Studies on Manufacture of Mineral Water with Wood Charcoals (목질탄화물을 이용한 미네랄수 제조에 관한 연구)

  • To evaluate wood charcoal as raw material for mineral water production, dissolution of inorganic ions from charcoal to water, pH and adsorption ability of chlorine in water were investigated as main variables. More potassium ion was dissolved in water as higher temperature manufactured charcoal but other ions showed no difference with different charcoal making temperatures. Highest dissolved cation was potassium followed by calcium and sodium. Among wood species, charcoal from Quercus variabilis and Platanus occidentalis showed significantly higher potassium content in water than that of larch, red pine and white pine. Other cations had similar pattern to the potassium but their difference was not apparent as much as potassium. pH value of water treated with charcoal was higher for wood charcoals from Platanus occidentalis (pH 8.5) and Quercus variabilis (pH 8.4) which contained higher inorganic cations. In chlorine removal in water by charcoal, all wood charcoals showed greater chlorine removal than that of the control, but softwood charcoals resulted in higher removal than those of hardwoods. There was no significant difference in the dissolution of cations and pHs between particle charcoal and whole charcoal. With easy of control, whole charcoal is better for mineral water making raw material than particle charcoal does.

Preparation of Asymmetric Folyethersulfone Hollow Fiber Membranes for Flue Gas Separation (온실기체 분리용 폴리이서설폰 비대칭 중공사 막의 제조)

  • It is well-known that polyethersulfone (PES) has high $CO_2$ selectivity over $N_2\;(or\;CH_4)$ and excellent pressure resistance of $CO_2$ plasticization among muy commercialized engineering plastics[1-4]. Asymmetric PES hollow fiber membranes for flue gas separation were developed by dry-wet spinning technique. The dope solution consists of PES, NMP and acetone. Water and water/NMP mixtures are used in outer and inner coagulants, respectively. Gas permeation rate (i.e., permeance) and $CO_2/N_2$ selectivity were measured with pure gas, respectively and the micro-structure of hollow fiber membranes was characterized by scanning electron microscopy. The effects of polymer concentration, ratio of NMP to acetone, length of air gap, evaporation condition and silicone coating were investigated on the $CO_2/N_2$ separation properties of the hollow fibers. Optimized PES hollow fiber membranes exhibited high permeance of $25\~50$ GPU and $CO_2/N_2$ selectivity of $30\~40$ at room temperature and have the apparent skin layer thickness of about $0.1\;{\mu}m$. The developed PES hollow fiber membranes, would be a good candidate suitable for the flue gas separation process.

  • Glucose 6-phosphate dehydrogenase of Leuconostoc mesenteroides which was purifid by an affinity chromatography was studied on the characterization, kinetics and chemical modification. The apparent molecular weight of the enzyme was 112,000 by the gel filtration method of Sephadex G-200 column. The optimum temperature of $NAD^+$-linked reation was 50$^{circ}C$ and the activation energy and the heat of inactivation were 8.36 kcal/mole and -58.2kcal/mole, respectively. The steady state kinetic study showed KG6P, Kemp, and CX KNADP to be 76.9 PM, 7.46${\mu}M$ and 7.14 ${\mu}M$, respectively, and KGGP, KNAD,and aKNm to be 53.7${\mu}M$, 115.2${\mu}M$ and 702.2${\mu}M$ for the $NAD^+$-linked reaction at pH 7.8, optimum pH. The pH dependent kinetic constants suggested that the two ionizing groups whose pKa is 7.2 .and pKb is 9.0-9.6 were involved in the enzyme-substrate interaction. Evidence by photooxidation and carboxymethylation of the enzyme suggested that the imidazole group of histidine with pKa group may participate in the catalytic site.

Effects of Moisture Content on Physical Properties of Extruded Cereal Flours (수분함량에 따른 곡류 압출성형물의 물리적 특성 비교)

  • The effects of moisture content on the physical properties of cereal extrudates were investigated. Cereal flours (rice, wheat, corn, barley, and oat) were extruded at a barrel temperature of $130^{\circ}C$, feed rate of 120 g/min, and various moisture contents (20, 22.5, 25, 27.5, and 30%). Proximate content, expansion index, specific length, bulk density, breaking strength, apparent elastic modulus, water absorption index (WAI), water solubility index (WSI), specific mechanical energy (SME) input, paste viscosity, and color values were analyzed. Expansion ratio of extruded corn flour was higher than that of other extrudates at low moisture content. Bulk density, specific length, and elastic modulus in all cereals decreased with an increase in moisture content. The WAI increased with an increase in moisture content, whereas WSI decreased. SME input of extruded corn flour was higher than those of other cereal flours at lower moisture content, whereas that of oat flour extrudate was lower than those of other cereals at higher moisture content. Lightness of extruded rice flour was lighter than those of other cereals while that of extruded barley flour was darker.

