인체용 항생제의 우선관리대상 선정과 물환경 중 오염농도 추정 (Prioritizing Human Use Antibiotics for Environmental Health Management and Estimating Their Environmental Concentrations in Korean Waterway)

  • Antibiotics are manufactured and used for specific physiological functions, hence they may exert adverse ecological consequences when they are in contact with nontarget organisms. In the last decade, many reports have been made on the occurrences of various antibiotic compounds in surface water, and their potential impact to the environment has become an increasing concern. This study was conducted to prioritize antibiotic substances with potential environment risk in Korea. Human use antibiotics with an EIC (Expected Introduction Concentration) value greater than $1{\mu}g/l$, US FDA's action limit criteria, were selected. In order to calculate a worst-case EIC for each substance, annual production volume (in kg) of each antibiotic substance was derived using the Korea Pharmaceutical Manufacturers Association (KPMA)'s monetary database. Sixteen substances were preliminarily selected. The EICs of the 16 antibiotic substances were refined with the excretion rate of the parent substances. Ten antibiotic substances were identified to have EIC-corrected greater than $1{\mu}g/l$, which include Amoxicillin ($15.8{\mu}g/l$), Cefaclor ($10.1{\mu}/l$), Roxithromycin ($4.2{\mu}g/l$), Cephradine ($4.5{\mu}g/l$), Cefatrizine ($2.6{\mu}g/l$), Cefadroxil ($3.3{\mu}g/l$), Aztreonam ($2.3{\mu}g/l$), Ceftazidime ($2.8{\mu}g/l$), Ribostamycin ($1.3{\mu}g/l$), and Ceftezole ($1.3{\mu}g/l$). Additional risk assessments for these antibiotic substances are suggested.

Production, Purification and Antifungal Activity of Antibiotic Substances Produced by Pseudomonas aeruginosa Strain B5

  • Pseudomonas aeruginosa strain B5 with antagonistic activity against Phytophthora capsici and Magnaporthe grisea, was isolated from pepper-growing soil. From the culture of P. aeruginosa strain B5 grown on King's medium B, antibiotic substances were purified using XAD-2 column chromatography. XAD-2 eluates inhibited not only the mycelial growth of P. capsid and M. grisea, but also the development of Phytophthora blight on pepper plants. The crude antibiotic substances were further purified by using silica gel column chromatography, Sephadex LH-20 column chromatography, thin layer chromatography on silica gel plates, and high performance liquid chromatography. Silica gel column chromatogrphy gave good separation of the four antibiotic substances. The pure antibiotics P1, P2, and P3 finally purified by preparative HPLC inhibited the mycelial growth of P. capsici, at concentrations from 7 to 10 $\mu g/ml$. Only P1 and P2 had antifungal activity against M. grisea at 8 $\mu g/ml$. P1 and P3 were highly inhibitory to the mycelial growth of Botryosphaeria dothidea and Botrytis cinerea at relatively low concentrations. However, the three antibiotics had no antifungal activity against Rhizoctonia solani. The chemical structures of these antibiotics are being identified.

  • Selective culture of actinomycetes from soil microbes and their antibiotic producing characters by agar-disk method were examined. Some of the organisms which produced antibiotics on agar disk did not produce antibiotics in liquid culture. Further examination indicated that production of antibiotic was dependent on the composition of medium. Many streptomycestes produced antibacterial substances in tryptic soy broth but others produced antifungal antibiotics in V-8 broth. Production of antibacterial substances by Streptomyces sp. was also dependent on the medium composition.

  • Among the Streptomyces isolated from soil, two strains which have antibacterial activity aginst various pathogenic bacteria are identified as Streptomyces globosus and Streptomyces albus subsp. according to I.S.P. methods and Bergy's Manual of Determinative Bacteriology. Morphological and physiological characteristics of them on several media were observed. Antibiotics from S. albus subsp. or S. globosus was identified asw tetracycline or streptomycin-like substances respectively by the paper chromatographic behavior in eight solvent systems of V. Betina. And it was revealed that these antibiotic substances are stable to temperature and weak acid.

항균기능정보가 각인된 플라스틱 저장용기의 항균 특성에 관한 연구 (A Study on the antibiotic properties of plastic containers templated with antibiotic functional information)

  • 본 연구에서는 새로운 개념의 항균처리 기술을 도입하여 항균 물질을 플라스틱 용기에 혼입하지 않고도 항균기능이 발휘되도록 하는 기술을 개발하고자 하였다. 기술의 요체는 항균기능정보를 물에 각인한 다음에 이를 플라스틱 제조용 원료 수지에 전사하여 최종 제품에서도 같은 기능이 나타나도록 한 것이다. 이러한 개념에 입각한 기능 정보 각인 기술의 실용화 가능성을 검토하기 위하여 시제품을 만들어 항균 실험을 수행한 결과 세균 증식 억제율이 75% 전후의 수준인 것으로 나타났다. 앞으로 이러한 기술이 보다 더 다듬어지고 그 작용기전이 밝혀진다면 더욱 뛰어난 제품이 개발될 가능성이 높다고 하겠다.

