Studies on the Efficacies of Water Extract of Propolis (프로폴리스 물추출물의 약효연구)

  • Propolis is a lipophilic, natural product prepared by mixing the exudates collected from various plants by honeybees with beeswax for the purpose of using to seal hive walls and to strengthen the borders of combs. Because of its versatile bioactivities, propolis has been attracting many investigators'interest. But the pharmacological studies on propolis has, to date, been exclusively performed for an alcohol extract, there is few information of water extract. Therefore, in this study, we investigated the various effects of water extract of Chinese propolis. The water extract of propolis and its fractions of organic solvents showed strong antioxidative activities, especially ether and ethylacetate fractions, and reduced the lipid peroxidation of rat liver in viro. Additionally the ether fraction of propolis (10 mg/ml) inhibited the activity of hyaluronidase by 50%. In vivo, the water extract of propolis considerably decreased s-GOT, s-GPT and s-LDH activities which represent for the hepatotoxicity induced by $CCl_4$ in rats, and prolonged the MST (Medium revival tinge) and ILS (Increasing in MST over control) by 18% in mice which inoculated with sarcoma 180 ascites cells. These results suggest that the water extract of propolis has various bioactivities as well as the alcohol extract.

  • The anti-oxidant activities of the various fractions from the herbs of Artemisia apiacea were investigated. The n- hexane and n-butanol fractions were found to cause significant free radical scavenging effects on DPPH. their scavenging potencies as indicated in $IC_{50}$ values. being 230.1 and 183.7 g/ml. respectively. The n-butanol fraction exhibited a significant decrease in serum transaminase activities elevated by hepatic damage induced by $CCl_{4}$-intoxication in rats. All fractions tested exhibited a lipid peroxidation causing a significant decrease in MDA production in TBA-reactant assay. (omitted)

Effects of $\beta$-Carotene Supplementation on Lipid Peroxide Levels and Antioxidative Enzyme Activities in Diabetic Rats ($\beta$-Carotene 첨가식이가 당뇨쥐의 지질과산화물 수준과 항산화효소 활성에 미치는 영향)

  • This study investigated the effect of dietary $\beta$-carotene supplementation on lipid peroxidation and anti oxidative enzyme activity as indices of oxidative stress in diabetic rats. Fifty Sprague-Dawley male rats aging 7 weeks were used as experimental animals, which were divided into the non-diabetic control group and the diabetic group. The diabetic group received an intraperitoneal injection with streptozotocin to induce diabetes. Then the diabetic rats were divided into four dietary groups which contained different amounts of $\beta$-carotene; 0%, 0.002%, 0.02%, or 0.2% of the diet. The diabetic rats were fed the experimental diets and the non-diabetic rats were fed the basal diet without $\beta$-carotene supplementation for 2 weeks and then sacrificed. The diabetic group had a significantly higher blood glucose level than the non-diabetic group. However, blood glucose level were not significantly changed by the level of dietary $\beta$-carotene supplementation. Compared to the non-diabetic control group, the diabetic control group indicated a significant increase of plasma thiobarbituric acid reactive substance (TBARS). Liver TBARS level also tended to be higher in diabetic control group, although it was not significant. The $\beta$-carotene supplementation did not reduce plasma TBARS level. However, Liver TBARS level was significantly decreased when 0.02% or more $\beta$-carotene was supplemented in the diet. The liver lipofuscin level in the diabetic control group was higher than in the non-diabetic control group, but the effect of $\beta$-carotene supplementation did not show any differences. Superoxide dismutase activity was significantly lower in the diabetic group, but it was increased in groups receiving 0.02% or more $\beta$-carotene. Compared to the non-diabetic control group, lower activities of catalase and glutathione peroxidase were observed in the diabetic control group, although it was not significant. Catalase and glutathione peroxidase activities tended to increase as the levels of $\beta$-carotene supplementation increased, although it was not statistically significant. Therefore, it seems that dietary $\beta$-carotene supplementation might reduce diabetic complications by partly decreasing the lipid peroxidation and increasing the activity of antioxidative enzyme in diabetes.

