제주 동백나무 겨우살이(Korthalsella japonica (Thunb.) Engl.)의 항산화 및 α-glucosidase와 pancreatic lipase 저해 활성 (In vitro antioxidant property and α-glucosidase and pancreatic lipase inhibiting activities of Jeju camellia mistletoe (Korthalsella japonica (Thunb.) Engl.) extracts)

  • 제주 동백나무 겨우살이 추출물의 항산화 활성을 DPPH 라디컬 소거능, 철이온 킬레이팅과 환원력 시험을 통해 조사한 결과, 메탄올과 에탄올 추출물이 열수 추출물보다 뛰어난 항산화 효능을 보였다. ${\alpha}$-Glucosidase와 pancreatic lipase 저해능 또한 메탄올과 에탄올 추출물이 우수한 활성을 보였다. 따라서, 제주 동백 나무 겨우살이의 메탄올 및 에탄올 추출물을 건강기능 의약식품 소재로 개발하기 위한 추가 연구가 진행되어야 할 것이다.

국내 및 미얀마에서 재배된 삼채뿌리의 항산화, α-Glucosidase 저해 및 Acetylcholinesterase 저해 활성 (Evaluation of antioxidant, α-glucosidase inhibition and acetylcholinesterase inhibition activities of Allium hookeri root grown in Korea and Myanmar)

  • 본 연구는 선행 연구된 삼채뿌리의 뛰어난 영양성을 토대로 기능성 식품소재로의 이용가능성을 확인하고 활용도를 높이고자 국내산 및 미얀마산 삼채뿌리의 항산화 및 생리활성을 측정하였다. 삼채뿌리의 총 폴리페놀과 총 플라보노이드 함량은 국내산 삼채뿌리가 각각 5.27 mgGAE/g, 0.35 mgQE/g, 미얀마산 삼채뿌리가 각각 4.80 mgGAE/g, 0.24 mgQE/g으로 국내산 삼채뿌리가 더 높은 함량을 나타냈다. 삼채뿌리의 항산화력을 측정한 결과 DPPH 라디칼 소거능은 미얀마산이 더 높은 활성을 보였지만, ABTS 라디칼 소거능과 $Fe^{2+}$ 킬레이팅 활성, 환원력은 국내산이 더 높은 활성을 보였다. ${\alpha}-Glucosidase$ 저해활성은 국내산의 저해활성이 미얀마산 삼채뿌리의 저해활성보다 더 높았으며 대조구인 acarbose에 비해서도 우수한 저해활성을 보였다. Acetylcholinesterase 저해 활성을 측정한 결과, 50 mg/mL 농도에서 국내산 삼채뿌리가 51.44%로 44.33%의 저해활성을 보인 미얀마산 삼채뿌리보다 더 높은 저해활성을 보였다. 본 연구를 통해 삼채뿌리 특히 국내산 삼채뿌리는 높은 항산화 활성, ${\alpha}-glucosidase$ 및 acetylcholinesterase 저해 활성을 보여 혈당조절제, 치매예방 및 개선제로써의 활용가능성이 큰 것으로 확인되었으며, 이로써 기능성 식품소재로의 활용이 증가될 것으로 판단된다. 하지만 향후 국내산 삼채의 우수성을 검증하기 위하여 여러 지역에서 재배된 삼채의 기능성 분석이 필요할 것으로 판단된다.

