The skin displays a highly active metabolism of polyunsaturated fatty acids (PUFA). Dietary deficiency of linoleic acid (LA), an 18-carbon (n-6) PUFA, results in characteristic scaly skin disorder and excessive epidermal water loss. Although arachidonic acid (AA), a 20-carbon (n6) PUFA, is metabolized via cyclooxygenase pathway into predominantly prostaglandin $E_2(PGE_2)$ and $PGF_{2{\alpha}}$, the metabolism of AA via the 15-lipoxygenase (15-LOX) pathway, which is very active in skin epidermis and catalyzes the transformation of M into predominantly 15S-hydroxyeicosatetraenoic acid (15S-HETE). Additionally, the 15-LOX also metabolizes the 18-carbon LA into 13S-hydroxyoctadecadienoic acid (13S-HODE), respectively. Interestingly, 15-LOX catalyzes the transformation of $dihomo-{\gamma}-linolenic$ acid (DGLA), derived from dietary gamma-linolenic acid, to 15S-hydroxyeicosatrienoic acid (15S-HETrE). These monohydroxy fatty acids are incorporated into the membrane inositol phospholipids which undergo hydrolytic cleavage to yield substituted-diacylglycerols such as 13S-HODE-DAG from 13S-HODE and 15S-HETrE-DAG from 15S-HETrE. These substituted-monohydroxy fatty acids seemingly exert anti-inflammatory/antiproliferative effects via the modulation of selective protein kinase C as well as on the upstream/down-stream nuclear MAP-kinase/AP-1/apoptotic signaling events.
Objectives : Polygoni Multiflori Ramulus has been widely used as a traditional medicinal herb for the treatment of insomnia, limb pain and itch. The extract of Polygoni Multiflori Ramulus (PMRE) is known to have a modulatory effect of many inflammatory responses. This study was performed to investigate the hepatoprotective effect of PMRE against arachidonic acid (AA) + iron-induced oxidative stress on HepG2 cell and carbon tetrachloride ($CCl_4$)-induced liver injury on mice. Methods : The effects of PMRE on cell viability was assessed by MTT assay. And flow cytometric analysis was performed to estimate the effects on mitochondrial function. To investigate its underlying mechanism, apoptosis-related proteins were analysed by using immunoblot analysis. In addition, ICR mouse were administrated (po) with the PMRE (30, 100 mg/kg) for 3 days and then, injected (ip) with $CCl_4$ (0.5 ml/kg body weight) to induce acute liver damage. The level of pro-caspase-3 was measured. Results : Treatment of PMRE increased relative cell viability, prevented a cleavage of poly (ADP ribose) polymerase and pro-caspase-3, and also reduced mitochondrial membrane permeability against AA + iron-induced oxidative stress. In addition, PMRE treatment decreased liver injury induced by $CCl_4$, as evidenced by increases in pro-caspase-3 level. Conclusions : These results demonstrate that PMRE has an ability to anti-oxidant and hepatoprotective effect against AA + iron-induced oxidative stress and $CCl_4$-induced liver injury.
Objectives : Ukgan-san plus Citri Pericarpium and Pinelliae Rhizoma (UCP) is used as a traditional herbal formula in Korea and Japan for treatment of fever, fever-induced convulsions, and liver dysfunction and so on. In this study, we investigated the cytoprotective effect and underlying mechanism of UCP against oxidative stress induced by cotreatment of arachidonic acid (AA) and iron. Methods : To evaluate the hepatoprotective effects of UCP against AA + iron-induced oxidative stress in HepG2 cell, cell viability and changes on apoptosis-related proteins were assessed by MTT and immunoblot analyses. The changes in intracellular reactive oxygen species (ROS), glutathione (GSH), and mitochondrial membrane permeability (MMP) were investigated against to the oxidative stress. Furthermore, to verify underlying molecular mechanism, NF-E2-related factor 2 (Nrf2) and its downstream target genes were examined by immunoblot analysis. Results : Treatment of UCP increased the cell viability and altered the expression levels of apoptosis-related proteins such as PARP, caspase-9, caspase-3, Bcl-2. UCP also inhibited the GSH depletion, excessive ROS production and mitochondrial dysfunction induced by AA + iron. In addition, the Nrf2 and the Nrf2 target genes activation were increased by UCP. Conclusions : These results indicated that UCP has the ability to protect against oxidative stress-induced hepatocyte damage, which may be mediated with Nrf2 pathway.