  • Asclepias curassavica. Linn, an erect, simple (or) much branched perennial herb with a somewhat woody base, belonging to the family Asclepiadaceae. It has been reported to have multiple pharmacological effect of anti-inflammatory, cardiotonic, anticancer, anthelmintic and to treat piles and gonorrhoea. It is to be expected that several activities might be related to a possible antioxidant action from this plant. The hydro alcoholic extract of Asclepias curassavica was tested in vitro for its antioxidant activities, such as DPPH radical, nitric oxide radical, superoxide anion radical, lipid peroxidation assay, hydroxyl radical, reducing power, and total phenol content. The extract exhibited scavenging potential with $IC_{50}$ value of $8.7\;{\mu}g/ml,\;198.4\;{\mu}g/ml\;and\;21.7\;{\mu}g/ml$ for DPPH, nitric oxide and superoxide anion radicals. The values were found to higher than those of Vitamin-C, rutin, and curcumin, as standards. The extract showed 50% protection at the dose of $134.2\;{\mu}g/ml\;and\;41.4\;{\mu}g/ml$ in lipid peroxidation as well as deoxyribose degradation, those values more to that of standard, vitamin E $(IC50\;values,\;119.2\;{\mu}g/ml\;and\;32.5\;{\mu}g/ml,\;respectively)$. The reducing power of the extract depends on the concentration and amount of extract. Since a significant amount of polyphenol could be detected by the equivalent to $0.0495\;{\mu}g$ of pyrocatechol from 1 mg of extract. It can be concluded that hydro alcoholic extract of aerial part of Asclepias curassavica could be considered as potent antioxidant, which makes it suitable for the prevention of human disease.

Biological Activities of Substance Extracted from the Fruit Body of Formitopsis rosea (잔나비버섯 자실체로부터 분리한 물질의 특성 및 생리활성)

  • This study was conducted to investigate the characteristics and biological activities of the substance extracted from the fruit body of Formitopsis rosea. The substance was extracted by hot water and then it was separated high and low molecular weight fraction by ethanol precipitation, The high molecular weight fraction was found to be a proteoglycan composed of glucose, mannose, galactose, xylose, fructose, ribose and amino acids. The amino acids of proteoglycan were mainly threonine, isoleucine, glycine, aspartic acid. Anticomplementary activity of the high molecular weight fraction was higher than that of the low molecular weight fraction. And also, the high molecular weight fraction enhanced phagocytic activity and nitric oxide production of macrophage. In enzymatic lipid peroxidation reaction with $ADP-FeCl_3-NADPH$, the lipid peroxidation was inhibited 82.4% and 86.6% by high and low molecular weight fractions, in nonenzymatic lipid peroxidation reaction, it was 83.0% and 84.6%, respectively.

Activities of Extract from Leonurus sibiricus Against UVB-Damage in HS68 Cell (자외선 B로 산화적 손상이 유도된 HS68 세포에 익모초 추출물의 효능 평가)

  • Objectives : The aim of this study is to investigate anti-inflammation of Leonurus sibiricus methanol extract against UVB-damage in fibroblast. The skin is continuously exposed to damage from environmental stresses. UV radiation causes a variety of biological effects especially on the skin, including inflammation and photoaging. Methods : In this study, we tried to search for Leonurus sibiricus which exhibit protective activities against UVB-induced cytotoxicity and oxidative cell death, NO and $PGE_2$ production. HS68 cells were exposed to UVB ($120mJ/cm^2$) and treated with various concentrations (0, 0.25, 0.5, 1, 2, 4, $8mg/m{\ell}$) of Leonurus sibiricus methanol extract for additional 24 h. Intracellular reactive oxygen species (ROS) levels generated by UV radiation were detected using a spectrofluorometer after DCF-DA staining. Also, HS68 cells were irradiated with UVB and then treated with Leonurus sibiricus methanol extract for 12 h. The lipid peroxidation was assayed by measuring the levels of 8-isoprostane secreted into the culture medium. Results : UVB-induced cytotoxicity and cell death were effectively suppressed by treatment of Leonurus sibiricus aqueous methanol extracts. Oxidative cell damage was mediated $PGE_2$ in UVB-induced HS68 fibroblast cell, which was significantly inhibited by treatment with Leonurus sibiricus extracts. Also, the protective effect of these extract seemed to be mediated by inhibited intracellular ROS generation and lipid peroxidation in dose-dependent manner. Conclusion : These results suggest that Leonurus sibiricus aqueous methanol extracts may have anti-aging effects new functional materials against oxidative UVB stress-mediated skin damages.

Effect of G009 on Lipid Peroxidation Induced by Peroxidizer in Rats

  • In order to elucidate the correlation between the lipid peroxidation and hepatotoxicity, the formation of malondialdehyde (MDA) in liver homogenate and serum, and the transaminase activities were determined in intoxicated by ascorbic acid-Fe$\^$2+/-ADP in rat. In a model of ascorbic acid-Fe$\^$2+/-ADP hepatotoxicity, G009, which was obtained from Ganoderma lucidum IY009, exhibited anti-lipid peroxidative effect in rat liver homogenate, and that MDA values of the liver homogenate decreased from 48.1% to 74.8% in comparision to controls (p<0.01) Also, the MDA formation in serum inhibited 66.5% at 100 mg/kg of G009. Serum levels of glutamic oxaloacetic transaminase(GOT) and glutamic pyruvic transaminase(GPT) in peroxidizer-induced rats treated with G009 was decreased compared with control. Especially, The formation of lipid peroxides in serum was related to GPT levels. These results that G009 has a protective effect on ascorbic acid-Fe$\^$2+/-ADP-induced hepatic injury through an inhibition of lipid peroxidation in liver.

