Habit characteristics of imperfect and perfect stage of D. bryoniae encountered on naturally infected seeds of cucumber and pumpkin were studied by the blotter method and compared with those grown on Difco potato dextrose agar (PDA), V-8 juice agar and water agar leaf medium (WALM). Most of the pycnidiospores obtained from each isolate of this fungus grown on PDA were non-septate and microtype. Non-septate pycnidiospores were predominanted in all isolates, but a macrotype of the non-septate and a number of uniseptate pycnidiospores were produced on V-8 juice agar and water agar leaf medium. On seed the pycnidiospores were mostly non -septate, but rarely uniseptate ones were also found. On radicle of cucumber seed, the pycnidiospores were non-septate and uniseptate but small percentage biseptate with somewhat constricted at septa. Pycnidiospores produced on V-8 juice agar and water agar leaf medium were similar to those produced on seeds. In the present investigation the perithecia were mostly globose to subglobose with apical papillate ostiole and whitish spore masses formed on the ostiole of perithecia, either on naturally infected seed or on culture media. The mature perithecia were dark brown to black. They were partially embedded or erumpent on seed coat and culture media. The perithecia varied in size within a much narrower range than the pycnidia. But perithecial formation of this fungus on PDA, V-8 juice agar, WALM and seed varied considerably depending upon isolate and culture media.
The purpose of present study was to compare the speed of coronal leakage before and after post space preparation using Streptococcus mutans. Forty straight extracted human teeth were selected. The crowns were removed to a uniform remaining root length 14 mm. Canals were enlarged by 06 taper $Profiles^{(R)}$ to a size $\#40$ as a master apical file. And these were filled with gutta percha point and $Tubuliseal^{(R)}$ sealer, using continuous wave technique. Groupings are as follows. Group 1 - These teeth were obturated without sealer. Group 2 - These teeth were obturated and covered the surface of the root completely with sticky wax. Group 3 - These teeth were obturated. Group 4 - These teeth were obturated and prepared for post space remaining 5 mm of gutta percha. The teeth were suspended in plastic tubes. The upper chamber received the bacterial suspension everyday to simulate clinical situation. The lower chamber consisted of BHI added Andrade's indicator. All roots in the positive control group (Group 1) turned yellow within 24 h and those of negative control group (Group 2) remained red throughout the experimental period (70 days) The samples of group 3 were contaminated within an average of 27.2 days. The samples of group 4 were contaminated within an average of 15.7 days, ranging from 9 to 22 days. There was significant difference between group 3 and group 4 statistically (p < 0.05).
The purpose of this study was to compare the centering abilities of four root canal instrument systems and the amounts of dentin removed after root canal shaping using them. The mesial canals of twenty extracted mandibular first molars having $10-20^{\circ}$ curvature were scanned using X-ray micro-computed tomography (XMCT)-scanner before root canals were instrumented. They were divided into four groups (n = 10 per group). In Group 1, root canals were instrumented by the step-back technique with stainless steel K-Flexofile after coronal flaring. The remainders were instrumented by the crown-down technique with Profile (Group 2), ProTaper (Group 3) or K3 system (Group 4). All canals were prepared up to size 25 at the end-point of preparation and scanned again. Scanned images were processed to reconstruct three-dimensional images using three-dimensional image software and the changes of total canal volume were measured. Pre-and post-operative cross-sectional images of 1, 3, 5, and 7 mm from the apical foramen were com pared. For each level, centering ratio were calculated using Adobe Photoshop 6.0 and image software program. ProTaper and K3 systems have a tendency to remove more dentin than the other file systems. In all groups, the lowest value of centering ratio at 3 mm level was observed. And except at 3 mm level, ProTaper system made canals less centered than the other systems (p < 0.05).
