Jellies enhancing physiological functions were prepared with using 5 and 10% dietary fiber isolated from ascidian (Halocynthia roretzi) tunic collected from recycled seafood waste. The texture development of the samples was examined with two-bite compression test and analyzed using five sorts ofrheological parameters with texture profile analysis as follows; hardness, adhesiveness, springiness, cohesiveness and gumminess. All the rheological parameters decreased in the fiber enhanced jellies. However, the springiness and cohesiveness increased slightly in the fiber enhanced jellis. Hunter L' and d' values increased in the fiber enhanced jellies and accordingly the color was light yellow. a' value showed green with the addition of fiber. As a result of sensory evaluations, the color and overall acceptability of 10% fiber enhanced jelly were significantly different at p<0.05. The 10% fiber enhanced jelly was noted as having high sensory scores and peferable acceptability.
Strawberry jams were prepared with addition of 1.0 and 2.0% dietary fiber isolated from ascidian (Halocynthia roretzi) tunic for recycling wastes of seafood source and developing new food products. The texture of the jams examined were using a back extrusion rig. All of the rheological parameters decreased in the fiber-enhanced jams. Viscosity profiles were also decreased in the fiber-enhanced jam samples. Hunter L' and a' values increased in the fiber-enhanced jams, accordingly revealing reddish color. As a result of sensory evaluations, the result of taste tests and overall acceptability were significantly acceptable. In the list of acceptability factors, except spreadibility, jam where 1% fiber was added, high sensory scores and preferable acceptability were noted.

In order to determine the utilization of ascidian tunic, which has been blamed for problems of costal environmental pollution when discharged into the sea after being used as a natural dietary pigment sources for rainbow trout(Oncorhynchus mykiss), fingerlings were fed on experimental diets containing acetone-extracts for 6 weeks. The amounts of acetone-extracts were 11,000mg/Kg and contained 50mg/100g wet tissues of carotenoid and $6\%$ of carotenoids were astaxanthin. From the results of feeding experiments, the growth rate in the extract group was a little higher than that of the control and pink groups after 6 weeks. The redness and yellowness of the fish skin and muscle in the extract group were similar to the pink group. Therefore, acetone-extracts of ascidian tunic were judged to be a natural dietary pigment source suitable as a substitute synthetic pigment for aquaculture use.

In this study, the immune-enhancing effects of purified polysaccharides from ascidian (Halocynthia roretzi) tunic were investigated. Crude polysaccharides (AP) were isolated by enzyme extraction (neutrase, $60^{\circ}C$, 15h), ethanol precipitation, and lyophilization. In addition, crude polysaccharides were further fractionated into unabsorbed fractions (APF-I, fraction No. 11~17) and absorbed fractions (APF-II, fraction No. 22~37) by DEAE-sepharose CL-6B column chromatography in order to isolate immune regulating polysaccharide. The major constituents in APF-I and APF-II were total sugar (66.62% and 27.03%), uronic acid (47.53% and 15.87%), hexosamine (16.62% and 46.79%), and protein (2.43% and 4.94%), respectively. APF-I increased production of nitric oxide (NO) and cytokines, such as tumor necrosis factor-alpha (TNF-${\alpha}$) and interleukin (IL)-6 in a dose-dependent manner. The mRNA expression levels of inducible NO synthetase, cyclooxygenase-2, TNF-${\alpha}$, and IL-6 were markedly increased as determined by polymerase chain reaction analysis. The above data led us to conclude that macrophage activation of purified polysaccharides was higher than that of crude polysaccharides. The polysaccharides isolated from ascidian tunic investigated herein are useful as natural immune enhancing agents.
The rheological properties of wheat flour dough and qualities of bread prepared with 0, 10, and 20% of dietary fiber slurry extracted and purified from ascidian (Halocynthia roretzi) tunic were investigated. Water absorption of the dough increased with the increase of dietary fiber slurry. Both arrival and development time of the dough with 10 and 20% dietary fiber slurry added were shorter than those of the control. An increase in the added amount of the dietary fiber slurry resulted in an increase of weakness. The dough's extensibility and resistance to extension were decreased, and the ratio of resistance to extensibility (R/E) decreased with the increase in the dietary fiber slurry. The maximum viscosity gradually decreased with the increase in the amount of dietary fiber slurry, while the temperature of gelatinization was not changed. Both loaf and specific volume of bread were slightly decreased with an increase in the amount of dietary fiber slurry. Overall preference scores by sensory evaluation and the quality characteristics of the bread with up to 20% dietary fiber slurry added was not significantly different from those of the control(p<0.05). The results indicated that the addition of the dietary fiber from ascidian tunic retarded staling and improved the shelf-life of the bread by enhancing the water holding capacity.
Properties of enzymatic hydrolysates from ascidian tunic were assessed on supernatant ratio, solid yields and solid concentration. The concentartion of solid and yields in the extracts were increased as the enzyme concentration raised from $100\;{\mu}l\;to\;1000{\mu}l$ during the extraction period. The optima concentration and reaction time of each enzyme on digestion were $400\;{\mu}l$ 60 minutes, through treated with Duncan's multiple test. The percent of yields of solid, protein and carotenoids for 60 minutes extraction at $400\;{\mu}l$ were $32.32\%,\;1.34\%\;and\;74.60\;mg\%$, respectively, in Viscozyme systems. The extracts were composed with many kinds of carbohydrates such as arabinose, ribose, xylose, galactose, glucose, N-acetyl-D-galactosamine, and N-acetyl-D-glucosamine. Aspartic and glutamic arid were noted as predominant amino acids in all parts. Amino acid profiles of various ascidian tunic part were similiar to each other, but most of essential amino acids content of inter coat was higher than that of root and tunic (body). About sixty six fatty acids components were observed, and their distribution among neutral and polar lipids was compared. The main fatty acids were found to be 14:0, 16:0, 16:1n7, 18:0, 18:1n9, 18:1n7, 18:2n6, 20:5n3, and 22:6n3.
The flow properties of sulfated polysaccharides purified from the ascidian Halocynthia roretzi tunic were investigated. The sulfated polysaccfarides produced aqueous solutions of low apparent viscosity with pseudoplastic flow behavior. The respective activation energies of TCA-treated and TCA-nontreated solutions were $2.2248{\times}10^4\;and\;1.442{\times}10^4J/kg{\cdot}mol$ at a 150L/s shear rate. The viscosity of the sulfated polysaccharides solution was increased by the addition of sugar, while it was not changed by the addition of NaCl.