The incorporation of docosahexaenoic acid(DHA) and arachidonic acid(AA) into brain and liver lipid has been compared in male pups from binth to 10 weeks old by feeding DHA-rich experimental diets or chow diets to dams from pregnancy in rats. The experimental DHA-rich diets contained 7g fish oil and 3g corn oil per 100g diet. There were three experimental groups, FO-I : Dams were fed DHA-rich diet during pregnancy and lactation, and their it pups fed the same diet until 10 weeks old. FO-II Dams fed chow diet during pregnancy and DHA-diet during lactation, and their pups fed the same DHA-diet until 10 weeks. FO-III : Dams fed chow diet during gestation and lactation, and then the pups fed DHA-diet after weaning. The relative % of DHA in hepatic lipid was about 12% with chow diets, but increased rapidly to 20-25% level when DHA-rich diets were supplied after weaning. The AA(%) of FO-III group was relatively high when a chow diet containing higher amount of linoleic acid was given, but there was no significant difference between the groups after feeding on a DHA-rich diet. When the DHA-rich diet was supplied from pregnancy(FO-I), the relative % of DHA in brain lipid was 13.7% at birth and continuously increased to a maximum level(17.2%) at 3-weeks and then was sustained until 5 weeks old. Similar levels of DHA incorporation were observed when DHA-rich diet was supplied from lactation(FO-II). However, the pups of FO-III group showed significantly lower levels of DHA incorporation(72%) at birth. These livels slowly increased and reached an 87% level of FO-I at 10 weeks when the pups ate DHA-rich diets after weaning. The relative % of AA in brain lipid was 10.4% in the FO-I group at birth, which was significantly lower than those of other groups, but there was no significant difference between groups after feeding DHA-rich diets in all groups. The Ah(%) level increased to maximum(11-12%) at 3-weeks and then was slightly reduced and was sustained at about 10% after S-weeks. Total amounts of DNA in the whole brain rapidly reached maximum level at 3-weeks and then was sustained at a constant level after S-weeks. DNA content was not significantly different between groups at birth, but it was significantly higher in FO-I and FO-II groups than in FO-III group at 3-weeks. However, DNA content in FO-III group was continuously increased to 80% level of FO-I at 10-weeks after feeding DHA-rich diet since weaning. In conclusion, the DHA(%) in whole brain was most effectively deposited when DHA-rich diet had been supplied during pregnancy and lactation in rats. However, DHA supplementation after weaning also improved the incorporaton of DHA into brain and content of DNA even though brain development was almost completed, which suggests that DHA supplementation might be necessary to improve brain development in humans during infancy as well as pregnancy and lactation. This study investigated the effect of different dietary fats and fibers on colon tumor incidence and cell proliferation, the levels of eicosanoids and polyamines in colonic mucosa of DMH-treated rats. The experiment was conducted on male Sprague Dawley rats using a 2 $\times$3 factorial design with two fats (corn oil and DHA-rich fish oil) and two fibers (cellulose and pectin) and a fiber-free control. The rats were find an experimental diet containing 15% (w/w) dietary fat and 6% (w/w) fiber for 25 weeks. Tumor incidence was Bower in rats fed fish oil as opposed to corn oil. The levels of arachidonic acid (AA) and eicosanoids ($PGE_2, and TXB_2$) in normal colonic mucosa were significantly lower in rats fed fish oil and there was a concomitant increase of docosahexaenoic acid (DHA). The levels of eicosanoids and AA in tumor tissues were significantly higher than those of normal colonic mucosa. The level of polyamines in normal colonic mucosa was not affected by dietary fats but was significantly lower than that in rumor tissues. Dietary fiber did not have a significant effect on rumor incidence and the levels of AA, eicosanoids and polyamines. Overall, fish oil rich in DHA reduced cell prolifiration and thus inhibited colon carcinogenesis through its effect on the distribution of AA and production of eicosanoids in normal colonic mucosa. However, its effect on colon carcinogenesis revealed a lack of consistency depending on the type of dietary fiber in diet.