  • Antibiotic residues of raw milks collected in Anyang area were tested by TTC-reduction test, EEC-4plate method and TLC(SOS-kit ) method to improve and monitor the quality of milk. Antibiotic substances were not detected from 100 raw milk samples, but sulfamethazine was detected from only one sample(1.3PPM) by SOS test. Unclassified anti-microbic substances were detected from 22 samples by EEC-4 plate method. EEC-4 plate test was useful to categorize the species of antibacterial substances and SOS test was useful to detect the sulfamethazine field screening test.

  • According to various negative effects of antibiotic growth promoters, the effects of nature-derived alternatives to antibiotic growth promoters are mainly investigated in livestock industry these days. The effects of nature-derived alternatives to antibiotic growth promoters are mainly investigated in livestock industry these days. This study was conducted to estimate the efficiency of natural-derived alternatives to antimicrobial and antioxidant such as oregano oil (OGO), sulfide type antimicrobial peptides derived from Bacillus or Yeast (SAPBY), antimicrobial peptides derived from yeast (APY). The lactic acid bacteria growth effects were found to be minimal. The antibacterial activity of SAPBY was minimal, but OGO and APY showed antibacterial activity against Escherichia coli and Salmonella Typhimurium, and APY also showed antibacterial activity against Staphylococcus. Antioxidant effects were observed in all SAPBY, OGO, and APY, and when comparing the radical scavenging rate at 10 mg·mL-1 for each sample, OGO showed the highest at 84.9%, followed by SAPBY at 48.7%. Therefore, the substances that can be used as natural antibiotics are SAPBY, APY, and OGO. It is believed that adding these substances to feed and feeding them to livestock will greatly contribute to improving the health of livestock and the productivity of safe livestock products. In further, research focused on various nature-derived alternatives to antibiotic growth promoter in livestock industry is needed.

  • 본시험은 한국산항생물질 또는 항생균의 개발이용을 목적으로 한 기초연구로서 주로 전년도의 계속연구와 아울러 토양시료로부터 새로히 항생세균을 분리하였다. 청주부근의 토양시료 500점으로부터 83점의 항생균을 분리하였으며 이들은 주로 gram음성균인 Escherichia coli에 유효한 것을 선별하였다. 이들에 대한 연구는 앞으로 계속할 것이며 수년간 계속 연구중인 우수항생균 JS7501은 그동안 수차에 걸쳐 재선별하였기 때문에 JS901로 개칭하여 본시험에 공시하였으며 항균작용법 위도 재확인하였으며 주로 추출을 목적으로 용매추출, 흡착, TLC, 이온교환 수지 등을 총동원하였으나 TLC에 의하여 활성이 확인된 4 spot를 분리할 수 있었다. JS7901의 배양배지로서는 Soytone Sugar배지가 가장 높은 항균력을 보였으며 Soytone보다는 Sugar가 활성물질 증가와 관계가 있었다.

Purification and Antifungal Activities of an Antibiotic Produced by Gliocladium virens G1 Against Plant Pathogens

  • This study was undertaken to separate and identify antifungla substances produced by Gilocladium virens G1, a biocontrol agent used for the control of plant diseases caused by Rhizoctonea solani. The culture of G. virens G1 effectively inhibited the growth of R. solani, Colletotrichum gloeosporioides, and Phytophthora capsici, but less that of Fusarium oxysporum. The n-hexane extract of the G. virens culture, which was used for the purification of responsible substances, strongly inhibited R. solani and C. gloeosporioides, but not P. capsici, although the n-butanol extract was effective on all of the pathogens tested. An antifungal substance was purified using the n-hexane extract by Silica gel column chromatography and HPLC. The substance was examined for purity by HPLC and for nature by UV spectrometry, which differed from known antibiotic compounds such as gliotoxin, viridin and gliovirin. The antifungal substance was very liphophilic based on its solvent-solubility and Rf values on TLC, and more inhibitory to C. gloeosporioides than other fungal pathogens tested.

  • 1. Of the 32 extracts from Genus of lishenes broth tested for antimicrobial activity, 28 inhibited at least one of the 3 test microorganisms used. 2. Twenty seven lichnes broth from 32 species tested were active against at least one of the Gram-positive bacteria M. pyogenes var, aureus 203 p, and twenty four lichenes broth from 32 Species tested were active against at least one of the Gram-positive bacteria Bacillus subtilis ATCC 6633. 3 Twenty five lichenes broth from 32 species tested were active against at least one of the Gram-negative bacteria Escherichia coli 0.126. 4. The antibiotic substances in lichenes were readily extracted by organic solvents.
