8, 2.943.5, 0.150.39, 0.411.05, and 4.206.15% (wet weight basis), respectively. Moisture and crude ash content increased after heat treatment; however, crude protein, crude lipid, and dietary fiber content were reduced after heating. The major minerals found in bamboo shoots were potassium, phosphorous, sulfur, magnesium, and calcium. In addition, glucose and fructose were abundant free sugars, while asparagine and tyrosine were the most abundant free amino acids. Approximately 70% of the total free fatty acids found in bamboo shoots were linoleic acid and linolenic acid. The ascorbic acid content was 6.60~17.56 mg/100 g (wet weight basis), and one phenolic compound, p-hydroxy benzoic acid, was 0.10.2% (wet weight basis) and detected by HPLC analysis. The intake of bamboo shoots for seven days significantly increased viable cell counts of Lactobacillus spp. and reduced viable cell counts of Bacteriodes spp. in feces (p<0.05). In our data, bamboo shoots may be useful in the food industry as high dietary fiber ingredients.

Deamidation on Glutaminyl and Asparaginyl Residues of Protein by Neutrase (Neutrase에 의한 glutamine과 aspargine 잔기의 탈아미드화)

  • Deamidation by neutrase on glutaminyl and asparaginyl residues of protein was examined. The optimum pH and temperature for BSA(bovine serum albumin) deamidation by neutrase were 10 and $20^{\circ}C$, respectively. The incubation for 3 hrs under the optimum condition removed 24% of amide groups and hydrolyzed 2.9% of peptide bonds. Deamidation by neutrase was superior to that by pronase, bromelain, or ficin. Deamidation degrees of egg albumin, soy protein isolate and casein by neutrase under the optimum condition were about 20%, 14% and 14%, respectively. However, relatively high degree of peptide hydrolysis was accompanied with casein deamidation.

Proximate Composition, Amino Acid, Mineral, and Heavy Metal Content of Dried Laver

  • Laver, a red algae belonging to the genus Porphyra, is one of the most widely consumed edible seaweeds. The most popular commercial dried laver species, P. tenera and P. haitanensis, were collected from Korea and China, respectively, and evaluated for proximate composition, amino acids, minerals, trace heavy metals, and color. The moisture and ash contents of P. tenera and P. haitanensis ranged from 3.66~6.74% and 8.78~9.07%, respectively; crude lipid and protein contents were 1.96~2.25% and 32.16~36.88%, respectively. Dried lavers were found to be a good source of amino acids, such as asparagine, isoleucine, leucine, and taurine, and ${\gamma}$-aminobutyric acid. K, Ca, Mg, Na, P, I, Fe, and Se minerals were selected for analysis. A clear regional variation existed in the amino acid, mineral, and trace metal contents of lavers. Regular consumption of lavers may have heath benefits because they are relatively low in fat and high in protein, and contain functional amino acids and minerals.

Single-nucleotide polymorphisms in prion protein gene of the Korean subspecies of Chinese water deer

  • Susceptibility to chronic wasting disease (CWD) in cervid species has been associated with polymorphisms in the prion protein gene (PRNP). The single nucleotide polymorphisms (SNPs) were found in the PRNP of the Korean subspecies of Chinese water deer via analyses of the DNA sequences obtained from 34 individual deer. Two SNPs were detected at codons 77 and 100. One SNP at codon 77 encoding Glycine was determined to be a silent mutation but the other SNP detected at codon 100 induced an amino acid change, from Asparagine to Serine. The prion protein (PrP) amino acid sequence of the water deer showed 98.8-99.2% homology with those of American elk, white-tailed deer and mule deer. The PrP of the water deer contained amino acid residues closely related with CWD-susceptibility. This study is the first to describe genetic variations in the PRNP of the Korean subspecies of Chinese water deer.

  • A fluorescence tagging agent, FMOC-Cl (9-fluorenylmethyl chloroformate) was used for the determination of 1-amino-oligosaccharide intermediates generated from glycoproteins by peptide-N $(N-acetyl-{\beta}-D-glucosaminyl)$ asparagine amidase (N-Glycanase, PNGase F). The derivatives were separated on an Amido 80 column by HPLC using a gradient system with 25 to 51% aqueous acetonitrile and monitored by a fluorometric detector. The detection limit of FMOC-amino-oligosaccharides was $0.05{\sim}1.5$ pmol with fluorometric detection at 278 nm.

Optimal Mixing Ratio of Seafood Sauce with Blue Crab (Portunus trituberculatus)

  • This study optimized the mixing ratios of seafood sauce with blue crab for maximal sensory attributes using a response surface methodology with central composite design. It evaluated sensory appeal, nutritional value, and some rheological properties. The optimal formulation appeared to be 252 g of tomato paste, 78 g of chopped garlic, and 519 g of blue crab block; this ratio resulted in a predicted sensory score of 5.7 (on a 7-point scale) for overall acceptability. Addition of blue crab block increased moisture, ash, and protein contents in various sauces, but fat contents were similar in every sauce. Major free amino acids such as glutamic acid, phospho serine, asparagine, and arginine composed about $60\%$ of the total amino acid content. Standard and optimized sauces were red and light yellow, and those color values were significantly higher than in American-style sauces. Optimal viscosity was half the value of American-style sauces; more blue crab block added to the sauce resulted in lower viscosity. Adding this sauce to seafood dishes will likely aid development of flavoring substances.

