Red rose petals are usually disposed but they are an abundant source of phenolics and traditionally used as food supplement and as herbal medicine. Of the Various phenolics, they are known to have anticancer, antioxidant, and anti-inflammatory properties. In this study, we investigated the anti-inflammatory effects of red rose ethanolic extracts (GRP) on lipopolysaccharide (LPS)-activated RAW 264.7 cells. The results demonstrated that pretreatment of GRP (500㎍/mL) significantly reduced NO production by suppressing iNOS protein expression in LPS-stimulated cells. Anti-inflammatory effects by red rose petals were observed in the following. Red rose petals inhibited the translocation of NF-κB from the cytosol to the nucleus via the suppression of IκB-α phosphorylation and also inhibited LPS-stimulated NF-κB transcriptional activity. These findings suggest that red rose petals exert anti-inflammatory actions and help to elucidate the mechanisms underlying the potential therapeutic values of red rose petals. Therefore, red rose petals could be regarded as a potential source of natural anti-inflammatory agents.
Hemorheological properties are easily modified by glucose-induced oxidation and glycation. Carnosine, a naturally occurring dipeptide ($\beta$-alanyl-LFull-size image-histidine), has been recently proposed to act as an antioxidant as well as a free-radical scavenger. In the present study, we investigate its protective and rejuvenating effects in erythrocytes that are exposed to glucose-rich plasma. Erythrocytes that were incubated in glucose solutions were treated with different concentrations of carnosine and for different incubation times. Their hemorheological alterations were examined. The results reveal that the presence of carnosine effectively prevented these rheological alterations in a concentration-dependent manner in glucose-rich media. It is proposed that moderate concentrations of carnosine might be further explored as potential therapeutic agents for pathologies that involve hemorheological modification.
The purpose of the present research was to investigate the physiological activities of the thermal treated eggplant on the skin. Five minute of thermal treatment at $100^{\circ}C$ had the highest polyphenol content of eggplant. However, below and over 5 min of thermal treatment time, they did not increase. When water and ethanol extracts were used, the maximum DPPH radical scavenging activities were obtained, 66.3 and 62.8%, respectively. Among various extracts, the acetone extract gave the highest cosmetic activity. Especially, when acetone extract (15.0 mg/mL) was used, the maximum inhibition activities of tyrosinase, elastase, and collagenase were obtained, 83.4, 78.2 and 62.5%, respectively. These results suggest the anti-wrinkle and whitening and effects of acetone extract were excellent. Therefore, it is should be considered as a promising candidate for novel cosmetic agents.
Xylene is a cyclic hydrocarbon and an environmental pollutant. It is also used in medical technology, paints, dyes, polishes and in many industries as a solvent; therefore, an understanding of the interaction between xylene and human lymphocytes is of significant interest. Biochemical assessment was used to demonstrate that exposure of lymphocytes to xylene induces cytotoxicity (at 6 hr), generates intracellular reactive oxygen species, collapse of mitochondrial membrane potential, lysosomal injury, lipid peroxidation and depletion of glutathione (at 3 hr). The findings show that xylene triggers oxidative stress and organelle damage in lymphocytes. The results of our study suggest that the use of antioxidant, mitochondrial and lysosomal protective agents can be helpful for individuals subject to chronic exposure to xylene.
Two flavonoids (1 and 2) and one phenolic acid (3) obtained from Erigeron annuus have recently been shown to have potent antioxidant activities. Aim of this study was to investigate the inhibitory effects of these components on $interferon-{\gamma}$ and lipopolysaccharide-induced nitric oxide productions in the mouse pheritoneal macrophage. Compounds 2 and 3 showed marked inhibitory activities against inducible nitric oxide synthase (iNOS) on the lipopolysaccharide and $interferon-{\gamma}-stimulated$ mouse pheritoneal macrophages without cytotoxicity. Therefore, these results suggest that the compounds could be effective anti-inflammatory agents as nitric oxide inhibitors in vivo.