Effects of the size of Mg powder on the formation of MgB2 and the superconducting properties

  • The effect of the size and shape of magnesium(Mg) powder on the formation of $MgB_2$ and the critical current density($J_{c,}$) of $MgB_2$ bulk was studied. As a precursor for the formation of $MgB_2$, Mg and $MgB_4$ powder, which was synthesized through the reaction of boron (B) with Mg powders, was used. $MgB_4$ was mixed with Mg powders of various sizes, pressed into pellets and heat-treated at $650^{\circ}C-750^{\circ}C$ in flowing argon gas. The XRD analysis of the heat-treated $MgB_2$ samples showed that the volume fraction of $MgB_2$ was the highest as 92.74 % when spherical Mg powder with an average size of $25.7{\mu}m$ was used, whereas the volume fraction was the lowest as 79.64 % when plate-like Mg powder with a size of $34.1{\mu}m$ was used. The superconducting transition temperature ($T_c$) of $MgB_2$ was not sensitive to the characteristics of the Mg powders used. All of the prepared $MgB_2$ samples showed a high $T_c$ of 38.3 K and a small superconducting transition width of 0.2 K-0.5 K. $J_c$ (5 K and 1 T) of $MgB_2$ was the highest as $3.93{\times}10^4A/cm^2$ when spherical Mg powder with a size of $25.7{\mu}m$ was used, whereas $J_c$ was the lowest as $2.18{\times}10^4A/cm^2$when plate-like Mg powder with a size of $34.1{\mu}m$ was used. The relationship between the $J_c$ of $MgB_2$ and the characteristics of the Mg powders used was explained in terms of the volume fraction of $MgB_2$ and the apparent density of the $MgB_2$ pellets.

Biosynthesis of Isoprenoids: Characterization of a Functionally Active Recombinant 2-C-methyl-D-erythritol 4-phosphate Cytidyltransferase (IspD) from Mycobacterium tuberculosis H37Rv

  • Tuberculosis, caused by Mycobacterium tuberculosis, continues to be one of the leading infectious diseases to humans. It is urgent to discover novel drug targets for the development of antitubercular agents. The 2-C-methyl-Derythritol-4-phosphate (MEP) pathway for isoprenoid biosynthesis has been considered as an attractive target for the discovery of novel antibiotics for its essentiality in bacteria and absence in mammals. MEP cytidyltransferase (IspD), the third-step enzyme of the pathway, catalyzes MEP and CTP to form 4-diphosphocytidyl-2-C-methylerythritol (CDP-ME) and PPi. In the work, ispD gene from M. tuberculosis H37Rv (MtIspD) was cloned and expressed. With N-terminal fusion of a histidine-tagged sequence, MtIspD could be purified to homogeneity by one-step nickel affinity chromatography. MtIspD exists as a homodimer with an apparent molecular mass of 52 kDa. Enzyme property analysis revealed that MtIspD has high specificity for pyrimidine bases and narrow divalent cation requirements, with maximal activity found in the presence of CTP and $Mg^{2+}$. The turnover number of MtIspD is $3.4 s^{-1}$. The Km for MEP and CTP are 43 and $92{\mu}M$, respectively. Furthermore, MtIspD shows thermal instable above $50^{\circ}C$. Circular dichroism spectra revealed that the alteration of tertiary conformation is closely related with sharp loss of enzyme activity at higher temperature. This study is expected to help better understand the features of IspD and provide useful information for the development of novel antibiotics to treat M. tuberculosis.

Rheological Properties of Hot-Water Extractable Concentrates of Boxthorn (Lycii Fructus) and Mixed Boxthorn (구기자 및 혼합구기자 열수 추출 농축액의 리올로지적 특성)

  • To improve utilization of boxthorn (Lycii Fructus), the boxthron and mixed boxthron (Lycii Fructus, Schizandrae Fructus, Corni Fructus, Zizyphi Fructus, Zingiberis Rhizoma, Cinnamami Cortex) hot-water extractable concentrates were prepared by vaccum evaporation and its rheological properties were investigated. The rheological properties of concentrates $(20{\sim}50^{\circ}Bx)$ followed power low model and showed a pseudoplastic behavior at the temperature range of $20{\sim}60^{\circ}C$. The apparent viscosity of $20^{\circ}Bx$, $30^{\circ}Bx$, $40^{\circ}Bx$ and $50^{\circ}Bx$ boxthron hot-water extractable concentrate was 0.0074 Pa s, 0.0175 Pa s, 0.0431 Pa s and 0.0988 Pa s, that of mixed boxthorn hot-water extractable concentrate was 0.0099 Pa s, 0.0328 Pa s, 0.0720 Pa s and 0.1940 Pa s at $20^{\circ}C$ and 1500 l/s, respectively. The yield stress of boxthron and mixed boxthron hot-water extractable concentrates ranged from 0.045 to 6.253 Pa and from 0.022 to 8.891 Pa, respectively. The activation energy for the flow of boxthron and mixed boxthorn hot-water extractable concentrates increased from 1.6182 to $2.0543{\times}10^7\;J/kg{\cdot}mol$ and from 1.7057 to $2.1462{\times}10^7\;J/kg{\cdot}mol$ with the concentrations of concentrates, respectively.

