감태 (Ecklonia cava) 추출물의 항고혈압 효과 (Antihypertensive Effect of Ecklonia cava Extract)

  • Ecklonia cava is brown alga which ow in the sea all over the world. It has been shown that Ecklonia cava has antioxidative, anticoagulant, and antiviral effect, lately. In this study, we examined the antihypertensiye effect of Ecklonia cava extract (Eca) on 2-kidney 1-clip Goldblatt Hypertensive Rats (GB-HT). In vitro assay, the angiotensin converting enzyme (ACE) activity was significantly inhibited by Eca (27-95% inhibition at $10-50\;{\mu}g/ml$). The systolic blood pressure of Eca-treated GH-HT significantly was decreased compared to that of control CB-HT (15% decrease at 50 mg/kg). The serum ACE activity was also inhibited 19% with 50 mg/kg Eca treatment. These results suggest that Eca can be used as antihypertensive functional foods and nutraceuticals.

  • Effect of BR-900317 on the angiotensin I-induced pressor response in pithed rats and the effects of its single oral administration on plasma angiotensin converting enzyme (ACE) activities in normotensive rats and on the cardiovascular system in hypertensive model rats (SHR, RHR), were compared with those of captopril. BR-900317 attenuated the angiotensin I-induced pressor effects in pithed rats. In a single oral dose administration study, BR-900317 inhibited the plasma ACE activities in a dose-dependent fashion. Duration of the action of BR-900317 was similar to that of captopril. BR-900317 produced antihypertensive effect in spontaneously hypertensive rats and dose-dependent antihypertensive effect in 2-kidney Goldblatt hypertensive rats without affecting heart rate. These results suggest that the main mechanism of the antihypertensive effect of BR-900317 is the suppression of angiotensin II production due to the inhibition of the ACE.

Production and Characterization of Antihypertensive Angiotensin I-Converting Enzyme Inhibitor from Pholiota adiposa

  • Angiotensin I-converting enzyme (ACE) inhibitors have generally been very useful to remedy or prevent hypertension. This study describes the extraction and characterization of an ACE inhibitor from the fruiting body of Pholiota adiposa ASI 24012, which can be used as an antihypertensive drug. The maximal ACE inhibitory activity $(IC_{50};0.25mg)$ was obtained when the fruiting body of Pholiota adiposa ASI 24012 was extracted with distilled water at $30^{\circ}C$ for 12 h. After the purification of ACE inhibitor with ultrafiltration, Sephadex G-25 column chromatography, and reverse-phase HPLC, an active fraction with an $IC_{50}$ of 0.044 mg was obtained. The purified ACE inhibitory peptide was a novel pentapeptide, showing very little similarity to other ACE inhibitory peptide sequences. The molecular mass of the purified ACE inhibitor was estimated to be 414 daltons with a sequence of Gly-Glu-Gly-Gly-Pro, and showed a clear antihypertensive effect on spontaneously hypertensive rats (SHR) at a dosage of 1 mg/kg.

수산폐기물인 미더덕껍질 용매 추출물의 항산화 및 항고혈압 활성 (Antioxidant and Antihypertensive Activities of Solvent Extract from Styela clava Tunic, Fishery Waste)

  • Styela clava tunic is generated in large amounts as a waste from S. clava processing plants and leads to environmental problems. It destroys the beach scenery and causes a bad smell and pollution by trashing on the seashore. Therefore, purpose of this study was to investigate antioxidant and antihypertensive activities of different solvent extracts from S. clava tunic for recycling of fishery waste. Antioxidant and antihypertensive activities of all extracts were concentration-dependent. Of extracts, hot water extract showed the highest DPPH radical scavenging activity with the lowest effective concentration ($EC_{50}$) value (0.733 mg/ml). Chloroform extract exhibited the highest metal chelation activity with the lowest $EC_{50}$ value (2.696 mg/ml). Autoclaved water extract showed the highest NO radical scavenging activity with the lowest $EC_{50}$ value (0.491 mg/ml) and n-hexane extract showed the highest reducing power ($A_{700}=1.897$ at 100 mg/ml). And n-butanol extract showed the highest SOD-like activity with the lowest $EC_{50}$ value (19.116 mg/ml) and ACE inhibition activity with the lowest inhibitory concentration($IC_{50}$) value (0.149 mg/ml). These results indicate that extracts obtained from S. clava tunic may potential candidate to reduce diseases caused by various oxidative stresses and hypertension.

