Anti-inflammatory and Anti-oxidative Effects of Alpiniae Oxyphyllae Fructus Hot Aqueous Extract in Lipopolysaccharide (LPS)-stimulated Macrophages

  • Objectives : Alpiniae oxyphyllae Fructus (AOF) is an herbal medicine, which has been used for the treatment of fatigue, chills, and poor physical conditions. The objective of this study was to investigate the anti-inflammatory and anti-oxidative effects of AOF hot aqueous extract. Methods : The cytotoxicity of AOF extract was evaluated using the MTT assay. Nitric oxide (NO) production was measured by the Griess reaction. Prostaglandin $E_2$ ($PGE_2$) production was measured by a commercial competitive enzyme immunoassay. Cytokine production (IL-1tion co6, and TNF- F- was measured by ELISA. The anti-oxidative effect of AOF extracts was measured by the DPPH method. Polyphenol and flavonoid contents were measured by Folin-Ciocalteu's phenol reagent and aluminum chloride, respectively. Results : AOF hot aqueous extract did not show toxicity at doses of 25, 50, 100, and $200{\mu}g/mL$. AOF extract significantly inhibited NO production at doses of 100 and $200{\mu}g/mL.PGE_2$ production was inhibited by AOF extract treatment at doses of 100 and $200{\mu}g/mL$. AOF extracts reduced IL-6 production in a dose-dependent manner. IL-1ent maTNF- F- 1ent mannerd IL-6 production in uction at doses of 100 and ${\mu}g/mL$. The DPPH free radical scavenging capability was above 50% at $200{\mu}g/mL$. Conclusion : This study suggests that AOF hot aqueous extract may exert anti-inflammatory and anti-oxidative effects in a dose-dependent manner. Further studies are required for validating the safety and efficacy of AOF.

Effects of taurine and ginseng extracts on energy metabolism during exercise and their anti-fatigue properties in mice

  • BACKGROUND/OBJECTIVES: Ginseng extract (GSE) and taurine (TR) are widely used antifatigue resources in functional foods. However, the mechanism underlying the antifatigue effects of GSE and TR are still unclear. Hence, we investigated whether GSE and TR have synergistic effects against fatigue in mice. MATERIALS/METHODS: L6 cells were treated with different concentrations of TR and GSE, and cell viability was determined using 2-(4-iodophenyl)-3-(4-nitrophenyl)-5-(2,4-disulfophenyl)-2H-tetrazolium. Oxidative stress was analyzed by immunocytochemistry using MitoTrackerTM Red FM and an anti-8-oxoguanine antibody. Respiratory gas analysis was performed to investigate metabolism. Expression of an activated protein kinase was analyzed using immunohistochemistry. Gene expression of cluster of differentiation 36 and pyruvate dehydrogenase lipoamide kinase isozyme 4 was measured using reverse transcription-polymerase chain reaction. Mice were orally administered TR, GSE, or their combination for 30 days, and then fatigue-related parameters, including lactate, blood urea nitrogen, and glycogen, were measured after forced swimming. RESULTS: TR and GSE reduced oxidative stress levels in hydrogen peroxide-stimulated L6 cells and enhanced the oxygen uptake and lipid metabolism in mice after acute exercise. After oral administration of TR or GSE for 30 days, the fatigue-related parameters did not change in mice. However, the mice administered GSE (400 mg/kg/day) alone for 30 days could swim longer than those from the other groups. Further, no synergistic effect was observed after the swimming exercise in mice treated with the TR and GSE combination for 30 days. CONCLUSIONS: Taken together, our data suggest that TR and GSE may exert antifatigue effects in mice after acute exercise by enhancing oxygen uptake and lipid oxidation.

