허혈재-관류 적출심장에서 Arachidonic Acid에 의한 산소래디칼 생성 및 심근손상에 대한 Cyclobuxine의 영향 (Effect of Cyclobuxine on Oxygen Free Radical Production and Cellular Damage Promoted by Arachidonate in Perfused Rat Hearts)

  • 흰쥐의 허혈-재관류 적출심장에서 arachidonic acid의 투여에 의해 촉진된 superoxide anion의 생성과 심근손상에 대한 cyclobuxine (스테로이드성 알카로이드)의 영향을 관찰하였다. 적출심장을 Langendorff 관류장치에 현수하고 0.5 ml/min의 저용량으로 45분간 관류한 후 정상관류 (7ml/min)로 복귀시켜 허혈-재관류 심장으로 사용하였다. 재관류 시 arachidonate (0.1과 $1.0\;{\mu}g/ml$)를 투여한 후 superoxide anion의 생성을 관찰하였고, 좌심실 내의 지질 과산화정도는 MDA의 량으로 측정하였으며, 심근손상의 지표로 lactic dehydrogenase (LDH)유리를 측정하였다. 한편 cyclobuxine (1.0과 $10\;{\mu}g/ml$)을 허혈 이전부터 전관류 과정 동안 투여하여 arachidonate에 의해 초래되는 손상에 대한 영향을 관찰하였다. Arachidonic acid는 용량적으로 superoxide anion의 생성을 증가시켰으며 이 작용은 superoxide dismutase (SOD 300 U/ml)와 cyclobuxine에 의해 현저히 억제되었다. Arachidonate를 투여하였을 때 좌심실 내의 malondialdehyde (MDA)의 생성이 현저히 증가되었으며 cyclobuxine은 MDA의 생성을 용량적으로 억제시켰다. 또한 arachidonate는 LDH의 유리를 증가시켰으며 arachidonate에 의한 LDH의 유리 증가는 SOD와 cyclobuxine에 의해 유의하게 억제되었다. 이상의 결과로 흰쥐의 허혈-재관류 심장에서 스테로이드성 알카로이드인 cyclobuxine이 arachidonate에 의한 반응성 산소대사물의 생성과 심근세포손상을 유의하게 억제하는 것으로 관찰되었다.

Gene Targeting of the Acyl-CoA Synthetase Specific to Arachidonate

  • The synthesis of acyl-CoA catalyzed by acyl-CoA synthetase (ACS, EC 6.2.1.3) from fatty acid, ATP, and CoA is a crucial reaction in mammalian fatty acid metabolism. In arachidonate metabolism, acyl-CoA synthetase(ACS) plays a key role in the esterification of free arachidonate into membrane phospholipids. Following its release by the action of calcium dependent phospholipase, free arachidonate is believed to be rapidly converted to arachidonoyl-CoA and reesterified into phospholipids in order to prevent excessive synthesis of eicosanoids. In previous studies, we have characterized five ACSs (designated as ACS1-5) with different tissue distribution. ACS1, ACS2, and ACS5 are similar in structure and fatty acid preference, and completely different from ACS3 and ACS4. The latter are arachidonate-preferring enzymes closely related in structure but expressed in different tissues: ACS3 mRNA is highly expressed in the brain and the mRNA for ACS4 is expressed in steroidogenic tissues including adrenal gland, ovary, and testis. To learn more about the potential function of ACS4 in arachidonate metabolism, we have produced knock-out mice for ACS4 gene. ACS4+/- females become pregnant less frequently and produce small litters with extremely low transmission of the disrupted alleles. Striking morphological changes including extremely enlarged uterine filled with numerous proliferative cysts of various size were detected in ACS4+/- females. Furthermore, marked accumulation of prostaglandins were seen in the uterus of heterozygous females. These results indicate that ACS4 is critical for the uterine arachidonate metabolism and heterozygous disruption of its gene lead to impaired pregnancy.