발아에 따른 콩류의 식후 혈당 상승 억제효능과 항산화 활성 (Enhancement of the Anti-hyperglycemic and Antioxidant Activities of Five Selected Beans by the Germination Process)

  • After a mixed carbohydrate diet, inhibition of ${\alpha}$-amylase and ${\alpha}$-glucosidase involved in the digestion and absorption of carbohydrates can significantly decrease the postprandial increase of blood glucose level. In the course of screening these useful enzyme inhibitors, we selected five kinds of bean, using an in-vitro enzyme inhibition assay method. To evaluate the effect of germination process on the functionality of the bean, we investigated the inhibitory activities of the water extracts of non-germinated bean and germinated bean against ${\alpha}$-amylase and ${\alpha}$-glucosidase, relevant to postprandial hyperglycemia. We also investigated the oxygen radical absorbance capacity(ORAC), total phenolics content, and postprandial blood glucose lowering effect in rats(Sprague-Dawley rat model). Most germinated beans showed significantly higher ${\alpha}$-glucosidase inhibitory activity, compared with non-germinated beans. Among germinated beans, Glycine max had the highest ${\alpha}$-glucosidase inhibitory activity(53.3%). The water extract of germinated Phaseolus vulgaris L. had the highest ${\alpha}$-amylase inhibitory activity(95.1%), followed by Glycine max(58.7%), and Glycine max L. Merr(54.1%). Furthermore, the five germinated beans also showed high antioxidant activities in ORAC assay. Results suggested that the germination process may improve and enhance the anti-hyperglycemia potential and antioxidant activity of the bean.

Antioxidant and Anti-diabetes Activities of Methanolic Extract and Fractions of Astragalus membranaceus Roots

  • The potential biological activities of methanol extract and 5 fractions (hexane, $CH_2Cl_2$, EtOAc, BuOH and water) from roots of Astragalus membranaceus were examined by 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging, hydroxyl radical (${\cdot}OH$) scavenging activity, reducing power assays, lipid peroxidation inhibitory activity, $\alpha$-amylase and $\alpha$-glucosidase inhibition assays. The EtOAc fraction showed high DPPH free radical scavenging activity ($EC_{50}=170.34\;{\mu}g/mL$), hydroxyl radical scavenging activity ($EC_{50}=32.14\;{\mu}g/mL$), lipid peroxidation inhibitory activity ($EC_{50}=52.46\;{\mu}g/mL$) and a concentration dependence, with OD value ranging from 0.234 to 0.345 (0.1 to 0.5 mg/mL), for reducing power. The EtOAc fraction has the highest total phenolic content ($142.13\;Gal\;{\mu}g/mg$) and the $CH_2Cl_2$ fraction has the highest flavonoid content ($71.63\;Que\;{\mu}g/mg$). Meanwhile, hexane and EtOAc showed certain $\alpha$-amylase and $\alpha$-glucosidase inhibition activities. These results suggest that the methanol extract and fractions from Astragalus membranaceus root have significant antioxidant and anti-diabetes activities, which could be used as a potential source of pharmaceutical materials.

큰방가지똥 추출물의 항당뇨 및 항고혈압효과 (Antidiabetes and Angiotensin Converting Enzyme Inhibitory Activity of Sonchus asper (L) Hill Extract)

  • In this study, we evaluated the bioactivities of methanol extract and its solvent fractions of Sonchus asper (L.) Hill. The EtOAc fraction of S. asper exhibited more strong antioxidant activity than other extracts as evidenced by the strongest 1,1-diphenyl-2-picryl hydrazyl (DPPH) radical scavenging activity with a $EC_{50}$ value at $33.55\;{\mu}g/mL$ and reducing power, the total polyphenol (180.71 mg GAE/g) and flavonoid contents (145.86 mg QE/g) of S. asper extract were higher than other extracts. The EtOAc fraction of the S. asper also showed 47.38% mushroom tyrosinase inhibition activity, 56.22% ${\alpha}$-glucosidase inhibition and 46.58% ${\alpha}$-amylase inhibition ratio at 1 mg/mL. Both methylene chloride and EtOAc fractions of methanol extract of S. asper effectively reduced of the 86.34% and 62.03% angiotensin I converting enzyme (ACE) activity at 2 mg/mL, respectively. These findings suggest that the EtOAc fraction of the S. asper could be a potential antioxidant in food additive, medicinal, and industry product.

