In the present study, the Lycium chinensis Miller was harvested at intervals of one month in order to distinguish outstanding species and to determine optimal harvest time. From these harvests, extracts were prepared from ethanol. The total usable sugar, betaine, and phenolic acid contents as well as electron donating ability and SOD liked activity of the extracts were then measured. While sugar content of the Lycium chinensis Miller showed no significant difference among the various species examined, usable sugar content of the crop harvested in November was higher than that of the crop harvested in August. The Lycium chinensis Miller was picked in August, September, October, and November and analyzed for betaine content. According to this analysis, betaine content was higher in the crop harvested in November than in that harvested in August. In particular, considerable difference in betaine content per species or harvest time was exhibited. The SOD-liked activity in all of the Lycium chinensis Miller extracts showed an alleviation effect of at least 90%. In addition, there was no significant difference according to either species or harvest time. On the other hand, SOD liked activity was higher in November than in August.

Objectives : This study is aimed to compare the changes in Antioxidative capacity of Liriopis Tuber by steaming process and to compare the effects in hyperlipidemia induced rats fed high cholesterol diet between Simvastatin and Liriopis Tuber by steaming process. Methods : The SD rats were divided into six groups: normal diet (Nor), high cholesterol diet (Veh), high cholesterol diet plus Simvastatin 5 mg/kg (Sim), high cholesterol diet plus LT0 extract 200 mg/kg (LT0), high cholesterol diet plus LT6 extract 200 mg/kg (LT6) and high cholesterol diet plus LT9 extract 200 mg/kg (LT9). We compared the total cholesterol (TC), triglyceride (TG), high density lipoprotein cholesterol (HDL), low density lipoprotein cholesterol (LDL) contents and reactive oxygen species (ROS) from each serums. Protein expression in liver tissues related to antioxidant and cholesterol was analyzed. Results : The Antioxidant activity of Liriopis Tuber increased by steaming process. In vivo, TC, TG, LDL-c, atherogenic index (AI) and cardiac risk factor (CRF) decreased and HDL-c increased with increasing steaming frequency. aspartate aminotransferase (AST), alanine aminotransferase (ALT), creatinine, and blood urea nitrogen (BUN) decreased with increasing steaming frequency. ROS decreased only in LT9, and SOD, catalase and glutathione peroxidase (GPx) increased with increasing steaming frequency. phospho-AMP-activated protein kinase (p-AMPK) increased and sterol regulatory element-binding protein 2 (SREBP-2), Phospho-Acetyl-CoA Carboxylase (p-ACC) and HMG-CoA reductase (HMGCR) decreased with increasing steaming frequency. Liver staining showed a decrease in hepatic fat accumulation of LT9. LT9 showed significant results in all experiments. Conclusions : LT9 showed significance of anti-lipid effect and improved fatty liver of hyperlipemia induced rats fed on high cholesterol diet, In conclusion, LP9 can be effectively used for the treatment of hyperlipidemia.
This study aimed to investigate the growth parameters and antioxidant activity of Cnidium officinale under controlled temperature and carbon dioxide levels during the cultivation period. The plants were cultivated for four months, each group being set at the average temperature of the cultivation area +1.8℃/445ppm(SSP1), +3.6℃/872ppm(SSP3), and +4.4℃/1,142ppm(SSP5), respectively. During the cultivation period, the growth, Top/Root ratio, and leaf weight ratio(LWR) of C. officinale slightly decreased in SSP3 and SSP5 compared to SSP1, while the root weight ratio(RWR) increased. The antioxidant activity and related phenolic compound content in the aerial parts of C. officinale increased proportionally with temperature and CO2 concentration. However, an adverse effect was observed in the high-concentration SSP5 group. Conversely, in the roots, the SSP5 group exhibited the highest antioxidant activity. This study suggests that it can be utilized as fundamental data necessary for understanding the correlation between environmental conditions and the growth as well as physiological activities of medicinal plants.
