부추 추출 성분의 항발암 효과 연구 (Anticarcinogenic Effects of Allium tuberosum on Human Cancer Cells)

  • 천연물을 이용하여 생리활성이 있는 성분을 추출 분리하여 그 유효 성분을 탐색함으로써 질병 예방 및 치료제로 이용하고자 하는 연구가 최근 활발히 진행되고 있다. 본 연구에서는 예부터 혈액 순환을 돕고, 지혈 작용에 효과가 있는 것으로 알려져 있는 식탁에 상용되는 야채인 부추를 이용하여 4종의 인체 암세포주인 HepG2, HeLa, MCF7 및 SK-N-MC를 이용하여 암세포 증식억제 및 암예방 QR 유도활성 효과를 측정하였다. MTT assay를 이용한 암세포 증식억제 실험 결과, 4종의 암세포주 모두 ethylether층인 ATMEE에서 아주 높은 농도 의존적이 암세포 증식억제 효과를 보였다. 즉 본 실험에 사용된 인체 암세포주 4종에서 시료의 농도가 $150\;{\mu}g/mL$일 때 각 세포주 모두 약 90% 이상의 높은 암세포 증식억제 효과가 나타났으며, 특히 신경아종세포주인 SK-N-MC 에서는 ethylether층의 시료 농도를 다른 층을 첨가시의 농도보다 적은 $100\;{\mu}g/mL$ 첨가에서도 이미 99.8%의 아주 높은 암세포 증식억제 효과를 나타내었다. 또 신경아종세포인 SK-N-MC에서는 ethylacetate층인 ATMEA의 경우에서도 최종 첨가 시료농도를 $200\;{\mu}g/mL$ 첨가했을 때 이미 91%의 높은 암세포 증식억제 효과가 나타났다. 암예방 quinone reductase(QR) 유도활성을 측정한 결과, 시료의 butanol 분획물 ATMB를 $50\;{\mu}g/mL$ 첨가했을 때 시료무처리구인 대조군을 1.0으로 한 경우 암예방 QR유도 효과가 아주 높은 3.92배의 활성이 나타났다. 본 연구 결과, 인체암세포 4종류에 대한 암세포 증식억제(cytotoxicity) 효과는 부추의 ethylether 분배층(ATMEE)에서 제일 뚜렷하였고, 암예방 QR 유도활성 효과는 butanol 분배층(ATMB)에서 가장 유도효과가 좋았다. 나아가 단계적인 생리활성 물질의 분리 동정이 계속 이루어져야 할 것으로 사료된다.

  • Panax ginseng C. A. Meyer has been the most highly recognized medicinal herb in the Orient. The prolonged administration of red ginseng extract significantly inhibited the incidence of hepatoma and also proliferation of pulmonary tumors induced by aflatoxin B$_1$and urethane. Statistically significant anticarcinogenic effects were observed in powders and extract of 6 year-dried fresh ginseng, 5 and 6 year-white ginseng and 4, 5 and 6 year-red ginseng by 9 week medium-term anticarcinogenicity test using benzo[a]pyrene (Yun's model).(omitted)

  • This study was carried out to investigate the effect of red ginseng extract on development of pepsinogen 1 decrased pyloric glands in experimental stomach carcinogenesis in Sprague-Dawley male rats. Sequential quantitative analyses (by ABC immunohistochemical staining) were made of pepsinogen 1 decreased pyloric glands (PDPG) after treating rats first with a single dose (160 mg/kg) N-methyl-N'-nitrosoguanidine (MNNG) and then with N-ethyl-N'-nitro-N-nitrosoguanidine (ENNG) (100ug/ml of drinking water) as a second gastric carcinogen (or promoter).