The effective extraction methods and chemical components of crude polysaccharides of ascidian tunics were investigated. Tow extraction conditions, autoclaving or enzyme treatment, were applied. The proximate composition of ascidian tunics was not much different between those dried in raw (containing pigments) and those acetone treated and dried (decolorized), showing $50\%$ of carbohydrate and $40\%$ of protein. It was possible to extract up to $10\%$ of crude polysaccharides from ascidian tunics regardless of the extraction methods, autoclaving or enzyme treatment. In case of the latter the extraction yield by neutrase was higher than that with alkalase (Novo co.) or mixture 2000 (Pacific chemical co.). The most effective enzyme concentration and extraction time appeared to be 24 hrs of extraction with $3\%$ neutrase. On the other hand, in autoclave treatment, 6 hrs extraction showed most desirable extraction yield, about $9.7\%$. The compositions of amino acid of decolorized ascidian tunic (acetone treated group) and the crude polysaccharide from the autoclaving (water solubles) or neutrase treatment (enzyme digestibles) were similar to each other. Histidine was the highest both in the neutrase and autoclave treatment group and the yield were $29.2\%,\;20.4\%$, respectively, followed by aspartic acid and glutamic acid. Among the minerals, the content of Ca was significantly high, followed by Mg and Na.
As a part of research to elucidate the pathogenesis of so called Soft Tunic Syndrome(STS), that caused mass mortalities in the cultured sea squirts, Halocynthia roretzi, the epidemiological and pathological analysis were done to both clinically normal and diseased groups of the farms of Tongyoung and Geoje coastal areas in southeast sea from February to July, 2008. In the histological finding of the tunic, most of individuals showed tunic softness syndromes that included the disarrangement and destruction of tunic fiber with the simultaneous presence of flagellates-like cells, recently suspected as main agents of tunic softness syndromes. Simultaneously, the intensive degenerative changes of the skeletal muscle of diseased sea squirts were recognized. The changes were characterized with the hyalinization and condensation of muscle fibril and hemocytic infiltration in the muscle fibers. Those were thought to be a kind of typical Zenker's necrosis as in the skeletal muscle of higher vertebrates. Besides of the diseased sea squirts, Zenker's necrosis of skeletal muscles were seen in the normal ones. Epidemiological inquiry for diseased groups revealed that the higher incidences of tunic softness syndrome were recorded in the fast growing groups and in the sites presuming the organic pollution. And Higher malondialadehyde(MDA) and glutathione peroxidase(GPx) activity were detected in the groups showing STS. Those results suggested that Zenker's necrosis of body muscles was a kind of"nutritional myopathy" by oxidative stress. Conclusively, it was considered that Zenker's necrosis of body muscles gives an important clue for elucidating pathogenesis of STS of cultured squirts. And it seems that the necrosis were caused by the oxidative stress to body muscle during abnormal rapid growth of sea squirts.
Economic loss by soft tunic syndrome of edible ascidian, Halocynthia roretzi has become a serious problem. Recently, it has discovered that the cause of this syndrome is infection by a protozoan parasite Azumiobodo hoyamushi. However, only a few studies have been conducted to control this parasitic disease. In a previous research, non-specific disinfectants have been found to be effective in controling the causative parasite. In an attempt to eradicate this causative parasite, organic acids were tested in this study to evaluate their in vitro and in vivo efficacy. In vitro tests showed that 8 different organic acids used in this study were moderately or highly effective with protozoan-killing effects ($EC_{50}=153{\sim}275{\mu}g/ml$). Despite weak in vivo penetration of organic acids into the tunic tissues, treatment with high concentration reduced the mortality of ascidian caused by infection the parasite, indicating that we might be able to develop a disinfection method using environmentally-friendly organic acids.
