Anticariogenic Effects of Unripe Apple Extract (애사과 추출물의 충치억제효과)

  • Anticariogenic activity of the unripe apple extract was studied by observing the inhibitory effects on GTase(glucosyltransferase) activity, cell adherence and acid production of Streptococcus mutans. Among the four S. mutans strains, S. mutans MT 8148 had the highest water-insoluble glucan forming activity. (+)-Catechin and tannic acid, the major components of the unripe apple polyphenols inhibited GTase activity by 60% at 1 mg/ml and 90% at 5 mg/ml. Tannic acid and unripe apple extract inhibited adherence ability of S. mutans by 50% and 30%, respectively. But the acid production of S. mutans was not influenced by the polyphenols. Disc diffusion test showed that the polyphenols have no antimicrobial activity against S. mutans, which indicates that the inhibition of GTase activity and cell adherence were not resulted from the cell growth inhibition. Our results convinced the possible application of the unripe apple extract as the anticariogenic food additives.

  • The methanolic extract of Commelina communis (aerial part) showed antibacterial activity against a cariogenic bacterium, Streptococcus mutans OMZ 176. The active principles were identified to be $\beta$-carboline alkaloids, 1-carbomethoxy-$\beta$-carboline, norharman and harman, which were bactericidal in the minimal inhibitory concentration (MIC) of $100\;\mu{g/ml}$ against the strain.

  • Natural extracts were isolated from 20 kinds of medicinal plants that have been traditionally widely used in Korea to develop treatments for dental caries. Medicinal plants showing antibacterial activity against a cariogenic bacterium, Streptococcus sobrinus KCOM 1157 were investigated. The natural extracts of Glycyrrhiza uralensis, Sophora flavescens, Coptis Chinensis, Mentha arvensis, Pinus densiflora, Spirodela polyrhiza showed antimicrobial activities against S. sobrinus KCOM 1157. The extracts of Pinus densiflora, Sophora flavescens, Coptis Chinensis among these medical herbs showed significant antimicrobial activity. These results suggested that the natural products of Pinus densiflora, Sophora flavescens, Coptis Chinensis could be the potential source of anticariogenic agent against dental pathogens including S. sobrinus.

Anticariogenic Activity and Glucosyltransferase Inhibition of Phenolic Compounds (페놀성 화합물의 항충치활성 및 Glucosyltransferase 억제효과)

  • Fourteen phenolic compounds(benzoic acid, p-hydroxybenzoic acid, protocatechuic acid, vanillic acid, syringic acid, gallic acid, caffeic acid, ferulic acid, (+)-catechin, quercetin, rutin, catechol, chlorogenic acid and L-ascorbic acid) were examined for their effects on the anticarigenic activity. Among tested samples, catechol was significantly inhibited the S. mutans, exhibiting an clear zone 18.5-19.5mm by 10 mg/disc level. The minimal inhibition concentration(MIC) of the phenolic compounds for Streptococcus mutans, M1 and M2 strain were determined as 2,000 ppm, whereas catechol was 1,000 ppm. The activity of glucosyltransferase(GTase) was significantly inhibited by catechol, at 10 ppm(58.7%), 50 ppm(60.7%) and 100 ppm(88.4%) and 500 ppm(89.6%), respectively. Among them, catechol showed most significant anticariogenic activity as well as inhibition of GTase activity.

Anticariogenic effects of Coptis chinensis Franch Extract (황련(Coptis chinensis Franch) 추출물의 항충치효과)

  • The effects of Coptis chinensis Franch(CCF) extract on the growth, acid production, cell adherence and glucosyltransferase(GTase) activity of Streptococcus mutans JC-2 were investigated. Methanol extract from CCF showed a strong inhibitory effect on the growth of S. mutans. The minimal inhibition concentration(MIC) of the methanol extract was determined as $130\;{\mu}g/mL$, whereas MIC of water extract was $200\;{\mu}g/mL$. MICs of berberine and palmatine were $50\;{\mu}g/mL$ and $110\;{\mu}g/mL$, respectively, showing stronger antimicrobial activities than the extracts of CCF. Antimicrobial activities of methanol extract, berberine and palmatine were not decreased by heating at $121^{\circ}C$ for 15 min, suggesting that the antimicrobial components including berberine and palmatine are heat-stable. Acid production of S. mutans was decreased by methanol and water extracts, berberine and palmatine. The activity of GTase was inhibited by methanol extract, berberine and palmatine at $300\;{\mu}g/mL$ with 23.2%, 46.1% and 17.1%, respectively, but was not inhibited by water extract. The water extract and palmatine at sub-MICs inhibited the adherence of S. mutans to glass surface by 59.2% and 41.7%, respectively. These results suggest that CCF extracts have anticariogenic effects and could be used as an anticariogenic food additive.

