Antioxidant, Antimicrobial, and Cancer Cell Proliferative Inhibition Activities of Propolis

  • A commercial propolis was investigated in terms of its antioxidant, antimicrobial, and antiproliferative activities. The contents of total phenol and flavonoid of propolis were 8.3 and 6.6 mg, respectively. The reducing power of the propolis increased with concentration increasing. 1,1-Diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity was shown at 82.70% in 1,000 ${\mu}g/mL$ of the propolis. 2,2'-Azinobis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging effect of antioxidant activity on the propolis was 35.64 g/sample. The propolis showed high antimicrobial activity against Bacillus cereus at all concentration of propolis. All of the cancer cell lines have 53-73% as effective growth inhibition. These results showed that the commercial propolis has potential antioxidant, antimicrobial, and cancer cell proliferative inhibition activities thus, propolis can be applied to the functional food, pharmaceutical, and cosmetic industry.

한국산 대나무의 추출방법에 따른 항균 및 항산화 특성 (Comparison of Antimicrobial and Antioxidant Activities by Different Extraction Methods in Korean Bamboos)

  • To develop potential cosmetic ingredients with antimicrobial and antioxidant activities of 4 Korean bamboo species (P. bambusoides, P. nigra var. henonis, P. pubescens and Sasa coreana) using three different extraction methods-water, ethanol and supercritical fluid extraction. Antimicrobial activities and DPPH assay have been examined. Among the antimicrobial activities against two test strains, Escherichia coli and Staphylococcus aureus, ethanol extracts of 3 bamboo trees, P. bambusoides, P. nigra var. henonis, and P. pubescens, showed stronger than those of supercritical extracts. However, 4 bamboo supercritical extracts showed dose-dependent increase in antioxidant activity by DPPH assay. These results suggest that water fraction of bamboo extracts may be useful for the cosmetic ingredient with low cytotoxicity.

  • Nine benzoxazolin-2-thione derivatives were sunthesized as the potential antimicrobial substances. These compounds were tested for the antimicrobial activities using Staphylococcus aureus and Escherichia coli by way of tube dilution method. The three compounds of I, II and VII omjoboted the growth of S.aureus at the concentration of 1${\mu}$g/ml, and III,VI,VIII and IX exhibited considerable antimicrobial activities against S. aureus at the concentration of 10.m.u.g/ml. As to the growth of E. Coli, VII VII and VIII inhibited at the concentration of 1${\mu}$g/,l. II,III and IV exhibited considerable antimicrobial activities against E. coli at the concentration of 10${\mu}$g/ml.

Antimicrobial Activity of Korean Medicinal Plants and Herbal Formulations

  • In this study, we screened twenty four extracts of eight medicinal plants and three extracts of the commercial product for their antimicrobial and antioxidant properties. Ethanol and ethyl acetate extracts were found active where as aqueous extracts were little or no active. Extracts of Sophora flavescens, Salvia miltlorrhiza and Glycyrrhiza uralensis showed strong activity againsttested organisms and could be the potential antimicrobial agent. The increase of antimicrobial and antioxidant activities of formulations might be due to synergic effect. The results also indicated that the activity of bamboo salt and herbal products can be enhanced by making appropriate formulations.

Antimicrobial efficacy and safety analysis of zinc oxide nanoparticles against water borne pathogens

  • Metal nanoparticles have been intensively studied within the past decade. Nano-sized materials have been an important subject in basic and applied sciences. Zinc oxide nanoparticles have received considerable attention due to their unique antibacterial, antifungal, and UV filtering properties, high catalytic and photochemical activity. In this study, microbiological aspects of scale formation in PVC pipelines bacteria and fungi were isolated. In the emerging issue of increased multi-resistant properties in water borne pathogens, zinc oxide (ZnO) nanoparticle are being used increasingly as antimicrobial agents. Thus, the minimum bactericidal concentration (MBC) and minimum fungal concentration of ZnO nanoparticles towards pathogens microbe were examined in this study. The results obtained suggested that ZnO nanoparticles exhibit a good anti fungal activity than bactericidal effect towards all pathogens tested in in-vitro disc diffusion method (170 ppm, 100 ppm and 30 ppm). ZnO nanoparticles can be a potential antimicrobial agent due to its low cost of production and high effectiveness in antimicrobial properties, which may find wide applications in various industries to address safety issues. Stable ZnO nanoparticles were prepared and their shape and size distribution characterized by Dynamic light scattering (35.7 nm) and transmission electron microscopic TEM study for morphology identification (20 nm), UV-visible spectroscopy (230 nm), X-ray diffraction (FWHM of more intense peak corresponding to 101 planes located at $36.33^{\circ}$ using Scherrer's formula), FT-IR (Amines, Alcohols, Carbonyl and Nitrate ions), Zeta potential (-28.8). The antimicrobial activity of ZnO nanoparticles was investigated against Bacteria and Fungi present in drinking water PVC pipelines biofilm. In these tests, Muller Hinton agar plates were used and ZnO nanoparticles of various concentrations were supplemented in solid medium.