In order to investigate of the Influence of $Mg^{2+}$, $Ca^{2+}$ on ${\alpha}$-linolenic acid converted into the eicosapentaenoic acid(EPA) and docosahexaenoic acid(DHA) forming in plasma lipid and in liver microsomes of rabbit, the animals were fed on the perila oil rich ${\alpha}$-linolenic acid or sardine oil rich EPA and DBA diet for 4 weeks were examined. In plasma, liver lipid, $Mg^{2+}$ was influenced on arachidonic acid(AA), EPA, DHA formative from ${\alpha}$-linolenic acid in perilla oil, but stearic acid was increased, $Ca^{2+}$ was Influenced on stearic acid increased and DHA was decreased. In phospholipid, $Mg^{2+}$, $Ca^{2+}$ was influenced on stearic acid increased and DHA was decreased in perilla oil.
In order to observe the effects of the feeds mixed with the lard and two vegetable seed oils on the fatty acid compositions of serum lipkid and lipoprotein, the oils mixed with 2.5% lard and various levels of perilla oil and evening promrose oil were administered to the male rats of the Sprague-DAwley for 4 weeks. In the fatty acid compositions of serum lipid and serum lipoprotein fractions, as the content so f mixed perilla oil decreased and the contents of mixed evening primrose oil increased, n -3 PUFa (polyunsaturated fatty acid) contents and ratio of EPA/AA (eicisapentaenoiidacid/arachidonic acid) tended to decrease, but n-6 PUFA contents and ratio of AA/PUFA tended to increase. Fatty acid compositions of serum lipid and serum lipoprotein fractions were influenced from the fatty acid composition of the test lipids.
본 연구에서는 흰쥐에 마이크로웨이브를 조사한 후 폐조직을 대상으로 폐조직 기능장애를 일으키는데 주된 역할을 하는 혈전생성능을 arachidonic acid(AA) cascade계를 통해 관찰하여 마이크로웨이브에 의한 폐혈관 기능장애와 그에 대한 녹차 catechin의 항혈전 효과를 규명하고자 하였다. 실험군은 마이크로웨이브를 조사하지 않은 정상군과 마이크로웨이브를 조사한 군으로 나누고 마이크로웨이브 조사 군은 다시 식이 중 catechin 공급수준에 따라 catechin을 넣지 않은 군(MW group), catechin을 0.25% 급여한 군(MW-0.25C group), catechin을 0.5% 급여한 군(MW-0.5C group)으로 나누었다. 식이와 음료는 자유 섭식시키면서 2주간 사육한 후 2.45 GHz 대역의 주파수를 15분간 1회 조사하였으며 마이크로웨이브 조사 후 6일째 동물을 희생시켜 본 실험에 사용하였다. 실험군의 실험동물 수는 각각 10마리로 실험하였다. $PLA_2$ 활성은 마이크로웨이브 조사로 30% 증가하였으며 MW-0.25C군은 15% 증가였으나 MW-0.5C군은 정상군 수준이었다. 인지질분자종의 변화를 관찰한 결과 lyso PE가 MW군에서 47% 증가되었으나 MW-0.25C군 및 MW-0.5C군에서는 각각 18%, 20% 증가되었다. $TXA_2$ 생성은 MW군에서 50%의 현저한 증가를 보였으나 catechin 공급군인 MW-0.25C군 및 MW-0.5C군은 정상군 수준이었다. $PGI_2$ 생성은 MW군에서 31%의 유의적인 감소를 보였으나 catechin 공급군인 MW-0.25C군 및 MW-0.5군은 정상군 수준이었다. 따라서 혈전생성지표인 $PGI_2/TXA_2$ ratio는 정상군에 비해 MW군에서 43% 유의적으로 감소되었으나 MW-0.25C군 및 MW-0.5C군은 정상군 수준이었다. 지질과산화물의 함량은 MW군에서 34% 유의적으로 증가하였으며 catechin 공급군은 정상군 수준이었다. 결론적으로 마이크로웨이브에 피폭된 흰쥐 폐조직에서는 AA cascade계의 율속 효소인 $PLA_2$ 활성의 증가와 혈전생성지표로 인식하는 $PGI_2/TXA_2$ ratio의 불균형이 초래되었으나 catechin은 TBARS 농도를 낮추면서 $PLA_2$ 활성을 저해시키고 AA cascade계를 개선시킴으로써 항혈전 작용을 나타내었다.