Development of Fermented Isotonic Beverage with Anticariogenic Activity using Bacteriocin-Producing Lactic Acid Bacteria (박테리오신 생성 젖산균을 이용한 항충치 활성을 지닌 발효이온음료 개발)

  • The fermented fruit and vegetable mixed broth was prepared by using bacteriocin-producing lactic acid bacteria to evaluate the possibility of developing isotonic beverage with anticariogenic activity. Optimum conditions were also established to produce bacteriocin by a mixed culture system consisting of Lc. lactis and Leu. mesenteroides in a fruit and vegetable mixture. Production of bacteriocin was not observed when both strains were simultaneously innoculated, but pH adjustment of the broth fermented by Leu. mesenteroides to nearneutral pH stimulated the production of bacteriocin by Lc. lactis. The concentration of sodium of the fermented broth was higher than those of commercial products. Color change of the fermented broth was not observed during storage. The fermented broth showed strong inhibitory effect against Streptococcus mutans which is an oral inhabitant with a cariogenic activity. Bacteriocin activity in the fermented broth was retained very stable for 4 weeks at 4$\^{C}$. The results indicated that bacteriocin-producing lactic acid bacteria can be used for the preparation of a Korean style thirst-quenching beverage containing bacteriocin.

  • The main purpose of this study was to compare the anticariogenecity of glass ionomer restorative material polymerized by argon laser versus visible light, The results from the present study can be summarized as follows; 1. Under the polarized light microscope, the specimens of laser-cured group showed the shallower lesion body than that of visible-light cured group, both in the stage of lesion initiation and progression. 2. Glass ionomer material cured by visible light showed shallower body of lesion than that of composite resin cured by argon laser at the stage of lesion progression(p<0.05). It was suggested fluoride released from the glass ionomer might have the additive anticariogenic effect. 3. Statistical difference between groups on depth of lesion body was evident after lesion progression (p<0.05). It was suggested that anticariogenic effect by argon laser was more effective at the stage of lesion progression than the lesion initiation. 4. The increment of lesion body during progression was highest in group IV (p<0.05). 5. Based upon the above-mentioned results of this study, it can be concluded that the advantage of anticariogenic effect and short curing time of argon laser in glass ionomer polymerization should be considered in children and adolescents whose caries activity is relatively higher.

  • The occurrence of dental caries is mainly associated with oral pathogens, especially cariogenic Streptococcus mutans. The aim of this study was to isolate and characterize lactic acid bacterium showing inhibitory activity against cariogenic Streptococcus mutans. As results, an isolate with strong inhibitory activity was obtained from Kimchi and it was identified as Lactobacillus sakei by API and 16S rRNA gene analyses. This strain secreted an inhibitory compound in cell growth medium and the activity of the compound was completely disappeared by proteinase K revealing the fact that the compound is proteinous substance, bacteriocin. Optimal culture condition for bacteriocin production by Lb. sakei K-7 was at pH 7.5 and $37^{\circ}C$ for 18 h. Oral administration of this isolate may give anticariogenic and probiotic effects on hosts.

  • For the purpose of developing of anticariogenic agents, P-phenylphenol derivatives were synthesized and determined their antibacterial activities against a cariogenic bacterium, Streptococcus mutans. Among synthetic compounds, 2-nitro-6-bromo-p-phenylphenol showed as potent antibacterial activity as magnolol and honokiol.

  • In a continuous study for the developing of the anticariogenic agents, 3,3'-diacyl-4,4'-biphenol derivatives (Fig. 2, 2 and 3) and 3,3'-bis-(1-hydroxyalkyl)-4,4'-biphenol (Fig. 3, 4 and 5) derivatives are synthesized successively from 4,4'-diphenol (Fig. 2, 1). The synthesized compounds are tested for their antibacterial activity against a cariogenic bacterium, Streptococcus mutans OMZ 176. The acyl derivatives, 2 and 3, do not show antibacterial activity, but the hydroxyalkyl derivatives, 4 and 5, reduced from the acyl group of 2 and 3, show the activity. The antibacterial activity of 2 and 3 may be inhibited due to intramolecular hydrogen bonding between the acyl group and the hydroxyl one (Fig. 4).
