Relationship Among Body Mass Index, Nutrient Intake and Antioxidant Enzyme Activity of Postmenopausal Women

  • To elucidate the relationship among body mass index, nutrient intake and blood antioxidant capacity in the postmenopausal period, 60 women residing in Iksan area were recruited. Body mass index (BMI) was calculated base on height and weight, and food and nutrient intakes were estimated by 24-hour recalls of 3 non-consecutive days. Parameters of antioxidant capacity including the activities of superoxide dismutase (SOD), glutathione peroxidase (GPx), catalase (CAT) and total antioxidant capacity (TA) were measured in fasting blood samples from the subjects. The average age, height, weight and BMI of the subjects were 65 years, 151.1cm, 59.5 kg and 26.0 m/kg$^2$, respectively. The macronutrient intake rate of carbohydrate : protein : fat were 65:17.5:17.5; the mean intakes of energy and protein were 1532.7 kcal (86.3% of RDA) and 67.1 g (122.0% of RDA) respectively. The mean intakes of phosphorus, vitamin A, niacin and vitamin C were higher than Recommended Daily Allowance (RDA) for Koreans. On the other hand, calcium and riboflavin intakes were only 84.6% and 70.4% of RDA. Among the parameters of antioxidant capacity, SOD activity was significantly lower in lean subjects (BMI<20) than in the normal or overweight subjects (BMI${\geq}$20) (p<0.05). TAs of the subjects with the highest intakes of vegetables and fruits were significantly higher than those of subjects with lower intakes (p<0.05). Antioxidant capacity was compared among subjects according to 3 different nutrient intake levels according percentage of RDA for Koreans for selected nutrients with the following results: The high protein and niacin groups exhibited significantly lower TA status than those of the other intake groups (p<0.05). In conclusion, the low BMI was associated with lower SOD activity in postmenopausal women. Higher consumption of fruits and vegetables was associated with higher TA. When protein and niacin intakes were excessive, SOD activity and TA tended to be low. SOD and TA, among antioxidant indexes, seemed to be mostly influenced by other factors. Therefore, more studies on the effects of nutritional intake and the activity of antioxidant enzyme should be conducted.

In Vitro and In Vivo Antioxidant Activity of Aged Ginseng (Panax ginseng)

  • Fresh ginseng roots were aged in an oven at $80^{\circ}C$ for 14 d. The in vitro and in vivo antioxidant activities of this aged ginseng, in comparison with those of the white and red ginsengs, were evaluated. In in vitro antioxidant assays, the ethanolic extracts from aged ginseng showed significantly higher free radical scavenging activity and reducing power than those of the white and red ginsengs. In in vivo antioxidant assays, mice were fed a high fat diet supplemented with white, red, or aged ginseng powders. High fat feeding resulted in a significant increase in lipid peroxidation and a substantial decrease in antioxidant enzymes activities in the animals. However, diet supplementation of ginseng powders, particularly aged ginseng, markedly reduced lipid peroxidation and enhanced the antioxidant enzymes activities. The results illustrate that the aged ginseng has greater in vitro and in vivo antioxidant capacity than the white and red ginsengs. The aged ginseng also showed considerably higher total saponin, phenolic, and flavonoid contents, indicating that its antioxidant capacity may have been partly due to its high levels of antioxidant compounds. This new ginseng product may be useful as a functional food with strong antioxidant potential.

A selective Assay To Measure Antioxidant Capacity in Both The Aqueous and Lipid Compartments of Plasma

  • The measurement of the total antioxidant capacity (TAC) of human plasma has been widely applied in nutritional science, for example to evaluate the antioxidant contribution of dietary components and to study, although indirectly, the bioavailability of dietary antioxidants. Several methods have been proposed for the measurement of TAC, most of them based on the ability of plasma to withstand the oxidative damage induced by aqueous radicals. Although plasma contains both hydrophilic and lipophilic antioxidants that interact through extensive cross-talk in most of the methods employed for the TAC measurement, the hydrophilic antioxidants such as ascorbic acid, uric acid, and protein thiols mainly contribute to the total antioxidant plasma capacity (almost 70%) while lipophilic antioxidants embedded in the lipoproteins (carotenoids, a-tocopherol, ubiquino1-10) participate only in a negligible amount (less than 5%). The present paper reviews the analytical methods used to assess the TAC and in particular focuses on new approaches that are capable of distinguishing the antioxidant capacity of both the aqueous and lipid compartments of plasma. The general principle of the method as well as some in vitro and ex vivo applications will be discussed within the text.