Paraquat가 Arachidonate Lipoxygenase의 대사에 미치는 영향 (The Effects of Paraquat on Arachidonate Lipoxygenase Metabolism)

  • Using mixed incubation of cultured endothelial cells, cultured fibroblasts, neutrophils activated with PMA and paraquat, the production of superoxide anion, $H_2O$$_2$and lipoxygenase metabolites (5-HETE and 12-HETE) of arachidonate was estimated. The results was as follows: 1. Neutrophils activated with PMA was produced superoxide anion,$H_2O_2$and lipoxygenase metabolites (5-HETE and 12-HETE) of arachidonate. 2. Fibroblasts did not alter the production of superoxide anion and $H_2O_2$ by neutrophils, it was markedly reduced by mixed incubation with endothelial cells. 3. Mixed incubation with endothelial cells significantly augmented the production of 5 or 12-HETEs, but fibroblasts did not. 4. Using mixed incubation of endothelial cells, fibroblasts, neutrophils and paraquat (50ug/m1 and 100ug/m1), the production of superoxide anion, $H_2O_2$and HETEs was significantly increased on both cells at low concentration (50ug/m1), but markedly reduced at high concentration (1001g/m1). 5. Paraquat showed concentration dependent effects on arachidonate lipoxygenase metabolism.

  • Acyl-CoA synthetase 4(ACS4) is an arachidonate-preferring enzyme aboundant in steroidogenic tissues and postulated to modulate eicosanoid production. Most of arachidonate present in cells is esterified predominantly in phospholipids. After its release by the action of calcium-dependent phospholipases, arachidonate can be converted to prostaglandins, thromboxanes, and leukotrenes via the cyclooxygenas and lipoxygenase pathways, respectively, depending on the cell type.(omitted)

  • This study was designed to elucidate the effects of iron, a well known catalyst of lipid peroxidation, on the contents of phospholipids, unsaturated fatty acids composed in phospholipid molecules and their derivatives, prostaglandins, and the composition changes of fatty acids contained in phospholipids. Iron decreased the contents of phospholipids and its components of unsaturated fatty acids. Catalytic action of iron decreased the composition rates of linoleate and linolenate composed in phospholipid molecules, while that of arachidonate was inclined to increase. The content of arachidonate was increased and that of prostaglandins was decreased without regard to increase the precursor of prostaglandins. It may be concluded that the decreases of prostaglandins and the increase of arachidonate are due to inhibition of the activities of enzyme systems responsible for prostaglandin synthesis by lipid peroxides produced by the catalyst of iron.

Inhibition of Arachidonate Release From Rat Peritoneal Macrophage by Biflavonoids

  • Biflavonoid is one of unique classes of naturally-occurring bioflavonoid. Previously, certain biflavonoids were found to possess the inhibitory effects on phospholipase $A_2$ activity and lymphocytes $ proliferation^1$ suggesting their anti-inflammatory/immunoregulatory potential. In this study, effects of several biflavonoids on arachidonic acid release from rat peritoneal macrophages were investigated, because arachidonic acid released from the activated macrophages is one of the indices of inflammatory conditions. When resident peritoneal macrophages labeled with $[^{3}H]$arachidonic acid were activated by phorbol 12-myristate 13-acetate(PMA) or calcium ionophore, A23187, radioactivity released in the medium was increased approximately 4.1-7.3 fold after 120 min incubation compared to the spontaneous release in the control incubation. In this condition, biflavonoids (10 uM) such as ochnaflavone, ginkgetin and isoginkgetin, showed inhibition of arachidonate release from macrophages activated by PMA (32.5-40.0% inhibition) or A23187 (21.7-41.7% inhibition). Amentoflavone showed protection only against PMA-induced arachidonate release, while apigenin, a monomer of these biflavonoids, did not show the significant inhibition up to 10 uM. Staurosporin (1 uM), a protein kinase C inhibitor, showed an inhibitory effect only against PMA-induced arachidonate release (96.8% inhibition). Inhibition of arachidonate release from the activated macrophages may contribute to an anti-inflammatory potential of biflavonoids in vivo.

