Action Pattern of Anti-Yeast Substance Originated from Rahnella aquatilis Strain AY2000 (Rahnella aquatilis AY2000균 유래의 항 효모물질의 작용양상)

  • For reduction of side effects by anti-fungal agents, a less toxic anti-fungal substance or a synergistic substance with a new mechanism is needed. The anti-yeast substance (AYS) originated from Rahnella aquatilis strain AY2000 is like to be a heterogeneous protein. The AYS inhibited the growth of Candida albicans in culture broth, and AYS-treated cells were arrested in each phase during cell cycle. Among AYS-treated cells, the population of the cells belonging to sub-G1 phase was not increased during cell cycle. Therefore, AYS has rather yeaststatic than yeastcidal effect to C. albicans. Moreover, with combination of itraconazole or fluconazole, AYS had a synergistic anti-yeast activity against Saccharomyces cerevisiae based on the analysis of fractional inhibitory concentration index.

Evaluation of Anti Fungal Efficacy on Different Formaldehyde Level of Wood-Based Composite Panels (목질판상제품의 포름알데히드 방출등급과 항균성 평가)

  • This study was carried out to investigate the susceptibility of wood-based composite panels exposed to mould and sap-stain fungi. Five wood deterioration fungi (three mould fungi, two sap stain fungi) were inoculated into two types of commercial wood-based composite panels (medium density fiberboard and particleboard), which have three class of formaldehyde emission. All wood-based composite panels were more or less susceptible to mould and sap stain fungi. The attacking mode of the fungi was highly dependent on formaldehyde emission. This study indicates that all wood-based composite panels, specially low formaldehyde emission class panels should be considered to prevent fungal deterioration when they are used for exterior and humid interior applications.

  • Two kinds of chemicals (RNF-3 and RNF-4) with high anti-mold effectiveness against stored fungi causing fungal discoloration was selected from among the six kinds of fluoride-based chemicals in the preceding study. In this study, the anti-mold test using 16 species fungi isolated from the softwood lumbers which were fungal discolored and field test in the sawmill was carried out to prove the feasibility for practical using of selected chemicals.For the isolated fungi, the RNF-3 consist of F and Cu showed high mycelial growth control in the PDA medium and fungicidal effectiveness in the japanese red pine (Pinus densiflora), korean pine (Pinus koraiensis) and radiata pine (Pinus radiata) when the concentration was 2% or more. However the RNF-4 consist of F only was not effective compared with RNF-3 because of mycelial growth in the PDA medium and wood treated with 2% or more chemical solution. The RNF-3 also showed a strong anti-mold effectiveness because there was no fungal discoloration for the radiata pine boards treated by 2% and 10 min. soaking in the field test. These results mean that RNF-3 can be used as domestic anti-stain chemicals for prevention of fungal discoloration of the softwood lumber

  • Past decade systemic mycoses caused by opportunistic human fungal pathogens, including Candida, Aspergillus, and Cryptococcus, have been a growing problem for both immunocompromised and immunocompetent individuals. Particularly, Cryptococcus neoformans has recently emerged as a major fungal pathogen, which can cause fungal pneumonia and meningitis that are lethal if not timely medicated. However, treatment for cryptococcosis has been difficult due to a lack of proper anti-cryptococcal drugs with fungicidal activity and less toxicity. In this review we introduced novel therapeutic methods for treating cryptococcosis by exploring pathogenomic signa1ing networks of C. neoformans with genome-wide transcriptome approaches as well as diverse molecular/genetic tools.

Syntheses and Biological Activities of Potential Antifungal Allylamine Compounds (항진균 알릴아민 화합물의 합성과 활성평가)

  • Structure-activity relationship studies of allylamine type of antimycotics were carried out to evaluate the effect of naphthyl and methyl portion of naftifine. Compounds with 3,4-difluorophenyl (2a-5a), 4-hydroxyphenyl (2b-5b), 3-nitro­phenyl (2c-5c), 4-chlorobenzothiazoly (2d-5d) and 5-methylfurfural (2e-5e) instead of naphthyl group, and with hydrogen (3a-3e), methyl (4a-4e) and ethyl (5a-5e) in the place of methyl in naftifine were synthesized and tested for their in vitro anti-fungal activities against five different fungi. Fourteen compounds (3a, 4a, 5a, 3b, 4b, 5b, 3c, 4c, 5c, 3d, 4d, 3e, 4e and 5e) showed significant anti-fungal activities against T. mentagrophytes. (E)-N-(3-Phenyl-2-propenyl)-3,4-difluoro-ben­zenemethaneamine(3a), (E)_N_(3_phenyl_2_propenyl)_4_hydroxy_benzenemethaneamine(3b) and (E)-N-ethyl-N-(3-phenyl­2-propenyl)-3-nitro-benzenemethaneamine (5c) displayed moderate anti-fungal activities against all five different fungi.