노간주나무(Juniperus rigida Sieb) 추출물의 항산화 활성 및 α-amylase와 α-glucosidase에 대한 저해활성 (Antioxidant activity and inhibition activity against α-amylase and α-glucosidase of Juniperus rigida Sieb extracts)

  • 본 연구에서는 노간주나무 추출물의 항산화 및 ${\alpha}$-amylase와 ${\alpha}$-glucosidase을 조사하여 항산화소재로 이용하고자 실험하였다. 노간주나무 추출물의 항산화효과를 살펴보기 위하여 총 폴리페놀 함량을 측정한 결과 열수추출물은 71.3mg/g, 에탄올추출물은 116.0 mg/g 함량을 포함하고 있었으며, 플라보노이드 함량은 열수추출물은 17.7 mg/g, 에탄올추출물은 76.4 mg/g 함량을 포함하고 있었다. ABTS 라디칼 소거능을 측정한 결과 $500{\mu}g/mL$ 농도에서 열수추출물은 76.4%, 에탄올 추출물은 79.3% 라디칼 소거능을 나타내었다. FRAP 값을 측정한 결과 열수추출물에서 1.83 mM를 에탄올추출물에서 1.77 mM로 열수추출물에서 더 높은 효과를 나타내었다. ACE 활성억제효과는 $500{\mu}g/mL$ 농도에서 열수추출물 75.39%, 에탄올 추출물 71.25%로 저해율을 나타내었다. 노간주나무 추출물의 antioxidant protection factor를 측정한 결과 열수추출물의 경우는 1.5 PF, 에탄올추출물의 경우는 2.1 PF를 나타내었다. TBARS 저해율은 $500{\mu}g/mL$ 농도에서 열수추출물과 에탄올추출물 각각 55.78%와 71.48%을 나타내었다. ${\alpha}$-glucosidase와 ${\alpha}$-amylase 저해효과를 측정한 결과 $500{\mu}g/mL$ 농도에서 각각 70%와 90% 이상의 저해효과를 나타내어 항산화 및 항당뇨 예방물질 소재로의 활용이 가능할 것으로 판단된다.

제비꽃 추출물의 항산화 활성 및 α-Amylase와 α-Glucosidase에 대한 저해 활성 (Antioxidant Activity and Inhibition Activity against α-Amylase and α-Glucosidase of Viola mandshurica Extracts)

  • 민간요법에서 화농성 피부질환이나 염증성 질환에 대해 효과가 있다고 알려진 제비꽃(Viola mandshurica)의 항산화 활성과 항당뇨 활성을 조사하였다. 제비꽃 10 g에 200 mL의 네 가지 용매(메탄올, 에탄올, 아세톤, 물)를 각각 가하여 추출한 다음, 농축하여 각각의 용매별 추출물을 얻었다. 이용매별 추출물의 총 페놀 함량은 메탄올 추출물이 34.49 mg/g 갈산 당량으로 가장 높았고, DPPH 라디칼과 ABTS 라디칼 소거능은 아세톤 추출물이 $1,000{\mu}g/mL$ 농도에서 각각 21.13%와 43.53%로 가장 높은 값을 보였다. 환원력의 경우에는 물 추출물이 $1,000{\mu}g/mL$ 농도에서 0.454의 값으로 가장 높은 활성을 보였다. 항당뇨 활성은 ${\alpha}$-amylase와 ${\alpha}$-glucosidase에 대한 저해 활성으로 측정하였는데 아세톤 추출물이 가장 활성이 높았으며 $1,000{\mu}g$/mL 농도에서 모든 효소 활성들을 저해하였다. 이상의 결과로부터 제비꽃 추출물은 항산화능과, 당뇨 관련 효소에 대한 저해능이 있음을 확인할 수 있었다.