  • Objective : Erysipelas is an acute inflammation caused by pyogenic bacteria. This mainly involves the upper part of dermis. It begins as erythematous patches with tenderness, followed by fever, headache, chills and fatigue etc. It may results in edema, obstruction of lymphatics and sepsis. So this experiment is carried out for test whether the Bojeasodok-um subtracted Scrophulariae Radix, Lasiosphaera seu Calvatia, Isatidis Radix added indigo Naturalis, Lithospermi Radix have an anti-inflammatory effect and have suppression effect on immunocyte in the state of inflammation which induced by Erysipelas. Method : Experimental animals made use of 4-5 week-age(weight 20-25g) ICR(male) mouse. In the breeding farm, the lighting time was controlled from 7:00 am till 7:00 pm, the temperature was controlled So we concluded that BS is prospected as an anti-inflammatory agent to cure inflammation induced bywithin 18-23$23^{\circ}C$ and water and food were not limited.The freezing lyophilization powder which were extracted from Bojesodok-Um divided low dose group(200mg/kg-BSL) and high dose group(500mg/kg-BSH) and after melting in water, it was orally administered to the mouse. Compared with inflammation induced group which were induced by triggering-inflammation reagent Carageenan and Zymosan and normal contrast group, we measured the edema decrement effect,macrophage and spleen cell activation. Result : 1. BS has suppress inflammatory reaction induced by Carageenan. 2. BS has suppress increasing activation of abdominal cavity macrophage in the Carageenan and Zymosan induced inflammation. 3. BS has suppress increasing activation of spleen cell in the Carageenan and Zymosan induced inflammation. Based on the above result, BS was improved its suppression effect to the inflammatory reaction through the suppression of spleen cell and macrophage activation. So we concluded that BS is prospected as an anti-inflammatory agent to cure inflammation induced by Erysipelas.

공진단(拱辰丹)의 성분 분석 및 갱산화(坑酸化) 작용에 미치는 영향 (An analysis of the Gongjindan's ingredients and its efficacy on anti-oxidation)

  • Objectives : For the congenital feeble, the Gongjindan is useful medicine. The experiment is to estimate the value of the Gongjindan as therapeutic agent preventing against aging with an analysis of the ingredients and the bio-activating effects by enzymologic methods. Methods : General ingredients' of the Gongjindan's extract were analyzed first and the quantitative analysis of a reducing sugar, a soluble protein, an amino acid and minerals was made. The Gongjindan, which is extracted, concentrated, and freeze drying with water, ethanol and chloroform, have got applied for the experiment. The effects on electronic donating ability, SOD-like activity, nitric oxide inhibition, xanthine oxidase inhibition, whitening effect have been investigated in the physiological activity measurement of function experiment. Results : The contained amino acid, in order of high amount, is Arginine, Alanine, Glutamic acid, Proline and the contained free amino acid is Glutamic acid, Leucine, Lysine, Phenylalanine. The derivative of free amino acid is ${\gamma}-Aminoisobutyric$ acid, Phosphoserine, Taurine. And the Gongjindan is containing 13 species of minerals in order of Ca>K>Na>Mg>Fe>AI>Mn. Then, to assure of the Gongjindan's capability of anti-oxidation, these following subjects-polyphenol, electronic donating ability, SOD-like activity, nitric oxide inhibition, xanthine oxidase inhibition, tyrosinase inhibition- are analyzed and show high activity especially the most in ethanol extracts. Conclusion : With this analysis of ingredients, the Gongjindan is containing many materials functioning as anti-oxidation, anti-aging, anti-fatigue, neurotransmitter and immune agent. Moreover, In every water, ethanol, chloroform extracts, the Gongjindan's capability of anti-oxidation is confirmed so that we can apply to patients' treatment clinically.

경옥고 섭취가 고등학교 축구선수의 운동수행능력 향상 및 피로 회복에 미치는 영향 (Effect of Kyungohkgo on Aerobic Capacity and Anti-fatigue in High School Soccer Players)