Antimicrobial Properties of Glass Surface Functionalized with Silver-doped Terminal-alkynyl Monolayers

  • Glass discs functionalized with alkynyl (GDA) terminated monolayers were prepared and incubated in $AgNO_3$ solution (GDA-Ag). The modified functional glass surfaces were characterized by X-ray photoelectron microscopy (XPS). The potential of GDA and GDA-Ag as antimicrobial surfaces was investigated. Anti-microbial efficacies of GDA against Escherichia coli, Pseudomonas aeruginosa, Staphylococcus aureus, Bacillus cereus, and Candida albicans was relatively low ranging from 4.67 to 17.00%. However, the GDA-Ag was very effective and its antimicrobial efficacy ranged from 99.90 to 99.99% against the same set of microbial strains except for C. albicans where it was 95.50%. The durability of the Ag bonded to the terminal alkynyl groups was studied by placing the GDA-Ag in PBS buffer solution (pH 7.4) for two weeks. Initially, the silver release was relatively fast, with 40.05 ppb of silver released in first 24 h followed by a very slow and constant release. To study the potential of GDA-Ag for medical applications, in vitro cytotoxicity of GDA-Ag against Human Embryonic Kidney 293 (HEK293) cell lines was studied using WST-assay. The cytotoxicity of the GDA-Ag was very low (5%) and was almost comparable to the control (blank glass disc) indicating that GDA-Ag has a promising potential for medical applications.

복분자 에탄올 추출물의 식중독 관련 위해 세균에 대한 항균활성 분석 (Antimicrobial Activity of the Ethanol Extract from Rubus coreanum against Microorganisms Related with Foodborne Illness)

  • This study analyzed the antibacterial activity of a Rubus coreanum (Bokbunja) ethanol extract. The antimicrobial activity was determined by disc diffusion, minimum inhibitory concentration (MIC), and growth inhibition methods with seven kinds of bacteria related to foodborne illness (Bacillus cereus, Escherichia coli, Escherichia coli O157:H7, Listeria monocytogenes, Pseudomonas aeruginosa, Staphylococcus aureus, and Salmonella typhimurium). In the results, disc diffusions of the ethanol extract from R. coreanum (9.8-17.5 mm at $4,000{\mu}g/disc$) clearly showed the antimicrobial activity of the extract against all tested microorganisms. Rubus coreanum promoted an inhibitory effect as follows: E. coli O157:H7 > P. aeruginosa > L. monocytogenes > E. coli > S. aureus > B. cereus ${\geq}$ S. typhimurium. In the MIC test, R. coreanum showed high antimicrobial effect against L. monocytogenes at 500 ppm. Moreover, the R. coreanum ethanol extract showed strong growth inhibition against microorganisms, similar to the MIC results. These results show that a R. coreanum ethanol extract has powerful antimicrobial activity against all tested microorganisms, suggesting that R. coreanum will be useful as a potential natural preservative.

Characteristics of Sophorolipid as an Antimicrobial Agent

  • Sophorolipid, a biosurfactant produced from Candida bombicola ATCC 22214, showed antimicrobial activity against Bacillus subtilis, Staphylococcus xylosus, Streptococcus mutans, and Propionibacterium acne at 4, 1, 1, 0.5 ppm, respectively. Also, 100 ppm of sophorolipid inhibited $50\%$ of cell growth of plant pathogenic fungus, Botrytis cineria. However, sophorolipid showed no effect on Escherichia coli, indicating that its selective antimicrobial activity depended on the cell wall structure. Treatment of B. subtilis with sophorolipid increased leakage of intracellular enzyme, malate dehydrogenase, indicating a possible interaction of sophorolipid with a cellular membrane. Comparing lactone-type and acid-type sophorolipids, the former showed a higher antimicrobial activity. Supplementing other surfactants showed no significant effects on the antimicrobial activity. Animal study showed that 5 g of sophorolipid per kg body weight by oral administration caused no toxicity, and sophorolipid induced no irritation on the skin. These results show potential use of sophorolipid as an active ingredient in healthcare products.

  • 관중의 근경, 감초의 뿌리, 고삼의 뿌리, 진피의 과피로 부터 에탄올로 추출물을 분리하였다. 이들 4종류의 에탄올 추출물을 이용하여 Streptococcus mutans에 대한 항균활성을 조사하였다. 그 결과 관중의 근경, 감초의 뿌리, 고삼의 뿌리는 Streptococcus mutans에 대하여 항균활성을 나타냈다. 그 중 관중과 고삼의 추출물 300ug/disk 농도는 Streptococcus mutans에 대한 뚜렷한 항균활성을 나타냈다.l 이 결과 관중의 추출물은 Streptococcus mutans에 대한 황균 활성물질로 잠재성이 있음을 알 수 있었다.

Overproduction of anthocyanin in ginseng hairy roots enhances their antioxidant, antimicrobial, and anti-elastase activities

  • Genetic engineering is a potential approach to improve secondary metabolism in plants. In order to elucidate the effect of production of anthocyanin pigment 1 (PAP1) overexpression on the bioactivity of ginseng, we analyzed its antioxidant, antimicrobial, and anti-elastase activities in this study. Our results showed that PAP1 overexpression increased the production of polyphenolic compounds including anthocyanins. The antioxidant, antimicrobial, and anti-elastase activities were stronger in anthocyanin-overproducing ginseng hairy roots (AOX) than in wild ginseng hairy roots. Using a different solvent system (0, 30, 70, and 100% (v/v) EtOH), we revealed that variations in the contents of the polyphenolic compounds were highly correlated with changes in the antioxidant and antimicrobial activities of AOX. The antioxidant, antimicrobial, and anti-elastase effects of AOX highlight genetic engineering as a powerful approach to enhance the therapeutic properties of plants. Our results show that AOX could potentially have various functional applications in the cosmetic and pharmaceutical industries.