  • Imperatorin, a biologically active furanocoumarin from the roots of Angelica dahurica (Umbelliferae), was mutagenic and induced transformation of mouse fibroblast cell lines, whereas it provided inhibiting effects on mutagenesis and carcinogenesis induced by various carcinogens. Furthermore, it has been suggested that imperatorin may have potential anticarcinogenic effects when administered orally in the diet. In addition to its anticarcinogenic properties, imperatorin has been shown to possess anticancer activities. We investigated the macro scale gene expression analysis on the HL-60 cells treated with imperatorin. Imperatorin (10μM) were used to treat the cells for 6h, 12h, 24h, 48h, and 72h. In a human cDNAchip study of 10,000 genes evaluated 6, 12, 24, 48, 72 hours after treated with imperatorin in HL-60 cells. Hierarchical cluster against the genes which showed expression changes by more than 2 fold. Three hundred eighty six genes were grouped into 6 clusters by a hierarchical clustering algorithm. Pathway analysis using gene microarray pathway prof Her that is a computer application designed to visualize gene expression data on screen representing biological pathways and groupings of genes.

DMBA로 유발한 Mouse 피부암에 대한 Astaxanthin이 함유된 난황의 항암효과 (Inhibition of DMBA-Induced Mouse Epidermal Carcinogenesis by Astaxanthin-Containing Egg Yolks)

  • Anticarcinogenic activity of astaxanthin-containing egg yolks (designate AEY) was investigated for 7,12-dimethylbenz[a]anthracene (DMBA)-induced two stage mouse epidermal carcinogenesis. Female ICR mouse (6-7 weeks of age) were house in a humidity-and-temperature-controlled facility and subjected to feed and water ad libitum. AEY (10 mg/0.2 ml acetone) was painted on the back of mice 7 days, 3 days and 5 min before DMBA treatment (50 nmole/0.2 ml acetone). One week later after DMBA treatment, 6 ${\mu}g$ tetradecanoyl 12-phorbol 13-O-acetate (TPA) dissolved in 0.2 ml acetone was applied on the mouse twice weekly over a period of 22 weeks. No sample was given to control mice. Control egg yolk (CEY) and astaxanthin-containing oil (designate AO) from Phaffia rhodozyma were used as positive controls. Mouse treated with AEY exhibited 10 tumors per mouse whereas control mouse exhibited 15 tumors per mouse, the fact that 33% reduction of tumor per mouse by AEY treatment. Tumor incidence was also reduced to 15% by AEY treatment when compared to that of control group. Such effects were also seen in CEY and AO treatment groups, but leaser extent. AO gave reduction of food intake and body weights relative to those of AEY and CEY, indicating toxicity of AO. These results suggest that AEY exhibits anticarcinogenic activity for DMBA-induced mouse epidermal carcinogenesis.

Antimutagenic and Anticarcinogenic Effect of Methanol Extracts of Sweetpotato (Ipomea batata) Leaves

  • The present study was conducted to investigate the antimutagenic potential of the methanolic extract from the leaves of sweet potato (Ipomea batatas, IB) with the SOS chromotest (umu test) and Salmonella typhimurium TA 98 and TA 100. The anticarcinogenic effects were also studied by calculation of the $IC_{50}$ on human cancer cell lines and investigating the function of gap junction in rat liver epithelial cells. The IB extract inhibited dose-dependently the ${\beta}$-galactosidase activity induced spontaneously at concentration of more than 200 mg/ml in S. typhimurium TA 1535/pSK 1002, and decreased significantly (p < 0.01) the ${\beta}$-galactosidase activities induced by mutagen 6-chloro-9-[3-(2-chloroethylamino)proylamino]-2-methoxyacridine dihydrochloride (ICR) at dose of more than 0.4 mg/0.1 ml. The IB extract showed no effect on the spontaneous reversions of S. typhimurium TA 98 and 100 but benzo(${\alpha}$)pyrene (BaP)-stimulated reversions were decreased dose-dependently (p < 0.01) at the concentration of more than 100 mg/ml. The $IC_{50}$ value of stomach cancer cells was lower than that of normal rat liver epithelial cells, but the values of colon and uterine cancer cell lines were similar to those of normal rat liver epithelial cells. The transfer of dye through gap junctions was not affected by treatment of the IB extracts at any concentration during treatment periods. The simultaneously treatment of IB extract and 12-O-tetradecanoylphorbol-13-acetate (TPA) effectively prevented the inhibition of dye transfer induced by TPA 1 hour after treatment at all exposed concentrations. The number of gap junctions was significantly (p < 0.01) increased by the treatment with IB extract at concentrations of more than 40 ${\mu}g$/ml. The inhibition of the expression of gap junction proteins by TPA (0.01 ${\mu}g$/ml) was recovered dose dependently by the simultaneous treatment of IB extracts. Our data suggest that Ipomea batatas has antimutagenic and anticarcionogenic activity in vitro.