Mineralogy and Geochmistry of the Sanjeon Au-Ag Deposit, Wonju Area, Korea (산전 금-은 광상에 관한 광물 및 지화학적 연구)

  • The Sanjeon Au-Ag deposit consists of three subparallel hydrothermal quartz-calcite veins which filled fault-related fractures (generally $N20^{\circ}$ to 35"W-trending and $70^{\circ}$ to $80^{\circ}$ SW-dipping) within quartz porphyry. The vein mineralization shows an apparent variation of mineral assemblages with paragenetic time: (1) early, white quartz + pyrite + arsenopyrite + brown sphalerite, (2) middle, white (vein) to clear quartz (vug) + base-metal sulfides + electrum + argentite, (3) late, calcite + pyrite + native silver. Mineralogic and fluid inclusion data indicate that gold-silver minerals were deposited at temperatures from 2l $0^{\circ}$ to $250^{\circ}$ with salinities of 4 to 5 wt. % equiv. NaCl and log fS2 values from -14.0 to -12.2 atm. The linear relationship between homogenization temperature and salinity data indicates that gold-silver deposition was a result of meteoric water mixing. Ore mineralization occurred at pressure conditions of about 70 bars, which corresponds to the mineralization depths of about 260 m to 700 m. There is a remarkable decrease of the calculated 1)180 values of water from 1.3 to -9.7%0 in hydrothermal fluid with increasing paragenetic time. This indicates a progressive increase of meteoric water influx in the hydrothermal system at the Sanjeon deposit. Oxygen-hydrogen, sulfur, and carbon isotope values of hydrothermal fluids indicate that the ore mineralization was formed largely from meteoric waters with the contribution of sulfur and carbon from a deep igneous source.

Fibrinolytic Enzyme Activity of Extract from Camellia japonica L. (동백나무 추출물의 혈전용해 효소활성)

  • The fibrinolytic activities of soluble proteins extracted from young leaves of Camellia japonica L. were studied. Fibrinolvity activity of extract from partitions of C. japonica L. showed 1.6-2.0 times higher than plasmin used as positive control. The fibrinolytic enzyme was confirmed directly from young leaves of C. japonica L. by a fibrin Plate and fibrin zymography. The protein was composed of a single polypeptide and its apparent molecular weight was found to be 45 kDa, as judged by SDS-polyacrylamide gel electrophoresis. The optimum pH and temperature for the fibrinolytic activity were pH 5.5 and $30^{\circ}C$, respectively. Also, the fibrinolytic activity was clearly inhibited by PMSF and TLCK, suggesting that it is a member of the trypsin-like serine protease. All these results suggest the protease is a fibrinolytic enzyme belong to a family of trypsin-like serine protease.

Purification and Characterization of Superoxide Dismutase in Sphingomonas sp. KS 301 (Sphingomonas sp. KS 301의 Superoxide Dismutase 정제 및 특성)

  • Sphingomonas sp. KS 301, which was isolated from oil contaminated soil, was shown to have five different SODs (SODI, II, III, IV, V) which can be separated by DEAE-Sepharose chromatography, and SOD III was finally purified in this study by ammonium sulfate precipitation, DEAE-Sepharose chromatography, Superose 12 gel filtration and Uno-Q1 ion exchange chromatography. The molecular weight of SOD III was 23 kDa as determined by SDS-PAGE and the apparent molecular weight of the native enzyme was estimated to be approximately 71 kDa by Superose-12 gel filtration chromatography. These data suggest that the purified SOD consists of at least two subunits. The specific activity of the SOD III was higher than Mn type or Fe type SOD of Escherichia coli by 5 fold. To determine the type of SOD III, inhibitory effects of $NaN_{3},\;H_{2}O_{2},\;KCN$ were examined. 10 mM $NaN_{3}$ was able to inhibit 56% of the SOD III activity, which indicates that this SOD is Mn type. The optimum pH of the SOD III was 7.0 and the optimum temperature was $20^{\circ}C$. N-terminal amino acid sequence of purified SOD III was most similar to those of Psudomonase ovalis and Vibrio cholerae among bacteria.
