D-라이보스 섭취가 해수면환경과 고지대환경에서의 운동지속능력 및 항산화능력에 미치는 영향 (Effects of D-ribose Supplementation on Run-to-exhaustion Time and Antioxidative Capacity under Sea Level or High Altitude Condition)

  • This study evaluated the effects of D-ribose supplementation on exercise time in a hypoxic chamber. Eight healthy young males participated in experiments under all four different conditions; placebo+normoxic, placebo+hypoxic, ribose+normoxic, and ribose+hypoxic. Subjects took 1 g per 10 kg body weight of ribose dissolved in drinking water 30 minutes before and immediately before running. We observed the run-to-exhaustion time, the maximum heart rate, and the changing pattern of the heart rate during exercise. The longest running time was achieved when subjects ran under normoxic condition with ribose supplementation. The shortest running time occurred when subjects ran under hypoxic conditions without ribose supplementation. We measured MDA and GPx to determine any changes in oxidative stresses or antioxidative systems. MDA was affected by the environmental conditions and the running time. The activity of GPx showed a significant difference only with the different environmental conditions of exercise. The results of this study indicate that ribose can be considered a possible ergogenic during exercise in specific conditions, but more detailed and well-controlled studies are needed to make a definitive conclusion.

Antioxidative Properties of Brown Algae Polyphenolics and Their Perspectives as Chemopreventive Agents Against Vascular Risk Factors

  • Several polyphenolic compounds and complex mixtures were isolated from brown algae species. The 1,1-diphenyl-2-picryhydarzyl (DPPH) radical scavenging activity and ferric reducing antioxidant power (FRAP) of these compounds were evaluated to determine their physiological usefulness as antioxidants for vascular protection. The antioxidative protection of low-density lipoprotein (LDL) was also evaluated and compared with that of catechin, because the generation of oxidized LDL is one of the most active and specific risk factors contributing to atherogenesis. Oral administration to rats of a commercially available sample ($VNP^{TM}$) containing 30% of these polyphenolic compounds and 70% dietary fiber revealed that the serum reducing capacity measured in terms of FRAP value was significantly elevated 30 min after the treatment, but declined rather quickly thereafter, indicating the good oral absorption of the compounds and their fast binding to the lumenal surface of the blood vessels. An eight-week, human, clinical trial (n=31) of $VNP^{TM}$ showed significant improvement in erectile function measured by IIEF (international index of erectile function) score. These results collectively demonstrated the usefulness of these polyphenolic compounds as fundamental chemopreventive agents against vascular risk factors originating from oxidative stress.

Anti-angiogenic, Anti-cell Adhesion Switch from Halophilic Enterobacteria

  • The halophilic enterobacteria, Enterobacteria cancerogenus, was isolated from the intestines of the fusiform fish (Trachurus japonicus) to yield a protein-like material termed PLM-f74. PLM-f74 was characterized by strong inhibition ratios to angiogenesis (82.8% at the concentration of $18.5{\mu}g/mL$) and elevated antioxidative capacities with low toxicity. The PLM-f74 is a glycoprotein comprised of saccharides and amino acids. PLM-f74 inhibited non-activated U937 monocytic cell adhesion to HUVECs activated with IL-$1{\beta}$ by 78.0%, and the adherence of U937 cells treated with the PLM-f74 and stimulated with IL-$1{\beta}$ to unstimulated HUVECs decreased by 102%. When both cell types were pretreated with PLM-f74, the adhesion of U937 cells to IL-$1{\beta}$ stimulated HUVECs was completely suppressed by 121% at a concentration of 18.5 ug/mL. PLM-f74 blocked signal pathways from VEGFR2, PI3K, ${\beta}$-catenin and VE-cadherin to NF-kB based on western bolt analysis. And also inhibited IL-1-stimulated HUVEC expression of the adhesion molecules, ICAM-1 by 40%, VCAM-1 by 60%, and E-selectin by 70% at the same concentration noted above. New anti-angiogenic and anti-cell adhesion materials showing elevated antioxidative capacities and non-toxicity may be expected from these results.

참나물 에탄올 추출물의 항산화 효과 (Antioxidative Effect of Pimpinella brachycarpa Ethanol Extract.)

