The Protective Effect of Eupatilin against Hydrogen Peroxide-Induced Injury Involving 5-Lipoxygenase in Feline Esophageal Epithelial Cells

  • In this study, we focused to identify whether eupatilin (5,7-dihydroxy-3',4',6-trimethoxyflavone), an extract from Artemisia argyi folium, prevents $H_2O_2$-induced injury of cultured feline esophageal epithelial cells. Cell viability was measured by the conventional MTT reduction assay. Western blot analysis was performed to investigate the expression of 5-lipoxygenase by $H_2O_2$ treatment in the absence and presence of inhibitors. When cells were exposed to 600 ${\mu}M$ $H_2O_2$ for 24 hours, cell viability was decreased to 40%. However, when cells were pretreated with 25~150 ${\mu}M$ eupatilin for 12 hours, viability was significantly restored in a concentration-dependent manner. $H_2O_2$-treated cells were shown to express 5-lipoxygenase, whereas the cells pretreated with eupatilin exhibited reduction in the expression of 5-lipoxygenase. The $H_2O_2$-induced increase of 5-lipoxygenase expression was prevented by SB202190, SP600125, or NAC. We further demonstrated that the level of leukotriene $B_4$ ($LTB_4$) was also reduced by eupatilin, SB202190, SP600125, NAC, or nordihydroguaiaretic acid (a lipoxygenase inhibitor) pretreatment. $H_2O_2$ induced the activation of p38MAPK and JNK, this activation was inhibited by eupatilin. These results indicate that eupatilin may reduce $H_2O_2$-induced cytotoxicity, and 5-lipoxygenase expression and $LTB_4$ production by controlling the p38 MAPK and JNK signaling pathways through antioxidative action in feline esophageal epithelial cells.

Antioxidant Activity of Medicinal Plant Extracts Used as Folk Remedies by Diabetic Patients

  • The aim of this study was to investigate the antioxidant effect of medicinal plants used by diabetic patients. Fourteen medicinal plants were selected and antioxidant activity such as 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity, inhibition effect of linoleic acid autoxidation and low density lipoprotein (LDL) oxidation, thiobarbituric acid (TBA) value were measured. The Cornus officinalis had DPPH radical scavenging activity of 84.79%, which was higher than the 78.79% for $\alpha$-tocopherol. Rosa rugosa Thunberg, Pueraria thundergiana Bentham, Artemisia princeps var. orientalis and Sasamorpha purpurascens Nakai also had high values. Extracts with higher DPPH radical scavenging activities had higher total phenol concentrations, and positive correlations between these parameters were found. Inhibitory activities of linoleic acid autoxidation, LDL oxidation and TBA value, used as indices of oxidative stress, were observed in most of the selected medicinal plants. The highest inhibitory activity for TBA value was observed in the extract of Pueraria thundergiana Bentham. Rosa rugosa Thunberg (75.50 %), Sasamorpha purpurascens Nakai (74.00 %), and Cornus officinalis (73.00 %) all had high antioxidant activity against linoleic acid autoxidation, similar to that of $\alpha$-tocopherol. The same 3 plants also had high in inhibitory activity against LDL oxidation. Therefore, we demonstrated that medicinal plants used as folk remedy by diabetic patients had antioxidant activity. Especially, Rosa rugosa Thunberg, Cornus officinalis, Pueraria thundergiana Bentham, Arlemisia princeps var. orientalis, and Sasamorpha purpurascens Nakai had high phenol concentrations which resulted in high values for DPPH radical scavenging. These same plants exhibited high values for inhibitory activities for TBA value, linoleic acid autoxidation and LDL oxidation, indices associated with lipid peroxidation.