This study was to investigate the effects of dietary linoleic acid(18:2\omega6, LA) and aipha-linolenic acid(18:3\omega3. \alpha-LNA) levels on brain development and fatty acid compositions of various lipid classes in the chicken embryo brain tissues. Thirty two ISA Brown layers, 52 weeks-old, were divided into four groups. Birds of each group were given corn-soybean meal based diets added with 1) safflower oil 8%, 2) safflower oil 6% + perilla oil 2%, 3) safflower oil 2% + perilla oil 6%, or 4) perilla oil 8%. Mter 15 days fed the diets. the layers were artificially inseminated to obtain fertile eggs. During the incubation. embryonic brains were sampled at 15th and 21st days. Fatty acid contents were quantitated by using heptadecanoic acid (17:0) as an internal standard. No significant differences in brain weight and in contents of various lipids such as phospholipid. triglyceride, cholesterol. cholesterol ester and free fatty acid in the tissues were found among the dietary groups (P<0.05). The ratios of AA/LA in the brain lipid classes were lowered as the dietary levels of perilla oil were increased. Higher LA was found in phosphatidylcholine(PC) than arachidonic acid (20:4\omega6. AA), meanwhile the level of LA was less than AA in phosphatidylethanolamine(PE). Docosahexaenoic acid(22:6\omega3, DHA) was the* major fatty acid in the tissue and its content in PE was 2.5~3 times higher than in PC. DHA level in the phospholipid reached at a peak (1.7~1.8 mg/brain) in dietary groups added with 6% or 8% perilla oil. suggesting that no more increase in that fatty acid level in the brain tissue could be obtained by consuming more \alpha-LNA, the major precursor of DHA.
본 연구에서는 제주재래돼지와 듀록 품종 사이에서 생산된 F2 참조축군에서 melanocortin-4-receptor (MC4R) 유전자형과 지방산 조성간의 관련성을 연구하였다. 전체 290 여두의 F2 자손을 이용하여 14개의 지방산 조성을 측정하였다. Taq I PCR-RFLP 방법을 이용하여 MC4R c.1426A>G (p.Asp298Asn)의 단일염기다형성(single nucleotide polymorphism, SNP)을 확인하였다. MC4R 세 가지 유전자형(AA, AB, BB)이 모두 발견되었고, 그 빈도는 각각 0.299, 0.542, 0.159로 확인되었다. AA 유전자형을 가진 개체에서 palmitic acid (C16:0, p<0.05), stearic acid (C18:0, p<0.01), eicosenoic acid (C20:1n9, p<0.05), saturated fatty acid (SFA, p<0.01) 함량이 GG 유전자형을 가진 개체보다 더 높은 것으로 확인되었다. 반면에 GG 유전자형을 가진 개체는 linoleic acid (C18:2n6, p<0.001), linolenic acid (C18:3n3, p<0.001), linolenic acid (C18:3n6, p<0.001), arachidonic acid (C20:4n6, p<0.001)과 같은 불포화지방산(unsaturated fatty acid) 함량이 AA 유전자형을 가진 개체보다 더 높은 것을 확인하였다. 제주재래돼지와 듀록 품종 사이에서 생산된 F2 참조축군에서 MC4R GG 유전자형이 포화지방산은 낮추고, 불포화지방산은 높이는 것으로 확인되었다. 본 연구 결과 MC4R의 유전적 다형성이 듀록과 제주재래돼지 교배 프로그램에서 육질 향상과, 고기내의 지방산 함량을 조절할 수 있는 유전적 표지 인자로 활용될 수 있을 것이라 사료된다.

1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