  • 볶음조건에 따라 치커리의 유리당 함량은 무처리구에서는 sucrose 4.78%, fructose 0.81% 및 glucose 0.53%이었는데 볶음온도가 증가하고 볶음시간이 길어질수록 감소하여 16$0^{\circ}C$ 40분 볶음처리구에서는 sucrose 2.21%, fructose 0.35% 및 glucose 0.20%로 전체 유리당 함량으로는 약 45.10% 잔존하였다 아미노산은 18종이 분리되었고, 그 중 유리아미노산 함량은 arginine이 504.49 mg%, asparagine이 500.78 mg%, glutamic acid가 97.82 mg%이었고, 충아미노산 함량은 arginine이 676.7 mg%, glutamic acid가 583.9 mg%, aspartic acid가 319.2 mg%, prolineol 297.4 mg%로 다른 것에 비하여 높게 나타났다 볶음온도가 증가하고 볶음시간이 길어질수록 아미노산의 함량은 감소하였고, 그 정도도 다르게 나타났다. 전체함량으로 보면 유리 아미노산의 경우 무처리구에서는 1728.6 mg%이었던 것이 16$0^{\circ}C$ 40분 처리구에서 4.37 mg%로 약 0.25%만 잔존하였고, 총아미노산의 경우 무처리구에서 3159.4 mg%이었던 것이 16$0^{\circ}C$ 40분 볶음처리구에서는 1732.5 mg%이었다.

  • Thermostable asparaginase was purified to homogeneity from mesophilic Strepfomyces lincolnensis M-20 by 30${\sim}$70% ammonium sulfate precipitation and asparagine-Sepharose CL 6B affinity column chromatography, The apparent molecular mass of L-asparaginase by SDS-PAGE was found to be 47 kDa, whereas by its mobility on Sephacryl S-300 column was around 180 kDa, indicating that the enzyme at the native stage acts as tetramer, The purified enzyme showed a single band on acrylamide gel electrophoresis. The optimum pH and temperature were pH 9.5 and 55${\circ}$C, respectively. Chemical modification experiments of purified asparagines implied the existence cystein residue located at or near active site. Purified asparaginase retained the 85% of the initial activity after incubation at 90${\circ}$C for 30 min. A correlation between themostability and resistance to proteolysis of commercial asparaginase and purified asparaginase from Strepfomyces lincolnensis M-20 was investigated. Purified thermostable asparaginase was resistant to trypsin and chymotrypsin treatment, while the commercial asparaginase was not themostable and was susceptible to proteolytic treatment with trypsin and chymotrypsin.

Differentiation of Roots of Glycyrrhiza Species by 1H Nuclear Magnetic Resonance Spectroscopy and Multivariate Statistical Analysis

  • To classify Glycyrrhiza species, samples of different species were analyzed by $^1H$ NMR-based metabolomics technique. Partial least squares discriminant analysis (PLS-DA) was used as the multivariate statistical analysis of the 1H NMR data sets. There was a clear separation between various Glycyrrhiza species in the PLS-DA derived score plots. The PLS-DA model was validated, and the key metabolites contributing to the separation in the score plots of various Glycyrrhiza species were lactic acid, alanine, arginine, proline, malic acid, asparagine, choline, glycine, glucose, sucrose, 4-hydroxy-phenylacetic acid, and formic acid. The compounds present at relatively high levels were glucose, and 4-hydroxyphenylacetic acid in G. glabra; lactic acid, alanine, and proline in G. inflata; and arginine, malic acid, and sucrose in G. uralensis. This is the first study to perform the global metabolomic profiling and differentiation of Glycyrrhiza species using $^1H$ NMR and multivariate statistical analysis.

  • Ethanol extracts of Mung Bean seeds and seedings were analysed by 2-dimensional and circular paper partion chromatography for Nitrogen compounds as a part of the study on the Amino acids metabolism. In the seeds, 18 ninhydrin positive substances were present, before germination, but the number increased to 21 after germination. There were 3 unknown substances and one of it formed newly after germination. After 2-days germination, the amount of amides, such as Asparagine and Glutamine. where increased very large which were very small amount before it. Those were accumulated more in dark place than in light and the amount of Asparigine were more than that of Glutamine. Through the germination, there were large amount of Glutarmic acid, Aspartic acid and Alanine which seems to be concerned in transamination reaction in seedings. Valine, Leucine, and Phenylalanine increased to considerable amount after germination. This is very remarkable fact as those Amino acids were reported to be concerned in transamination reaction recently. ${\gamma}$-amino butyric acid was detected in both Cotyledon and Embroy through the germination. It seemed that there is no any Nitrogen Metabolism in the unbroken seed even if it is preserved very long period.
