근육식품에서 지방산화와 관련된 항산화 효소 (Antioxidant Enzymes in Relation to Oxidative Deterioration of Muscle Foods)

  • Antioxidant enzymes such as catalase (CAT), glutathione peroxidase (GSH-Px) and superoxide dismutase (SOD) are known to inhibit oxidative reactions by incativating compounds responsible for the formation of ree radicals. SOD transforms superoxide radical into hydrogen peroxide which is precursor to active free radicals. CAT reduces hydrogen peroxide to water. GSH-Px reduces hydroperoxides to corresponding alcohols. Antioxidant enzyme activities of muscle are different by animal species age, stress and exercise, muscle type and part, conditions of post mortem, storage and processing which are related to oxidative deterioration I muscle foods as well as oxidative defence in living systems. Antioxidant enzyme systems are enhanced rather than weakened in aging skeletal muscle. Red muscle contains higher antioxidant enzyme activity than white muscle. The antioxidant enzyme activities of poultry are higher in leg than in breast, and those of beef are higher in redder and more unstable muscles. It is clear that the effectiveness of the antioxidant enzyme in muscle foods seems to be influenced by meat processing operations. Both GSH-Px and CAT are inactivated by heat processing NaCl also influence the efficiency of the antioxident enzymes since its presence diminishes their catalyitc activity.

규칙적인 운동습관이 남자 성인의 적혈구내 항산화효소활성과 혈장 항산화능력(TRAP)에 미치는 영향 (Effects of Regular Physical exercise Habits on the Activities of Erythrocyte Antioxidant Enzyme and Plasma Total Radical-trapping Antioxidant Potential in Health Male Subjects)

  • In the present work we investigated the effect of regular physical exercise on the activities of erythrocyte antioxidant enzyme, plasma total radical-trapping antioxidant potential(TRAP) and plasma level of lipid peroxidation(malondialdehyde, MDA) in 64 healthy male, aged 34-67 years. The study population were divided in two groups: small amount of exerciser(exercise time less than 10min/d) and moderate amount of exerciser(exercise time more than 20min/d) according to their physical exercise habits measured by a questionnaire. Erythrocyte superoxide dismutase(SOD), glutathione peroxidase(GSH-Px) and catalase(CAT), plasma TRAP, as well as plasma MDA were determined. Erythrocyte GSH-Px and plasma TRAP were higher in moderate amount of exercisers than those in small amount of exercisers by 17% and 26%, respectively. No significant differences were observed in erythrocyte SOD, CAT and plasma MDA between the two groups. Mean exercise time was positively correlated with the erythrocyte GSH-Px activity and plasma TRAP significantly. The results would sugest that regular moderate exercise enhances antioxidant defences against reactive oxygen species and may increase the likelihood of a healthier life span.

Effects of Multivitamin-Mineral Supplementation, at Nutritional Doses, on Plasma Antioxidant Status, Erythrocyte Antioxidant Enzyme Activities and Lipid Peroxidation during Pregnancy

  • The aim of this study was to evalute the effect of multivitamin-mineral supplementation during pregnancy on plasma levels of antioxidants, erythrocyte antioxidant enzyme activities, and lipid peroxidation. A controlled, semi-randomized, prospective trial was performed by comparing the supplement group, which received multivitamin-mineral tables once daily for 10 weeks, with the control group. Plasma levels of $\beta$-carotene, tocopherol, coenzyme Q10, ascorbate, folate, zinc, and selenium and malondialdehyde (MDA), as well as the activities of superocxide dismutase(SOD) and glutathione peroxidase(GSH-Px) in erythrocytes were measured initially (20 wk gestation) and at the end of the intervention (34 wk gestation). In the control group, plasma ascorbate and selenium levels decreased and tocopherol levels increased. In the supplement group, a significant increase in plasma $\beta$-carotene(46%), conenzyme Q10 (42%), and zinc (24%) was observed after 10 weeks of supplementation. No changes were observed in the plasma levels of MDA, and erythrocyte GSH-Px activity, while SOD activity increased in both control group and the supplement group during the intervention. These data suggest that multivitamin-mineral supplementation during pregnancy produced moderate increases in plasma $\beta$-carotens, coenzyme Q10, and zinc concentrations but the enhancement of those plasma antioxidants had on direct on the plasma level of MDA, erythrocytes SOD or GSH-Px activities.