  • 참나물의 생리활성 효과를 구명하기 위하여 in vitro에서 용매별 분획물의 항산화효능을 검토하였다. 참나물 에탄올추출물의 추출 수율은 12.01%이었으며, 이를 n-hexane, chloroform, ethylacetate, n-butanol, water로 계통 분획한 수율은 water 분획이 1.90%로 가장 높았으며, n-hexane, n-butanol, chloroform, ethylacetate 순이었다. In vitro에서 참나물 에탄올 추출물을 계통 분획하여 DPPH radical에 대한 자유기 소거능을 측정한 결과 n-hexane 분획이 $41\;{\mu}g/ml$로 가장 강한 항산화 활성을 나타내었다. 또한 Rancimat로 측정한 항산화지수도 n-hexane 분획이 1.98로 가장 높게 나타났으며, 이는 기존의 항산화제인 BHT가 1.97로 유사한 활성을 나타내었다. 아질산염 소거능에서도 모든 pH에서 n-hexane 분획이 가장 높게 나타났으며, pH 1.2에서 가장 우수하였다. 지질과산화물 생성 억제효과는 n-hexane 분획이 88.40%로 가장 높았으나, 양성대조군으로 사용한 BHT의 억제율보다도 낮았다. Linoleic acid에 대한 항산화효과를 지질과산화물가로 측정한 결과도 분획 중 n-hexane의 항산화력이 가장 우수하였고, ethylacetate, chloroform 순이었다. 이상의 결과 참나물은 식품으로의 이용가치뿐만 아니라 참나물 추출물의 in vitro 에서 항산화력의 우수함을 통해 천연 항산화제로서의 효과를 기대해 본다.

거대배아미 에탄올 추출물의 항산화활성 및 항변이원성 (Antioxidative and Antimutagenic Activity of Ethanolic Extracts from Giant Embroynic Rices)

  • 신선찰거대배아미, 화청거대배아미 및 남풍거대배아미 등 거대배 돌연변이 계통 쌀 3종류 및 일반미의 70% 에탄올 추출물을 제조하여, 이들의 항산화 활성 및 항변이원성을 비교 검정하였다. 거대배아미 추출물의 항산화 활성은 DPPH radical 및 Fenton 반응에 의해서 유도되는 hydroxy radical의 소거활성, hypoxanthine/xanthine oxidase system에서 생성되는 활성산소종인 superoxide radical의 소거활성, linoleic acid자동산화에 대한 지질 과산화 억제활성 및 토끼 적혈구 막지질의 과산화 억제활성 둥으로써 검정하였으며, 항변이원성은 E. coli PQ 37 균주를 사용하여 화학적 변이원 mitomycin C에 대한 변이원성 억제효과를 SOS chromotest에 의해서 검정하였다. 일반미 품종에 비해서 거대배아미 품종의 항산화 활성 및 항변이원성이 모두 높았으며, 거대배아미 품종 중에서는 남풍거대배아미가 가장 효과적인 경향이 있었다. 남풍거대배아미의 DPPH radical, superoxide radical과 hydroxyl radical 소거활성, 그리고 지질과산화 억제활성은 일반미보다 각각 2.3배, 3.3배, 1.7배 및 2.5배 정도까지 더 높았다.

감잎, 녹차의 건분 및 물, 에탄올추출물이 노령쥐의 지방대사와 항산화능에 미치는 영향 (Effects of Dried Leaf Powders, Water and Ethanol Extracts of Persimmon and Green Tea Leaves on Lipid Metabolism and Antioxidative Capacity in 12-Month-Old Rats)