  • Isoniazid was discovered in 1950's and since then it has been widely used as a synthetic bactericidal agent in the treatment of tuberculosis. However, the adverse effect of isoniazid has been reported to show significant hepatotoxicity in approximately 1-2% of patients. Nitrofurantoin {1-(5-nitro-2-furfurylideneamino)-hydantoin} is a synthetic nitrofuran that is commonly used for the treatment and prophylaxis of urinary tract infections, but its use is associated with liver cirrhosis and fatal liver necrosis. Therefore, studies for natural products with protective effect on the isoniazid- and/or nitrofurantoin-induced hepatotoxcity would be valuable as the potential therapeutic use. 107 plants sources were collected at Mt. Baekdu, and extracted with methanol. These extracts had been screened for the protective effects against isoniazid- and/or nitrofurantoin-induced cytotoxicity in HepG2 cells at the both 100 and $300{\mu}g/ml$. Five methanolic extracts, Acanthopanax senticosus, Acer mono, Asparagus schoberioides, Fagopyrum tataricum, Potentilla centigrana, showed significant protective effects against isoniazidinduced hepatotoxicity. Two methanolic extracts, Acer mono and Leonurus artemisia, showed significant protective effects against nitrofurantoin-induced cytotoxicity in HepG2 cells.

In-Jin-Ho-Tang as a potential anti-cancer drug by induction of apoptosis in HepG2 cells

  • Hepatocellular carcinoma is the world's most common primary malignant tumor of the liver. In-Jin-ho-Tang (IJHT) has been used as a traditional Chinese herbal medicine since ancient times, and today it is widely used as a medication for jaundice associated with inflammation of the liver. In-Jin-Ho-Tang is a drug preparation consisting of three herbs: Artemisiae Capillaris Herba (Artemisia capillaries $T_{HUNS}$, Injinho in Korean), Gardeniae Fructus (Gardenia jasminodes $E_{LLIS}$, Chija in Korean) and Rhei radix et rhizoma (Rheum palmatum L., Daehwang in Korean). This study investigated whether or not methanol extract of IJHT could induce HepG2 cancer cell death. Cytotoxic activity of IJHT on HepG2 cells was measured using an XTT assay, with an $IC_{50}$ value of $700{\mu}g/ml$ at 24 h Apoptosis induction by IJHT in HepG2 cells was verified by the cleavage of poly ADP-ribose polymerase, and a decrease in procaspase-3, -8, -9. Treatment of IJHT resulted in the release of cytochrome c into cytosol, loss of mitochondrial membrane potential (${\Delta}{\Psi}_m$), decrease in anti-apoptotic Bcl-2, and an increase in pro-apoptotic Bax expression. Thus, IJHT induced apoptosis in HepG2 cells via activation of caspase and mitochondria pathway. These results indicate that IJHT has potential as an anti-cancer agent.

The Development of Functional Food with Plant Extracts for Enhancing Growth Rate (생약추출물을 이용한 키 성장 기능성 식품 개발)

  • It has been reported that various kinds of chinese herbs have an activity of promote growth rate in both animals and human. To investigate the growth promoting effect of the selected plants, fish and pigs were used as experimental animals. In fish, Eleutherococcus senticosus extract and Artemisia capillasis extract were found to be most effective. And these plant extracts were given to pigs. The result showed that plant extracts-fed pigs were significantly increased their body weight gain at 7.06% of average daily gain, as compared to control. The verify this results, secreted growth hormone (GH)levels and insulin-like growth factor-1 (ICF-1) levels into blood were measured. This result indicated that GH and IGF-1 levels in the blood in plant extracts-fed pigs were higher that those of control. To confirm growth promotion effect on human, we manufactured the mixture of these plant extracts, and coated this mixture onto rice, named as $Kiwoomi^{TM}$. When we administered $Kiwoomi^{TM}$ to elementary students, it was found to be effective in growth promotion. This result showed that $Kiwoomi^{TM}$-treated elementary students significantly increased their growth rate (about 2.14 times), as compared to untreated children. Taken together, it is suggested that this functional rice ($Kiwoomi^{TM}$) might be helpful for growing children without any side effects.

