Effects of Twin Birth on the Occurrence of Postpartum Disorders, Culling and Reproductive Performance, and its Risk Factors in Dairy Cows (젖소에서 쌍태분만이 산후질병 발생, 도태 및 번식능력에 미치는 영향과 쌍태 위험 요인)

  • This study determined the effects of twin birth on subsequent postpartum disorders, culling and reproductive performance, and its risk factors in dairy cows. Detailed data related to reproduction, health, and calving events, including dates of artificial insemination (AI) and calving, hormonal treatment prior to conception, cow parity, twin birth, retained placenta, metabolic disorders, and endometritis were collected from 1,717 individual calvings on 9 dairy farms. The incidence rate of twin birth was 3.4%. Gestation length was shorter in cows with twin birth ($270.6{\pm}2.0$ days) than cows giving birth to singletons ($279.5{\pm}0.2$ days, P < 0.01). The incidence of retained placenta (47.5 vs. 16.0%), metabolic disorders (18.6 vs. 3.8%) endometritis (62.7 vs. 28.2%), and culling rate (32.2 vs. 16.5%) was greater in cows with twin birth than in cows giving birth to singletons, respectively (P < 0.01). The hazard of pregnancy (adjusted hazard ratio, AHR) by 210 days in milk analyzed using Cox proportional hazards model was affected by calving season, in that the rate of pregnancy for cows calved during spring was less than for cows calved during winter (AHR = 0.80; P = 0.01); this was also true for endometritis (AHR = 0.46, P < 0.01). AHR was not affected by twin birth. Logistic analysis demonstrated that increasing the cow parity (P < 0.01) and hormonal treatment prior to conception (adjusted odds ratio, AOR = 1.84, P < 0.05) increased the risk of twin birth. These results demonstrate that twin births are responsible for severe economic losses through the increased occurrence of postpartum disorders and culling; thus, appropriate management for the deleterious impact of twin birth, and/or reducing the risk factors by herd control regarding cow parity and the use of reproductive hormones prior to conception might be necessary.

Effects of Feeding of Whole Crop Barley Silage on the Reproductive Performances of Holstein Heifers (청보리 사일리지 급여가 Holstein 젖소 미경산우의 번식 성적에 미치는 영향)

  • This study was carried out between 2008 and 2009 in four dairy farms to investigate the effect of feeding of whole crop barley silage on the reproductive performance of Holstein heifers. Two diets, mixed hay or whole crop barley silage separately from concentrate were fed 6-month old Holstein heifers (=37). In control group (=CON), heifers (n=16) were fed 6 kg (/head) mixed hay and 4 kg (/head) commercial diet. In whole crop barley silage group (=WBS), heifers (n=21) were fed 10 kg (/head) whole crop barley silage, 4 kg (/head) mixed hay and 2 kg (/head) commercial diet. To manage body weight gain, the body condition score of heifers were measured every month. The results obtained were as follows: 1. Body weight in CON and WBS heifers at 13-, 14-, 15- and 17-month old were $340{\pm}17.9$ and $342{\pm}13.6\;kg$, $356{\pm}15.7$ and $366{\pm}14.7\;kg$, $382{\pm}13.1$ and $387{\pm}14.4\;kg$, and $429{\pm}15.0$ and $417{\pm}10.3\;kg$, respectively. 2. Body condition score in CON and WBS heifers at 9-, 12-, 15- and 17-month old were $2.88{\pm}0.04$ and $2.80{\pm}0.04$, $2.88{\pm}0.04$ and $2.80{\pm}0.04$, $2.89{\pm}0.08$ and $3.00{\pm}0.07$, and $2.89{\pm}0.08$ and $3.00{\pm}0.07$, respectively. 3. Average age of sexual maturity in CON and WBS heifers were $437.3{\pm}9.9$days and $939.6{\pm}12.5$days, WBS group heifers were significantly shorter (p<0.05) than CON group heifers. 4. First-service conception rates in CON or WBS group were 81.3% (13/16) and 66.7% (14121), respectively, and cumulative conception rate to 2nd artificial insemination were 87.5% for CON and 85.7% for WBS group. Conception rate was not different between treatments.

