Development of Natural Seasoning from Alaska Pollack Skin Gelatin Using Continuous Three-Step Membrane Reactor (연속식 3단계 막 반응기를 이용한 명태피 젤라틴으로부터의 천연조미료 개발)

  • The hydrolysates of three kinds [FSEH(first step enzymatic hydrolysate), SSEH(second step enzymatic hydrolysate), and TSEH(third step enzymatic hydyolysate)] were prepared by continuous hydrolysis of Alaska pollack(Theragra chalcogramma) skin gelatin with three-step membrane enzyme reactor. The molecular weight distributions of FSEH, SSEH, and THSE are 9,500∼4,800Da, 6,600∼3,400Da, and 2,300∼900Da, respectively. The contents of amino acid having sweet taste (glycine, proline, serine, alanine, hydroxyproline, glutamic acid, and aspartic acid) were about 70% of total amino acid being in the three kind hydrolysates. We also tried preparing of natural seasonings (complex seasoning and enzymeatic hydrolysale sauce) using the hydrolysates. From the results of sensory evaluations, complex seasoning containing TSEH was nearly equal to shellfish complex seasoning on the market. The mixture sauce which was made by mixing of 80% enzymatic hydrolysis sauce and 20% fermented soy sauce, was at least similar to the tradition soybean sauce in product quality, too.

  • The flavor constituents and functionality of small kingfish were studied as affected by two stage enzyme hydrolysis (TSEH). Total free amino acid contents in water extract, autolytic extract and TSEH of small kingfish were 541.3 mg%, 8,245.3 mg% and 6,636.6 mg%, respectively. Major free amino acids in TSEH were hydroxyproline, glutamic acid, proline, leucine, phenylalanine, lysine, arginine. As for nucleotides and other bases, IMP, TMAO and total creatinine were principal components in TSEH. And the major inorganic ions in TSEH were Na, K, P, Cl and $PO_{4}$. Also TSEH of small kingfish revealed very higher Angiotensin-I converting enzyme inhibition effect than those of water and autolytic extract.

Comparision of Chemical Components of Angelica gigas Nakai and Angelica acutiloba Kitagawa (토당귀와 일당귀의 화학성분 비교)

  • Chemical components of domestic Angelica gigas Nakai and Angelica acutiloba Kitagawa were analyzed. Proximate analysis of each species showed crude protein 18.1% and 13.4%, crude lipid 8.9% and 4.3%, crude fiber 8.6% and 9.4%, crude ash 7.4% and 8.2%, and carbohydrate 57.0% and 64.7%, respectively. Contents of potassium which was found to be the most abundant mineral in both species were 2,740.0 mg% and 2,582.8 mg%, and those of sucrose were 0.4 % and 0.3% respectively while neither fructose nor glucose were detected in each species. Major fatty acids in Angelica gigas Nakai and Angelica acutiloba Kitagawa were linoleic acid (60.8% and 59.5%), palmitic acid (17.4% and 15.3%), oleic acid (8.8% and 7.7%) (respectively) but there was no significant difference between two species. Arginine was revealed as the most abundant amino acid in both species with 2,599.8 mg% in Angelica gigas Nakai and 1,543.4 mg% in Angelica acutiloba Kitagawa. Angelica gigas Nakai and Angelica acutiloba Kitagawa also were shown to contained 10.5 mg% and 12.2 mg% $vitamin\;B_1$, 0.1 mg% and 0.04 mg%, $vitamin\;B_2$, 4.3% and 0.8% decursin, and 988.0 mg% and 900.0 mg% tannin, respectively.

  • 본 연구는 육성돈에 있어 인분해효소인 phytase의 첨가가 환경친화성사료개발을 위한 기초자료로 사용하기 위해 영양소 이용률을 측정하였다. 대조구는 옥수수-대두박의 육성돈 사료이고, 처리구는 대조구 사료에 Phytase의 첨가 수준을 500 unit 및 1,000 unit을 첨가한 구로 3×3 Latin Square 방법으로 실시하였다. 건물과 질소에 대한 소화율에서는 각 처리구간에 유의적인 차이가 없었다(P>0.05). 회분, 칼슘 및 인의 소화율의 경우 phytase 첨가구가 대조구에 비해 높은 수준을 보였다(P<0.05). 돼지가 섭취한 인의 양은 phytase 첨가구가 대조구에 비해 증가하였다. 또한 인의 흡수량에서 phytase 첨가구가 대조구에 비해 높았다(P<0.05) 분과 뇨의 총 인 배설량은 유의적인 차이를 나타내지 않았다. 칼슘의 섭취량은 처리구가 대조구에 비해 높았다. 칼슘의 분 배설량은 각 처리구간에 별 다른 차이를 보이지 않았으나(P>0.05), 뇨의 배설량은 처리구가 대조구보다 적은 양이 배설되었다. 소화된 아미노산 중 cystine, isoleucine, tyrosine, phenylalanine은 처리구가 대조구에 비해 소화율이 현저히 증가된 것을 알 수 있었다(P<0.05). Asparagine, threonine, serine, methionine, leucine의 경우 phytase를 500 unit 첨가한 구가 높은 소화율을 나타내었다(P<0.05). 한편 lysine, histidine, arginine 등의 다른 아미노산은 처리구가 대조구보다 약간의 소화율 증가를 보였으나 통계적 유의차는 나타나지 않았다(P>0.05). 결론적으로 인분해효소 phytase를 사료내 첨가하므로서 분중 영양소를 줄일 수 있는 환경친화성 사료를 개발 가능하다고 사료된다. 체결홈이 형성된 체결부를 갖는 안경집과 이것에 결합되는 체결구가 마련되어 개폐되는 커버가 형성된 케이스로 구성된다.로 금형설계 및 제작기간을 단축하고자 한다.ere demonstrated in rats after i.c.v., intraperitoneally and orally administration, respectively. Effect of Yijin-tang on Gastric Motility between Normal Intact and Partial Pyloric Obstructed Rats (이진탕(二陳湯)이 정상 및 위 유문부가 부분폐색된 흰 쥐의 위 운동성에 미치는 효과)

