Invasive Candida infections (ICI) have become the third most common cause of late-onset infection among premature infants in the neonatal intensive care unit (NICU). Risk factors include birth weight less than 1,000 g, exposure to more than two antimicrobials, third generation cephalosporin exposure, parenteral nutrition including lipid emulsion, central venous catheter, and abdominal surgery. Candida colonization of the skin and gastrointestinal tract is an important first step in the pathogenesis of invasive disease. Strict infection control measures against the infection should be done in the NICU. The following practices are likely to contribute to reducing the rate of ICI: (1) restriction of broad-spectrum antibiotics, antacids and steroid; (2) introduction of early feeding and promoting breast milk. Fluconazole prophylaxis may be an effective control measure to prevent Candida colonization and infections in individual units with high incidence of fungal infection. In addition, there is a need of further data including the development of resistant strains and the effect on long-term neurodevelopmental outcomes of infants exposed to drugs before the initiation of routine application of antifungal prophylaxis in the NICU.
The polyene antibiotics including nystatin, pimaricin, amphotericin and candicidin are a family of most promising antifungal polyketide compounds, typically produced by rare actinomycetes species. The biosynthetic gene clusters for these polyenes have been previously investigated, revealing the presence of highly homologous biosynthetic genes among polyene-producers such as polyketide synthase (PKS) and cytochrome P450 hydroxylase (CYP) genes. Based on amino acid sequence alignment among actinomycetes CYP genes, the highly-conserved regions specific for only polyene CYP genes were identified and chosen for degenerate PCR primers, followed by the PCR-screening with various actinomycetes genomic DNAs. Among tested several polyene non-producing actinomycetes strains, Pseudonorcardia autotrophica strain was selected based on the presence of PCR product with polyene-specific CYP gene primers, and then confirmed to contain a cryptic novel polyene hydroxylase gene in the chromosome. These results suggest that the polyene-specific hydroxylase gene PCR should be an efficient way of screening and isolating potentially-valuable cryptic polyene antibiotic biosynthetic genes from various microorganisms including rare actinomycetes.
Pulmonary aspergillosis is a relatively rare disease in Korea. Immune suppressive patients, resulting from misuse or long term therapy of antibiotics, steroid and anticancer chemotherapeutics tend to be vulnerable to pulmonary aspergillosis. This study is made to illustrate the clinical features, preoperative diagnosis and surgical role in the management is this diseases. In retrospective study of operative cases from May 1980 through July 1986, 10 cases were analysis. Hemoptysis and blood tinged sputum were the most common chief complaints. Major underlying pathology was cavitary lesion or bronchiectasis caused by pulmonary tuberculosis [5 cases]. 2 cases were disseminated form due to immune suppression. Anatomic location of lesion was in the upper lobe in half case. Systemic antifungal agent is helpful for invasive aspergillosis and Resection is the treatment of choice for localized fungus balls and symptomatic localized forms.
Peptide synthetases produced from various microorganisms are multifunctional enzyme complexes and their substrates are recognized and activated by adenylation domains. To identify the substrate specificity of the peptide synthetase isolated from Bacillus subtilis 713, known to produce an antifungal peptide, two adenylation domains containing the minimal functional portion were expressed and purified. ATP-ppi exchange experiments and kinetic studies revealed that the two adenylation enzymes had a substrate specificity to $_{L}-lysine{\;}and{\;}_{L}-tryptophan$, respectively. In addition, based on a signature sequence comparison, the substrate of the third domain was predicted to be L-glutamic acid. These results suggest that this peptide synthetase is novel because there has been no previous report on a peptide synthetase that uses $_{L}-lysine,{\;}_{L}-tryptophan,{\;}and{\;}_{L}-glutamic$ acid as substrates in that order.
A 1-year-old castrated mixed-breed dog presented with diffuse, purple lesions arranged in an irregular patchy pattern, with a slight elevation on the right hindlimb extending from the tarsus joint to the upper region of the thigh. Dermatological examinations and fungal and bacterial cultures revealed no infectious agents. The therapeutic response to antibiotics and antifungal agents was negative. A histopathology examination of the lesion revealed vascular proliferation with vasodilation and numerous varying-sized vessels. Mast-cell-dominated perivascular cuffing was also noted. The dog was diagnosed with cutaneous angiomatosis due to diffuse lesions and the histopathology findings of hemangioma.