  • The effects of dried leaf powders and water and ethanol extracts of persimmon and green tea on lipid metabolism, lipid peroxidation and antioxidative enzyme activity were investigated in 12-month-old rats. Forty-nine male Sprague-Dawley rats weighing 520$\pm$19g were blocked into seven groups according to body weight. Rats were raised for four weeks with control(no tea leaf powder or extracts) and experimental diets containing either 5%(w/w) dried leaf powders of persimmon(Diospyros kaki Thunb) or green tea(Camellia sinensis O. Ktze), or water or ethanol extract from equal amounts of each dried tea powder. Food intakes of all tea diet groups were higher than that of control. Weight gains and food efficiency ratios of all tea diet groups were not significantly different from those of control. All tea diets decreased plasma triglyceride level, especially, green tea powder and persimmon ethanol diets were more effective than other diet. All the tea diet groups showed decrease in liver triglyceride level, and persimmon powder and ethanol extract increased fecal triglyceride excretion. Plasma cholesterol levels of all the tea diet groups were not significantly different from the control, but control. Fecal cholesterliver cholesterol concegroups were significantlntrations of all tea y lower than that of ol excretions of persimmon powder, green tea ethanol extract, persommon ethanol extract and green tea ethanol extract groups were significantly higher than that of control. Plasma and liver thiobarbituric acid reactive substance(TBARS) concentrations of all the tea diet groups were lower than that of control. Especially, plasma TBARS concentrations of green tea powder and persimmon ethanol extract groups were sinificantly low. Red blood cell(RBC) superoxide dismutase(SOD) activities of persimmon ethanol extract and green tea water extract groups were increased, and RBC catalase activities of all experimental groups were not significantly different. RBC glutathione peroxidase(GSH-px) activities of persimmon ethanol extract, persimmon water extract and green tea powder groups were increased. Liver SOD activities of all the tea diet groups except green tea ethanol extract group were higher than that of control. Liver catalase activities of all experimental groups were not significantly different, and liver GSH-px activity of green tea powder group was significantly higher than that of control. In conclusion, dried leaf powders, and water and ethanol extracts of persimmon and green tea were effective in lowering lipid level, inhibiting lipid peroxidation and increasing antioxdative enzyme activities in 12-month-old rat. Green tea leaf powder with high contents of flavonoids and water soluble dietar fiber was most effective in lowering plasma triglyceride, cholesterol and TBARS level. Clostridium perfringens KCTC 5014의 성장 억제를 위한 항산화 천연산물의 효과 (The Growth Inhibiting Effect of Enterobacteria, Clostridium pefringens KCTC 5014, by the Combination of Natural Products)

  • C.perfringens의 성장은 선택되어진 천연산물의 한 종류 및 이들의 조합에 의하여 특징적으로 성장이 저해되었으며, 이것은 항산화력이 강한 천연산물들을 free radical의 제거와 동시 에 C.pefringens의 성장 억제 물질로 사용할 수 있고 이를 이용하여 기능성 항산화 제제의 제조가 가능하리라 사료된다. 한 종류의 천연산물, 백작약, 백출, 음양곽, 석창포, 오미자, 기 및 황련 등에 의하여 성장이 저해되었고, 황기는 40%의 제어율을 보여, 이 세균의 대사를 제어하는 성분이 있거나, 세포막의 수송 환경의 변화에 의한 물질의 전달이 제어되는 것으로 추정된다. 그리고 두 종류 천연산물의 조합인 황금과 감초, 황금과 상백피, 황백과 감초 및 황금과 황기등의 조합에 의해서 저해되었고, 황백과 감초의 조합에서 64%의 제어 효과를 보였다. 세 종류의 천연산물 조합인 황금과 황백 및 구기자, 황금과 황백 및 음양곽의 조합에 의하여 70%의 제어율을 보였고, 황백, 음양곽, 황기 및 감초의 조합등의 4종류의 천연산물 조합에 의해서도 성장이 저해되었으며, 75%의 제어율을 보였다. 4종류의 조합의 항산화력은 황백, 음양곽, 황기 및 감초 조합의 AUC가 581.89로써 대조군보다 4.4배 정도 강하였으며, 항산화력 이 강한 조합에서 제어가 잘되는 것으로 나타났다. Hydroxyl radical의 소거 활성은 황백, 음양곽, 황기 및 감초의 조합이 300배의 희석액에서 93.49%로서 최상의 소거율을 보였고, 방향족의 함량 및 총 phenol할량은 가공된 녹차보다 10배 정도 높은 것으로 나타났다. 이것은 항산화력이 강한 천연산물들을 이용하여 free radical의 제거와 동시에 C. pefringens의 성장 저해 물질로 사용할 수 있으며, 생체의 기능을 향상시킬 수 있는 기능성 제제로의 응용 가능성을 보였다.