Objective: This study aimed to investigate the effects of dietary supplementation with Agaricus blazei polysaccharide (ABP) at varying concentrations on the performance, egg quality, blood biochemistry, intestinal morphology, and microflora of quail. Methods: The study involved a total of 2,700 Korean quails, which were randomly divided into three groups. The measured variables encompassed productive performance, egg parameters, carcass parameters, serum metabolites, immune response parameters, antioxidative properties, and gut microbiome. Results: The addition of ABP did not have a significant effect on average daily feed intake. However, it was found to increase the average daily egg weight and egg production rate, reduce the feed-egg ratio. There were no significant impacts on egg quality measures such as egg shape index, egg yolk index and color, egg yolk and protein content. However, ABP supplementation significantly increased the Hough unit (p<0.01) and decreased the rate of unqualified eggs (p<0.01). Regarding serum parameters, the inclusion led to an increase in total protein concentration (p<0.05) and a reduction in low-density lipoprotein cholesterol (p<0.05). There were no significant effects observed on immune indicators such as immunoglobulin A (IgA) and IgM. ABP supplementation increased the levels of serum antioxidant indicators, including glutathione peroxidase, total superoxide dismutase (p<0.05), and total antioxidant capacity colorimeter (p<0.05). Furthermore, ABP supplementation significantly elevated the intramuscular fatty acid content in quail meat. Additionally, ABP supplementation demonstrated a significant improvement in the diversity of gut microbiota and induced alterations in the composition of the gut microbiota. Conclusion: The findings of this study indicate that dietary supplementation of ABP enhanced production performance and antioxidant capacity while increasing the levels of polyunsaturated fatty acids in quail muscle.
The purposes of this study were to investigate effects of Bambusae Caulis in Liquamen(BCL) on antioxidant activities and inhibitory activities of HMG-CoA reductase of in vitro, and lipid metabolism in rats fed the high cholesterol diet in vivo. Sprague-Dawley male rats weighing l00$\pm$10 g were devided into five groups ; normal group(NOR), the high cholesterol diet administered group(1 $\%$ cholesterol and 0.25$\%$ sodium cholate)(CON), 5$\%$ BCL administered group (5BL), the high cholesterol diet and 5$\%$ BCL administered group (5BCB) and the high cholesterol diet and 10\$\%$ BCL administered group (10BCB), respectively. In antioxidative activities of BCL using Rancimat in vitro, 1.25 diulent and original solution were more excellent activities than the control group, and in inhibiting activities of HMG-CoA reductase, BCL was shown inhibitory effects compared with the control, in dose dependent manners, especially 57.9$\%$ in original solution and 36.0$\%$ in 1.25 diulent. The growth rate of the control group was higher than the normal group, wheras the group given 5$\%$ BCL and 10$\%$ BCL were gradually decreased, especially the most excellent effect in 10$\%$ BCL. Serum levels of total cholesterol, LDL-cholesterol, triglyceride and free cholesterol were significantly decreased, whereas levels of HDL-cholesterol and phospholipid were increased, but not significantly. BCL administered group was increased in HDL-cholesterol/total cholesterol ratio and lowered antherogenic index. The activities of AST in serum were rather lowered in the BCL administration group than the cholesterol diet group, but not in ALT and ALP. The hepatic contents of total cholesterol were lowered significantly than control group, but not in triglyceride. Therefore, it might be expected that BCL is believed to be a possible protective or curative effects for fatty livers and hyperlipidemia-induced by a hi~h cholesterol diet.

