  • 돼지 정액의 동결보존은 돼지 정자가 내동성이 약한 특성을 가지고 있어 아직 만족할만한 결과를 얻지 못하여 액상정액을 이용하여 인공수정을 실시하고 있는 실정이다. 돼지 정액의 동결보존을 효과적으로 이룩한다면 돼지 인공수정의 효율성을 증대시킬 수 있고, 우수한 종돈의 보호 및 국제적 돼지 품종의 교류를 증진시킬 수 있을 것이다. 본 실험은 돼지 정액 동결시 동결온도와 시간, 항산화제로 알려진 Taurine의 농도별 첨가, 동해보호제 및 융해조건이 돼지 정액의 동결 융해후 생존성에 미치는 영향을 검토하고자 수행되었다.

Recycling of $CO_2$-Silicate Bonded Sand

  • Once-used $CO_2$-silicate bonded sand from domestic foundry is mostly discarded in a reclaimed land because of its bad collapsibility and reproduction properties. So this causes serious environmental problem. We can get 82% recovery of silica from used sand by scrubbing reclamation process in this research. When we repeat the reclamation-recycling of the foundry sand, artificial silica sand is broken down below 2-cycles, but natural silica sand does not destroyed when used repeatedly more than 10-cycles and have a good property of recycling with little change of its size.

  • Intense artificial selection has been imposed to Luzhong mutton sheep population in the past years. Improvements on growth and reproductive performance are two breeding goals in the present herd. Although some progresses were phenotypically observed possibly due to inbreeding induced by strong selection in terms of these traits, the genomic evaluation was poorly understood. Therefore, a high-density SNP array was used to characterize the pattern of runs of homozygosity (ROH), estimate inbreeding and inbreeding depressions on early growth performance and litter size based upon ROH, and scan positive selection signatures of recent population. Consequently, a low inbreeding level was observed which had negative effects on litter size, but not on early growth performance. And 160 genes were under selection, of which some were reported to be linked to several traits of sheep including body weight, litter size, carcass and meat quality, milk yield and composition, fiber quality and health, and the top genes were associated with growth (growth hormone [GH]- growth hormone receptor [GHR]- Insulin-like growth factor 1 [IGF1] axis) and litter size (bone morphogenic proteins [BMPs]-associated). The effectiveness of previous breeding measures was highlighted, but purging selection was proposed to alleviate the inbreeding depression on litter size, providing some genomic insights to breeding management of Luzhong mutton sheep.

Effect of Pregnancy Rate Following Timing of Artificial Insemination after Estrus of Hanwoo Female

  • This study was conducted to investigate optimal time of artificial insemination (AI) to Hanwoo female after natural estrus. AI was occurred 12 and 24 hours after natural estrus in both heifer and multiparous recipient then pregnancy and parturition rates were estimated. Results indicated that AI performed at 24 hours after natural estrus showed significant (p<0.05) higher pregnancy rate in both heifer and multiparous recipient groups with significantly (p<0.05) higher abortion rate. However, there are no significant differences of parturition rate, twin birth and sex ratio in both heifer and multiparous recipient groups. Therefore, our results may suggest that performance of AI at 24 hours after natural estrus promise higher pregnancy rate than AI at 12 hours after natural estrus in both heifer and multiparous recipient.

Ovarian Cycle, the Biological Minimum Size and Artificial Spawning Frequency in Female Meretrix petechialis (Bivalvia: Veneridae) in Western Korea

  • The ovarian cycle, the biological minimum size, and artificial spawning frequency by artificial spawning induction of the female hard clam, Meretrix petechialis, were investigated by histological observations and morphometric data. The ovarian cycle of this species can be classified into five successive stages: early active stage, late active stage, ripe stage, partially spawned stage, and spent/inactive stage. The spawning period was from June to September, and the main spawning occurred between July and August when the seawater temperature exceeds over $20^{\circ}C$. The biological minimum size (shell length at 50% of first sexual maturity) in females were 40.39 mm in shell length (considered to be two years of age), and all clams over 50.1 mm in shell length sexually matured. In this study, the mean number of the spawned eggs by spawning induction increased with the increase of size (shell length) classes. In case of artificial spawning induction for the clams > 40.39 mm, the number of spawned eggs from the clams of a sized class was gradually decreased with the increase of the number of the spawning frequencies (the first, second, and third spawning). In the experiments of artificial spawning induction during the spawning season, the interval of each spawning of this species was estimated to be 15-18 days (approximately 17 days).

