Antimutagenic Effects of Juices from Edible Korean Wild Herbs

  • The mutagenic and antimutagenic activities of juices from 20 common edible wild herbs found in Korea were investigated using the spore-rec assay and Ames test. The juices of Hemerocallis fulva and Capsella bursapastoris exhibited a little induction or inhibition of mutagenesis in the presence of selected metal ions, but juices of most edible wild herbs did not affect on the mutagenesis in the spore-rec assay. In the other hand, all of the juices strongly inhibited the mutagenesis induced by benzo[a]pyrene, 2-amino-fluorene, and 3-amino-1,4-dimethyl-5H-pyridol tested on Salmonella typhimurium TA98 or TA100 in the presence of S-9 mix. The antimutagenic effects increased as the concentration of the mutagens increase. The results suggest that concentration of samples or types of various mutagen interact to affect the antimutagenic potential of the juices in the TA98 and TA100 strain.

  • The antimutagenic effects of buchu kimchi and Chinese cabbage kimchi and theri cytotoxic effects against human cancer cell line were investigated in the Salmonella typhimurium system and MTT assay, respectively. Leek and Chinese cabbage were aslo evaluated in the same system. Buchu kimchi was fermented at 15 $^{\circ}C$ for 4 days . Buchu kimchi samples showed somewhat higher antimutagenic effects against aflatoxin B1(AFB1) than CHinese cabbage kimchi in Salmonella typhimurium TA100 strain. There was no difference onthe antimutagenic activity according to the length of fermentation . Leek exerted stronger antimutagenicity against AFB1 than Chinese cabbage in the Ames assay. In MTT assay, 6-day fermented buchu kimchin revealed the highest cytotoxicity against AGS human gastric adenocarcinoma cells in which 62% and 82% of the inhibition were observed wiht the addition of 100ug, 400ug/well, respectively. Buchu kimchi samples caused 60~70% inhibition on the proliferation of HT-29 at 400ug/well. Leek exhibited higher antiproliferative effect against both AGS cells and HT-29 cells than Chinese cabbage in MTT assay. From these results, it is considered that buchu kimchi has stronger antimutagenic and in vitro anticancer effects than Chinese cabbage kimchi and the high inhibition rate of buchu kimchi probably results from leek, the major ingredient of buchu kimchi .

  • The purpose of this study is to research the antimutagenic and antioxidative effects of water dropwort and small water dropwort by Ames test and by measuring malondialdehyde(MDA) production. Water dropwort and small water dropwort were extracted with methanol and then further fractionated to hexane, chloroform, ethyl acetate, butanol and water, stepwise. The methanol extracts from both samples reduced the mutagenicities by aflatoxin $B_1(AFB_1)$ in Salmonella typhimurium TA 98. The production of MDA also decreased when the methanol extracts were added to the system. The hexane, chloroform and ethyl acetate fractions revealed higher antimutagenic activities against $AFB_1$ than the butanol and water fractions. Among the five fractions, the ethyl acetate fraction showed the highest level of antioxidant activity. From the results of the experiment, water dropwort and small water dropwort seem to be good antimutagenic and antioxidative sources of food.

Anticancer and Antimutagenic Activities after Simulated Digestion of Ethanol Extracts from White, Red and Yellow Onions

  • The beneficial effects of digested onion extracts have been assessed by antimutagenic and anticancer activities by Ames test and SRB test. The total phenolic acids and flavonoids in onion extracts were determined. Red and yellow onions contain more phenolic acids and flavonoids than those in the white onion. Digested, extracts showed antimutagenic activity and anticancer activity, and it appears that the antimutagenic activity of digested extracts of onion against mutagens and anticancer activities were related to their phenols and flavonoids contents. Moreover, the extracts inhibited the proliferation of four human tumorigenic cell lines such as HT-29 (colon), MCF-7 (breast), DU-145 (prostate) and HepG2 (liver), in a dose-dependent manner. Phenolic acids and flavonoids caused oxidative damage to the cancer cell lines and induced apoptosis. Generally, red onion extracts showed effective antimutagenic and anticancer activity, and the digested red onion extracts elicited stronger antimutagenic activity than those of the onion extracts without digestion.

  • A wide array of natural products have been identified to possess potenital cancer chemopreventive properties. Of particular interest is Polyporus umbellatus frs.. that exhibits antimutagenic and cytotoxic effects. The crud drug "chorei" prepared from the dried fruit body of Polyporus umbellatus is used for kidney and other such disease. In the study, we are isolating the minor components of Chorei and examined their antimutagenic and cytotoxic effects.(omitted)

  • The antimutagenic effects of insoluble dietary fibers(IDF) extracted from some green-yellow vegetables(kale, carrot, spinach, broccoli and soybean sprout) and soybean by binding the carcinogens of MeIQ (2-amino-3,4- dimethyl-imidazo(4,5-f) quinoline) and Trp-P-2(3-amino-1-methyl-5H-pyrido[4,3-b] indole) in Salmonella tylhimirium TA100 and TA98 were studied. All of the insoluble dietary fiber samples which binded MeIQ exhibited high antimutagenic effects by removing the mutagen. Among the samples, IDFs from kale and soybean showed strong binding capacity fo the carcinogen and revealed about 90% of the antimutagenic activity. the IDF samples showed somewhat lower binding capacity to the Trp-p-2. The lignin which extracted from kale, soybean and carrot, and the cellulose strongly removed the mutagenicity of MeIQ by the binding. Among the samples, the level of lignin in kale revealed the highest(about 10%), and it seemed that the higher content of lignin in kale is one of the reasons to increase its antimutagenic effect.