  • This study was designed to investigate the effects of Kyungohkgo to improve aerobic capacity and eliminate exercise-induced fatigue in high school soccer players. Twenty four subjects were participated and randomly assigned into two groups [KG, Kyungohkgo group (n=12); PG, Placebo control group (n=12)]. Two groups were completed treadmill exercise protocol using graded exercise test at before and after experimental treatment of 4 weeks. The $VO_2$max and endurance time were measured by gas analysis and heart rate (HR) was measured by polar system at pre, post 0, post 5, post 15, post 30 and post 60 minutes. Blood samples were collected to analyze blood components. 1. The $VO_2$max was significant increased in the group of after intake Kyungohkgo compared to the group of after intake placebo (p<.05). 2. The HR was significant decreased in the group of after intake Kyungohkgo compared to the group of after intake placebo during recovery time at post 5 mins(p<.05), 30 mins(p<.01), 60 mins(p<.01). 3. Weight, body mass index, percent body fat, anaerobic threshold, endurance time, blood lactate concentrate, lactate dyhydrogenase, creatine kinase, serum glutamic oxaloacetic transaminase, serum glutamic pyruvic transaminase, other energy sources(total-cholesterol, triglyceride, high density lipoprotein-cholesterol, low density lipoprotein-cholesterol, creatinine) and electrolyte (Na, K, Cl) were shown no significant differences between groups. These results suggested that Kyungohkgo can be used as ergogenic aids to improve aerobic capacity and eliminate exercise-induced fatigue.

단기(短期) 파극(巴戟) 투여(投與)가 생쥐의 근피로(筋疲勞) 및 혈액(血液) 성분(成分)에 미치는 영향(影響) (Effect of Morindae officinalis Radix extract on muscle fatigue and blood component)

  • Purpose: This study was conducted to investigate the muscle anti-fatigue effects of Morindae officinalis Radix liquid extract. Methods: 4-6 weeks old ICR rats are used in the study, and we administered the extracts powder of Morinda officinalis concentration of 1, 10, 100mg/0.3 ml to each to rats(MO group) once a day for each 30 and 60 days. After 30 and 60 days, we examined the plasma concentraion of lactate dehydrogenase, glucose. And we measured the persistent time of swimming exercise test and conducted grip strength test Results: We found that there are no significantly differences between the control and the treatment group in the plasma concentration of activity level of lactate dehydrogenase. And we found that in 30days group, level of glucose concentration is significant. In swimming exercise test, as administration time and concetration growed up, swimming time is increased. And in grip strength test, as concentraion is growed up, grip strength is increased. Conclusion: This study show that Morinda officinalis is effective against muscle fatigue.

메꽃(Calystegia pubescens Lindl.) 추출물의 항균, 항산화 및 항염증 효과에 관한 연구 (Study on the Anti-bacterium, Antioxidant and Anti-inflammatory Effect of Calystegia pubescens Lindl. Extracts)

  • Calystegia pubescens Lindl. (C. pubesens)는 전통적으로 이뇨, 피로 및 혈당 강하 효과로 사용되어 온 한국 토종 식물이다. C. pubesens는 이전 연구에서 항산화 및 미백 효과를 보였지만 항균 및 항염증 특성에 대한 연구가 제한적이고 기능성 소재로서의 입증이 부족하다. 본 연구에서는 70% 에탄올로 추출한 C. pubesens의 잎(CPL)과 줄기(CPS) 추출물의 항균, 항산화 및 항염증 효과를 조사했다. CPL은 CPS보다 여드름균, 황색포도상구균, 대장균과 녹농균에 대해 우수한 항균 효과를 보였다. 또한 CPL은 CPS보다 DPPH 및 ABTS+ 라디칼 소거 분석에서 더 높은 항산화 활성을 나타냈다. 따라서 CPL은 CPS에 비해 우수한 항균 및 항산화 효과를 보였다. 이러한 결과를 바탕으로 RAW264.7 대식세포를 이용한 CPL의 항염증 효과를 확인하기 위한 추가 연구를 진행하였다. 지질다당류(LPS) 자극에 의해 생성된 산화질소(NO)는 CPL에 의해 감소되었다. 또한, LPS에 의해 증가된 iNOS의 단백질 및 mRNA 발현은 CPL에 의해 감소되었으며, 이는 NO 생산과 동일한 경향을 나타내었다. 결론적으로 본 연구는 CPL이 CPS에 비해 우수한 항균 및 항산화 효과를 가지고 있음을 확인하였으며, LPS에 의해 유도된 iNOS 발현 및 NO 생성 억제를 통해 CPL의 항염증 효과를 입증했다. 본 연구 결과를 바탕으로 CPL의 기능성 소재로서의 가능성을 제시한다.