Acute Toxicity of Crude Anti-fungal Compounds Produced by Lactobacillus plantarum AF1 (Lactobacillus plantarum AF1이 생성한 조항진균 물질의 마우스에 대한 급성독성)

  • We investigated the acute toxicity from a single dose of crude anti-fungal compounds produced by Lactobacillus plantarum AF1, a lactic acid bacterium isolated from kimchi, on ICR male and female mice in vivo. The test article was orally administered once to both sexes of mice. The mortality rate, clinical findings, autopsy findings, and body weight changes were monitored daily for 14 days. In the oral acute toxicity test, male and female mice were gavaged with four doses (5, 50, 300 or 2,000 mg/kg) of the crude anti-fungal compounds. The oral $LD_{50}$ of the crude anti-fungal compounds was higher than 2,000 mg/kg. No significant changes in general conditions, body weights, clinical signs, or appearance of gross lesions were observed. In conclusion, our results suggest a low toxicity and no-adverse-effects from crude anti-fungal compounds produced by Lactobacillus plantarum AF1 up to 2,000 mg/kg via the oral route.

Effects of Boiled Solution from Chrysanthemum Cineriaefolium Visiani Inunction Therapy on Dermatomycosis in Rabbits (토끼의 피부진균증에 대한 제충국화자비액의 도포치료효과)

  • This study was carried out to investigate the anti-fungal effect of Pyrethri Flos Boiled Solution (PFBS) from Chrysanthemum Cinerariaefolium Visiani on Microsporum canis, Microsporum gypseum and Trichophyton Mentagrophytes with 0.5 ml, 1ml, 2 ml. In comparative drugs, on Sabouraud's glucose agar medium to 2 ml respectively and PFBS on the rabbit skin lesions, in vivo experiments, were observed. The results obtained were as follows : PFBS slightly suppresed the growth of the fungi in 0.5 ml and markedly it in 1 ml and 2 ml. In comparative drugs, benzylbenzoate represented similar anti-fungal action in 2 ml compared with PFBS. While, salicylic acid and boric acid represented less suppression of growth of the fungi than PFBS in 0.5 ml. The effects of PFBS on the rabbit skin lesions inoculation with dermatophytes was almost normally recorved about Microsporum gypseum, Trichophyton mentagrophytes except Microsporum canis after 8 days.

Review of Randomized Controlled Trials of Oriental Medicine for Recurrent Vulvovaginal Candidiasis (재발성 칸디다성 외음질염의 한약치료 효과에 관한 무작위 대조 연구 문헌고찰)

  • Objectives: The purpose of this study is to review and evaluate the effectiveness of oriental medicine per oral for recurrent vulvovaginal candidiasis (RVVC). Methods: We searched articles from Korean journal databases including Journal of Korean Obstetrics and Gynecology, Korean studies Information Service Studies, and Chinese National Knowledge Infrastructure, Cochrane Library, PubMed. Searched keywords were "재발성 칸디다성 외음질염", "recurrent vulvovaginal candidiasis", "㚆发性外阴阴道假丝酵母菌病", "中藥". Results: 10 randomized controlled trials with 700 patients were identified and reviewed. 8 studies compared combination of oriental medicine and anti-fungal agent with anti-fungal agent, and 6 of them reported that treatment group (TG) showed statistically higher total effective rate or cure rate and lower recurrence rate. One study compared oriental medicine and anti-fungal agent and TG showed statistically higher total effective rate and lower recurrence rate after 1 month. One study compared combination of oriental medicine and lacto bacillus capsule with lacto bacillus capsule, and TG showed statistically higher total effective rate and lower positive fungul test rate after 6 months. No severe adverse response was reported. Conclusions: Our review found that oriental medicine per oral is effective for alleviating symptoms, lowering recurrence rate and positive fungal test in RVVC patients. We recommend standardized randomized controlled trial guideline should be made to obtain stronger evidence and well designed trials with larger sample sizes are needed.

Draft Genome Sequence of Alternaria alternata JS-1623, a Fungal Endophyte of Abies koreana

  • Alternaria alternata JS-1623 is an endophytic fungus isolated from a stem tissue of Korean fir, Abies koreana. Ethyl acetate extracts of culture filtrates exhibited anti-inflammatory activity in LPS induced microglia BV-2 cell without cytotoxicity. Here we report a 33.67 Mb sized genome assembly of JS-1623 comprised of 13 scaffolds with N50 of 4.96 Mb, and 92.41% of BUSCO completeness. GC contents were 50.97%. Of the 11,197 genes annotated, gene families related to the biosynthesis of secondary metabolites or transcription factors were identified.