녹차 건분이나 항산화 비타민 보충이 9개월령과 12개월령 흰쥐의 부위별 뇌조직에 미치는 항산화 효과 (Effects of Green Tea Powder or Antioxidant Vitamin Supplementation on Lipid Peroxidation and Antioxidant Enzyme Activities in 9 Month- and 12 Month-old Rat Brain Regions)

  • 본 연구에서는 녹차 건분 식이나 항산화 비타민 보충식이가 뇌 조직의 항산화능 증진효과를 가지고 있는지를 보고자, 생후 9개월령과 12개월령 Sprague-Dawley종 실험동물에게 3% 녹차 건분식이와 항산화 비타민 보충식이를 3주간 공급하였다 희생 후 뇌 조직을 대뇌, 소뇌, 선조체, 해마로 나누어 malondialdehyde (MDA)의 함량과 catalase, superoxide dismutase (SOD) , glutathione peroxidase (GSH-Px)의 활성을 측정하였다. 본 실험 결과를 종합하여 볼 때, 3% 녹차 건분 식이의 공급은 선조체 내 SOD활성과 대뇌 내 GSH-Px활성을 12개월령에서 증가시키는 것으로 나타났고, 녹차군과 항산화 비타민군의 항산화능에는 서로 유의적인 차이가 없었다. 가령에 따라 부위별 뇌조직의 MDA 함량은 증가하였고, GSH-Px 활성은 감소하는 것으로 나타났다 이러한 결과는 3% 녹차 건분 식이의 항산화능 증진효과가 녹차 건분에 함유된 항산화 비타민에 기인한 것일 수 있으며 , 가령에 따라 항산화 효소 활성의 변화가 효소의 종류에 따라 달라질 수 있다는 것을 보여준다.

Correlation between Antioxidant Enzyme Activity, Free Iron Content and Lipid Oxidation in Four Lines of Korean Native Chicken Meat

  • This study was conducted to observe the association between antioxidant enzyme activity, free iron content and lipid oxidation of Korean native chicken (KNC) meat during refrigerated storage. Four lines of KNC (Yeonsan ogye, Hyunin black, Hoengseong yakdak and Hwangbong) were raised under similar conditions. A total of 16 roosters were randomly sampled and slaughtered at the age of 12 mon. The breast and thigh meats were stored aerobically for 10 d at 4℃. Although thigh meat had higher antioxidant enzyme activity, it was more susceptible to lipid oxidation and released more iron during storage than breast meat. Aerobic refrigerated storage for 10 d significantly decreased the activity of antioxidant enzymes and increased the amount of free iron and malondialdehyde. The activities of superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) were negatively correlated with lipid oxidation, whereas that of catalase was not. The amount of free iron was positively associated with lipid oxidation. We concluded that chicken line did not affect strongly on antioxidant enzyme activity and lipid oxidation in breast meat of KNC. However, the thigh meat of Hwangbong and Hyunin black had higher SOD and GSH-Px activity, respectively, and lower malondialdehyde contents than that of other chickens. SOD, GSH-Px and free iron play significant roles in meat lipid oxidation during refrigerated storage.

Indoxyl sulfate, homocysteine, and antioxidant capacities in patients at different stages of chronic kidney disease

  • BACKGROUND/OBJECTIVES: Increased levels of uremic toxins and decreased antioxidant capacity have a significant impact on the progression of chronic kidney disease (CKD). However, it remains unclear whether they interact with each other to mediate the damage of kidney function. The purpose of this study was to investigate whether uremic toxins (i.e., homocysteine and indoxyl sulfate [IS]), as well as glutathione-dependent antioxidant enzyme activities are dependently or independently associated with kidney function during different stages of CKD patients. SUBJECTS/METHODS: One hundred thirty-two patients diagnosed with CKD at stages 1 to 5 participated in this cross-sectional study. RESULTS: Patients who had reached an advanced CKD stage experienced an increase in plasma uremic toxin levels, along with decreased glutathione peroxidase (GSH-Px) activity. Plasma homocysteine, cysteine, and IS concentrations were all positively associated with each other, but negatively correlated to GSH-Px activity levels after adjusting for potential confounders in all CKD patients. Although plasma homocysteine, cysteine, IS, and GSH-Px levels were significantly associated with kidney function, only plasma IS levels still had a significant association with kidney function after these parameters were simultaneously adjusted. In addition, plasma IS could interact with GSH-Px activity to be associated with kidney function. CONCLUSIONS: IS plays a more dominant role than homocysteine and GSH-Px activity in relation to kidney function.