Optimization of Electrofusion Condition for the Production of Korean Cattle Somatic Cell Nuclear Transfer Embryos

  • This study was designed to determine the effect of electric field strength, duration and fusion buffer in fusion parameters on the rate of membrane fusion between the somatic cell and cytoplast for Korean cattle (HanWoo) somatic cell nuclear transfer (SCNT) procedure. Following electrofusion, effect of 5 or $10\;{\mu}M$ $Ca^{2+}$-ionophore of activation treatment on subsequent development was also evaluated. Cell fusion rates were significantly increased from 23.1% at 20 V/mm to 59.7% at 26 V/mm and 52.9% at 27 V/mm (p<0.05). Due to higher cytoplasmic membrane rupture or cellular lysis, overall efficiency was decreased when the strength was increased to 30 V/mm (18.5%) and 40 V/mm (6.3%) and the fusion rate was also decreased when the strength was at 25 V/mm or below. The optimal duration of electric stimulation was significantly higher in $25\;{\mu}s$ than 20 and $30\;{\mu}s$ (18.5% versus 9.3% and 6.3%, respectively, p<0.05). Two nonelectrolyte fusion buffers, Zimmermann's (0.28 M sucrose) and 0.28 M mannitol solution for cell fusion, were used for donor cell and ooplast fusion and the fusion rate was significantly higher in Zimmermann's cell fusion buffer than in 0.28 M mannitol (91.1% versus 48.4%, respectively, p<0.05). The cleavage and blastocyst formation rates of SCNT bovine embryos activated by $5\;{\mu}M$ $Ca^{2+}$-ionophore was significantly higher than the rates of the embryos activated with $10\;{\mu}M$ of $Ca^{2+}$-ionophore (70.0% versus 42.9% and 22.5% versus 14.3%, respectively; p<0.05). This result is the reverse to that of parthenotes which shows significantly higher cleavage and blastocyst rates in $10\;{\mu}M$ $Ca^{2+}$-ionophore than $5\;{\mu}M$ counterpart (65.6% versus 40.3% and 19.5% versus 9.7%, respectively; p<0.05). In conclusion, SCNT couplet fusion by single pulse of 26 V/mm for $25\;{\mu}s$ in Zimmermann's fusion buffer followed by artificial activation with $5\;{\mu}M$ $Ca^{2+}$-ionophore are suggested as optimal fusion and activation methods in Korean cattle SCNT protocol.

Spawning Periodicity and Behavior of Amphiprion melanopus and Development of Mass Hatching System for Clownfishes (Cinnamon Clownfish Amphiprion melanopus의 산란 주기, 산란 행동 및 Clownfish류의 부화장치 개발)

  • The marine ornamental industry has become a multi-billion dollar industry these days. As developing, however, this industry has been criticized for the indiscriminate captures and the destruction of the surrounding environment. To circumvent these problems, it is suggested to breed the organisms artificially. While clownfishes Amphiprion sp. and Premnas sp. are the most famous ornamental organisms in the trade, few studies are yet available on the culture and commercial production of these fishes. These studies were performed to investigate the spawning periodicity, behavior and the habits during egg incubation, and to provide the information on the mass hatching system. The spawning periodicity and frequency were different in 4 pairs under the constant condition, temperature, salinity and photoperiod. On the contrary, the male's behaviors for egg incubation are almost same in the all. The egg-fanning activity of the male increased as the developing eggs reaching to the hatching day. Based on the above results, we designed a new artificial hatching system, the rotating type (RT), and compared it with the aeration type (AT) and spray type (ST) that were previously described. RT showed higher hatching rate of 87.3% than AT (74.4%) and ST (60.5%). Also, there were no significant differences in the hatching rate regardless of the number (2, 3, 5) of hatching plates. We suggest RT may accommodate various number of hatching plates and constitute a better hatching system for clownfishes.