Determination of Phenolic Compounds Responsible for Allelopathy in Upland Weeds (밭 잡초(雜草)중에 존재(存在)하는 Allelopathy 관련(關聯) Phenol 화합물(化合物)의 검색(檢索))

  • Phenolic compounds present in upland weeds (Artemisia asiatica Nakai, Capsella bursa-pastoris (L.) Medik, Portulaca orleracea L. and Trifolium repens L.) which have shown allelopathic activity were determined using paper chromatography (PC) and high performance liquid chromatography (HPLC). Effect of the determined phenolic compounds on germination and post-germination growth of test plants was also investigated. Kinds of phenolic compounds determined by PC in the four weed species were greater in the aqueous extract than in the methanol extract. Ferulic acid was found in both extracts of the weeds studied, whereas benzoic acid was that the weeds commonly contained hydroquinone, p-hydroxybenzoic, ferulic and cinnamic acids. Out of the phenolic compounds determined by PC and HPLC p-hydroxybenzoic, cinnamic and ferulic acids inhibited germination and post-germination growth of radish and sesame. Inhibition of the phenolic compounds on the radicle growth was greater than on the germination of the test plants.

Quality characteristics of Sikhye prepared by different ratio of Seomaeyaksuk (Artemisia argyi H) extract (섬애약쑥 추출물의 첨가 비율에 따른 식혜의 품질특성)

  • This study was conducted to increase the functionality of Korean traditional beverage, Sikhye. The Sikhye were prepared by supplementation up to 20% of Seomaeyaksuk extract (SE) and saccharification for five hours (h). During the saccharification, turbidity of SE-added Sikhye (SES) was the highest at two hours of saccharification and then lowered gradually. L vlaue of SES did not show significant difference feduring saccharification, and both a and b value of Sikhye were increased as saccharification time passed and SE addition ratio was increased. The pH before saccharification was not significantly different by SE addition ratio but it was increased to 5.68-5.73 at 5 h saccharification. Soluble solid, total sugar and reducing sugar contents were enhanced as saccharification time and SE addition ratio were increased. The total phenolic compounds contents and DPPH radical scavenging activity showed a similar trend by saccharification time and SE addition. In sensory test, the evaluations of 5% SES and control were not significant different, but 20% SES group showed significant lower evaluation than other groups because strong color and mugwort flavor. From all of these results, physicochemical characteristics and antioxidant activity of SES was improved by SE addition but sensory characteristics was lower. Acceptable addition ratio of SE in Sikhye is determined to be within 15%.

Changes in Compositions of Fatty Acids According to Drying Methods of Mugwort (Artemisia asiatica Nakai) (쑥의 건조방법(乾燥方法)에 따른 지방산(脂肪酸) 변화(變化)에 관하여)

  • This study was carried out to investigate changes in the compositions of fatty acids according to the different drying methods of raw mugwort. Raw mugwort from Kang-wha was dried with four methods such as sun-drying, shady sun-drying, heated oven-drying and freeze-drying. Total contents and fatty acid compositions of the ethyl ether extracts from the dried mugworts were examined. Total contents of the ethyl ether extracts showed remarkable differences with drying methods; freeze-dried mugwort had the highest content (5.60%), while oven dried mugwort revealed the lowest content (2.45%). Eleven fatty acids and four unknown peaks were identified by gas-liquid chromatography; major fatty acids were linoleic acid, linolenic acid and palmitic acid. Fatty acids of mugworts dried by 4 different methods were the same in kinds, but different in their compositions. The content of polyunsaturated fatty acids $(C_{18:2}+C_{18:3})$ was the highest and the loss of low volatile fatty acids was the least in mugwort dried by the freeze-drying method. The volatile fatty acids considerably decreased in oven-dried mugwort. However, the contents of polyunsaturated fatty acids were a little higher in oven-dried mugwort than in sun-dried and shady sun-dried mugwort, and the lowest in sun-dried mugwort.