The Polyoxin complex is an antifungal antibiotics produced by Streptomyces cacaoi var. asoensis that exhibit marked and selective activity against pytopathogenic fungi. They incorporate carbamoylated dipeptides attached to the sugar moiety. Controlled alkaline hydrolysis of polyoxins result in several products. one of which has been identified as (+)-(2S. 3S, 4S)-2-amino-3. 4. 5-trihydroxypentonic acid(polyoxamic acid). 원목과 톱밥배지에서 푸른곰팡이의 오염과 성장은 표고버섯의 생장을 심하게 저해할 수 있다. 표고버섯 재배에서 푸른곰팡이병의 방제를 위하여 농약과 항생제와 같은 화학 약품의 사용은 일반적으로 허용되지 않는다. 본 연구에서는 푸른곰팡이병의 방제를 위하여 느타리버섯에서 갈반병을 일으키는 세균성 병원균을 분리하고 이들의 펩티드 독소를 분리하였다. Pseudomonas tolaasii 균주들은 항진균 활성을 갖는 톨라신 및 이와 구조적 유사체인 다양한 펩티드 독소를 분비한다. 이러한 펩티드들은 표고버섯의 생장에는 영향을 주지 않는다는 것이 알려져 있다. 펩티드 독소들을 Trichoderma harzianum H1 푸른곰팡이에 처리하였을 때, 푸른곰팡이의 성장을 저해하였다. 펩티드를 분비하는 20 종의 P. tolaasii 균주 중에서 강, 중, 약의 항진균 활성을 가진 균주의 수는 각각 8, 5, 7 종으로 나타났다. 표고버섯을 재배하는 동안에, 펩티드 독소를 함유한 세균 배양액에서 세균을 제거한 배양원액을 톱밥배지 표면에 자란 푸른곰팡이 균사에 분무하였다. 배양원액은 푸른곰팡이의 성장은 억제할 수 있는 반면, 표고버섯 생장과 생산에는 영향을 주지 않았다. 펩티드 독소를 함유하는 배양원액은 곰팡이의 흰색 균사체를 노란색의 마른 딱지로 변화시켰고, 이것은 펩티드 독소가 강력한 항진균 활성과 살균 활성을 갖고 있음을 보여준다.
여러 토양에서 분리한 400여 개의 방선균 균주에 대해 4가지 식물병원성 진균에 대한 항진균 활성을 조사하였으며 그 중 Streptomyces costaricanus HR391 균주는 PDB 배지에서 식물병원성 진균인 Fusarium oxysporum f. sp. raphani, F. oxysporum f. sp. niveum, F. oxysporum f. sp. lycopersici와 Rhizoctonia solani의 균사 생장을대조군과 비교하여 각각 26.5, 26.2, 21.2와 23.8% 저해하였다. S. costaricanus HR391은 항진균물질인 siderophore를 $98{\mu}M$을 생성할 뿐만 아니라 유화활성을 나타내며 막지질을 파괴할 수 있는 생물계면활성제인 rhamnolipid와 lipopeptide인 iturin A와 surfactin를 생성하였다. 또한 진균 세포막을 분해할 수 있는 chitinase와 glucanase 활성도 나타내었으며 병원균의 막을 파괴하는 AMP와 항생물질 phenazine도 분비하였다. 이외에도 식물생장 촉진활성을 갖는 zeatin, gibberellin과 indole acetic acid 같은 식물호르몬도 생성하였다. 이와 같이 항진균 활성을 나타내는 S. costaricanus HR391 균주는 다양한 종류의 항진균 물질의 상승작용과 더불어 높은 생물계면활성이 본 균주의 항진균 활성에 큰 역할을 하는 것으로 보이며 친환경적인 생물학적 항진균제로서 활용이 가능할 것으로 기대된다.
Allergic bronchopulmonary aspergillosis (ABPA) is a hypersensitivity lung disease due to bronchial colonization of Aspergillus fumigatus that occurs in susceptible patients with asthma or cystic fibrosis. A 10-year-old girl was referred to the Department of Pediatric Pulmonology for persistent consolidations on chest radiography. Pulmonary consolidations were observed in the right upper and left lower lobes and were not resolved with a 4-week prescription of broad-spectrum antibiotics. The patient had a history of atopic dermatitis and allergic rhinitis but no history of asthma. She had no fever but produced thick and greenish sputum. Her breathing sounds were clear. On laboratory testing, her total blood eosinophil count was $1,412/mm^3$ and total serum IgE level was 2,200 kU/L. Aspergillus was isolated in the sputum culture. The A. fumigatus-specific IgE level was 15.4 kU/L, and the Aspergillus antibody test was also positive. A chest computed tomography scan demonstrated bronchial wall thickening and consolidation without bronchiectasis. An antifungal agent was added but resulted in no improvement of pulmonary consolidations after 3 weeks. Pulmonary function test was normal. Methacholine provocation test was performed, revealing bronchial hyperreactivity ($PC_{20}=5.31mg/mL$). Although the patient had no history of asthma or bronchiectasis, ABPA-seropositivity was suspected. Oral prednisolone (1 mg/kg/day) combined with antifungal therapy was started. Pulmonary consolidations began decreasing after 1 week of treatment and completely resolved after 1 month. This is the first observed and treated case of seropositive ABPA in Korean children without previously documented asthma.