Analysis of Frozen-Thawed Sperm Characteristic in Alpha 1,3-Galactosyltransferase(GalT) Knock-out Cloned Miniature Pig (Alpha 1,3-Galactosyltransferase (GalT) Knock-out 복제 미니돼지 정액의 동결-융해 후 정액 성상 분석)

  • The purpose of this study was undertaken to evaluate of cryopreservation efficiency in ${\alpha}$ 1,3-galactosyltransferase knock-out(GalT KO) cloned miniature pig sperm. To compare ability of frozen-thawed sperm characteristics, three different pig strains (GalT KO) cloned miniature pig, PWG miniature pig and Duroc were used. The ejaculated semen from the three pig species was diluted with same volume extender and added to LEY solution for freezing. The diluted semen was placed in 0.5 ml straws, and freezing was initiated by exposing the straws to liquid nitrogen ($LN_2$) vapours for 10 min before placing them into $LN_2$ for cryopreservation. After thawing, the sperm ability were assessed for viability (SYBR-14/PI staining), abnormality (Rose Bengal staining), and acrosome status (intactness, intensity and capacitation) (chlorotetracycline, CTC staining). The viability of frozen-thawed GalT KO pig sperm had no significant difference as compared with Duroc and PWG miniature pig sperm. However, The CTC pattern of frozen-thawed GalT KO cloned miniature pig spermatozoa showed significantly lower rates in F pattern and AR pattern (p<0.05) and significantly higher rates in B pattern than Duroc and PWG miniature pig (p<0.05). The abnormality of GalT KO cloned miniature pig sperm was significantly lower as compared to Duroc and PWG miniature pig sperm (p<0.05). In conclusion, GalT KO cloned miniature pig semen can be cryopreserved successfully and used for artificial insemination reasonably.

Analysis of the Relationship between MHC-DRB1 Gene Polymorphism and Hydatidosis in Kazakh Sheep

  • The objective of this work was to analyze the relationship between ovine major histocompatibility complex (MHC) DRB1 gene polymorphism and genetic resistance to hydatidosis in Kazakh sheep. The Ovar (ovine MHC) class II DRB1 second exon was amplified by polymerase chain reaction (PCR) from DNA samples of 702 Kazakh sheep, including 302 sheep with hydatidosis and 400 health controls. PCR products were characterized by the restriction fragment length polymorphism (RFLP) technique using five restriction enzymes, i.e., MvaI, HaeIII, SacI, SacII and Hin1I, yielding 14 alleles and 28 genotypes. Comparing the frequency of genotypes in hydatidosis sheep with the control group, it was found that the genotype frequencies of MvaIbc, Hin1Iab, SacIIab, HaeIIIde, HaeIIIdf and HaeIIIdd in control sheep were significantly (p<0.01) higher than in hydatidosis sheep, indicating that a significant correlation existed between these genotypes and resistance to hydatidosis. Genotype frequencies of MvaIbb, SacIIaa, Hin1Ibb and HaeIIIef in sheep with hydatidosis were extremely significantly (p<0.01) higher than in the control group, and the genotype frequency of HaeIIIab was significantly higher (p<0.05), indicating that a marked correlation existed between these genotypes and susceptibility to hydatidosis. By way of analyzing haplotype with these resistant genotypes, the hydatidosis resistant haplotype MvaIbc-SacIIab-Hin1Iab of Kazakh sheep was screened out, and then verified through artificial hydatid infection in sheep. The results indicated that the infection rate of sheep with the resistant haplotype of hydatidosis was significantly lower (p<0.01) than without this resistant haplotype. It showed that the genic haplotype MvaIbc-SacIIab-Hin1Iab of Ovar-DRB1 exon 2 was the resistant haplotype of hydatidosis in Kazakh sheep.

  • Treatment conditions for the induced diploid gynogenesis, a maternal-exclusive form of artificial parthenogenetic reproduction, were optimized in bagrid catfish (Leiocassis ussuriensis, Siluriformes). Optimal amounts of ultraviolet (UV) irradiation for the genetic inactivation of spermatozoa in bagrid catfish and Pseudobagrus fulvidraco were proven to be ranged from 3,600 to 4,800 $ergs/mm^2$ based on the examination of viability of embryos and haploid incidence. Haploid embryos were restored to diploidy by preventing the extrusion of the second polar body using cold shock treatment. Thermal treatments (4 or $6^{\circ}C$ for 30, 40 or 50 min) were carried out 3, 5 or 7 min post insemination. Best scores for embryo viability (38.6% of total eggs taken) and incidence of normal diploidy (87.9% of hatched larvae) were observed at the embryo group treated at $4^{\circ}C$ for 40 min, 5 min after insemination. Restoration of gynogenetic diploidy was confirmed based on the absence of haploid syndrome, cell size and/or nucleolar organizing region (NOR) counts.

