  • The apparent Michaelis constant Km of extracellular adenine deaminase from Streptomyces sp. J-350P was 5.8$\times$10$^{-5}$M. The activation energy or the enzyme was calculated from Arrhenius plots for adenine and the value was 3.13 Kcal/mole. The purine analogues, 6-chloropurine, 2,6-diaminopurine, 6-bromopurine, 4-aminopyrazolo[3,4-d] pyrimidine, 6-iodopurine, and 8-bromoadenine were substrates for the enzyme. 6-Dimethylaminopurine was a competitive inhibitor of the enzyme. The enzyme was inhibited by 0.1mM of Fe$^{3+}$, Ag+, and Hg$^{2+}$ and 1 mM of $\alpha$, $\alpha$'-dipyridyl, Penta-chiorophenol, and p-chloromercuribenzoate.

Physicochemical Characteristics of Cephalosporin Derivative, CKD-604 : Stabilization and Solubilization in Aqueous Media (세팔로스포린계 유도체 CKD-604 물성연구 : 수용액중에서의 안정화 및 가용화)

  • To formulate the parenteral delivery of a new cephalosporin derivative, 7-${\beta}$-[(2)-2-(2-arninothiazol-4-yl)-2methoxyiminoacetamido]- 3- [(2,3-cyclopenteno-4-carbamoyl-l-pyridinium)methyl]- 3-cephem-4-carboxylate sulfate( CKD604), the stability and solubility of CKD-604 in various aqueous media were investigated. The degradation kinetics of CKD-604 in aqueous solutions (ionic strength 0.1, pH 1-8) were studied at $37^{\circ}C$. The observed degradation rates followed pseudo first order kinetics. The pH-rate profile exhibited a minimum degradation rate at pH 5. The Arrhenius activation energy was 14.2 kcal/mol in pH 5 buffer solution. Excellent agreement between the cephalosporins' theoretical pH-rate profile and the experimental data indicated that the degradation pathway of CKD-604 could be predicted according to the general pathway of cephalosporins. The solubility of CKD-604 was 8.16 mg/ml at $25^{\circ}C$. To enhance the solubility and adjust the suitable pH, CKD-604 was solubilized by using sodium ascorbate, ascorbic acid and urea. The compositions were obtained to satisfy optimum pH and concentration, and the total amount of additives was several times of the active ingredient, CKD-604.

  • In this paper, EPR (ethylene propylene rubber) insulation material was accelerated degradation test at $121^{\circ}C$, $136^{\circ}C$, $151^{\circ}C$, and experiment the typical EAB (elongation at break) at mechanical characteristics analysis. It is shown that the failure-time at the point of 50% of the initial value of Elongation rate to obtain the activation energy. The failure-time was shown each 5,219 hr, 3,165 hr, and 668 hr at three temperatures. In order to derive the activation energy, Arrhenius methodology was applied. Also, we got the Arrhenius plot from three accelerated temperatures. The activation energy values got 0.98 eV from EAB test. The experimental data were evaluated for estimating the probability density, and the suitable distribution by using statistical program MINITAB. It is shown that EAB data by the acceleration thermal degradation is most suitable for the Weibull distribution.

  • The drying characteristics of fluidized bed drying with different drying conditions using naked barley were carried out. This fluidized drying mechanism of naked barley was consisted of consecutive two falling rate parts, first falling rate period and second falling rate period without showing constant rate period. The drying rate constant was increased with decreasing charged amount and relative humidity and increasing air temperature and air velocity. Since the drying rate constant expressed by Arrhenius type equation in the falling rate period showed good linearity, the falling rate period was considered as the controlling step. The activation energy of first falling step was 1,900 cal/gmol, while for second falling step the values showed 2,500 cal/gmol.

Computation of Ionic Conductivity at NASICON Solid Electrolytes (II) Effects of mid-Na Sites on Na1-Na2 Conduction Paths (NASICON 고체 전해질의 이온 전도도 계산 (II) Na1-Na2 전도 경로에 미치는 mid-Na의 영향)

  • The ionic conductivity of NASICON solid electrolytes was simulated by using Monte Carlo Method (MCM). There were included two conduction paths: (1) Na1-Na2 and (2) Na1-Na2 including Na2-Na2. We assumed that mid-Na ions provde an additional driving force for Na mobile ions due to the interionic repulsion between Na1 and Na2 ions. The inflection point of vacancy availability factor, V has been shown at nearby x=2, the maximum mid-Na ions. The inflection point of vacancy availability factor, V has been shown at nearby x=2, the maximum mid-Na sites are occupied. The effective jump frequency factor, V has been shown at nearby x=2, the maximum mid-Na sites are occupied. The effective jump frequency factor, W increased rapidly with the composition at low temperature, but decreased at high temperature region. On Na1-Na2 conduction path, the minimum of charge correlation factor, fc and the maximum of $\sigma$T were appeared at x=2.0. this indicated that mid-Na ions affect on the high ionic conductivity behavior. At the whole range of NASICON composition, 1n $\sigma$T vs. 1/T* plots have been shown Arrhenius behavior but 1n (VWFc) vs. 1/T* have been shown the Arrhenius type tendency at x=2, which mid-Na is being the maximum. The results of MCM agreed with the experimental one when the chosen saddle point value was 6$\varepsilon$ : 3$\varepsilon$. Here the calculated characteristic parameter of materials, K and the phase transition temperature were -4.001$\times$103 and 178$^{\circ}C$ (1/T*=1.92, 1000/T=2.22), respectively.

