Antibacterial activity and toxicity of Halymenia durvillei red seaweed from Kayangan island, South Sulawesi, Indonesia

  • This study aimed to determine the antibacterial activity and toxicity of methanol and hexane extracts of Halymenia durvillei red seaweed which were found abundantly in Kayangan island, South Sulawesi. The antibacterial activity of the crude extract was tested against five gram-negative bacteria, namely Escherichia coli, Salmonella typhi, Pseudomonas aeruginosa, Aeromonas hydrophila, and Vibrio harveyi at a dose of 200 g/disk. Extract toxicity was tested on Artemia salina larvae at concentrations of 1,000, 500, 250, 125, 62.5, and 31.25 ㎍/mL. The results showed that the methanol and hexane extracts of H. durvillei had the highest activity against S. thypi and A. hydrophila, respectively, with inhibition zones of 26.2 mm and 21.0 mm. On the other hand, the two extracts did not show activity against E. coli and P. aeruginosa, respectively. The toxicity of the methanol extract of H. durvillei was twice as high as that of the hexane extract with half-maximal inhibitory concentration of 98.24 and 184.21 ㎍/mL, respectively. Thus, the methanol and hexane extracts of red seaweed H. durvillei have the potential as new antibacterial agents respectively against the pathogenic bacteria S. typhi and A. hydrophila, but also have the opportunity to be developed into antitumor herbal compounds.

흰 쥐의 감염된 전층피부결손에 대한 항균연고들의 항균효과 및 창상치유 효과의 비교 (Comparison of Antibacterial Ointments for Antibacterial and Wound Healing Effect on Infected Full Thickness Skin Defect in the Mouse)

  • Purpose: Many topical agents had been used for contaminated wound treatment. Although antimicrobial ointments were widely used as topical agents, their comparative antibacterial and wound healing effects were largely unreported. The purpose of this study was to compare antibacterial effects and wound healing effects of common topical ointments on infected full thickness skin defect in the mouse. Methods: One full thickness skin defects in the mice (n=60) were developed on the back and left open for twenty-four hours. Sixty mice were divided into four groups: group B (dressing with Bactroban$^{(R)}$, n=15), group I (dressing with Iodosorb$^{(R)}$, n=15), group T (dressing with Terramycin$^{(R)}$, n=15), group G (control group, dressing with dry gauze, n=15). The size of wound defects and the grades of wound healing were evaluated at 4, 7, 10 days, and antibacterial effect was evaluated with restricted zone in Mueller Hinton agar by disk diffusion method. After the wound was left open for twenty-four hours, many Staphylococcus aureus was cultured. The wound defect size was decreased in order of Bactroban$^{(R)}$ (B), Iodosorb$^{(R)}$ (I), Terramycin$^{(R)}$ and gauze dressing group in all days, but difference among experimental groups was not statistically significant. The grade score of wound healing was increased in order of Bactroban$^{(R)}$, Iodosorb$^{(R)}$, Terramycin$^{(R)}$ and gauze dressing group, and the difference was statistically significant. Antibacterial effect for was increased in order of Bactroban$^{(R)}$, Iodosorb$^{(R)}$, Terramycin$^{(R)}$ and gauze dressing group, and the difference was statistically significant. Conclusion: Topical antimicrobial ointments application was effective against wound infections by S. aureus. Bactroban$^{(R)}$ may be an optimal topical treatment for infected wounds according to this study. However, further study is necessary to evaluate the clinical efficacy of antimicrobial ointments and to search for the mechanisms that explain their effects.

주요 임상 내성균에 대한 어성초 에탄올 추출물의 항균효과 (Antimicrobial Activity of Houttuynia cordata Ethanol Extract against Major Clinical Resistant Microorganisms)