Radical scavenging effect and protective activity against oxidative stress of Acer okamotoanum were investigated. A. okamotoanum was extracted with methanol (MeOH) and then fractionated with n-BuOH, ethyl acetate (EtOAc), methylene chloride and n-hexane fractions. The MeOH extract and fractions showed strong 1,1-diphenyl-2-picrylhydrazyl and superoxide radical scavenging activity. Among the MeOH extract and fractions, the EtOAc fraction showed the strongest radical scavenging activity. In addition, total phenolic and flavonoid contents of EtOAc fraction was higher than other extract and fractions. Furthermore, we investigated the neuroprotective effect of the MeOH extract and fractions from A. okamotoanum against oxidative stress under cellular system using C6 glial cell. The C6 glial cells showed a decrease in cell viability and high production of reactive oxygen species (ROS) by the treatment of amyloid $beta_{25-35}$ ($A{\beta}_{25-35}$). However, with the treatment of the MeOH extract and fractions, it significantly increased the cell viability and inhibited the overproduction of ROS by $A{\beta}_{25-35}$. In particular, the EtOAc fraction led to significantly increase the cell viability and decrease the generation of ROS against oxidative stress by $A{\beta}_{25-35}$. The current study indicated that A. okamotoanum demonstrated antioxidative and neuroprotective effects. In particular, the EtOAc fraction which attributed a strong protective activity against oxidative stress.

In this study, the antioxidative effects, inhibitory effects on tyrosinase and elastase and components of Castanea crenata leaf were investigated. The free radical (1,1-diphenyl-2-picrylhydrazyl radical, DPPH) scavenging activity ($FSC_{50}$) of extract / fractions of Castanea crenata left was in the order: 50% ethanol extract ($13.6{\mu}g/mL$) < ethyl acetate fraction (6.2) < aglycone fraction (2.1). Reactive oxygen species (ROS) scavenging activities ($OSC_{50}$ of extract / fractions from Castanea crenata leaf extract / fractions on ROS generated in $Fe^{3+}$-EDTA/$H_2O_2$ system were investigated using the luminol-dependent chemiluminescence assay. The order of ROS scavenging activity was in the order: aglycone fraction (0.8) < 50% ethanol extract (0.5) < ethyl acetate fraction (0.3). The scavenging activity ($IC_{50}$ for ${O_2}^{{\cdot}\;-}$ (superoxide anion radical) generated by NBT method was in the order: ethyl acetate fraction (145.5) < aglycone fraction (65.5). The protective effects on the rose-bengal sensitized photohemolysis of human erythrocytes were investigated. The ethyl acetate fraction exhibited the most prominent cellular protective effect (${\tau}_{50}$, $191.9{\pm}12.2\;min$ at $10{\mu}g/mL$). The inhibitory effect of aglycone fraction ($9.1{\mu}g/mL$) on elastase was higher than oleanolic and ($13.7{\mu}g/mL$). And the inhibitory effect of aglycone fraction ($21.6{\mu}g/mL$) on tyrosinase was higher than arbutin ($226.2{\mu}g/mL$). But 50% ethanol extract rarely exhibited the inhibitory activity on tryosinase and elastase. Flavonoids were contained in Castanea crenata left (96.3 mg / 100 g dried Castanea crenata leaf). And flavonoids contained in ethyl acetate fraction were kaempferol, quercetin, quercitrin, and so on. Quercitrin is the most abundant component. These results indicate that extract / fractions of Castanea crenata can function as antioxidants in biological systems, particularly skin exposed to UV radiation by scavenging free radical and ROS, Castanea crenata leaf extract/ fractions could be used as new cosmeceutical for whitening and anti-wrinkle products.

The purpose of this study was to evaluate antioxidant and antigenotoxic effects of Omija (Schizandra chinensis B.) extracted with various solvents (acetone, ethanol, and methanol). The total polyphenol content (TPC) of methanol extract (ME), ethanol extract (EE) and acetone extract (AE) from Omija were 1183.3, 1009.4, and 747.3 mg/100 g (garlic acid equivalents: GAE), respectively. Antioxidant effects of the Omija extracts was measured by DPPH radical-scavenging activity (RSA) and superoxide dismutase (SOD)-like activity. The $IC_{50}$ for DPPH RSA was in the order of EE $(1411.1\;{\mu}g/mL)$=AE $(1462.0\;{\mu}g/mL)$>ME $(1585.0\;{\mu}g/mL)$. The $IC_{50}$ for SOD-like activities was the highest in ME $(905.7\;{\mu}g/mL)$=EE $(970.3\;{\mu}g/mL)$>AE $(1579.4\;{\mu}g/mL)$. The antigenotoxic effect of Omija on DNA damage induced by $H_2O_2$ in human leukocytes was evaluated by comet assay. $H_2O_2$ induced DNA damage was effectively protected by all of the Omija extracts. Aectone extract of Omija showed the highest antigenotoxic effect ($IC_{50}$ value of AE is $14.6\;{\mu}g/mL$) followed by EE, and ME (21.4 and $34.4\;{\mu}g/mL$), respectively. As a result, we propose that Omija (Schizandra chinensis B.) can serve as a new natural source enriched with potent antioxidant and antigenotoxic agents.