남자 대학생의 적혈구 항산화 효소 활성 및 혈장 TRAP수준에 대한 운동량 및 기타 관련 요인의 영향 (The Effects of Exercise and Other Relating Factors on the Activity of Erythrocyte Antioxidant Enzymes and Plasma TRAP Levies in Male College Students)

  • This study was conducted to evaluate the effect of regular exercise and other relating factors on the activities of erythrocyte antioxidant enzymes and plasma total radical-trapping antioxidant potential (TRAP) in 61 healthy male college students. The study population were divided in two groups ; small amount of exerciser (exorcise time less than 30min/d) and moderate amount of exerciser (exorcise time more than 30min/d) according to their physical exercise habits measured by a questionnaire. Dietary intake of vitamin C and vitamin E, Plasma lipid Profiles, erythrocyte superoxide dismutase(SOD), glutathione peroxidase(GSH-Px) and catalase activities, as well as plasma TRAP levels were determined. Plasma TRAP level was significantly higher in moderate amount of exercisers than that in small amount of exercisers. No significant differences were observed in erythrocryte SOD, catalase and GSH-Px between the two groups. Mean exercise time was positively correlated with the plasma level of TRAP significantly, and amount of alcohol consumption was negatively correlated with the erythrocyte SOD activity, Dietary vitamin C and I intakes did not correlated with either erythrocyte enzyme activities or plasma TRAP levels. There were positive correlations between plasma HDL-cholesterol, and erythrocyte GSH-Px or plasma TRAP levels. Plasma vitamin C concentrations was negatively correlated with plasma TRAP levels and erythrocyte SOD activity, however plasma vitamin C concentration was positively correlated with erythrocyte GSH-Px activity, The results would suggest that regular moderate exercise, nonsmoking, high HDL-cholesterol and high plasma vitamin E concentration enhance antioxidant defences against reactive oxygen species and may increase the likelihood of a healthier life span.

Protective Effects of the BuOH Fraction from Laminaria japonica Extract on High Glucose-induced Oxidative Stress in Human Umbilical Vein Endothelial Cells

  • This study investigated the protective effect of the butanol (BuOH) fraction from Laminaria japonica (BFLJ) extract on high glucose-induced oxidative stress in human umbilical vein endothelial cells (HUVECs). Freeze-dried L japonica was extracted with distilled water, and the extracted solution was mixed with ethanol then centrifuged. The supernatant was subjected to sequential fractionation with various solvents. The BuOH fraction was used in this study because it possessed the strongest antioxidant activity among the various solvent fractions. To determine the protective effect of the BFLJ, oxidative stress was induced by exposing of HUVECs to the high glucose (30 mM) or normal glucose (5.5 mM) for 48 hr. Cell viability, lipid peroxidation, glutathione (GSH) concentration, and antioxidant enzyme activities such as catalase, superoxide dismutase (SOD), glutathione peroxidase (GSH-px), and glutathion reductase (GSH-re) were measured. Exposure of HUVECs to high glucose for 48 hr resulted in a significant (p<0.05) decrease in cell viability, SOD, GSH-px and GSH-re and a significant (p<0.05) increase in thiobarbituric acid reactive substances (TBARS) formation in comparison to the cells treated with 5.5 mM glucose or untreated with glucose. BFLJ treatment decreased TBARS formation and increased cell viability, GSH concentration, and activities of antioxidant enzymes including catalase, SOD, GSH-px, and GSH-re in high glucose pretreated HUVECs. These results suggest that BFLJ may be able to protect HUVECs from high glucose-induced oxidative stress, partially through the antioxidative defence systems.