홍경천(紅景天)추출물(KH101)이 강제유영 흰쥐의 피로회복에 미치는 영향 (Effects of Rhodiola rosea (KH101) on Anti-fatigue in Forced Swimming Rats)

  • Objectives : Rhodiola rosea has been used in herbal medicine to treat various conditions, such as antimelancholia, antifatigue, improvement of work competence and prevention of altitude sickness. In this study, we investigated effects of Rhodiola rosea extract (KH101) on fatigue in forced swimming rats. Methods : Sprague-Dawley rats were induced with fatigue by forced swimming, then rats in each group were treated with KH101. We observed changes of glucose, LDH and cortisol in serum and LDH, glycogen, hexokinase, citrate synthase MDH, SDH and CK in muscle. Results : Obtained results were as follows: 1. Continuance times of exercise significantly increased in all groups at day 1, in the 50 mg/kg concentration group at day 2, in all groups at day 3 and in the 50 mg/kg conc. group at day 4. 2. In serum, glucose significantly decreased in all concentration groups. 3. In the soleus muscle, LDH significantly decreased in the 50 mg/kg concentration group. HK significantly decreased in the 100 mg/kg conc. group. SDH significantly increased in the 100 mg/kg conc. MDH were significantly decreased in all conc. groups. 4. In the gastrocnemius muscle, HK significantly decreased in all concentration groups, while MDH significantly increased all conc. groups. Conclusions : It is concluded that the KH101 has and anti-fatigue effect in rats. Additional studies are needed to find the mechanism of the association between each single herb.

양파(Allium cepa L.)의 항고지혈 및 항산화 활성에 관한 연구 (Studies on Antihyperliperlipemic and Antioxidant Activity of Allium cepa L.)

  • This Study was attempted to investigate the effect of Allium cepa L. (onion) on the activities of GOT and GPT, the levels total lipids, triglyceride, total cholesterol, phospholipid and ${\beta}-lipoprotein$ in the serum and the increase ratio of body and liver weight in the experimentally induced hyperlipemic rats. The activities of S-GOT and S-GPT were significantly decreased in dose of onion juice 3 ml and 5 ml/head as compared to the high lipid-diet control group. The levels of total lipids. Triglyceride, total cholesterol and phospholipids were significantly decreased in dose of onion juice 3 ml and 5 ml/head as compared to the control group, respectively. However, the level of ${\beta}-lipoprotein$ was significantly decreased in dose of onion juice 3 ml/head. The levels of triglyceride and total cholesterol in the liver were significanatly decreased in dose of onion juice 3 ml/head. Increase ratio of the body and liver weight were significantly decreased in dose of onion juice 3 ml/head. Anti-fatigue activity of onion juice were studied in mice using the swimming performance method. The potencies of anti-fatigue acti-vities was significantly found in 1.2 ml/head. p.o. of onion juice. The methanol extract and juice extract of onion was significantly found to have an antioxidative activity on the air oxidation of linoleic acid as compared to the 3-butyl-4-hydroxyanisole and ${\alpha}-tocopherol$.

경옥고(瓊玉膏)의 성분 분석 및 추출물별 항산화(抗酸化) 효능 비교 (An Analysis of the Gyungokgo's Ingredients and a Comparison Study on Anti-oxidation Effects According to the Kinds of Extract)

The derivative of free amino acid is Phosphoserine, Carnocine, ${\gammer}$-Aminoisobutyric acid. And the Gyungokgo contains 14 species of minerals, K>Na>Ca>Mg>Fe>Al>Mn. Then, to assure of the Gyungokgo's anti-oxidation, these following subjects -polyphenol, electronic donating ability, SOD-like activity, nitric oxide inhibition, xanthine oxidase inhibition, tyrosinase inhibation- are analyzed and show high activity especially the most in chloroform extracts, (every ingredients written by the order of high amount) Conclusions : The Gyungokgo contains many materials functioning as anti-oxidation, neurotransmitter, anti-fatigue and immune agent.