한국산 영지의 혈압 강하 성분에 관한 연구 (Studies on Antihypertensive Components of Ganoderma lucidum in Korea)

  • 한국에서 재배된 영지의 혈압 강하 작용을 검색한 결과, 두 종류의 영지 자실체에서 모두, 용량을 증가시킴에 따라 심박수, 수축기 혈압 및 확장기 혈압을 강하시킴을 관찰하였다. 또, 이러한 혈압을 강하시키는 유효 성분은 fraction B에 많이 포함되어 있음을 확인할 수 있었는데, 선천성 고혈압 쥐에 체중당 10mg을 정맥내 투여했을 때, 대조군과 비교하여 유의한 차이를 가지고 혈압을 강하시켰다. 혈액 관류 심장 표본에서의 영지의 작용을 관찰해 본 결과, sample J의 경우는 용량에 따라 심박수 및 심실 수축력을 증가시켰으나 sample K의 경우 용량을 증가시킴에 따라 심박수 및 심방 수축력을 오히려 감소시켰으며, 심실 수축력은 거의 변화가 없었고, 또, 관상동맥 혈류량을 용량의존적으로 증가시켰다. 심장에 대한 위의 모든 반응은 fraction A와 B에서만 나타났고, fraction C는 영향을 끼치지 못했다.

Cinnarizine을 Propranolol이나 Metoprolol과 병용할 때의 혈압(血壓) 강하(降下) 효과(效果)에 관한 약리학적(藥理學的) 연구(硏究)(II) -적출(摘出) 평활근(平滑筋)에 대한 효과- (Pharmacological Studies on the Antihypertensive Effects of Cinnarizine Coadministered with Propranolol or Metoprolol(II) -Effects on the Isolated Smooth Muscle-)

  • In our former report we observed that cinnarizine influenced the antihypertensive effect of propranolol beneficially, but not of metoprolol in SHR and normal cat. Cardiac contractilities and smooth muscle relaxations induced by above drugs were measured to elucidate their mechanism of action. In cinnarizine and propranolol treated group, both of negative inotropic and ${\beta}-blocking$ activity of propranolol in perfused rat hearts were increased and propranolol induced contraction in isolated arterial and trachea smooth muscle of the guinea pig was antagonized comparing to propranolol alone treated group. However, in the cinnarizine and metoprolol treated group, no significant differences in activity on the above were observed compared to metoprolol alone treated group.

각종 미강 추출물들의 항고혈압성 엔지오텐신 전환효소 저해 활성 (Antihypertensive Angiotensin I-Converting Enzyme Inhibitiory Activity of Various Extracts from Some Rice Brans)

  • 각종 미강 추출물들의 항고혈압성 엔지오텐신 전환효소 저해 활성을 측정하고 이들의 추출 최적조건을 검토하였다. 각종 미강 추출물들중에서 일품벼 미강의 물 추출물이 77%의 가장 높은 안지오텐신 전환효소 저해활성 보였다. 일품벼 미강중의 안지오텐신 전환효소 저해물질은 물을 1:10(w/v)으로 미강 분말에 첨가한 후 $30^{\circ}C$에서 12시간 추출하였을 때 가장 많이 용출되었다.

Biological activity of peptides purified from fish skin hydrolysates

  • Fish skin waste accounts for part of the solid waste generated from seafood processing. Utilization of fish skin by bioconversion into high-grade products would potentially reduce pollution and economic cost associated with treating fish processing waste. Fish skin is an abundant supply of gelatin and collagen which can be hydrolyzed to produce bioactive peptides of 2-20 amino acid sequences. Bioactivity of peptides purified from fish skin includes a range of activities such as antihypertensive, anti-oxidative, antimicrobial, neuroprotection, antihyperglycemic, and anti-aging. Fish skin acts as a physical barrier and chemical barrier through antimicrobial peptide innate immune action and other functional peptides. Small peptides have been demonstrated to possess biological activities which are based on their amino acid composition and sequence. Fish skin-derived peptides contain a high content of hydrophobic amino acids which contribute to the antioxidant and angiotensin-converting enzyme inhibitory activity. The peptide-specific composition and sequence discussed in this review can be potentially utilized in the development of pharmaceutical and nutraceutical products.

배 첨가가 막걸리의 품질과 생리 기능성에 미치는 영향 (Effect of Pears on the Quality and Physiological Functionality of Makgeoly)

  • This study was conducted to develop a high value pear Makgeoly that possessed the physiological functionality of pears and rice. To accomplish this, the effects of the addition of pears on the alcohol fermentation of Korean traditional rice wine, Makgeoly were investigated. The total acidity contents were unchanged for 9 days of fermentation after the addition of pear to the mash, however, it decreased by approximately 0.13% after 9 days. Additionally, the ethanol contents increased to 16.0% after 5 days of fermentation, after which they did not change significantly. The residual reducing sugar and free sugar content were decreased after 7 days and 5 days of fermentation, respectively. The total acceptability was the best in the Makgeoly that contained grated pear after 10 days of fermentation. Additionally, antihypertensive angiotensin Iconverting enzyme inhibitory activity was found to be greater than 80% in all pear Makgeoly, but the fibrinolytic activity, SOD-like activity and antioxidant activity were weakened. Based on these results, Makgeoly that was brewed by the addition of 20% grated pear into the fermented broth from fermentation at $20^{\circ}$ for 10 days has the potential to become a new functional Korean traditional Makgeoly with high acceptability and antihypertensive properties.