한국인 백내장환자의 항산화 체계에 관한 연구 (A Study on Antioxidant System in Cataract Patients)

  • Cataracts are the leading cause of blindness worldwide and are characterized by increased opacity of the lens that significantly diminishes visual acuity. It has been suggested that increased risk of lens opacities are associated with age, exposure to sunlight, diabetes, smoking, and poor nutrition. Antioxidant nutrients have born demonstrated to protect the lens membrane and protein against damage due to oxidative stress. The purpose of this study was to investigate the antioxidant system in the blood of cataract patients. The status of the blood antioxidant system was evaluated based on the levels of antioxidant vitamins and minerals as well as glutathione peroxidase (GSH-Px) and malondialdehyde (M7A) activity in 34 patients with cataracts (17 male and 17 female) and 45 control subjects (20 male and 25 female). After adjusting for age, the results showed significantly lower levels of antioxidant vitamins such as lycopene (M : p < 0.05, F: p < 0.01), zeaxanthin (F: p < 0.01), ${\gamma}$-tocopherol (F: p < 0.01) and ascorbic arid (M: p < 0.05) in the cataract patients than in the control subjects. In contrast, the concentration of cryptoxanthin (F : p < 0.07) showed a significantly higher value in the cataract patients. The serum level of the antioxidant mineral Zn (M : p < 0.01) was found to be significantly lower in the cataract patients while the ratio of cu/zn appeared significantly higher (M : p < 0.05). Significantly higher (M : p < 0.01, F: p < 0.05) concentrations of MDA in serum was found in the cataract patients as compared to the control subjects. GSH-Px activity was significantly lower (F: p < 0.05) in 71e cataract patients. In conclusion, the antioxidant system may play an important roll in cataract creation. Further studies are needed to clarify the mechanisms underlying these findings and to establish preventive measures with an emphasis on antioxidant nutrition for cataract patients.

한국 남성 관상동맥질환자의 혈청 항산화 무기질 수준과 효소 활성 (Levels of Serum Antioxidant Minerals and Enzyme Capacities of Korean Male Patients with Coronary Artery Disease)

  • Increased oxidative stress contributes to the progression of atherosclerosis. We measured serum antioxidant mineral concentrations, capacities of serum antioxidant enzymes and fasting lipid profile in 97 male patients with coronary artery disease (CAD) and 21 male controls. Nutrient intake was assessed by the semi-quantitative food frequency method. CAD patients were divided into single-vessel disease (SVD, n=66) and multi-vessel disease (MVD, n = 31) groups on the coronary angiography. The ratio of serum LDL- to HDL-cholesterol elevated with an increasing number of diseased vessels compared to the control (control < SVD < MVD, p < 0.05). Patients with SVD and MVD had higher levels of serum lipoprotein (a) than the control (p < 0.05). The mean intake of carbohydrate, protein and cholesterol was higher in MVD patients and the intakes of vitamins C and E were lower in MVD and SVD patients than in the control (p < 0.05). Serum copper (Cu) and zinc (Zn) levels were higher in MVD and SVD patients than in the control (Cu: control $75.8{\pm}5.07$, SVD $99.2{\pm}2.90$, MVD $100.1{\pm}2.32{\mu}g/dL$, p<0.01; Zn: $76.8{\pm}5.36$, $119.0{\pm}5.95$, $129.1{\pm}2.70{\mu}g/dL$, p < 0.01). And the ratio of Zn to Cu was higher in SVD and MVD patients than in the control (control $0.78{\pm}0.06$, SVD $0.88{\pm}0.05$, MVD $0.99{\pm}0.04$, P < 0.05). The activity of glutathione peroxidase (GSH-Px) was lower in MVD than in SVD and the control (control $35.13{\pm}1.34$, SVD $35.30{\pm}1.01$, MVD $31.00{\pm}1.04 U/mg$ protein, p < 0.05). The ratio of the activities of superoxide dismutase (SOD) to GSH-Px was higher in MVD than in control and SVD (p < 0.05). In groups with CAD, serum Cu and Zn concentrations and their ratio were changed compared to the control. GSH-Px activity was decreased and the ratio of SOD to GSH-Px was increased in the patients with MVD. The balances between the activities of SOD and GSH-Px should also be considered a risk factor in CAD patients.