Background: N-acetylcysteine(ACE) is used both orally and intravenously in a variety of experimental pathologies resembling human disease states which exhibit endothelial toxicity as a result of oxidative stress, including acute pulmonary oxygen toxicity, septicemia and endotoxin shock. Despite these observations in vivo, it is not certain how this thiol drug produces its protective effects. ACE is a cysteine derivative which is able to direct1y react with oxygen radicals and may also act as a cysteine and glutathione(GSH) precursor following deacetylation. In this paper, we tried to know whether the therapeutic doses of ACE can modify the inflammatory function of the neutrophils and can increase the glutathione level of plasma in chronic obstructive pulmonary disease(COPD) patients. In addition, the effect of ACE to the purified neutrophil in terms of superoxide release and glutathione synthesis were observed. Method: Firstly, we gave 600mg of ACE for seven days and compare the release of superoxide, luminol-enhanced chemiluminescence from the neutrophils, neutrophil chemotaxis, and plasma GSH levels before and after ACE treatment in COPD patients. Secondly, we observed the dose dependent effect of ACE to the purified neutrophil's superoxide release and GSH levels in vitro. Results: 1) Usual oral therapeutic doses(600mg per day) of ACE for seven days did affect neither on the neutrophil's superoxide release, chemiluminescence, chemotaxis, nor on the plasma GSH concentration in the COPD patients. 2) ACE decreases the purified neutrophil's superoxide release and increase the GSH production in dose dependent fashion in vitro. Conclusion: Despite the fact that oral ACE treatment did not affect on the neutrophil's inflammatory function and plasma GSH concentration in COPD patients in usual therapeutic doses, it decreases the superoxide release and increases the GSH production from the isolated neutrophils in high molar concentrations. These findings suggest that to obtain an antioxidative effects of ACE, it might be needed to increase the daily dosage of ACE or therapeutic duration or change the route of adminisration in COPD patients.

In vitro activities of Codonopsis lanceolata (CL) 70% ethanol extract and its fractions (hexane, chloroform, ethyl acetate, butanol and water) were examined by total polyphenol content, reducing power, 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonate) (ABTS), 2,2-diphenyl-$\beta$-picrylhydrazyl (DPPH), and oxygen radical absorbance capacity (ORAC) assays. The ethyl acetate fraction from CL ethanol extract (CLEA) showed the highest total polyphenol content (22.7 mg/g) among five fractions, and also exhibited an excellent reducing power (0.42~1.27 at $250\sim1,000\;{\mu}g/mL$). CLEA at $100\sim400\;{\mu}g/mL$ concentrations had 27.7~70.3% of ABTS radical scavenging activity and the highest DPPH radical scavenging activity (81.6% at $400\;{\mu}g/mL$). CLEA had dominantly higher $ORAC_{{ROO}{\cdot}}$activity compared to other fractions. CLEA and butanol fraction had significantly higher $ORAC_{{OH}{\cdot}}$ activities than 70% ethanol extract, hexane, chloroform and water fractions. The CLEA exhibited the highest antioxidant activity in CL 70% ethanol extract and its fractions. Thus, effect of CLEA treatment on antioxidant gene expression under the oxidative stress conditions by a high fat diet in animal model was studied by microarray and RT-PCR methods. The 31 antioxidant genes were expressed but the genes were not up-regulated at least a two-fold by CLEA treatment. We concluded that CLEA does not have an indirect antioxidant effect but a direct antioxidant effect by up-regulation of antioxidant genes in high fat diet-induced obese mice.