루테인과 후코이단 병용 처리에 의한 AAPH 유도 세포 손상 억제 (Protective Effect of Co-treatment of Lutein and Fucoidan Against AAPH-Induced Damage in THP-1 Cells)

  • This study was designed to investigate the protective effect of the combination of fucoidan and lutein against AAPH-induced oxidative stress in THP-1 cells. The combination of fucoidan and lutein existed significant antioxidant effect on AAPH-damaged THP-1 cells by using lipid peroxidation and cellular antioxidant capacity assay. Fucoidan($1\;{\mu}g/m{\ell}$) and lutein($10\;{\mu}g/m{\ell}$) did not affect at all the viability of THP-1 cells, but protected the AAPH-damage of THP-1 cells at the same concentration. The viability of THP-1 cells was 0% with 1 mM AAPH alone, the protective effect of fucoidan($1\;{\mu}g/m{\ell}$) and lutein($10\;{\mu}g/m{\ell}$) was 37% and 36%, respectively. The combination of fucoidan($1\;{\mu}g/m{\ell}$) and lutein($10\;{\mu}g/m{\ell}$) exhibited significant inhibitory effect of lipid peroxidation using TBARS assay and cellular antioxidant capacity using DCFH-DA assay. In lipid peroxidation, the TBARS value of 1 mM AAPH alone was $0.8{\pm}0.03\;nM$ MDA, its of the combination of fucoidan($1\;{\mu}g/m{\ell}$) and lutein($10\;{\mu}g/m{\ell}$) was $0.2{\pm}0.05\;nM$ MDA. In cellular antioxidant capacity, the combination of fucoidan($1\;{\mu}g/m{\ell}$) and lutein($10\;{\mu}g/m{\ell}$) exhibited significant cellular antioxidant capacity of 76%, whereas quercetin($10\;{\mu}M$) as positive control exhibited the cellular antioxidant capacity of 32%. These results indicate that the cotreatment of fucoidan and lutein protects against AAPH-induced THP-1 cell damage by inhibiting lipid peroxidation, increasing cellular antioxidant capacity.

Total antioxidant capacity of the Korean diet

  • BACKGROUND/OBJECTIVES: The objective of this study was to measure and/or estimate the total antioxidant capacity of the Korean diet. MATERIALS/METHODS: Eighty-one plant foods that were expected to exhibit rather high antioxidant activities were selected from the Korean diet using the Fifth Korean National Health and Nutrition Survey (KNHANES V). These foods were categorized into 11 food groups: cereals, potatoes, legumes, nuts, vegetables, kimchies, mushrooms, fruits, fruit juices, sea weeds, and oils. The foods were mixed in the proportions specified in traditional Korean recipes and analyzed. The measured indicators for antioxidant capacities were total phenolics, 2,2-diphenyl-1-picrylhydrazyl (DPPH), oxygen radical absorbance capacity (ORAC), and Trolox equivalent antioxidant capacity (TEAC). RESULTS: Total phenolics were high in the fruit juices, nuts, vegetables, and fruits; and the average DPPH, ORAC, and TEAC values were high in the vegetables, fruits, fruit juices, and nuts. The correlation coefficient between the content of total phenolics of each food and the in vitro antioxidant capacity was relatively high at 0.851. The intake of total phenolics per capita per day in the Republic of Korea was estimated to be 127 mg. The total dietary antioxidant capacity (TDAC) values, which were obtained from the total antioxidant capacity of each food, taking into account the intake of each food, were 20,763, 54,335, and $876.4{\mu}mol$ of Trolox equivalents using the DPPH, ORAC, and TEAC methods, respectively. The food group that contributed the most to the Korean TDAC was cereals at 39.7%, followed by fruits and vegetables at 27.8% and 13.9%, respectively. The contribution of legumes, nuts, fruit juices, and mushrooms was quite minimal at less than 2% each. CONCLUSIONS: The content of total phenolics and the antioxidant capacity of the Korean diet are significantly correlated and the high contributing food groups are cereals, fruits, and vegetables.

ORAC Assay 에 의한 인삼의 항산화 활성 연구 (Determination of the Antioxidant Capacity of Korean Ginseng Using an ORAC Assay)

  • 인삼의 여러 생리 활성 가운데 항산화 정도를 알아보기 위하여 백삼(6 년근), 백삼(5 년근), 피부백삼(5 년근), 피부백삼 (4 년근) 의 80% 에탄올 엑기스, 에틸아세테이트 분획, 수포화 부탄올 분획, 물 분획을 얻은 후 LC/Mass 를 사용하여 사포닌 함량을 조사하고 기존의 여러 가지 항산화 작용 측정 방법들의 오류를 없애고 더욱 정확한 결과를 낼 수 있는 대처 방안으로 선정된 ORAC Assay에 의해 항산화 활성을 검토하였다. 인삼 중 사포닌은 ginsenoside Rg 1 과 Rb1 이 주요 성분으로 다량 함유하고 있었으며, Rc, Rb2, Re 등이 뒤를 이었고, 그밖에도 Rd, Rg3, Rh1가 널리 분포하고 있었다. 피부백삼 5 년근의 경우 에탄올 엑기스와 수포화 부탄올 분획에서 다른 인삼 분획에 비해 높은 함유량을 보였으나 실험의 한계상 인삼 재배기간과 인삼 종류별 각각의 분획에 대한 사포닌 함량 비교는 어려웠다. 인삼의 각 분획별 항산화 활성은 80% 에탄올 엑기스, 에틸아세테이트 분획, 수포화 부탄올 분획, 물 분획 모두에서 나타났고 비교적 전체 유기 용매 분획의 값이 비슷하였으며, 수층 분획이 다소 낮은 활성을 보였다. 검체 인삼들의 각 용매추출 분획 상호간의 유의성 비교에서는 모든 인삼 검체의 에틸아세테이트 분획에서만 유의성을 나타내었다(p>0.05). 인삼 중 항산화 활성은 에틸아세테이트 층으로 이행되는 폴리페놀 계통 성분이나 일부 비극성의 사포닌에 의한 것으로 추측되고 있으나 모든 분획에서 나타난 것으로 보아 이들 외에 산성 다당체, 당 단백질, 수용성 다당류 등 다른 생리활성 물질에 대한 연구가 요구된다.