  • The crystallization behaviors of slag-based glass in $CaO-MgO-Al_2O_3-SiO_2(-Na_2O)$ system have been studied. The mother glass containing 16.50 CaO, 7.50MgO, 19.70Al2O3, 50.80SiO3 and 2.09wt% $Na_2O$ was prepared by using Korean domestic raw materials such as granulated slag, serpentine, sea sand and etc. The glass-ceramics composed of major crystalline phase diopside was produced by the heat treatment in a temperature range from 850$^{\circ}$ to 9$25^{\circ}C$ for 0-6hr. The composition and morphology of diopside phase formed in the system were examined by X-ray diffraction analysis and electron microscopy. The kinetic measurements such as J.M. A plot and Arrhenius plot indicated that the process of nucleation of the initially formed diopside phase could be described from the view point of instantaneous nucleation. It was also demonstrated that the linear crystal growth of diopside phase was proceeded by short range diffusion of $Mg^{2+}$ and $Ca^{2+}$ ion. The microstructures of the resulting glass-ceramics were consisted of leafroidal shaped crystalline aggregations.

Enhancement of Hydroxylamine Reactivity of Bacteriorhodopsin at High Temperature

  • Recent denaturation experiments of bacteriorhodopsin (bR) in the dark and under illumination at high temperatures revealed that irreversible thermal bleaching occurs above ~ 70°C and the preceding reversible structural changes in the dark above 60°C are closely related to irreversible photobleaching observed in the same temperature range (Yokoyama et al. (2002). J Biochem. 131,785). In this study, structural properties of bacteriorhodopsin (bR) at high temperatures were extensively probed by hydroxylamine reactivity with the Schiff base in the dark and hydrogen-deuterium (H-D) exchange in the peptide groups. In the Arrhenius plot from kinetics measurements of the hydroxylamine reaction, a good linear relationship between the reaction time constant and the inverse of the absolute temperature was observed below 60°C, while significant increase started above 60°C, suggesting that remarkable increase in water accessibility of the Schiff base in the temperature region. FT-IR spectroscopic studies on the H-D exchange suggested increase in the deuterium exchanges rate of the peptide hydrogen in the same temperature region.

Kinetic Measurements of Irreversible Photobleaching of Bacteriorhodopsin in A High Temperature State

  • Irreversible photobleaching of bacteriorhodopsin (bR), namely denaturation induced by illumination of visible light, was investigated by absorption kinetic measurements. The denaturation kinetics revealed that light illumination significantly enhanced the structural decay of bR. The kinetic analyses showed that the molecular structure of bR denatures according to a single-exponential decay, whereas irreversible photobleaching has two decay components. The decay constant of the slow component of photobleaching is almost same as that in the dark. An Arrhenius plot of the denaturation kinetic constants for the fast and slow components showed similar activation energies of approximately 19 kcal/mol.

  • BiSrCaCuO superconducting thin films have been fabricated by co-deposition using IBS(Ion Beam Sputtering) method. Despite setting the composition of thin film Bi2212 or Bi2223, in both cased, Bi2201, Bi2212 and Bi2223 phase were appeared. It was confirmed the obtained field of stabilizing phase was represented in the diagonal direction of the right below end in the Arrhenius plot of temperature of the substrate and $PO_3$ and it was distributed in the reeone.

Synthesis of Silicon Nitride from Ethyl Silicate(I) (Ethyl Silicate로부터 Silicon Nitride의 합성(I))

  • Mixtures of carbon and silica (about 0.46${\mu}{\textrm}{m}$) which was synthesized by the hydrolysis of ethyl silicate, the molar ratio of silica/carbon was fixed as 1/10(weight ratio : 1/2), were nitrided in the temperature range 135$0^{\circ}C$~150$0^{\circ}C$. The phse of the product Si3N4 was $\alpha$ phase and the morphology was hexagnoal prism and the nitridation reaction was completed in 5 hrs at 150$0^{\circ}C$ or 7hrs at 145$0^{\circ}C$. The reaction rate above 150$0^{\circ}C$ was diffusion-controlled, following Jander equation. Activation energy Q was derived from the Arrhenius plot and the value was about 101kcal/mol. Axis ratio of Lattice constants(c/a) was 0.726 and unit volume was $\AA$3, the larger the molar ratio of carbon/Alkoxide was, the smaller the particle size of $\alpha$Si3N4 was.