Biological Activities of Water and Ethanol Extracts from Two Varieties of Rubus coreanus Miquel Fruits

  • The potential biological activities of water and ethanol extracts from two varieties of Rubus coreanus Miquel fruits produced in the Gochang-gun (GR) and Hoengseong-gun (HR) regions of Korea were examined. The hydroxyl radical ($\cdot$OH) scavenging activity, reducing power, lipid peroxidation inhibitory activity, and antiproliferative activity on cancer cells by these extracts were examined, and $\alpha$-amylase and $\alpha$-glucosidase inhibition assays were also performed. The EtOH extract from GR showed high hydroxyl radical scavenging activity ($EC_{50}=119.47{\pm}5.13\;{\mu}g/mL$), lipid peroxidation inhibitory activity ($EC_{50}=213.45{\pm}3.14\;{\mu}g/mL$) and a concentration dependence, with OD values ranging from 0.15 to 0.56 (50 to 200 ${\mu}g/mL$), for reducing power. The EtOH extract from GR has the highest antiproliferative activities on MDA-MB-231 and HepG2 cancer cells among four extracts. Meanwhile, all extracts showed certain $\alpha$-amylase and $\alpha$-glucosidase inhibition activities. These results indicate that extracts from two varieties of R. coreanus fruits have significant antioxidant, anti-diabetic and anti-tumorigenic activities, which suggest these extracts could be a potential source for pharmaceutical.

Lactobacillus plantarum MKHA15와 Leuconostoc mesenteroides MKSR을 첨가한 발효 채소 주스의 특성 (Characteristics of Vegetable Juice Fermented with Lactobacillus plantarum MKHA15 and Leuconostoc mesenteroids MKSR)

  • The objective of this study was to develop fermented vegetable juices that possess antidiabetic and antioxidant activities. Lactobacillus plantarum MKHA15 (MKHA15) and Leuconostoc mesenteroides MKSR (MKSR) were applied to ferment onion, cabbage, and tomato juices at $37^{\circ}C$ and $30^{\circ}C$ for 72 h, respectively, and their functionality was tested using the 12 h hour-fermented juice by MKHA15, and 48 h hour-fermented juice by MKSR. Inhibition of ${\alpha}$-glucosidase activity was observed in all fermented juices. The onion juice fermented by MKHA15 showed significantly higher ${\alpha}$-glucosidase inhibition activity compared to other juices. All juices showed more than 70% inhibition of ${\alpha}$-amylase activity. The DPPH radical scavenging activity of onion juice fermented by MKSR showed significantly lower activity than cabbage and tomato juices; however, no difference was observed between the types of starter cultures. The SOD-like activity of cabbage juice fermented by MKSR was the highest among the fermented juices. The juices fermented by MKHA15 showed higher reducing power than those by MKSR. Therefore, we believe that cabbage, onion and tomato juice fermented by MKHA15 and MKSR would be useful in probiotic juices, as they possess antidiabetic and antioxidant activities.

Antioxidant and Antidiabetic Activities of Eucommia ulmoides Bark

  • Eucommia ulmoides bark extracts by cold water, boiling water, 100% EtOH, 70% EtOH, 100% MeOH, 70% MeOH and $CHCl_3$ were assayed for their medicinal effects. The antioxidant activity of the extracts ranged from $IC_{50}$ 125.2 to $IC_{50}\;872.7{\mu}g/ml$ in the 1,1-diphenyl-2-picrylhydrazyl (DDPH) free radical-scavenging assay, and cold water extracts had the highest antioxidant activity. $CHCl_3$ extracts had the highest inhibitory effect on angiotensin I-converting enzyme (ACE) giving inhibition of up to 56.4% at a concentration of 1 mg/ml. Extracts in 100% EtOH had the greatest inhibitory effect on $\acute{a}-amylase$ activity ($IC_{50}=174.6{\mu}g/ml$), and 70% MeOH extracts had the greatest inhibitory effect on ${\alpha}-glucosidase$ activity ($IC_{50}=14.0{\mu}g/ml$). Taken together, these results provided the in vitro evidence on the ACE, amylase and glucosidase inhibitory actions of E. ulmoides bark that form the pharmacological basis for its antihypertensive and antidiabetic action.