  • 다제내성균의 증가는 사용할 항생제가 적거나 심지어 선택할 항생제가 없기 때문에 감염환자의 치료에 커다란 위협이 되고 있다. 최근 다양한 천연 추출물이 내성세균에서 항균효과 및 상승효과를 보인다는 보고가 많다. 어성초(Houttuynia cordata)는 항균, 항바이러스 및 항산화 효과를 보이는 전통적 약초이다. 이에 저자들은 임상의 주요 내성균에 대한 어성초의 항균효과를 알아보고, 이들의 적절한 항균효과 측정방법을 평가하고자 한다. 우선 현재 가장 많은 문제를 야기하는 주요 내성균인 methicillin-resistant Staphylococcus aureus (MRSA), vancomycin-resistant Enterococcus faecium (VRE), carbepenem-resistant Acinetobacter baumannii(CRAB) 각각 10 균주를 선택하여, CLSI 기준으로 디스크 확산법 및 미량액체배지희석법으로 감수성 시험을 하였다. CLSI 기준으로 시행한 디스크 확산법에서 $4,096{\mu}g/mL$에서 $30,000{\mu}g/mL$ 까지의 농도에서 모든 균의 억제대는 관찰되지 않았다. 그러나 액체배지 희석법에서는 MRSA, VRE, CRAB의 $MIC_{90}$이 각각 $4,096{\mu}g/mL$, $8,192{\mu}g/mL$, $4,096{\mu}g/mL$를 보였다. 결론적으로 어성초는 MRSA, VRE, CRAB 등 주요 임상 내성균에서 우수한 항균효과를 보이며 천연물의 항균효과를 평가에 디스크 확산법보다 액체배지 희석법이 유용하다고 할 수 있다. 만약 디스크 확산법으로 항균효과를 측정하려면 천연물에 대한 균 접종 농도의 변경 등 새로운 기준이 요구된다.

소목추출물의 젖소 유방염 원인균에 대한 항균효과 (Antibacterial Effect of Caesalpinia sappan Extract against Mastitis Pathogens from Dairy Cows)

  • To find alternative agents for bovine mastitis, the extracts of 30 selected herbal medicine were screened for their antibacterial activity against mastitis pathogens. The extracts of 9 herbal medicine, Terminaliae chebula, Cinnamonum loureirii, Paeonia japonica, Inula britannica, Caesalpinia sappan(C. sappan), Salix babylonica, Alpinia katsumadai, Polygonum aviculare and Coptidis japonica showed strong antibacterial activity. Among of them, C. sappan extract was found to be the most active against all bacteria in this experiment. Methanol extract of C. sappan and brazilin, the main component of C. sappan, had inhibitory activity against a variety of pathogens, however they had less of an inhibitory effect on gram negative bacteria. Minimal inhibitory concentrations of brazilin against gram positive pathogens was lower than that of methanol extract. These results showed that the extract of C. sappan has a potential as a therapeutic agent for bovine mastitis.

Broad Spectrum Antibacterial Activity of Allium cepa, Allium roseum, Trigonella foenum graecum and Curcuma domestica

  • In many parts of the world Allium cepa, Allium roseum, Trigonella foenum graecum and Curcuma domestica are extensively used as food and are popular in herbal medicine. The four were screened against 26 pathogens and all exhibited broad-spectrum anti-bacterial activity. The aqueous as well as fractionated methanol extract of Allium cepa and A. roseum demonstrated broader level of activity against most of the organisms. On the other hand the unfractionated methanol extracts as well as the fractions of both Trigonella foenum graecum and Curcuma domestica showed broad spectrum of activity. Fractionation was found to improve their level of activity. In both cases the ethyl acetate fractions exhibited higher level of activity. All the materials tested were inactive against any of the four moulds. Allium cepa, Allium roseum, Trigonella foenum graecum and Curcuma domestica are proposed as non toxic, safe, broad spectrum antibacterial agents.

Momordica charantia and Allium sativum : Broad Spectrum Antibacterial Activity

  • In the Asian sub-continent Momordica charantia and Allium sativum are extensively used as food and are popular in herbal medicine. The two were screened against 15 pathogens and both exhibited broard spectrum antimicrobial activity. As compared to the standard antibiotics, M. charantia demonstrated broader and higher level of activity against most of the organisms. On the other hand A. sativum showed comparable activity to the standard antibiotics. Both M. Charantia and A. sativum are proposed as non toxic, safe, broad spectrum antibacterial agents.

충치균, Streptococcus mutans OMZ176에 대한 황금의 항균활성성분 (Antibacterial Constituents from Scutellariae Radix against Streptococcus mutans OMZ176)

  • To develop anticariogenic agents, Scutellariae Radix was tested for its antibacterial activity against Streptococcus mutans OMZ176 which is known as a strong cariogenic bacterium. The antibacterial activity was evaluated with disk plate method and minimal inhibitory concentration (MIC). The ethyl acetate fraction showed significant activity. By means of activity-guided fractionation, baicalein and ganhuangenin were isolated as the active principles. The MICs of these compounds were $100{\mu}g/ml$ and $200{\mu}g/ml$, respectively.