Proanthocyanidins (PACs), also named condensed tannins, are polymers of flavan-3-ols such as (+ )-(gallo)catechin and (-)-epi(gallo)catechin. A proper analysis of the PACs, with difficult challenges due to their complex structures, is crucial in studies of cancer chemoprevention. Cancer is a leading cause of mortality around the world. Many experimental studies have shown that dietary PACs are potential chemopreventive agents that block or suppress against multistage carcinogenesis in both in vitro and in vivo models. Cancer chemoprevention by dietary PACs has been shown effective through different mechanisms of action such as antioxidant, apoptosis-inducing, and enzyme inhibitory activities. Good sources of dietary PACs are nuts, fruits, beans, chocolate, fruit juice, red wine, and green tea. The chemopreventive potential of dietary PACs should be considered together with their bioavailability in humans. The safety issues regarding carcinogenesis and gastrointestinal disorder are also reviewed.
Sesamol, one of constituents isolated from sesame seed (Sesamum indicum L.) has been known to have natural antioxidant property. Nicotinic acid and clofibric acid have been used for treatment of hyperlipidemia. Sesamol derivatives prepared by conjugation with nicotinic acid (SJ-301) and clofibric acid (SJ-302) were expected to have various biological activity caused by synergistic effect of sesamol. Thus, this study were carried out to investigate the synthesis of sesamol derivatives from seasamol and antihyperlipidemia drugs and their biological activities. As a result, SJ-301 showed a stronger antihyperlipidemia activity in vivo and inhibitory effect on $TNF-{\alpha}$ release in vitro compared to the original agents such as sesamol, nicotinic acid and clofibric acid.
Green tea and tea polyphenols have been studied extensively as cancer chemopreventive agents in recent years. Epigallocatechin-3-gallate (EGCG) is widely recognized as a powerful antioxidant and a free radical scavenger. The purpose of this study was to evaluate the protective effects of green tea catechins (GTC) on the Bleomycin- and Cyclophosphamide-induced cytotoxicity. Cell viability was measured by MTT assay. In the protective effect of GTC, the cell viability was significantly increased by the treatment of GTC. Furthermore, GTC showed the higher protective effect than EGCG and vitamin E. These results suggest that GTC has the protective effect which is related to the prevention of cancer. Our studies show that the continuous presence of EGCG can reduce radical-induced DNA damage in Chinese hamster lung fibroblast cells (CHL cells).
Oxygen radicals are produced in many pathophysiologic states whether the event is a causal factor of illness or is a result of their progression. Oxygen radicals including superoxide anions. hydrogen peroxide are thought to be involved in a number of type of acute. and chronic pathologic condition in the brain and neural tissue. So the antioxidants have recently received much attention as therapeutic agent for the treatment of neurodegenerative disease. In this study. we describe synthesis of a series of chromenone derivatives as antioxidant agents. The target compounds are designed to include the structural frature of caffeic acid. flavoniod. and focopherol known as antioxidants.
In this study, a preliminary evaluation of the antioxidant and anti-inflammatory activity of the Ficus erecta var. sieboldii (Miq.) King (FES) leaf extract has been performed to assess its potential as a natural resource for food and medicinal materials. FES was extracted using 70% EtOH and then fractionated sequentially using n-hexane, $CH_2Cl_2$, EtOAc, and n-BuOH. To screen for antioxidant and anti-inflammatory agents effectively, the inhibitory effect of the FES extracts on the production of oxidant stresses (DPPH, xanthine oxidase, and superoxide) and pro-inflammatory factors (NO, iNOS, COX-2, $PGE_2$, IL-6, and $IL-1{\beta}$) in the murine macrophage cell line RAW 264.7 activated with lipopolysaccharide (LPS) was examined. Among the sequential solvent fractions of FES, the $CH_2Cl_2$ and EtOAc fractions showed decreased production of oxidant stresses (DPPH, xanthine oxidase and superoxide), and the hexane and $CH_2Cl_2$ fractions of FES inhibited the production of pro-inflammatory factors (NO, iNOS, COX-2, and $PGE_2$). The $CH_2Cl_2$ fraction also inhibited the production of pro-inflammatory cytokines ($TNF-{\alpha}$, IL-6, and $IL-1{\beta}$). These results suggest that FES has a significant effects on the production of oxidant stresses and pro-inflammatory factors and may be used a natural resource for antioxidant and anti-inflammatory agents.