  • Objectives : This study was aimed to investigated the effect of Yijin-tang on gastric motility and its mechanism of action in normal intact and partial pyloric obstructed rats. Methods : Gastric emptying was measured by the number of glass beads expelled from the stomach (containing one hundred of glass beads. ${\phi}1mm$) in 1 hour or 2 hours after glass beads and test drugs (normal saline. Yijin-tang 90mg/kg. Yijin-tang 270mg/kg) administration in normal intact and partial pyloric obstructed rats. In another series of experiments to evaluate the mechanism of Yijin-tang 270mg/kg under delayed conditions, normal intact rats were treated with atropine sulfate (1mg/kg,s.c.), cisplatin (10mg/kg,i.p.), quinpirole HCI (0.3mg/kg,i.p.) and NAME (NG-nitro-L-arginine methyl ester. 75mg/ kg,s.c.), respectively. Partial pyloric obstructed rats were modified by wrapping the nonabsorbable rubber ring (D :6mm, W:4mm, T: 1mm) around the 1st portion of the duodenum for 8 weeks. The myoelectrical activity of the gastric smooth muscle was recorded by a bipolar electrode placed at the abdominal surface in normal intact and partial pyloric obstructed rats. The gastric myoelectrical activity was measured for 30 minutes before and after orogastric administration of each solution (normal saline, Yijin-tang 270mg/kg) and expressed as dominant frequency, percent of normogastria and power ratio. Results : Yijin-tang improved gastric emptying more than normal saline in normal intact(p<0.001) and partial pyloric obstructed rats(p=0.002). Under the delayed gastric emptying induced by atropine sulfate, cisplatin, quinpirole HCI and NAME. Yijin-tang enhanced gastric emptying significantly in the cisplatin treated group(p<0.001). but didn't in other treated groups. Administration of Yijin-tang 270mg/kg has no significant effect on the myoelectrical activity of the gastric smooth muscle in both normal intact rats and partial pyloric obstructed rats. Conclusions : Yijin-tang seems to stimulate the gastric motility through suppressing the 5HT3 receptor and promoting the antroduodenal flow. We expect that Yijin-tang would be effective especially in dysmotility-like functional dyspepsia with partial pyloric obstruction or the side effects of cisplatin such as nausea, vomiting, abdominal discomfort, and delay of gastric emptying.

Change of amino acids contents of Gastrodia elata Blume with harvest times and seed tuber (수확시기와 자마의 특성별 천마의 아미노산 함량 변화)

  • Gastrodia elata Blume, an achlorophyllous orchid plant, has been used in traditional medicine and harvests in spring and fall. Here we investigated the quantitative changes of amino acids in G. elata by harvest times and seed tubers. In the results, we found that there was not big difference in contents of total amino acids but the distribution of amino acids differed depending on harvest times. 19% of total amino acids were asparagin and valine, an essential amino acid, occupied 10% within total amino acids in the tubers harvested in October. Among amino acids in the tubers harvested in April, serine occupied 23% and arginine, an essential amino acid, occupied 10% within total amino acids in tuber. Interestingly, the use of sexually propagated seed tubers produced high concentration of total amino acids compared to vegetatively propagated seed tubers. As for sexually propagated seed tubers, essential amino acids contents similar to the tubes harvested in October and nonessential amino acids contents similar to the tubers harvested in April. In this study, we found that amino acids contents in G. elata tubers altered depending on various cultivation practices. Therefore if these results can be applied to food industry, the value of G. elata as a natural food resource will be enhanced to a great extent.