Antibacterial Activity of Acanthoic acid Isolated from Acanthopanax koreanum against Oral and Skin Microfloras

  • The (-)-pimara-9 (11), 15-dien-19-oic acid, acanthoic acid was extracted from the roots of Acanthopanax koreanum using bioassay-guided isolation of a MeOH extract. Acanthoic acid was assayed against Streptococcus mutans and Staphylococcus epidermidis causing dental caries and opportunistic pathogen. The minimum inhibitory concentration (MIC) of acanthoic acid against Streptococcus mutans and Staphylococcus epidermidis was 2 and 4 ${\mu}g/mL$, respectively, which was much lower than those of other natural antimicrobial agents such as 8 ${\mu}g/mL$ of tanshinone IIA. Acanthoic acid also significantly inhibited the growth of other cariogenic bacteria such as Streptococcus sobrinus and Streptococcus sanguis, and Streptococcus grodenii in the MIC range of 4${\sim}$32 ${\mu}g/mL$. Our findings suggest that acanthoic acid could be employed as a potential antibacterial agent for preventing dental caries and skin infections.

강원지역의 젖소 유방염 감염율 및 원인균에 관한 연구 (Studies on Infection Rate and Causative Agents of Bovine Mastitis in Kangweon Area)

  • A total of 2,024 quarters of 515 dairy cattle in Kangweon area were examined for incidence of subclinical mastitis. Milk samples from cattle infected with subclinical mastitis were studied bacteriologically and the bacterial strains isolated were further examined for sensitivity to 12 antibacterial agents. And the status of carrying out the mastitis control program in 28 dairy farms was examined. The results obtained were summerized as follows ; 1. A total of 308(59.8%) of 515 cattle and 656(32.4%) of 2,024 quarters were found to be infected with subclinical mastitis. 2. The 277 strains of etiological agents were isolated from 358 subclinical quarters. These were identified as Staphylococcus aureus(14.4%), other staphylococci(36.5%), Streptoccus agalatiae(8.7%), other streptococci(30.7%), Bacillus spp.(1.8%), Corynebacterium spp.(1.4%) and coliform(0.7%). 3. The 109 strains of streptococci and 141 strains of staphylococci were examined for sensitivity to 12 antibacterial agents. All the strains of streptococci were sensitive to penicillin, ampicillin and cephalothin, and they were also sensitive to erythromycin(88.1%), clindamycin(83.5%), enrofloxacin(75.2%), trimethoprim+sulfamethoxazole(67.9%), The strains of staphylococci were sensitive to cephalothin(97.2%), gentamicin(83.0%). enroflozacin(80.9%), trimethoprim+sulfamethoxazole(78.0%), erythromycin(71.6%) and clindamycin(71.6%). But all the strains resisted to colistin. 4. In the 28 dairy farms examined, condition of udder before washing was dirty in most of the farms (89.7%). Hygiene of milking equipment was only good in the 5 farms(17.9%). Teat preparation before milking was good in the 6 farms(21.4%). The farms in which teat dipping after milking was conducted were 46.4%. Dry cow treatment for the complete herd was carried out in most of the farms(89.3%) but mastitis checking was only carried out in the 8 farms(28.6%) irregularly.

Screening and Evaluation of Antibacterial Metabolites from Entomopathogenic Fungi

  • Entomopathogenic fungal species have been investigated for their potential use as biological control agents owing to their natural role as insect pathogens. These fungi produce a wide range of secondary metabolites with high therapeutic values, such as antibiotics and cytotoxic substances. To evaluate the antibacterial activity of entomopathogenic fungi, 10 isolates from Korean soil were selected and tested for their activity against Escherichia coli by using fungal culture filtrates. Antibacterial activity was assessed using a two-step process: (1) a screening assay for the selection of fungal isolates and (2) a quantitative assay to evaluate the activity of select fungi. Although 4 fungal isolates were selected through the screening assay, only 3 fungal isolates, from Beauveria bassiana and Metarhizium anisopliae, showed high antibacterial activity according to the quantitative assay. The antibacterial activity of selected fungal culture filtrates was stable when exposed to heat and proteolytic enzyme treatments, which indicated that the antibacterial compound is not a protein. These entomopathogenic fungal metabolites might be useful as a source for bacterial control and in the pharmaceutical industry.
