A Study on the Antimicrobial Activity and the Pharmacological Activities of matrial Isolated from Coptis Radix (황련(Coptis Radix)으로부터 분리된 물질의 항균효능 및 화장품 약리활성에 대한 연구)

  • This study evaluated antimicrobial efficacy and antioxidant effect of fraction isolated from Coptis Radix and confirmed its possibility as a cosmetic material. The extracts of isolated from Coptis Radix conducted an antimicrobial activity against Staphylococcus aureus, Staphylococcus epidermidis, Escherichia coli, Candida albicans by disc diffusion method and measure clear zone. As a result, it was confirmed that antimicrobial activity against S. aureus and candida. A was observed in all samples except Fr 1. The activity of 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging and The activity of 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) cation radical scavenging were determined by antioxidant assay according to the concentrations (50, 125, 250) ${\mu}g/mL$ of extracts of isolated from Coptis Radix. As a result, DPPH radical scavenging activity of Fr 1, 2, 3, 4 at $250{\mu}g/mL$ was 11.4%, 30.3%, 42.0% and 53.1%, respectively and $ABTS^+$ radical scavenging activity was 28.6%, 96.2%, 98.6% and 97.1% at the same concentration, respectively. Fr. 3 and 4 showed higher radical scavenging activity than the positive the control group BHT at the same concentration. In the WST assay results of measuring the cytotoxicity of Coptis Radix, except for Fr. 4, Fr. 1, 2 and 3 did not show toxicity. As a result, the fractions isolated from Coptis Radix can be regarded as a cosmetic material having antimicrobial activity and antioxidant ability.

Development of Antimicrobial Plant Extracts and its Application to Cosmetics (항균활성을 갖는 천연물 개발과 화장품 응용에 관한 연구)

  • This study is focused on finding new natural materials that have antimicrobial activity. We found that 3 plants extracts, Magnolia sieboldii K. Koch (M. sieboldii K. Koch), Rhus chinensis (R. chinensis) and Metasequioa glyptostroboides (M. glyptostroboides), have antimicrobial activities by disk diffusion method. We determined the Minimum Inhibitory Concentation (MIC) of each and found that 0.3 ~ 0.35 % of M. glyptostroboides essential oil and 0.35 ~ 0.4 % of M. sieboldii K. Koch extracts inhibited fungal growth and 0.45 ~ 0.5 % of R. chinensis extracts inhibited bacterial growth. We isolated compounds from extracts and verified what have antimicrobial activity. As a result we found that caryophyllene oxide and caryophyllene isolated from M. glyptostroboides, costunolide and dehydrocostus lactone from M. sieboldii K. Koch and ethyl gallate, ethyl-3-gallate from R. chinensis have antimicrobial activities. In accordance with antimicrobial activity, O/W cosmetic emulsion containing mixture of 3 plants extracts showed preservative efficacy against both bacteria and fungi. Based on the above data we suggest that extracts from M. siebodii K. Koch, R. chinensis, and M. glyptostroboides, replace chemical synthetic preservatives and be applied as a natural preservative.

Phenolic Compounds, Antimicrobial Effects and Tyrosinase Inhibition Activities of Cucumber Grown Greenhouse According to Cultivars and Growth Stages (품종별, 생육단계별 시설재배 오이의 페놀화합물 함량과 항균효과 및 Tyrosinase 억제활성)

  • The contents of total phenolics and flavonoids, antimicrobial and tyrosinase inhibition activities of five growth stages with four greenhouse-grown cucumber extracts were investigated. Total phenolic content was high in Jangjukcheongjang, Janghyeongnakhap and five growth stage(24~27 cm). The content of total flavonoid did not differ between cultivar or growth stages. Among the four cucumber cultivars, the extract of Janghyeongnakhap showed a relatively strong antimicrobial effect against Staphylococcus aureus. The inhibition zone against Staphylococcus epidermidis of the samples tested in this experiment was 8~12 mm. And the antimicrobial effects against Malassezia furfur was high in Jangjukcheongjang, and showed the highest by the inhibition zone of 14mm in three(17~20 cm) growth stage. The tyrosinase inhibition activity of cucumber extracts showed relatively high activity in Jangjukcheongjang and Sinjoeunbaekdadagi, followed by Janghyeongnakhap. From these results, we confirmed that the extract of cucumber has high antimicrobial and whitening efficacy, and that in the future, the cucumber will be increase the availability in the field of high-value cosmetic materials.