Antioxidant and inflammation inhibitory effects from fruiting body extracts of Ganoderma applanatum

  • In this study, the antioxidant and anti-inflammatory effects of methanol extract (ME) and hot water extracts (HE) from the fruiting bodies of Ganoderma applanatum were investigated. The 1,1-diphenyl-2-picryl-hydrazy (DPPH) radical scavenging activity of 2.0 mg/mL ME (94.83%) was comparable to that of butylated hydroxytoluene (96.97%), the reference standard. The hydroxyl radical scavenging activities of ME and HE were similar to that of BHT at 2.0 mg/mL, whereas lipid peroxidation activity of the ME and HE were significantly lower than that of BHT. High-performance liquid chromatography analysis showed that the G. applanatum fruiting bodies contained nine phenolic compounds, which might contribute to antioxidant and anti-inflammatory activities. The survival rate of RAW 264.7 macrophages treated with 2.0 mg/mL ME and HE were 65.23 to 68.12% at 2.0 mg/mL, thereby indicating that the extracts were slightly cytotoxic at the concentration tested. The extracts also inhibited the nitric oxide (NO)-mediated expression of inducible nitric oxide synthase (iNOS) protein in lipopolysaccharide-induced RAW 264.7 macrophages and carrageenan-induced paw edema in rats. The study results demonstrated that the fruiting bodies of G. applanatum possessed good antioxidant and anti-inflammatory activities, which might be used to develop novel anti-inflammatory agents.

Protective effects of Seoritae Chungkukjang added with green tea powder against 3-morpholinosydnonimine-induced oxidative stress (녹차 첨가 서리태 청국장의 3-morpholinosydnonimine에 의한 산화적 스트레스 개선 효과)

  • To increase antioxidative activity of Chungkukjang, the protective effect of Seoritae Chungkukjang (SC) added with green tea powder against oxidative stress was evaluated under the cellular system using LLC-$PK_1$ cells. The treatment of 3-morpholinosydnonimine showed increase in lipid peroxidation, and decrease in endogenous anti-oxidant enzymes activity and cell viability. The methanol extract of SC inhibited lipid peroxidation by 70.9%, and significantly increased cell viability up to more than 33.2%. In addition, it enhanced superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) activities. Particularly, the addition of green tea in SC exerted protective effect against oxidative stress by ONOO- through elevation in activities of SOD and GSH-Px, and inhibition of lipid peroxidation. More addition of green tea showed stronger protective activity. These results suggest that the addition of green tea to SC leads to the increase in the antioxidative effect of Chungkukjang through elevation in antioxidative enzyme activities and protection from lipid peroxidation.

Anti-wrinkle Effect of Cambodian Phellinus linteus Extracts (캄보디안 상황버섯 추출물의 주름개선 효과 연구)

  • The skin of human is constantly being exposed to environmental irritants such as ultraviolet, smoke and chemicals. These irritants cause free radicals and reactive oxygen species which leave serious damages on the cells of skin. The water and ethanol extracts of Cambodian Phellinus linteus were investigated for the activities of anti-lipid peroxidation and anti-wrinkle effects to apply as a functional ingredient for cosmetic products. As the result of evaluation of liquid oxidation rate by add $Fe^{2+}$ and $Cu^{2+}$ to Cambodian Phellinus linteus extracts, Cambodian Phellinus linteus ethanol extracts were higher than Cambodian Phellinus linteus water extracts in the chealting ability of $Fe^{2+}$ and $Cu^{2+}$. The Cambodian Phellinus linteus ethanol extracts exhibited that anti-lipid peroxidation higher than butylated hydroxytoluene (BHT) at the concentration of 0.1 mg/ml, 0.5 mg/ml and 1 mg/ml. Cambodian Phellinus linteus water and ethanol extracts showed a higher inhibitory effect on $Fe^{2+}$-induced lipid peroxidation compared to $Cu^{2+}$-induced lipid peroxidation. In the case of anti-wrinkle effect, the elastase inhibition activity of Cambodian Phellinus linteus ethanol extracts was 50.7%, and it is higher than urosolic acid at the concentration of 0.01 mg/ml. Also, in collagenase inhibition activity, Cambodian Phellinus linteus water extract showed low effect, but Cambodian Phellinus linteus ethanol extract was about 50% at a 0.1 mg/ml. concentration. These results proved that Cambodian Phellinus linteus had anti-lipid peroxidation and anti-wrinkle effect. Therefore, Cambodian Phellinus linteus could be useful as an anti-wrinkle cosmetic ingredient.
