Effects of Morin Pretreatment on the Pharmacokinetics of Diltiazem and Its Major Metabolite, Desacetyldiltiazem in Rats

  • The purpose of this study was to investigate the effect of morin, a flavonoid, on the pharmacokinetics of diltiazem and one of its metabolites, desacetyldiltiazem in rats. Pharmacokinetic parameters of diltiazem and desacetyldiltiazem were determined after oral administration of diltiazem (15 mg/kg) in rats pretreated with morin (1.5, 7.5, and 15 mg/kg). Compared with the control group (given diltiazem alone), pretreatment of morin significantly increased the absorption rate constant $(K_a)$ and peak concentration $(C_{max})$ of diltiazem (p<0.05, p<0.01). Area under the plasma concentration-time curve (AUC) of diltiazem in rats pretreated with morin were significantly higher than that in the control group (p<0.05, p<0.01), hence the absolute bioavailability $(AB\%)$ of diltiazem was significantly higher than that of the control group (p<0.05, p<0.01). Relative bioavailability $(RB\%)$ of diltiazem in rats pretreated with morin was increased by 1.36- to 2.03-fold. The terminal half-life $(t_{1/2})$ and time to reach the peak concentration $(T_{max})$ of diltiazem were not altered significantly with morin pretreatment. AUC of desacetyldiltiazem was increased significantly (p<0.05) in rats pretreated with morin at doses of 7.5 and 15 mg/kg, but metabolite-parent ratio (MR) of desacetyldiltiazem was decreased significantly (p<0.05), implying that pretreatment of morin could be effective to inhibit the CYP 3A4-mediated metabolism of diltiazem. There were no apparent changes of $T_{max}$ and $t_{1/2}$ of desacetyldiltiazem with morin pretreatment. Collectively, the pretreatment of morin significantly altered pharmacokinetics of diltiazem, which can be attributed to increased intestinal absorption as well as reduced first-pass metabolism. Based on these results, dose modification should be taken into consideration when diltiazem is used concomitantly with morin or morin-containing dietary supplements in clinical setting.

A Study on the Bioavailability of Calcium in Eggshell Powder Fortified Ramen in the Growing Rats (성장기 쥐에서 난각분 강화라면의 칼슘 이용성에 관한 연구)

  • This study has investigated the bioavailability of calcium in eggshell powder (ESP) fortified ramen. Wean-ling SD rats were maintained for 2 weeks on 0.05$\%$ low calcium diet then assigned randomly to one of 5 groups. Animals were provided for 4 weeks one of 3 different experimental diets containing 0.15$\%$ calcium as $CaCo_{3}$, NFDM, or ESP fortified ramen. Two control groups were fed either 0.05$\%$ calcium or general ramen containing 0.08$\%$ calcium. The body weight, diet intake, food efficiency ratio (FER), bone growth, calcium contents of bones, and apparent absorption were measured. Experimental results show that regardless of calcium sources 0.15$\%$ calcium groups increased body weight, weight and length, calcium content, and the strength of two bones (tibia and femur) significantly compared to 0.05$\%$ calcium group. The apparent absorption rate of calcium also showed similar results supporting the bioavailability of ESP fortified ramen is not inferior to either NFDM or $CaCO_{3}cdot$ The results indicate that ESP fortified ramen is a proper mediate for calcium fortification in Korea.

Effects of Feed Moisture and Barrel Temperature on Physical and Pasting Properties of Cassava Starch Extrudate (수분주입량과 배럴온도에 따른 카사바 전분 압출성형물의 물리적 특성)

  • Considering the importance of cassava as food crops in humid tropics, the effect of feed moisture (20, 25%) and barrel temperature (110, $130^{\circ}C$) on physical properties (piece density, expansion, mechanical properties, color, water solubility index, water absorption index) and pasting properties of extruded cassava starch was investigated. The feed moisture used during extrusion processing had a significant effect on extrudates SME input, specific length and piece density at (p<0.05) while effect on cross-sectional expansion index, apparent elastic modulus and breaking strength in bending shown significantly at p<0.1. Furthermore, the interaction effect of feed moisture and barrel temperature gave a significantly affected the SME input and piece density (p<0.1), specific length (p<0.05) and on redness (p<0.01). The increase in water injection rate led to increase in piece density, apparent elastic modulus, breaking strength in bending, cold peak viscosity, breakdown and final viscosity and decrease in cross-sectional expansion index and specific length. It was found that the extrusion cooking process did not affect the value of color L, color b, water solubility index and water absorption index. Thus, the results of this study can be useful to some extent in developing extruded cassava starch as human and animal feeds.