Effect of Glutathione on Lead Induced Modulation of NO Synthesis in RAW 264.7 Cell (RAW 264.7 Cell에서 납에 의한 NO 생성의 조절에 미치는 Glutathione의 효과)

  • Objectives : To evaluate the elect of glutathione(GSH) on lead induced modulation of nitric oxide(NO) synthesis, and to examine how lead modulates NO production in macrophages. Methods : This study was observed in a culture of RAW 264.7 cells, which originated from a tumor in a Balb/c mouse that was induced by the Abelson murine leukemia virus. The compounds investigated were lead chloride, N-acetyl-cystein(NAC), and Buthionine Sulfoximine( BSO). Results : ATP synthesis in RAW 264.7 cells was unchanged by each lead concentration exposure in a dose dependent manner. The NO synthesis was decreased when exposed to lead($PbCl_2$) concentration $0.5{\mu}M$. The presence of $300{\mu}M$ NAC, used as a pretreatment in the culture medium, caused the recovery of the lead induced decrease in NO synthesis, but in the presence of $300{\mu}M$ BSO as a pretreatment, there was no recoverey. Pretreatment with NAC and BSO had no affect on ATP synthesis at any of the lead concentrations used. Conclusions : These results indicated that GSH has a protective effect toward lead toxicity, and suggested that the inhibition of NO production in macrophage due to lead toxicity may be related to cofactors of iNOS (inducible nitric oxide synthase)

Investigation of the Gene Encoding Isotocin and its Expression in Cinnamon Clownfish, Amphiprion melanopus (Cinnamon clownfish Amphiprion melnaopus의 이소토신 유전자 구조와 삼투압 조절이 미치는 영향)

  • Isotocin (IT), a nonapeptide homolog of oxytocin in mammals, has been suggested to be involved in physiological processes including social behaviors, stress responses, and osmoregulation in teleost fish. To study its structure and function, the gene encoding the IT precursor was cloned from the genomic DNA and brain cDNA of the cinnamon clownfish, Amphiprion melanopus. The IT precursor gene consists of three exons separated by two introns, and encodes an open reading frame of 156 amino acid (aa) residues, comprising a putative signal peptide of 19 aa, a mature IT protein of 9 aa, a proteolytic processing site of 3 aa, and 125 aa of neurophysin. Tissue-specific analysis of the IT precursor transcript indicated its expression in the brain and gonads of A. melanopus. To examine its osmoregulatory effects, the salinity of the seawater (34 ppt) used for rearing A. melanopus was lowered to 15 ppt. Histological analysis of the gills indicated the apparent disappearance of an apical crypt on the surface of the gill lamella of A. melanopus, as pavement cells covered the surface upon acclimation to the lower salinity. The level of Na+/K+-ATPase activity in the gills was increased during the initial stage of acclimation, followed by a decrease to its normal level, suggesting its involvement in osmoregulation and homeostasis. The only slight increase in the level of IT precursor transcript in the A. melanopus brain upon low-salinity acclimation suggested that IT played a minor role, if any, in the process of osmoregulation.

Chemical Composition of Bangah(Agastache rugosa O. Kuntze) Herb (방아의 화학성분에 관한 연구)

  • Bangah (Agastache rugosa O. Kuntze), one of the wild plant grown in Korea and used for deodorizing some of the fish foods because it's strong and unique flavor, was determined for chemical composition in leaf, flower and stem. The highest protein content of 14.7% was found in leaf and lipids was in the range of $5.25{\sim}7.74%$. Total sugar content in leaf and flower were 27.53% and 14.86%, reducing sugar content were 11.80% and 2.75%, respectively. Fifteen fatty acids in bangah were identified and the major fatty acids were linolenic, palmitic, lauric and linoleic acids. Seventeen total amino acids were determined in leaf and flower. Glutamic acid, aspartic acid and leucine were high in each portion. The major free amino acids were histidine, serine, proline and glutamic acid. Most of minerals were high in leaf and flower than those in stem of bangah except for Ca and Fe. Especially K was higher in leaf and flower than stem by 4 times.

Nutritional Components of Zespri Green Kiwi Fruit (Actinidia delicosa) and Neuronal Cell Protective Effects of the n-hexane fraction (제스프리 그린 키위의 주요 영양성분 및 헥산 분획물의 신경세포 보호효과)

  • The physiological characteristics of kiwi (Actinidia delicosa) fruit were analyzed, which inclued its nutritional composition, in vitro-antioxidant activities, and neuronal cell protective effects. The most abundant components of mineral, amino acid, and fatty acid were found to be potassium (K), glutamic acid, and a-linolenic acid, respectively. The major free sugars were fructose, glucose, and sucrose. In addition, ${\beta}$-carotene and vitamin C contents were $1.35{\mu}g/100mL$ and 29.21 mg/100 g, respectively. The 2,2'-azinobis-(3-ethylbenothiazline-6-sulfonic acid) (ABTS) radical-scavenging activity of the n-hexane fraction obtained from the kiwi extract was 10.52% at a concentration of $1000{\mu}g/mL$. The malondialdehyde (MDA) inhibition of the n-hexane fraction was found to increased in a dose-dependent manner. The intracellular reactive oxygen species (ROS) accumulation after hydrogen peroxide ($H_2O_2$) treatment of PC12 cells was significantly reduced in the presence of the n-hexane fraction compared to PC12 cells treated with $H_2O_2$ only. Moreover, in the a MTT assay, the n-hexane fraction showed in vitro-protective effects against $H_2O_2$-induced neurotoxicity.
