Studies on the Efficacy of Combined Preparation of Crude Drugs(XXXVIII) -Effect of Gamiinjinoryung-San on Hepatic Damage- (생약복합제제의 약효연구(제38보). -가미인진오령산이 실험적 간장해에 미치는 영향-)

  • Gamiinjinoryung-San is composed of eight crude drugs including Artemisiae capillaris Herba which is widely used for the treatment of acute jaundice, acute and chronic hepatitis at the Oriental hospital of Kyung-Hee Medical Center. This study was conducted to investigate the effects of water extract of Gamiinjinoryung-San on the liver function. The results obtained were as follows; the extract markedly reduced LDH and ALP activities, but slightly decreased GOT and GPT activities elevated with D-galactosamine in rat serum. The liver protective activities were shown in $CCl_4-intoxicated\;rats$. The extract prevented the prolongation of sleeping time in $CCl_4-intoxicated\;rats$. The bile flow and the biliary bile acid secretion were significantly increased in normal rats.

  • In oriental medicine, Artemisiae asiatica (AA) has been used as analgesic, antiinflammatory agent, and coagulatory agent. Furthermore, eupatilin, a kind of flavonoids, is known as the active principle component of AA. This study was undertaken to determine the gastric antiulceration of AA and to elucidate its mechanism. AA showed the inhibitory effect on gastric ulceration induced by EtOH/HCl and aspirin. To elucidate its mechanism, the effect of AA on lipidperoxide level in gastric mucosa, microsome lipidperoxidation, iron -dependent lipidperoxidation, and neutrophil activation were examined. It is suggested that the antiperoxidative and antineutrophil activity of AA play important roles as a possible mechanism. These results suggest that AA might have gastric antiulceration activity due to antilipidperoxdative and antineutrophil activity.

  • Objectives : The leaves of Artemisia argyi L. have been used for the treatment of bleeding-related diseases in traditional korean medicine. But the immunological activities with macrophage have not been sufficiently reported. This study is to investigate the immunological bioactivities of the herbal acupuncture solution obtained from leaves of Artemisia argyi L. (AAAS). Methods & Results : Against Nicotine and Acetaldehyde, AAAS increased significantly the production of hydrogen peroxide (H2O2) within mouse macrophage Raw 264.7 cells above the concentration of 10 ${\mu}g/m{\ell}$. AAAS increased significantly the production of nitric oxide (NO) in Raw 264. 7 cells above the concentration of 100 ${\mu}g/m{\ell}$ against EtOH. And AAAS increased significantly the production of nitric oxide (NO) in Raw 264. 7 cells above the concentration of 200 ${\mu}g/m{\ell}$ against Nicotine, Acetaminophen, and Acetaldehyde. Conclusions : These results suggest that AAAS could be thought to have the immunological activities related with the production of hydrogen peroxide and NO in macrophage.

  • Nitric oxide (NO) can mediate numerous physiological processes, including vasodilation, neurotransmission, cytotoxicity, secretion and inflammatory response. The regulation of NO production by inducible NO synthase (iNOS) is considered to be the possible target of the development of anti-inflammatory agent, based on the observation that NO can activate cyclooxygenase, which results in the synthesis of prostaglandins. In an effort to screen new inhibitor of NO production from about 352 species of herbal extracts, we found 9 species with 50% or more inhibitory effect on NO production. Especially, the dose-dependent inhibition of NO production in lipopolysaccharide-treated macrophages by two of the herbal extracts (Artemisiae asiaticae Herba and Saussureae Radix) was due to the decrease in the expression of iNOS.

  • Objectives: To investigate the effect of fermented extract of Artemisiae Iwayomogii Herba, Curcumae Longae, Crataegi Fructus and Salviae Miltiorrhizae Radix (FMH) on anti-inflammation associated with dyslipidemia and anti-oxidation in RAW264.7 and HUVEC cells. Methods: The total polyphenols, total flavonoids, DPPH radical scavenging activity, ABTS radical scavenging activity, and cytotoxicity of FMH were measured. RAW264.7 cells treated with FMH were tested for production of NO, and for cytokine and LTB4 levels and HUVEC cells treated with FMH were examined for production of cDNA of genes related to inflammation. Results: 1. FMH contained polyphenols and flavonoids. The DPPH and ABTS radical scavenging activity of FMH increased in a concentration-dependent manner. 2. FMH treatment inhibited the production of nitric oxide (NO), cytokines, and LTB4 in RAW264.7 cell when compared to the untreated control group. 3. FMH decreased the transcription of pro-inflammatory genes, whereas it increased transcription of anti-inflammatory genes, in HUVEC cells. Conclusion: FMH is effective as an antioxidant and for treatment and prevention of dyslipidemia, atherosclerosis, ischemic heart disease, stroke, and other cardiocerebrovascular diseases.

Beneficial Effect of DA-9601, an Extract of Artemisiae Herba, on Animals Models of Inflammatory Bowel Disease

  • This study was conducted to investigate the effect of DA-9601, an extract of Artemisiae Herba, which is known to possess mucoprotective action either by free radical scavenging effect or increase of mucus secretion, against animal models of inflammatory bowel disease (IBD) induced by trinirobenzene sulfonic acid (TNBS) or other noxious agents. Experimental colitis was induced by intracolonic administration of TNBS in 50% ethanol, or 1 ml of 7% acetic acid solution (AA), by subcutaneous injection of indomethacin (INDO) in rats, or by supplementing drinking water with 5% dextran sodium sulfate (DSS) in albino mice. DA-9601 was treated orally for 4 to 7 days. Animals were euthanized 1 day after the last treatment for morphological and biochemical analysises. All the noxious agents including TNBS, AA, INDO and DSS elicited severe colitis. The animals treated with DA-9601 showed a consistent, dose-related reduction in the severity of colitis, grossly and histologically. The reduction was significant (p<0.05) after administration of DA-9601 at dose range of 10 mg/kg or above. In TNBS-induced colitis, the rats receiving DA-9601 showed significantly decreased mucosal myeloperoxidase (MPO) and thiobarbituric acid-reactive substances (TBA-RS), when compared to control and mesalazine groups. Mucosal proinflammatory cytokine levels were also decreased after DA-9601 treatment. In conclusion, DA-9601 ameliorated macroscopic and histologic scores in experimental colitis either through decreasing oxidative stress or by attenuating cytokines involved in inflammation. DA-9601 could be a promising drug for the therapy of IBD.