Effects of Foot Baths on Stress and exhaustion for high school senior students -A comparative study on the aroma oil and fermentation extract- (족욕이 수험생의 스트레스 및 피로에 미치는 효과 -아로마 오일과 발효추출물의 비교연구-)

  • In the twenty first century, people experience a lot of stresses by psychological, social and environmental impetus. If the stress is excessive, it can become a problem and finally lead to diseases. Foot bath makes the body warm starting with the feet, controls the blood circulations and autonomic nervous systems. This study aims at examining the effects of stress and fatigue of senior students preparing for college entrance examination by foot bath with the aroma essential oil and the fermentation extracts. The experimental period was from March 30 to April 18, 2009. The study objects were 21 senior students of C Girls' High School in Cheonan. They were divided into 3 groups and treated 20 times. The first group was a foot bath group with Artemisia Herbal fermentation extracts. The second group was a foot bath group with the blending aroma essential oil of grapefruit, cypress and cedarwood. The third group was not treated. In the case of difference of physical and psychological stress, there was a reduction of 18.43 and 16.71 for the fermented extract group and 10.71 and 9.43 for the aroma group, while a reduction of 0.43 and 0.29 for the control group, respectively. This shows that foot baths using fermented extracts and aroma had lower physical stress compared to the control group (p<.01). For the difference of fatigue, there was a reduction of 19.14 for the fermented extract group, 11.57 for the aroma group and 0.07 for the control group, showing that foot baths accompanied by fermented extracts and aroma used in this study having lower fatigue compared to the control group (p<.01). The fermented extract group (M=55.71) had the highest level of satisfaction after foot baths, followed by the aroma group (M=49.14) and the control group (M=33.29). Duncan's post hoc test showed that there was a statistically considerable difference between the tests groups and control groups (p<.01). Through the results of this study, it was found that foot baths using bathing supplements were very effective for reducing stress and fatigue.

Studies for Antibiotic Free Chicken Production Using Water Extracts from Artemisia capillaris and Camellia sinensis (인진쑥 및 녹차 추출물을 이용한 무항생제 닭고기 생산 연구)

  • Two experiments were conducted to determine whether water extracts from Artemisia capillaries (A. capillaries) and Camellia sinensis (C. sinensis) could be used as alternatives to antibiotic growth promoters in broiler feed. The experiment 1 was verified their chemical composition, extracts yields, total phenolic compounds concentration, antioxidant activity, antimicrobial activity, and chicken splenocytes proliferation through in vitro test. The extract yields of A. capillaries and C. sinensis were 26.5 and 16.8%, respectively. Total phenolic compounds concentrations of them expressed as gallic acid equivalent were 15.28 and 26.74 mg/mL, respectively. Electron donating abilities of them expressed as $SC_{50}$ showing 50% DPPH radical scavenging were 0.30 and 0.06 mg, respectively. Bacterial inhibitory rates of them against Escherichia coli, Staphylococcus aureus, and Salmonella Typhimurium were ranged from 42.1 to 52.3% and from 21.6 to 33.7%, respectively. And, these extracts increased proliferation of chicken splenocytes. Especially, A. capillaris was more excellent than Echinacea and Concanavalin A known as T-cell stimulator. The experiment 2 was investigated their effects on growth performance, relative organ weight, cecal microflora, blood biochemical parameters, and splenic cytokines mRNA expression in broiler chicks. Four hundred eighty 1-day-old male broiler chicks (Ross 308) were divided in to 4 treatment groups with 4 replicates of 30 birds in each group: NC (control, no antibiotics), PC (avilamycin, 10 ppm; salinomycin, 60 ppm), AC (A. capillaries, 100 ppm), and CS (C. sinensis, 100 ppm); treatments were administered through water supplementation. Final body weight was significantly higher in all treated groups than in NC (p<0.05). Cecal Salmonella numbers were significantly or somewhat decreased in all treated groups than in NC (p<0.05). The relative weights and lengths of the small intestine were more significantly decreased in the PC and AC groups than in the other groups. Cecal Salmonella numbers were significantly or somewhat decreased in all treated groups than in the NC group (p<0.05). The contents of total cholesterol, aspatate aminotransferase, and alanine aminotransferase in blood serum were more significantly decreased in all treated groups than in NC (p<0.05). In conclusion, these results suggested the possibility that these extracts could serve as alternatives for antibiotic growth promoters.