A Study on Sodium and Potassium Balance of College Women in Seoul (서울지역 일부 여대생의 나트륨과 칼륨평형에 관한 연구)

  • The purpose of this study was to investigate Na and K balances in healthy adult women. Anthropometric assessments, biochemical analysis of blood, 3-day dietary flood records and collections of 3-day food, 24-hr urine and faces were performed to evaluate intakes and excretions of Na and K in 20 college women living in Seoul. The mean BMI and blood pressure of the subjects were 21.08 and 110.25/67.50mmHg, respectively. Mean daily intake of energy was 1578.84kcal, 79% of Korean RDA. Also, daily intakes of Na and K ware 120.86mEq and 44.20mEq. The urinary and fecal excretions of Na were 99.88 and 4.45mEq/day, and those of K were 30.41 and 8.66mEq/day, respectively. The body retention, retention rate, and apparent absorption of Na were 17.11mEq, 13.23%, and 96.31%, and those of K were 5.82mEq, 8.69%, and 80.12%, respectively. The urinary and fecal Na/K ratio were 3.48 and 0.52. There were significantly positive correlations between 1) urinary Na, K excretions and intakes of Na or K, 2) urinary K and BMI, 3) serum K and serum globulin, and 4) urinary Na excretion and serum haptoglobin level, respectively. The results of this study show that Na intake was higher and K intake was lower than those of other advanced nations. Therefore, nutrition education show instruct people to reduce Na intake and to increase K intake.

The Effects of Dietary Ca Levels on Ca and Skeletal Metabolism in Ovariectomized Rats of Different Age (난소를 절제한 나이가 다른 흰쥐에서 식이 칼슘 수준이 골격 대사에 미치는 영향)

  • To investigate the effect of dietary Ca levels on metabolic changes of Ca and skeleton in postmenopausal women, 10-month-old ovariectomized female rats were compared with 2 month old rats. The rats were fed either 0.2% or 1.2% Ca diets for 16 weeks. Food intake and weight gain as higher in rats fed high Ca diets and in ovariectomized rats. Apparent Ca absorption as higher, and Ca balance was lower in the low Ca groups. Vertebrae density was higher in old rats or those fed a high Ca diets. The old rats and ovariectomized rats showed decreased bone formation, increased bone resorption and kidney function deterioration resulting in increased urinary Ca excretion. Contradictory to the above observation, old rats and ovariectomized rats still showed higher bone mass and bone ash content. Therefore aging was not fully onging in 10-month-old rats. Bone weights, mineral contents, and mineral/wt ratio were lower in ovariectomized rats. Dietary Ca level did not affect urinary Ca excretion, urinary protein excretion, GFR, serum alkaline phosphatase, or urinary hydroxyporline excretion. This means that dietary Ca level did not influence kidney function or bone turnover. However Ca content and the ash content of femur, 4th vertebra, and scapula were increased in high Ca groups. Therefore, it is considered that decreased bone formation and accelerated bone resorption may account for the increased osteoporotic risk in women in menopause after middle age. However, Ca metabolism can be improved and bone components can be maintained if Ca is supplemented.

Physical Properties of Foamed Concrete up In the Manufacturing Waste Expanded Poly-Styrene (폐스티로폼의 가공 형태에 따른 기포콘크리트의 물리적 특성 변화)

  • This study is focusing on mixing the foamed concrete incorporated by waste expanded polystyrene(W-EPS), investigating the physical properties and offering a proper quality control method to the field engineers. Two types of W-EPS (type A and type B) were studied. Type A (B) had globular (crushed) shape and diameter of 3-5 (1-2) mm. The results show that the flow was suddenly reduced with increasing mixing quantity of two types, but it satisfies KS F 4039 until 60 % of mixing rate. In general, the absorption rate was suddenly reduced with increased mixing quantity of two types especially, in type A. Apparent specific gravity was 0.36∼0.53 and reduced with increasing mixing quantify of type A. But it increased in case of type B. Compressive strength and heat conduction rate increased with mixing with W-EPS than non-mixing W-EPS but reduced with mixing too much W-EPS. Based ong the results, it is believed that mixing with W-EPS can improve the recycle of industrial wastes and produce the high quality foamed concrete.

Study on Crashworthiness of Icebreaker Steel: Part I Steel Properties (쇄빙선 강재의 내충격 특성에 관한 실험적 연구: 제1부 강재 특성)

  • This paper presents a study on the crashworthiness of the scaled-down stiffened panels used on a Korean icebreaker. In order to validate the crashworthiness of the panels, this paper provides various mechanical properties such as the results of a CVN test, quasi-static tensile test, and high-speed tensile test at arctic temperatures. Two types of steels (EH32 and FH32) were chosen for the material tests. CVN tests revealed that the two steels were equivalent up to −60℃ in terms of their impact energy absorption capacity. However, the toughness of FH32 was significantly superior to that of EH32. EH32 showed slightly higher flow stresses at all temperature levels compared to FH32. The improvement ratios of the yield strengths, tensile strengths, plastic hardening exponents, etc. for FH32, which were obtained from quasi-static tensile tests, showed an apparent ascending tendency with a decrease in temperature. Dynamic tensile test results were obtained for the two temperatures levels of 20℃ and −60℃ with two plastic strain rate levels of 1 s−1 and 100 s−1. A closed form empirical formula proposed by Choung et al. (2011;2013) was shown to be effective at predicting the flow stress increase due to a strain rate increase.