The berry thinning was a useful practice to reduce bunch size and to alter bunch shape for improving fruit quality. The grape cv. 'Muscat Bailey A' bunches were removed to find berry thinning effect on the bunch, in the apical end of the main stem about 4~6 laterals or conventional treatment and then compared a quality of fruit and wine between the treatments. Bunch weights on the different berry thinning treatment were in the range of 300 to 650g and conventional treatment was 550 to 750g. As a result, according to decreased lateral number of bunch in the treatments, total soluble sugar was increased but total acid was decreased. Our results was shown that wine color and taste components such as total anthocyanin, polyphenol and tannin were depended mainly by berry thinning. Also Berry thinning treatments of bunches was ranked higher sensory score than conventional ones.
Cordyceps pruinosa grows upon dead pupae of Lepidoptera and produces one or $3{\sim}4$ club-shaped stromata per host. The stromata have distinct club-shaped head and long stalk. The length of stromata varies from $1{\sim}3\;cm$. Apical head consists of densely crowded semi-immersed perithecia, which are $360{\sim}400\;{\times}\;180{\sim}200\;{\mu}m$ in size. Asci are $150\;{\mu}m$ in length and $2.8{\sim}3\;{\mu}m$ in diameter. Ascospores, which are $124{\sim}141\;{\mu}m$ in length, have thin thread-like structures in the middle with part-spores attached on both sides. Each ascospore does not separate into part-spores after dispersal, but each part-spore germinates and together develops a colony. The imperfect form produces phialides of $15{\sim}24\;{\times}\;2{\sim}3\;{\mu}m$ size, with spherical or spindle shaped conidia of $4{\sim}6\;{\times}\;1.8{\sim}2.4\;{\mu}m$ size, The anamorph was identified as Mariannaea elegans Samson. YMA and SDAY agar media with pH 7 was produced abundant mycelial growth with high density. Best mycelial growth was observed when dextrin was used as a carbon source. Lactose, saccharose and sucrose also produced high mycelial growth. Peptone, yeast extract and tryptone produced abundant mycelial growth, when used as nitrogen sources. Highest mycelial growth and density was observed when C/N ratio was 1 : 1 at the concentration of 12.5 g/l each. $KH_2PO_4$ was the best mineral source for mycelial growth. Highest mycelial dry wt. was produced in YM and SDAY broths. Optimum inoculum for 100 ml of liquid broth was 6 mycelial discs. Similarly, optimum liquid culture period was 7 days.
Flow cytometry (FCM) was used to measure the ploidy level of three different sports from 'Campbell Early' ($Vitis$$labruscana$) grape. Results of the study showed different ploidy levels. FCM analysis for 'Campbell Early' grape which contains 2C DNA diploid cells showed single peak around 35-40 while 'Kyoho' grape with 4C DNA tetraploid cells had a different level of 70-80. However, analysis of the sports displayed a histogram with 2 peaks containing both 2C and 4C nuclei. There was no difference in histograms of 2C DNA flesh and pericarp; on the other hand, 4C DNA flesh type of sports had a different histogram from that of the 2C DNA pericarp. Chromosome numbers of diploid ('Campbell Early'), tetraploid ('Kyoho'), and three sports were counted under the microscope. 'Campbell Early' and 'Kyoho' have 38 and 76 chromosomes, respectively. Three different sports are mixoploids with mixtures of diploid and tetraploid cells. Microscopic observations of shoot apical meristems in sports from 'Campbell Early' grape were carried out to determine the type of plant chimera. 'Campbell Early' grape (diploid) and 'Kyoho' grape (tetraploid) showed that both had 2 tunica layers covering corpus cells, while the three different sports had tunica layers showing mostly oblique division. Most cells from 'Kyoho' grape were larger than 'Campbell Early' grape. Cells from L-2 and L-3 layers of the three sports were similar to 'Kyoho' grape in size, although all cells in L-1 surface layer were uniform in size like 'Campbell Early' grape. Results of FCM analysis indicated that both normal and polyploid cells could be intermixed in sports and could become mixoploidy consisting of diploid and tetraploid. All sports used in the tests were periclinal chimera plants with two distinct L-1 and L-2 cell layers. The result of this study suggests that all three sports which originated from 'Campbell Early' grape might be 2-4-4 type chimera formation.
