Complete Genome Sequences and Evolutionary Analysis of Cucurbit aphid-borne yellows virus Isolates from Melon in Korea

  • Complete genome sequences of 22 isolates of Cucurbit aphid-borne yellows virus (CABYV), collected from melon plants showing yellowing symptom in Korea during the years 2013-2014, were determined and compared with previously reported CABYV genome sequences. The complete genomes were found to be 5,680-5,684 nucleotides in length and to encode six open reading frames (ORFs) that are separated into two regions by a non-coding internal region (IR) of 199 nucleotides. Their genomic organization is typical of the genus Polerovirus. Based on phylogenetic analyses of complete nucleotide (nt) sequences, CABYV isolates were divided into four groups: Asian, Mediterranean, Taiwanese, and R groups. The Korean CABYV isolates clustered with the Asian group with > 94% nt sequence identity. In contrast, the Korean CABYV isolates shared 87-89% sequence identities with the Mediterranean group, 88% with the Taiwanese group, 81-84% with the CABYV-R group, and 72% with another polerovirus, M.. Recombination analyses identified 24 recombination events (12 different recombination types) in the analyzed CABYV population. In the Korean CABYV isolates, four recombination types were detected from eight isolates. Two recombination types were detected in the IR and P3-P5 regions, respectively, which have been reported as hotspots for recombination of CABYV. This result suggests that recombination is an important evolutionary force in the genetic diversification of CABYV populations.

  • Cucumber mosaic virus (CMV) belongs to genus Cucumovirus. The Cucumovirus group contains three distinct members: CMV, Tomato aspermy virus (TAV), and Peanut stunt virus (PSV). The type member, CMV is the most widespread and most studied. CMV is isometric particles about 30 nm in diameter. The genome of CMV is divided into three RNAs. In addition, RNA extracted from virus particles contains a fourth RNA that is a subgenomic RNA generated from RNA3. RNA1 and RNA2 are each encapsidated in separate particles, whereas RNAs3 and 4 are coencapsidated in a third particle. Hence, inoculation by three particles, transmitted either mechanically or by the aphid vector, is required to infect plants.(중략)

  • Hibiscus syriacus were surveyed to study the growing temperature, relative humidity and growing responses shown at different elevations of Kangwon Province, and results of the assessment are summarized as follows 1. Growing responses as represented by leaf length, leaf width, shoot length, number of flowers and flowering duration, were decreased as the elevation increased, while the degree of defoliation was increased. 2. The important factor determining the growing responses to the elevation was the maximum and minimum temperatures, particularly those of July, August, and September. 3. The aphid occurrence on Hibiscus syriacus decreased as the elevation increased. Leading factor determining the population was relative humidity which increased as the elevation increased.

  • An aphid species, Glyphina betulae( Linnaeus, 1758), which has been known only in western Paleoarctic, is recognized from Mt. Baekdu-san, North Korea. Besides the descriptions and illustration for apterous viviparous female and alate viviparous female, brief biology, host plahts and distribution are discussed. This is the first record of the genus Glyphina in Korean Peninsula.

  • One of the major aims in studying plant viruses is to minimise the development of symptoms in infected plants. With the advent of in vitro transcript mediated research on plant viruses, substantial progress has been made. This article describes the biology of a plant specific RNA virus, Johnsongrass mosaic virus (JGMV), important to Australian sorghum and corn agriculture and, in particular, at a molecular level which of the RNA sequences in its genome that make it possible for the virus to move from cell to cell, and eventually spread systemically throughout the entire plant. The JGMV has caused considerable yield losses in maize and sorghum over a number of years in Australia. Incidents where 100% of the crop has been infected are on record. The use of this virus is convenient under laboratory conditions because it can be readily transmitted by mechanical inoculation with infected leaf sap, which obviates the need for maintaining aphid colonies. The JGMV is a single stranded positive sense RNA virus.

  • Sensitivities of insecticides and their seasonal variation were investigated against Myzus persicae. Some aphicides such as malathion were not effective on Myzus persicae in Chinese cabbage field. Difference of seasonal sensitivity was from I time to 16 times.

Insecticidal Activity of Extracts Isolated from Syzygium Aromaticum

  • This study separated the crude extract (70% ethanol) of and its three fractions (hexane, chloroform and ethyl acetate extracts) on the basis of polarity indexes, and examined for their insecticidal activities against aphid (Uroleucon lactucicola). For crude extraction, the 70% ethanol extract showed the best extract yield (58.0%) and insecticidal activity (69.0%) among the various concentrations tested (water, 30% ethanol, 50% ethanol, 70% ethanol and 95% ethanol). The major chemical compounds of different fractions (hexane, chloroform and ethyl acetate extracts) were identified as eugenol by head space-GC-MS analysis. The hexane extract showed the highest eugenol content (43.7%) and insecticidal activity (80.0%). The insecticidal activity is accordingly believed to be attributable to the eugenol component. This may provide a useful starting point for the development of bio-pesticides.

  • Turnip mosaic virus (TuMV) was isolated from Rorippa islandica showing mild mosaic symptom in growing field of Chinese cabbage and radish. Identification of the virus was based on host range, transmission by aphids, electron micrograph, serological reaction and hybridization detection. The virus systemically infected on Chenopodium quinoa, Nicotiana clevelandii, N. glutinosa, Brassica rapa, B. campestris subsp. pekinensis and Raphanus sativus, whereas showed local infection on C. amaranticolor, Gomphrena globosa and Tetragonia tetragonoides. The virus was transmitted by aphid (Myzus persicae). The virus particle was filamentous with 720$\times$12 nm in length, and reacted positively with an antiserum of TuMV in agar gel double duffusion test. In slot-blot hybridization using the digoxigenin(DIG)-labeled RNA probe, TuMV-RNA could be detected in sap of R. islandica infected with the virus. This is the first report of a natural infection of that virus on R. islandica.

  • This study on Pemphigidae was based on the specimens collected from May 1989 to April 1993, at 16 localities of southern part of Korean Penninsula. As a result, 11 species of 7 genera were identified. Of these, four species (Colophina zelkovae, Prociphilus chaenomelis, Tetraneura pumilae, T. iriensis) are new to science. These four species were described and drawn.

  • Symbionin, ahomologue of E. coli GroEL, produced by an intracellular symbiont of the pea aphid , has molecular chaperone activity bothin vitro and in vivo, and it is able to tarnsfer its high-energy phospholy group to other compounds through its autophosphorylation and phosphotransferase activity. The symbionin is a novel histidine protein Kinase and a senor molecular of the two-component pathway.