  • In patients with fully edentulous jaw, treatment of complete dentures should be carried out in many stages when following the conventional methods. Therefore there were disadvantages such as multiple visits to dental clinic is inevitable. In addition, errors caused by polymerization shrinkage, which happens during the fabrication of denture, and difficulties in reproduction of damaged or lost denture were considered as disadvantages. But nowadays, computer-aided design and computer-aided manufacturing (CAD/CAM) system is widely used in dentistry and it has begun to expand its spectrum in manufacturing complete dentures. Using CAD/CAM system to fabricate complete dentures can reduce the number of patient's visit and clinical chair time, since taking impression, recording jaw relation, and selection of artificial teeth are performed at the same time during the first visit, and delivering of dentures during the second visit is possible. In addition, because 3D-Printing technology is used, errors by polymerization shrinkage can be reduced. Among the companies that fabricate complete dentures using CAD/CAM system, DENTCA CAD/CAM denture (DENTCA Inc., Los Angeles, CA, USA) is the most commercialized company. In this case, we treated patients of complete dentures using conventional complete denture method and DENTCA CAD/CAM denture system in the same patient. We would like to report this case because we have achieved good results not only in functional aspects of pronunciation, chewing, and swallowing but also in aesthetic aspects.

The Expression of Plasma Progesterone and Estradiol-17$\beta$ Level before Parturition in the Recipients Pregnant by Hanwoo SCNT Embryos (체세포 복제란 이식 한우의 분만 전 혈장 Progesterone과 Estradiol-l7$\beta$ 농도 변화)

  • This study was performed to analyze the characterization of plasma hormonal levels during pregnancy in the Hanwoo recipients pregnant by artificial insemination (AI) or somatic cell nuclear transfer (SCNT) embryos. The synchronized recipients pregnant by SCNT embryos produced by Hanwoo fetal fibroblast cells (n=8) and by AI (control, n=5) were used. The plasma hormonal levels were measured by RIA (P4 and E2) and ELISA (cortisol), respectively. In control, the increase of E2 and the decrease of P4 were occurred immediately before the initiation of parturition. The expression pattern of plasma P4 was similar in both groups from 50 to 10 days before parturition, however, it did not decrease even at the expected date of labor in the SCNT recipients. The plasma cortisol was expressed a lower level during pregnancy in the SCNT recipients. But, the cortisol was increased in the cow aborted around 100 days of pregnancy (n=1). Based on these results, it can be postulated that the failure of the hormonal changes immediately before parturition in the SCNT recipients may be one of the most important reasons for a delayed parturition in clone calving.

Optimization of Estrus Synchronization Protocol for Target Breeding to Decrease Voluntary Waiting Period in Lactating Cows

  • Effective estrus detection and artificial insemination (AI) are necessary for profitable management of dairy herd. In current study, 45 crossbred lactating cows have been selected with the complaint of unobserved oestrus for more than sixty days postpartum. All cows had functional corpus luteum as examined by transrectal ultrasonography. Cows were treated with $PGF_2{\alpha}$ analogue and AI was performed with observed oestrus and then single dose of GnRH was administered. Similar synchronization protocol has been repeated after 14 days in cows that did not repose to first treatment. Remaining cows received additional $PGF_2{\alpha}$ after 14 days of second treatment and timed AI was performed following GnRH administration. Among 45 cows, 28.89% showed estrus after first treatment and 78.79% responded to second hormonal intervention. A higher conception rate (88.89% vs 26.66 and 72.72%) was observed in cows after triple administration of $PGF_2{\alpha}$ and timed AI. We noticed a significant differences in body condition score (BCS, 1~5 scale), postpartum period, and daily milk production between cows that either responded of non-responded following first and second hormonal treatment. In addition, there was a significant positive correlation between daily milk production and BCS, age and postpartum days, milk production and estrus/BCS, and milk production/BCS/estrus and conception rate. Depending upon the findings we conclude that hormonal intervention with $PGF_2{\alpha}$ and GnRH enhances postpartum ovarian cyclicity and help decreasing the days open of dairy herd. Therefore, this finding might provide an excellent guideline for target breeding system for profitable dairy herd management.