  • The antimutagenic effects of juices from green pepper(GP), red pepper leaf(RPL), red pepper(RP) and sweet pepper(SP) were examined by the Ames method using Salmonella typhimurium TA100. The juice supernatants of GP, RPL and RP showed antimutagenic activities against afltoxin B1(AFB1) in Salmonella typhimurium TA 100. The juice supermatants of GP and RPL also exhibited the inhibitory effects(p<0.05) to the mutagenicities induced by N-methyl-N'-nitro-N-nitrosoguanidine(MNNG) and 4-nitro-quinoline-1-oxide(4-NQO). The juice of RP showed antimutagenic activities against indirect mutagen of AFB1, however, the activity was reduced at higher concentration(5.0%), furthermore, as the adding concentration of sample increased to 5.0%, it exhibited slight comutagenicith on direct mutagen of MNNG. Theantimutagenic activities of GP and RPL juices were reduced significantly after heating at 100℃ for 20min, supposing that the antimutagenic compound(s) in the juices were heat labile.

Antimutagenic and Anticancer Effects of Salted Mackerel with Various Kinds of Salts (소금의 종류에 따른 염장 고등어의 항돌연변이효과 및 암세포 성장억제효과)

  • Raw meat of mackerel (Scomber japonicus) was salted by refined, sun-dried, bamboo, and KC1-added bamboo salts. Antimutagenic activity on N-methyl-N'-nitro-N-nitrosoguanidine (MNNG) in Ames test and growth inhibitory effects of AGS human gastric and HT-29 human colon adenocarcinoma cells were investigated using methanol extracts of the salted mackerels. Mackerel salted sun-dried, bamboo, and KC1-added bamboo salts used increased the antimutagenic activities against MNNG, however, the sample treated with refined salt reduced the antimutagenic activity. Inhibitory effects of the salted-mackerels on the growth of human cancer cells were increased as dose dependent pattern. Mackerel salted with refined salt activated the growth of AGS human gastric adenocarcinoma cells, but mackerel salted with sun-dried, bamboo, and KC1-added bamboo salts kept or increased anticancer effect compared to the raw mackerel. Mackerel salted with KC1-added bamboo salt led to the highest antimutagenic and anticancer activities. These results suggest that antimutagenic and anticancer effects of mackerel during manufacturing of the salted-mackerel could be enhanced by using different kind of salts such as bamboo, or KC1-added bamboo salts.

Antimutagenic and Antigenotoxic Effects of Ligularia fischeri Extracts (곰취 추출물의 항돌연변이성 및 유전독성억제효과)

  • The antimutagenic and antigenotoxic effects of ethanol, methanol, water and non-heating ethanol extract of Ligularia fischeri were investigated using Ames test and micronucleus test. Four solvent extracts by themseleves did not induce mutagenesis. The four extract of 200㎍/plate showed approximately 84.7%, 77.1%, 72.5% and 71% inhibitory effect on the mutagenesis induced by N-methyl-N'-nitro-N-nitrosoguanidine(MNNG) and 67.9%, 66.8%, 64.6% and 56% inhibition on the mutagenesis by 4-nitroquinoline-1-oxide(4NQO) against TA100 strain, whereas 70.2%, 60.9%, 61.9% and 52.8% inhibitions were observed on the mutagenesis induced by 3-amino-1,4-dimethyl-5H-pyrido[4,3-b]indol(Trp-P-1) in the presence of 200㎍/plate. TA100 strain was more sensitive than TA98 strain by four kinds of extracts on antimutagenesis. The effects of Ligularia fischeri extracts on the frequencies of micronucleated poly chromatic erythrocytes(MNPECs) induced by MNNG were investigated in the bone marrow. Ten, 20, 40 and 80mg g/kg of each extract were administered to animals immediately after injection of MNNG and the exposure time was 36 hours. Inhibitory effects of Ligularia fischeri ethanol extracts were 12%, 35.3%, 58.8%, and 57%, in the presence of 20, 40, 60 and 80mg/kg, respectively whereas methanol extracts showed 15.5%, 32.7%, 50.8%, and 57.9% inhibitory effects, respectively. Both extracts showed enhanced antimutagenic and antigenotoxic effects. These results showed a good correlation between antimutagenic effects in in vitro and in in vitro assay.

  • The antimutagenic effects of 46 kinds of medicinal plants that have been used as traditional folk antitumor agents in Korea were studied by using Ames mutagenicity test. Most of the methanolic extracts from the plants which were used in this experiment showed strong antimutagenic activity toward aflatoxin B1(AFB1) in Salmonella typhimrium TA100 and TA98. However, N-methyl-N'-nitro-N-nitrosoguanidine(MNNG) induced mutagenicity was not blocked by adding the methanolic extracts of the plants except persimmon leaves (Diospyros kaki Thunberg)and Elaeagnus umbellata.