Effects of Beverage Using Herbs on the Antimicrobial, Anticancer and Antiallergy Activities (약초 음료의 항균효과, 항암효과 및 알레르기 억제효과 검증)

  • This study was conducted to investigate the effect of beverage (beverage HC and beverage PG) using herbs on antimicrobial activity, proliferation of hepatic cancer cell (Hep3B) lines and sarcoma 180 (S-180) and antiallergy, respectively. Beverage PG showed higher antimicrobial activity than beverage HC against Staphylococcus aureus and Pseudomonas aeruginosa. Beverage HC and PG showed the tumor suppressive effect in mice injected with S-180 cells. The growth-inhibitoy ratio against tumor cells were $66\%\;for\;10\%$ beverage HC, $61\%\;for\;10\%$ beverage PG. In an anti-cancer test using Hep3B cells, beverage PG showed higher anti-proliferating effect than beverage HC. Beverage PG showed growth-inhibitory effect of $69.2\%\;at\;100\%$ beverage PG. Beverage PG inhibited histamine release from rat peritoneal mast cells (RPMC) activated by compound 48/80. In conclusion, these results suggest that beverage using herbs have an antimicrobial activity, anti-proliferating effect against Hep3B cell and S-180 tumor and will be beneficial in treatment of allergic reaction.

Contemporary Update of Mouth Rinse (구강양치액의 최신 경향)

  • Introduction : Inadequate oral health control is a major risk of oral diseases. Regular home-based care is essential to maintain good oral hygiene. In particular, mouthrinses can support conventional tooth brushing in reducing accumulation of oral plaque. Effect : Antimicrobial mouthrinses are used as part of daily oral care to reduce plaque and gingivitis. Mouthrinses contains fluoride could help remineralization of enamel and dentin. The most common molecules contained in mouthrinses are chlorhexidine, essential oils, cetyl pyridinium chloride, triclosan, hyaluronic acid. Currently, chlorhexidine is the most efficacious compound, with both antiplaque and antibacterial activities. Similar results are reported for essential oils and cetyl pyridinium chloride, although with a somewhat reduced efficacy. Considering the adverse effects of chlorhexidine and its time-related characteristics, this molecule may best be indicated for acute/short term use, while essential oils and cetyl pyridinium chloride may be appropriate for long-term, maintenance treatment. Conclusion and suggestion : Antimicrobial mouthrinses are safe and effective, and when used in conjunction with brushing and flossing, they are an important method of reducing plaque and gingivitis. To improve compliance, dental health care professionals should adapt oral health care recommendations to fit patients' specific needs.

The Dyeing Properties and Functionality of Water Lily(Nymphaea tetragona) Leaves Extract as a New Natural Dye Resource(1): Dyeing of Cotton Fiber (새로운 천연염료로서 수련 잎 추출색소의 염색성과 기능성(1): 면섬유 염색을 중심으로)

  • In this study, the efficacy of water lily(Nymphaea tetragona) leaves as a new natural dye resource was investigated. For this purpose, the colorants from water lily leaves were extracted in methanol, evaporated, and powdered. Dyeing onto cotton fiber was carried out to study the effects of dyeing conditions, mordant type and mordanting method on dye uptake, color change, and colorfastness. FTIR analysis supported that hydrolyzable tannins and chrolophyll were contained in the extracted colorants. The colorants showed good affinity to cotton fiber showing Y Munsell color. Pre-mordanting method gave better results in terms of dye uptake than post-mordanting method. By mordanting, dyed fabrics exhibited various colors such as green, khaki, brownish yellow, dark brown, dark gray and so on. Colorfastness to washing and rubbing was relatively good showing 4-5 rating. The light fastness was improved 1-2 rating by Fe mordanting. The dyed cotton fabric showed antimicrobial activity.

Inhibitory Activity against Helicobacter pylori of Isolated Compounds from Pinus koraiensis Siebold et Zucc Leaves

  • A phenol substance was extracted from Pinus koraiensis Siebold et Zucc leaf extracts and its biological efficacy was measured. The highest content of the phenol substance contained in Pinus koraiensis Siebold et Zucc leaves was 13.5 mg/g, which was obtained when it was extracted with 80% ethanol. At a concentration of 200 mg/mL, the phenolic substances extracted with 80% ethanol and water showed antimicrobial activities against Helicobacter pylori, producing clear zones of 10 and 12 mm diameter, respectively. Pinus koraiensis Siebold et Zucc. leaf extracts were separated using a Sephadex LH-20 column and 4 fractions were obtained (fractions A-D). Fractions C and D showed the greatest inhibitory activity against Helicobacter pylori producing 10.1 and 12.3 mm clear zones, respectively. These two fractions were purified using a Sephadex LH-20 and MCI-gel column ($H_2O{\rightarrow}100%$ ethanol). Purified compounds A and B were identified as syringic acid and compound C was identified as p-coumaric acid based on $^1H$-nuclear magnetic resonance (NMR), $^{13}C$-NMR, and fast atom bombardment mass spectrometry spectra. When two or more purified compounds were mixed, a synergistic effect of anti-Helicobacter pylori activity was evident. This result indicates that extracts of Pinus koraiensis Siebold et Zucc leaves could be considered a functional food because of their high antimicrobial properties.