빈도에 따른 복합운동프로그램이 노인들의 체내 총항산능 및 활성산소 유리기에 미치는 영향 (The Analysis of Old Person's Total Antioxidant Capacity and Free Oxygen Radical According to Exercise Frequency of Combination Exercise)

  • Purpose:The purpose of this study was analyze the effect combination exercise program on plasma total antioxidant capacity and free oxygen radical according to exercise frequency of elderly. Age range of subject is 65~70 years. Methods:Subjects divided into three groups: control group(0-time), week-3 times group(3-times), week-5 times group(5-times). Each group were performed combination exercise during 8 weeks. Results:The results of each group by ANOVA free oxygen radical have a change(p<.05). We performed comparison of total antioxidant capacity and free oxygen radical pre-test and post-test on each groups. Total antioxidant capacity was increased in the 3-times and 5-times groups but there have not significant. Total antioxidant capacity is improve better than to 5-times. Free oxygen radical is significant increased in the 0-time (p<.05). But 5-times but 3-times is no significant decrease. Free oxygen radical is improve better than to 3-times. Conclusion:In the old people, exercise frequency week 3 times doing appeared effectively in improvement of the free oxygen radical and 5 times doing appeared effectively in improvement of the total antioxidant capacity. So rightly must apply a exercise frequency well in characteristic of old person each person.

기허(氣虛), 혈허(血虛) 동물모델에서의 활성산소 및 항산화력 연구 (A Study on Active Oxygen and Antioxidant capacity of Qi Deficiency and Blood Deficiency Animal Model)

  • Background and Objective : There are a lot of theories that explained the aging process, and the oxidative stress is one of the important theory that explained the aging process. The aim of this study was to investigate active oxygen and antioxidant capacity of Qi deficiency and Blood deficiency animal models. Material and Methods : Sprague-Dawley rats were divided into three groups: normal group, Qi deficiency group and Blood deficiency group. The Qi deficiency animal model was induced through restriction of food (12g/kg/day) for 20 days. Blood deficiency animal model was induced by bleeding from tail vein(0.4ml/time) 8 times. The normal animal model was kept without any intervention. The oxidative stress was observed by measuring the active oxygen and antioxidant capacity. Results and Conclusion : 1. Active oxygen was significantly increased in the Qi deficiency group and Blood deficiency group. (P=0.061) 2. Antioxidant capacity was increased in the Qi deficiency group and Blood deficiency group. But there is no significant difference. (P=0.113)

만성 뇌졸중 환자의 항산화능에 영향을 주는 요인에 관한 연구 (Antioxidant Capacity and Associated Factors during the Chronic Phase after Stroke)

  • Purpose: The purpose of this study was to investigate antioxidant capacity and relating factors including gender, obesity, lifestyle factors, and nutrient intake in chronic stroke patients. Methods: A total of 188 chronic stroke patients who visited a medical center in Seoul participated in the study. A structured questionnaire was used to assess their clinical characteristics and lifestyles. Blood samples were collected for ferric reducing ability of plasma (FRAP) for antioxidant capacity. Dietary intake of the patients were obtained for 2 days by 24 hours recall method. Results: The mean FRAP was $392.0{\mu}mol/L$. According to the hierarchical multiple regression analysis, the factors related to high FRAP were male sex (p=.036), alcohol drinking (p=.013), and calorie intake deficiency (p=.005). Conclusion: We found that antioxidant capacity was related to lifestyle factors including alcohol drinking, and calorie intake in chronic stroke patients. A tailored strategy is needed to increase antioxidant capacity according to gender and lifestyles in the chronic phase of stoke patients.

Influence of Roasting Treatment on the Antioxidant Activities and Color of Burdock Root Tea

  • The major trend in the antioxidant market is the growing consumer demand for natural antioxidants. Tea, one of the most widely consumed beverages in the world, is an easy way to obtain antioxidant components from a natural source. Our objective was to develop burdock root tea (BRT) with potent antioxidant activity and good color quality. In order to obtain maximum antioxidant activity and quality, the effect of roasting was determined. The antioxidant capacities and total phenolic contents of BRT increased as roasting increased. The color of BRT became darker with increased roasting, extraction time, and amount of burdock roots. Color of BRT was also positively correlated with total antioxidant capacity. Roasting significantly enhanced the total antioxidant activities and color quality of BRT. These results suggest that roasting BRT increases beneficial antioxidant components from burdock roots.