무, 양파의 시료제조 방법에 따른 흰쥐의 지방대사와 항산화능에 관한 연구 (Effect of Dry Powders, Ethanol Extracts and Juices of Radish and Onion on Lipid Metabolism and Antioxidative Capacity in Rats)

  • This study was performed to investigate the effects of dry powders, ethanol extracts and juices of radish and onion on lipid metabolism, lipid peroxidation and antioxidative enzyme activity in rats. Forty-nine male Sprague-Dawley rats weighing 157$\pm$6g were blocked into seven groups according to body weight and raised for four weeks with diets containing 5%(w/w) dry powders of two different vegetables consumed frequently by Korean-radish(Raphanus sativus L.) and onion(Allium cepa L.), ethanol extracts and juices from equal amount of each dry powder. All the powders, ethanol extracts and juices of radish and onion decreased total lipids, triglycerides and total cholesterol concentrations in plasma and liver. Above all, onion ethanol extract decreased them most remarkably. It was thought that organosulfur compounds and flavonoids extracted from onion by ethanol inhibited biosynthesis and absorption of lipid and promoted degradation of lipid. Radish powder also decreased them by increasing fecal excretions of total lipids, triglycerides and total cholesterol most effectively. Catalase and glutathine peroxidase(GSH-px) activities in red blood cell(RBC) were most remarkably increased by radish powder and onion powder respectively. Superoxide dismutase(SOD), catalase and GSH-px activities in liver were most remarkably increased by onion ethanol extract, radish powder and onion ethanol extract respectively. Xanthine oxidase(XOD) activities in liver were most effectively decreased by ethanol extracts of radish and onion. Thiobarbituric acid reactive substance (TBARS) levels in plasma and liver of experimental groups were significantly lower than those of controls. Above all, onion powder decreased them most effectively. It was thought that vitamin E and high flavonoids in onion powder inhibited lipid peroxidation, promoting liver and RBC SOD, catalase and GSH-px activities and inhibiting XOD activities effectively. Flavonoids in onion ethanol extract inhibited lipid peroxidation, promoting three antioxident enzyme activities and inhibiting XOD activities most remarkably. Also flavonoids and high vitamin C in radish powder inhibited lipid peroxidation, promoting liver and RBC catalase most remarkably and inhibiting XOD activities. In conclusion, radish and onion were effective in lowering lipid levels and inhibiting of lipid peroxidation in animal tissue. From these data, radish and onion can be recommended in the treatment and prevention of diseases such as cardiovascular disease and cancer and in delaying aging. As ethanol from onion were most effective in lowering lipid level and promoting three antioxident enzymes, and inhibited lipid peroxidation as did we should try to utilize onion skin which is discarded though reported to have abundant flavonoids. Peroxyl Radical Scavenging Capacity of the Flavonolignan Silybin, Ginkgo Biloba Extract EGb 761, American Green Tea and a Series of Germacranolides

  • We report on the applicability oj a method recently developed in our laboratory for measuring the antioxidant potential of isolated chemicals and extracts derived from natural products. Peroxyl radicals generated by thermal homolysis of 2,2'-azobis-amidinopropane (ABAP) oxidize $\alpha$-keto-${\gamma}$-methiolbutyric acid (KMBA) to ethylene, which is monitored by gas chromatography. Inhibition of ethylene formation in the presence of antioxidants that compete with KMBA for peroxyl radicals is the basis of the Total Oxyradical Scavenging Capacity Assay (TOSCA; Winston et al., 1998). Antioxidative activities of water-soluble extracts of American green tea, the anti-hepatotoxic flavonolignan from milk thistle (Silybum marianum) silybin, Ginkgo biloba extract EGb 761, and a series of naturally occuring sesquiterpene lactones (all ger-macranolides found in in fungi, liverworts, and plants) were studied. The specific TOSC value per $\mu$M silybin was 5.2, which is essentially comparable to that of Trolo $x^{ⓡ}$, a water-soluble vitamine E analog. Tea and Ginkgo extracts exhibited potent peroxyl radical scavenging capacity with values, respectively of =1700 and 1000 $\mu$mols Trolo $x^{ⓡ}$ equivalent per gram dry matter. The known anti-inflammatory activity of some germacranolides prompted study of their antioxidant capacity. None of the lactones exhibited antioxidant capacity toward peroxyl radicals comparable to Trolo $x^{ⓡ}$; costunilide, the most lipophilic, had a TOSC value = to glutathione. The potential role of peroxyl radicals in lipidperoxidation, other cellular damage, and var-ious disease states suggest a possible preventive role for silybin, green tea and Ginkgo biloba in oxidative stress caused by these free radical species.ecies.