Physiological Responses of Cirsium setidens and Pleurospermum camtschaticum under Different Shading Treatments (피음처리에 따른 고려엉겅퀴와 누룩치의 생리반응)

  • This study was conducted to investigate the chlorophyll contents, photosynthetic characteristics and chlorophyll fluorescence of Cirsium setidens and Pleurospermum camtschaticum by shading treatment. Two species were grown under non-treated (full sunlight) and three different shading condition (88~93%, 65~75% and 45%~55% of full sunlight) for the experiment. Total chlorophyll content, photochemical efficiency (Fv/Fm), specific leaf area (SLA), and net apparent quantum yield were increased with elevating shading level but decreased dark respiration under the low light intensity. Therefore, light absorption and light utilization efficiency were improved under the low light intensity. 45~55% of full sunlight in C. setidens and 65~75% of full sunlight in P. camtschaticum showed best maximum photosynthetic rate, net apparent quantum yield and photochemical efficiency. On the other hand, non-treated showed lower maximum photosynthetic rate, photochemical efficiency, and chlorophyll content than treated ones. These results suggest that growth of P. camtschaticum adapted to 65~75% of full sunlight and C. setidens adapted to 45~55% of full sunlight.

Effect of Microbial Phytase on Performance, Nutrient Absorption and Excretion in Weaned Pigs and Apparent Ileal Nutrient Digestibility in Growing Pigs

  • Two experiments were conducted to evaluate the efficacy of Trichoderma reesei derived phytase for pigs fed diets with fixed calcium to total phosphorus ratios (1.5:1). In Exp. 1, 280 weaned pigs (initial BW of $10.32{\pm}1.94$ kg) were allocated to one of five dietary treatments on the basis of weight and gender in a randomized complete block design. Treatments were the low phosphorus (0.6% Ca, 0.4% total P and 0.23% available P) diets supplemented with 0, 250, 1,000, or 2,000 FTU phytase/kg of diet and a positive control diet (PC; 0.85% Ca, 0.58% total P and 0.37% available P). The treatments were applied to seven pens with eight pigs per pen, half male and half female. In Exp. 2, six barrows fitted with ileal T-cannula (initial BW = $35.1{\pm}1.6$ kg) were assigned to three dietary treatments with a double $3{\times}3$ Latin square design. The dietary treatments were the low-phosphorus diet (0.53% Ca, 0.34% total P and 0.14% available P), the low phosphorus diet plus 1,000 FTU phytase/kg and a positive control diet (0.77% Ca, 0.50% total P and 0.30% available P). In Exp. 1, there were linear increases (p<0.01) in weight gain, phosphorus absorption, bone strength, calcium and phosphorus content of fat-free dried bone and plasma phosphorus concentrations with increasing dose rate of phytase. The performance of pigs fed the diets with 250, 1,000, or 2,000 FTU of phytase/kg did not differ from pigs fed the PC diet. Pigs fed diets with 1,000 or 2,000 FTU of phytase/kg did not differ from pigs fed the PC diet in bone characteristics. The apparent digestibility of dry matter, crude protein, ash and energy was not affected by dietary treatment. However, pigs fed the PC diet excreted more fecal phosphorus (g/d, p<0.01) and fecal phosphorus per BW gain (g/kg) than pigs fed the diets with phytase. Phytase linearly decreased (p<0.01) fecal phosphorus excreted per BW gain (g/kg), plasma calcium concentration as well as plasma and bone alkaline phosphatase activity. In Exp. 2, phytase supplementation in the low-P diet increased (p<0.05) the apparent ileal digestibility (AID) of Ca, P, leucine, lysine, phenylalanine, alanine and cysteine, tended to AID of crude protein, isoleucine, threonine, asparagine and serine. In conclusion, the novel phytase originated from Trichoderma reesei is effective in releasing Ca, P, and amino acids from corn soy based diet for pigs.

Evaluation of Urinary Nitrogen Excretion from Plasma Urea Nitrogen in Dry and Lactating Cows

  • Data of 42 balance measurements from dry and lactating Holstein cows and blood and urine samples from 24 Japanese Black cows were collected to evaluate the potential for predicting urinary nitrogen (N) excretion from plasma urea nitrogen (PUN). Similar positive correlations were obtained between N intake and apparent N absorption in dry and lactating cows. The regression equations of N intake on urinary N excretion varied in dry and lactating cows, and the difference of urinary N excretion between dry and lactating cows was due to the N secretion into milk. Highly positive correlations were observed between urinary N contents and urinary urea N in Japanese Black cows, and urinary urea N increased with increasing PUN. There were positive correlations between N intake and PUN in dry and lactating cows, but PUN and urinary N excretion in lactating cows were higher than in dry cows. There were positive correlations between PUN and urinary N excretion per BW in dry and lactating cows. Although urinary N excretion could be calculated as (N clearance rate of kidneys)PUNBW, high N clearance rate of kidneys, such as 2.08 L/d/kg BW, may be suitable to calculate urinary N excretion in lactating cows, compared with 1.33 L/d/kg BW in dry cows.