메탄올로 추출한 여주 분획성분의 항균 및 항발암 효과 (The Effects on Antimicrobial and Anticarcinogenic Activity of Momordica Charantia L.)

  • 중국에서 약용 및 음용식물로 사용되고 있는 여주 (Mo-mordica charantia L.)를 이용하여 추출, 분획한 후 항균, 항암 및 암예방 효과를 살펴보았다 여주의 MCU층을 제외한 모든 층에서 항균력을 보였으며 특히 Staphylococru aureus의 MCMEE층에서 가장 높은 항균활성을 나타내었다 실험에 사용한 각균에 대한 여주 분획물의 암세포 증식 억제 효과를 MTT assay로 실험한 결과, 3종의 인체 암세포 HepG2, HeLa 및 MCF극에서 모두 여주의 ethyleth-er 분획층인 MCMEE와 ethylacetate 분획층인 MCMEA 층에서 아주 높은 암세포 증식억제 효과를 나타내었다. HepG2세포를 이용하여 암예방 QR 유도 활성을 측정한 결과, 다른 분회층에 비해 비극성 용매층인 hexane 분획층 MCMH에서 아주 높은 QR 유도를 활성시키는 것으로 나타났다. 본 실험 결과, 여주에는 식품 보존과 항균제로서의 개발가능성이 보이며. 항발암 효과를 보기위한 암세포 증식 억제 효과는 ethylether층 MCMEE과 ethylacetate층 MCMEA층에서 월등하였고, 비극성 용매층인 hexane 분획층에는 암발생을 예방하는 quinone reductase inducer가 있는 것으로 사료되며. 나아가 단계적인 생리활성 연구의 분리 동정이 계속 이루어져야 할 것으로 사료된다.

Platform Technology for Food-Grade Expression System Using the genus Bifidobacterium

  • Bifidobacterium spp. is nonpathogenic, gram-positive and anaerobic bacteria, which inhabit the intestinal tract of humans and animals. In breast-fed infants, bifidobacteria comprise morethan 90% of the gut bacterial population. Bifidobacteria spp. are used in commericial fermented dairy products and have been suggested to exert health promoting effects on the host by maintaining intestinal microflora balances, improving lactose tolerance, reducing serum cholesterol levels, increasing synthesis of vitamins, and aiding the immune enchancement and anticarcinogenic activity for the host. These beneficial effects of Bifidobacterium are strain-related. Therefore continued efforts to improve strain characteristics are warranted. in these respect, development of vector system for Bifidobacterium is very important not only for the strain improvement but also because Bifidobacterium is most promising in serving as a delivery system for the useful gene products, such as vaccine or anticarcinogenic polypeptides, into human intestinal tract. For developing vector system, we have characterized several bifidobacterial plasmids at genetic level and developed several shuttle vectors between E. coli and Bifidobacterium using them. Also, we have cloned and sequenced several metabolic genes and food grade selection marker. Also we have obtained bifidobacterial surface protein, which will be used as the mediator for surface display of foreign genes. Recently we have succeeded in expressing amylase and GFP in Bifidobacterium using our own expression vector system. Now we are in a very exciting stage for the molecular breeding and safe delivery system using probiotic Bifidobacterium strains.