In order to get information on the ecology of rice false smut, germination ability and pathogenicity of sclerotia and chlamydospores of the pathogen, environmental conditions affecting the disease outbreak and varietal resistance have been investigated. 1. The degree of outbreak of rice false smut was higher in the upland rice in comparison with the paddy field rice in respect to the number of affected grains per ear, the size and weight of smut balls formed on affected grains as well as the ratio of sclerotial formation produced on smut balls. 2. Germination percentage and days required for germination of overwintered sclerotia placed on the soil surface in July were 81% and 19 days, respectively, while those of overwintered sclerotia treated in May were 60-70% and 41 days. Sclerotia placed on the soil surface or under 1 cm depth of the soil surface and incubated at $25-30^{\circ}C$ were germinated well, whereas those placed under 3 cm or 5 cm depth of the soil surface were not germinated at all. Germinability and stroma productivity of sclerotia were reduced when the sclerotia were cutted into small pieces. 3. The average number of stroma formed on a sclerotium was six and that of perithecia formed in a stroma was about 50 to 140. 4. Percentage of germination of chlamydospores on the yellow balls was very high and was decreased as the color of the balls being darken with maturation. 5. Panicle of rice plants were successfully infected by injection inoculation with suspention of ascospores and chlamydospores of the pathogen to the sheaths at the booting stages, while seeding infection by spraying with suspensions of chlamydospores was unsuccessful. 6. More number of infected grains was distributed on basal parts of an affected ear than that of infected ones distributed upper parts of the ear, when the affected ear was divided into five parts from its basal portion to the apical of the ear. 7. The occurrence of the disease was more severe in the late maturing varieties of rice in comparison with the early maturing varieties. 8. When the level of nitrogen applied was increased, the incidence of disease increased, and the infection percentage of the disease was increased as the transplanting date was delayed. 9. The weight of panicles and 1000 kernels and the ratio of ripenness were reduced, and the contamination degree of grains with chlamydospores were increased as the number of smut balls per panicle were increased.
Spontaneous pneumothorax is the sudden collapse of a lung usually caused by air leakage from a subvisceral pleural blob. Responses to closed thoracostomy, thoracentesls and simple observation are usually prompt and effective. But in some cases, these are unsucceful and open thoracotomy is indicated. A clinical evaluation was performed on 242 cases(236 patients) of open thoracotomy in spontaneous pneumoth rax who were admitted and treated at department of Thoracic and Cardiovascular Surgery. Masan Samsung General Hospital during the past 9 years from January 1988 to December 1996. The results were as follows 1. The sex ratio was male predominance(M:F=11.7:1) 2. The most common age group were 2nd, 3rd decades(2nd=29.3%, 3rd=30.2%). 3. The most common chief complaints were chest pain and dyspnea(chest pain=41.7%. dryspnea= 36.8). 4. The etiologic factors of spontaneous pneumothorax were primary spontaneous pneumothorax(86.4%), tuberculosis(9.1%), COPD(3.7%) and pleuritis(0.8%). 5. The site of spontaneous pneumothorax was 52.1% in right, 45.4% in left and 2.5% in both. 6. The common indications of open thoracotomy were recurrence(44.2%), persistent air leakage(31.8%) and inadequate expansion(15.7%). 7. The operative procedures were bullectomy or mechanical pleurodesis through posterolateral thoracotomy or median sternotomy. 8. The most frequent location of bulla or blob were apical segme t oi RUL(35.1%) and apicoposterior segment of LUL(41.3 %). 9. The number of bulla or blob were mainly 1 to 5(88%), and there were no significant differences among operation indications. 10. The size of bulla or bleb were mainly below 5cm(81%)and small bulla($\leq$1cm) were predominant in recurrence group but large bulla(>5cm)were predominant in persistent air leakage and inadequate expansion group. 11. The pleural adhesion was seen in 54.5%.(Recurrence group 64.1%,Persistent air leakage group 51.9%,Inadequate expansion group 47.4%).