Antioxidant and Anti-dyslipidemic Effect of Artemisiae iwayomogii Herba, Curcumae longae Radix, and Plantaginis Semen Complex Extract(ACP) on HepG2 Cells (HepG2 cell에서 한인진, 울금, 차전자 추출물(ACP)의 항산화 및 항이상지질혈증 효과)

  • This study was performed to investigate the antioxidant and antidyslipidemic effects of Artemisiae iwayomogii Herba, Curcumae longae Radix and Plantaginis Semen complex extract(ACP) on HepG2 cells. We measured total polyphenols, total flavonoids, radical scavenging activity, and ABTS radical scavenging activity of ACP to evaluate its antioxidant activity. HepG2 cells were treated with ACP. Then, we evaluated ROS production; intracellular GSH content; GPx, GR, SOD, and catalase activities; free fatty acids and MDA levels; and mRNA expression levels of ACAT1 and HMG-CoA reductase. Results: ACP contains polyphenols and flavonoids and increased the DPPH and ABTS radical scavenging activities in HepG2 cells in a dose dependent manner. Also, ACP significantly reduced ROS production in HepG2 cells compared to the control group and significantly increased the GSH content, and elevated the enzyme activities of GPx, GR, and catalase in HepG2 cells compared to the control group. In addition, ACP reduced the mRNA expression of ACAT1 and HMG-CoA reductase in HepG2 cells compared to that in the control group. Conclusion: These results suggest that ACP has an antioxidant effect and may suppress the expression of dyslipidemia - associated genes and thus may be useful for the improvement of dyslipidemia.

The Effects of Artemisiae Capillaris, Ponciri Fructus and Cartaegi Fructus in Obese Rats Induced by High Fat Diet (고지방 식이로 유도된 비만 흰쥐에 인진(茵蔯), 지실(枳實), 산사가 미치는 영향)

  • Objectives : This study was designed in order to determine the effects of Artemisiae Capillaris, Ponciri Fructus and Cartaegi Fructus so as to care the obesity and obesity-related diabetes. Methods : After induction of obesity by high fat-diet in rats, the each extracts of three medicinal stuffs were administrated with continuous high fat-diet for 4 weeks. And the changes of lipid metabolism, liver function and blood levels of glucose and insulin were examined. Results : Blood concentrations of triglyceride, free fatty acid and LDL-cholestrol were increased in obese group compared with standard dieted group. By treatment of Ponciri Fructus or Cartaegi Fructus with high fat-diet, their concentrations of glyceride and free fatty acid were significantly decreased. Blood level of LDL -cholesterol was decreased in all stuffs treated groups, however atherogenic index was significantly decreased in treatment of Cartaegi Fructus with high fat-dieted group. Conclusion : High fat-dieted with treatment of Ponciri Fructus or Cartaegi Fructus, fatty degeneration was remarkably diminished.

Antitumor Activity of Bupleuri Radix and Artemisiae capillaris Herba and Synergistic Effect with Anticancer Drugs (시호(柴胡), 인진(茵蔯)의 간암세포(肝癌細胞)에 대한 항암활성(抗癌活性) 및 항암제(抗癌劑)와의 상승작용(相乘作用))

  • In order to prove the antitumer effect of Bupleuri Radix(BR) and Artemisiae capillaris Herba(ACH) experimently, studies were done. The antitumer effect against hepatic cancer such as Hep G2, PLC & Hep 313, and also th synergastic action was evaulatcd in the combined treatment with anticancer drugs using chiefly for liver cancer, such as mitomycin(MMC), cisplatin(CPT) and 5-fluorouracil(5-FU). The results were obtained as follows: 1. IC50 against Hep G2, Hep 3B and PLC was 15.5ug/ml, 25.4ug/ml, 31.25ug/ml in Mitomycin (MMC), 92.5ug/ml, 50.2ug/ml, 62.5ug/ml in cisplatin(CPT) and 125ug/ml in 5-fluouracil(5- FU) respectively. 2. The antitumor effect was shown in the all concentrations of ACH, BR and below 55%-Cytotoxic effect against Hep G2 as compared with the date of control was shown in the concentration of $10^{-4}g/ml$ above of BR but not in ACH and also BR and ACHI revealed the synergistic effect with MMC. 3. The antitumor effect was shown in the concentration of $10^{-5}g/ml$ above of ACH, BR and below 55%-Cytotoxic effect against Hep 3B as compared with the data of control was shown in the concentration of $10^{-5}g/ml$ above of ACH but not in BH and also BR & ACH revealed the svnergistic effect with MMC. 4. The antitumor effect was shown in the all concentrations of ACH, BR and 55%-Cytotoxic effect against PLC as compared with the data of control was shown in the concentration of $10^{-5}g/ml$ above of ACH but not in BR and also ACH revealed the synergistic effect with MMC. From the above results it was concluded that Artemisiae capillaris had antitumor effect against PLC, Hep 3B, Bupleuri Radix against Hep G2 and also MMC showed the most synergistic effect in the anticancer drugs.