허혈-재관류 적출심장에서 Arachidonic Acid에 의한 산소라디칼 생성 및 심근손상 (Arachidonate-induced Oxygen Radical Production and Cellular Damage in Ischemic-Reperfused Heart of Rat)

  • 허혈심근의 재관류시 arachidonic acid가 반응성 산소대사물의 발생원으로서 심근 손상에 미치는 영향을 검토하였다. Langendorff 관류장치를 이용하여 흰쥐 적출심장을 0.5ml/min의 저용량으로 관류 (45분)한 후 정상관류 (7 ml/min)로 복귀 시키므로써 실험적인 허혈-재관류 심장을 만들었다. 재관류시 Na arachidonate $(10^{-7}{\sim}10^{-2}{\mu}g/ml)$를 투여한 후 superoxide anion 생성을 관찰하고, 심근 손상의 지표로 lactic dehydrogenase(LDH)유리를 측정 하였으며 이들에 대한 각종 arachidonic acid 대사 억제 약물의 영향을 비교 검토하였다. Superoxide anion 생성은 SOD-억제성 ferricytochrorme C 환원 반응을 이용하였다. 연구성적은 다음과 같다. 1) 저용량 관류후 재관류시 ferricytochrorme C환원은 superoxide dismutase (SOD, 300 U/ml) 및 indomethacin (60 nmole/ml), ibuprofen $(30\;{\mu}g/ml)$에 의하여 억제되었다. 2) Na arachidonate는 용량의존적으로 ferricytochrorme C 환원을 증가 시켰으며 반응성 산소대사물 제거효소인 superoxide dismutase (SOD, 300 U/ml)에 의하여 현저히 억제되었다. 3) Na arachidonate $(10^{-3}\;{\mu}g/ml)$에 의한 superoxide anion 생성은 cyclooxygenase 억제약물인 indomethacin (60 nmol/ml), lipooxygenase 억제약물인 nordihydroguaiaretic acid$(NDGA,\;0.1\;{\mu}mole/ml)$, arachidonic acid의 substrate inhibitor인 eicosatetraynoic acid $(ETYA,\;1\;{\mu}g/ml)$에 의하여 현저히 억제되었다. 4) Na arachidonate는 LDH 유리를 증가시켰으며 SOD에 의하여 유의하게 억제 되었다. 5) Na archidonate에 의한 LDH 유리증가는 indomethacin, NDGA, ETYA에 의하여 유의하게 억제 되었다. 이상의 결과로 흰쥐의 허혈-재관류심근에서 arachidonic acid는 그 대사 과정에서 반응성 산소대사물을 발생하고 이는 심근세포손상에 부분적으로 기여할 수 있을 것으로 여겨졌다.

Effects of the Constituents of Paeonia lactiflora Root on Arachidonate and NO Metabolism

  • In order to establish the anti-inflammatory cellular mechanism of the paeony root(Paeonia lactiflora, Pall, Paeoniaceae), the constituents including paeoniflorin, albiflorin, (+)-catechin, paeonol, benzoic acid and methyl gallate were evaluated for their effects on arachidonate and NO metabolism. Among the compounds tested, only paeonol weakly inhibited cyclooxygenase-2-mediated $PGE_2$ production from LPS-treated RAW 264.7 cells. (+)-Catechin and methyl gallate weakly inhibited inducible nitric oxide synthase-mediated NO production from the same cell line. In particular, methyl gallate significantly inhibited 5-lipoxygenase from RBL-l cells with an $IC_{50}$ of 8.4 ${\mu}M$. These results suggest that the inhibition of these components on arachidonate and NO metabolism may contribute at least in part to anti-inflammatory mechanism of the paeony root.

  • Biosynthsis of prostaglandin E2 (PGE2), the most common prostanoid with potent and diverse bio-activities, is regulated by three sequential enzymatic steps composed of phospholipase A2, cyclooxygenase (COX), and prostaglandin E synthase (PGES).