Sexual Maturation and Artificial Spawning of the Hard Clam, Meretrix Iusoria (Bivalvia: Veneridae) on the West Coast of Korea

  • Reproductive cycle with the gonadal phases, first sexual maturity, artificial spawning amount by the size and spawning interval of the hard clam, Meretrix lusoria were investigated by histological observations and morphometric data by artificial spawning induction. Meretrix lusoria is dioecious and oviparous. The reproductive cycle of this species can be classified into five successive stages: early active stage (January to March), late active stage (February to May), ripe stage (April to August), partially spawned stage (June to September), and spent/inactive stage (September to February). The spawning period was from June to September, and the main spawning occurred between July and August when the seawater temperature exceeds over $20^{\circ}C$. Percentage of first sexual maturity of female and male clams ranging from 40.0 to 45.0 mm in shell length was over 50%, and all clams over 50.0 mm in shell length sexually matured. Female and male clams ranging from 40.0 to 45.0 mm in shell length are considered to be two years old. Therefore, we assume that the hard clams of both sexes begin reproduction from two years of age. The mean number of the spawned eggs increased with the increase of size (shell length) classes. In case of artificial spawning induction, the number of spawned eggs from the clams of a sized class was gradually decreased with the increase of the number of the spawning frequencies (the first, second, and third spawnings). In the experiments of artificial spawning induction during the spawning season, the interval of each spawning was estimated to be 15-18 days (average 17 days).

Study on Cryopreservation of Epididymal and Ejaculated Semen in Korean Native Canine and Subsequent Pregnancy Rate after Artificial Insemination

  • This study was carried out to investigate the general characteristics and viability of sperm after freezing and thawing and the pregnancy rates after artificial insemination with thawed semen. The rates of viable sperm after slow and rapid freezing were 87.4±3.85% and 70.8±4.45%, respectively which were significantly lower than that of fresh semen control (91.7±3.45%). The mean concentration of epididymal sperm after dilution in 1.0 ml saline and. 3.0 ml extender in a various concentrations of cryoprotectants was 124.5±48.3 x 10/sup 6/ (range of 45 x 10/sup 6/ to 280 X 10/sup 6/ /ml). There was a significant difference not in the percentage of acrosome-reacted sperm, but in the percentage of capacitated sperm, between fresh and frozen-thawed epididymal semen. When frozen-thawed after diluting with tris-buffer extender containing glycerol, DMSO and ethylene glycol with concentration of 2 to 6%, the rates of epididymal sperm exposed to different cryoprotectants ranged from 14.4±4.7% to 20.7±5.8%, 17.8±5.2% to 36.5±4.9%, and 14.4±4.6% to 18.5±5.3%, respectively which were lower compare to fresh semen control. The pregnancy rate after artificial insemination with frozen semen was 70.6%, whereas that with fresh semen was 90.0% in dogs with naturally induced estrus.

Short Term Storage and Cryopreservation of Trumpet Shell Charonia sauliae Sperm

  • With the purpose to estimate the possibility of short-term storage and cryopreservation for sperm of Charonia sauliae, which is a potential preparation for its artificial reproduction and further research, in this study, protocols for short-term storage and cryopreservation of trumpet shell sperm was optimized. The effects of different immobilizing solutions, dilution ratios were estimated for short-term storage. And the effects of different cryoprotectant extenders and freezing rates were estimated for cryopreservation in terms of motility and survival of sperm. The results indicated that the artificial sea water of 350 mOsmol/kg is a better immobilizing solution and sperm which was diluted at a ratio of 1:1 (v/v) had higher motility and survival rate during short-term storage. The effect of 5% dimethyl sulfoxide was significantly better than those of other cryoprotectant extenders. And a freezing rate of $-20^{\circ}C\;min^{-1}$ showed better effect than other freezing rates. In conclusion, this study optimized some key factors of the short-term and cryopreservation of C. sauliae sperm, which can provide valuable data for germ-plasm conservation and artificial propagation of C. sauliae.

Effects of Tobacco Leaves and Artificial Food on the Growth of Cigarette Beetle, Lasioderma serricorne F. (Coeoptera : Anobiidae) (담배잎과 인공먹이가 궐련벌레 (Lasioderma serricorne F.) 발육에 미치는 영향)

  • Survival, growth and reproduction of the cigarette beetle, Lasiodema sewicome F., were studied on flue-cured (NC82), air-cured (Br.21), aromatic (Izmir) tobacco and artificial diet (wheat feed 95% + yeast 5%). Cigarette beetle adults occurred from first generation in the flue-cured and aromatic tobacco, but no adults emerged in air-cured tobacco after the 2nd generations. The head capsule widths of larvae of the cigarette beetles L sewicome F.1 reared on different diets at 2$0^{\circ}C$, $25^{\circ}C$ and 3$0^{\circ}C$ were measured. The head capsule development was slower at 2$0^{\circ}C$ on the same diets than $25^{\circ}C$ and 3$0^{\circ}C$, and slower in the flue-cured and aromatic tobacco than the artificial diet at 2$0^{\circ}C$ and 2S$^{\circ}C$, At $25^{\circ}C$ the larval head capsule developed more rapidly in the aromatic tobacco than the flue-cured tobacco. However, no larval head development was noticed in the air-cured tobacco. The correlation between population densities of cigarette beetle,1 sewicome F., and sugar content of tobacco leaves was positive, but negative in nicotine contents. The regression between densities and sugar content was Y : 22.79 X + 57.29 (r=0.891**), and between nicotine content was Y : -141.31X + 321.40(r=-0.917**).