Lettuce leaves에 접종된 Foodborne Pathogen에 대한 Aerosolized Peroxyacetic Acid의 살균 효과

  • As an alternative to gaseous sanitizer which having penetrating and diffusing capacities, aerosolsanitizer’s effect on killing pathogens was investigated. To test the efficacy of aerosolized sanitizer, peroxyacetic acid was aerosolized($5.42-11.42\mum$) by nebulizer(Royal-G Enterprise, ShenZhen,China) in a model cabinet against artificially inoculated target microorganisms on lettuce. Lettuceleaves were inoculated with a cocktail of three strains each of Escherichia coli O157:H7, Listeriamonocytogenes, and Salmonella Typhimurium and treated with sanitizer aerosol for 10 min, 30 and60 min in a model aerosol cabinet at room temperature($22\pm2^\circC$). After treatment, surviving cellsincluding injured cells were enumerated on appropriate selective agar or using the over-lay agar method, respectively. Inoculated lettuce leaves exposed to antimicrobial aerosol for 10 min experienceda 0.8 log reduction in E. coli O157:H7, a 0.3 log reduction in Salmonella Typhimurium and a 2.5 logreduction in L. monocytogenes when compared to the control. After 30 min treatment, the threepathogens were reduced in number of CFU by 2.2, 3.3, and 2.7-log and after 60 min, the reductions were 3.4, 4.5, and 3.8-log, respectively. Aerosolization can be new antimicrobial agents deliverysystem in food sanitizing.

ANTIMICROBIAL EFFECT OF ROOT CANAL CEMENTS ON MICROORGANISMS FROM INFECTED ROOT CANALS (수종 근관 충전재의 항균 효과에 관한 연구)

  • The purpose of this study was to evaluate the antimicrobial effect of root canal cements such as AH-26, Sealapex, Canals and Apatite root sealers(Type I, II, III) and to determine the efficacy of their activities. S.mutans(AHT), S.sanguis(NCTC 9811) and B.gingivalis(381) were streaked on blood agar and the PVC tybes filled with root canal cements were applied on. Then the microorganisms were cultured for 48 hours, anaerobically. B.cereus(KCTC 1012) was streaked on nutrient agar, PVC tubes were applied on and were cultured for 48 hours, aerobically. The inhibition zones of root canal cements were measured with vernier caliper. The data statistically analyzed, and the results were as followed. 1. Apatite root sealers(Type I, II, III) showed no inhibition zones. 2. AH-26, Sealapex and Canals had inhibition zones with varying degrees. The inhibition zone of AH-26 was greatest and followed by Canals and Sealapex(P<0.01). 3. As time goes by after mixing the root canal cements, AH-26, Canals and Sealapex showed significantly reducing inhibition zones(P<0.01). 4. There were the least inhibition zones of all the root canal cements on S.mutans and followed in such order as; B.gingivalis, S.sanguis and B.cereus(P<0.01).

Synthesis of Azole-containing Piperazine Derivatives and Evaluation of their Antibacterial, Antifungal and Cytotoxic Activities

  • A series of azole-containing piperazine derivatives have been designed and synthesized. The obtained compounds were investigated in vitro for their antibacterial, antifungal and cytotoxic activities. The preliminary results showed that most compounds exhibited moderate to significant antibacterial and antifungal activities in vitro. 1-(4-((4-chlorophenyl) (phenyl)methyl)piperazin-1-yl)-2-(1H-imidazol-1-yl)ethanone and 1-(4-((4-Chlorophenyl)(phenyl)methyl)piperazin-1-yl)-2-(2-phenyl-1H-imidazol-1-yl)ethanone gave remarkable and broad-spectrum antimicrobial efficacy against all tested strains with MIC values ranging from 3.1 to $25\;{\mu}g/mL$, and exhibited comparable activities to the standard drugs chloramphenicol and fluconazole in clinic. Moreover, 2-((4-((4-chlorophenyl)(phenyl)methyl)piperazin-1-yl)methyl)-1H-benzo[d]imidazole was found to be the most effective in vitro against the PC-3 cell line, reaching growth inhibition values (36.4, 60.1 and 76.5%) for each tested concentration: $25\;{\mu}g/mL$, $50\;{\mu}g/mL$ and $100\;{\mu}g/mL$ in dose-dependent manner. The results also showed that the azole ring had noticeable effect on their antimicrobial and cytotoxic activities, and imidazole and benzimidazole moiety were much more favourable to biological activity than 1,2,4-triazole.
