Screening of Medicinal Plants on the Anti-inflammatory Effect and Active Component (전남지역 자생식물들의 소염활성검색 및 활성화합물)

  • The anti-inflammatory effect of the methanolic extracts of some medicinal plants was investigated on the hydrolysis of N-benzoylarginine-p-nitroanilide by trypsin in vitro, the writhing syndrome induced by acetic acid in mice, and the carrageenin-induced paw edema in rats. Among tested medicinal plants, the methanoilc extract of Angelica keiskei, Rosa rugosa and Cudrania tricuspidata showed significant inhibitory effect. And we also found that adenosine, isolated from A. keiskei was identified as one active components.

  • A rare flavonol glycoside identified as $eupalitin-3-O-{\beta}-D-glucoside$ (I) was isolated from Tephrosia spinosa (Leguminosae) and its anti-inflammatory activity was evaluated against carrageenin induced paw edema. The compound exhibited significant activity when compared to the standard drug indomethacin.

  • Synurus deltoides was previously found to possess significant anti-inflammatory activity especially against chronic inflammation, and strong analgesic activity in vivo. In this study, new anti-inflammatory formulation containing S. deltoides extract as a major ingredient was prepared and in vivo activity was evaluated. The plausible action mechanism was also investigated. The new formulation (SAG) contains 1 part of S. deltoides extract, 0.9 part of Angelica gigas extract and 0.9 part of glucosamine sulfate (w/w). SAG inhibited dose-dependently edematic response of arachidonic acid (AA)- and 12-O-tetradecanoyl 13-acetate (TPA)-induced ear edema in mice, which is an animal model of acute inflammation. SAG showed 44.1 % inhibition of AA-induced ear edema at an oral dose of 50 mg/kg. In an animal model of chronic inflammation, SAG clearly reduced the edematic response of 7 -day model of multiple treatment of TPA (38.1 % inhibition at 200 mg/kg/day). Furthermore, SAG (50-800 mg/kg/day) as well as S. deltoides extract (285 mg/kg/day) significantly inhibited prostaglandin $E_2$ production from the skin lesion of the animals of 7-day model. These results were well correlated with in vitro finding that SAG as well as S. deltoides extract reduced cyclooxygenase (COX)-1- and COX-2-induced prostanoid production, measured in mouse bone marrow-derived mast cells. Therefore, these results suggest that SAG possesses anti-inflammatory activity in vivo against acute as well as chronic inflammatory animal models at least in part by inhibition of prostaglandin production through COX-1/COX-2 inhibition. And COX inhibition of SAG is possibly contributed by S. deltoides extract among the ingredients. Although the anti-inflammatory potencies of SAG were less than those of currently used anti-inflammatory drugs, this formulation may have beneficial effect on inflammatory disorders as a neutraceutical.

Study on the Effect of Chaenomelis Fructus.Herba Chelidonii.Clematis Florida Thunb on Inflammation and Liver Function of Rats with Arthritis (관절염 백서의 염증과 간기능에 대한 목과.백굴채.위영선(木瓜.白屈菜.威靈仙)의 효능연구)

  • The purpose of this study is to observe the effects of Chaenomelis Fructus Herba Chelidonii Clematis Florida Thunb mixed water extract on the inflammatory edema induced by Found's complete adjuvant and on different blood volume, and then to demonstrate its efficiencies as well as its safety. We injected Chaenomelis Fructus Herba Chelidonii Clematis Florida Thunb mixed water extract on the inflammatory edema induced by Found's complete adjuvant once daily at the same time for a week, then observed the paw-edema, the counts of the blood WBC, Serum total protein, Serum total bilirrubin, SGOT and SGPT The result obtained is as follows : Inflammatory edema on the right plantar in both sample groups showed a significant reduce compared with that in the control group. The counts of WBC in both sample groups showed a significant decrease compared with that in the control group. The volume of serum total protein in the Sample A group showed a significant decrease compared with that in the control group. The volume of serum total bilirrubin, GOT and GPT in both sample groups was not changed compared with that in the control group. As a result of this experiment, it is concluded that Chaenomelis Fructus Herba Chelidonii Clematis Florida Thunb mixed water extract showed that therapeutic effect on the anti-inflammation on arthritis; otherwise it showed non-toxic effect on the liver function.

The Effects of Triamcinolone Acetonide by Iontiophoretic Transdermal Delivery on Inflammation Induced Rat (백서에서 유발된 염증에 대한 Triamcinolone Acetonide 이온도입 경피투과 효과)

  • This study investigated the effects of triamcinolone acetonide by iontophoretic transdermal drug delivery on anti-inflammatory action into the rats and which had carrageenan-induced hyperalgesia and edema in the feet, trauma-induced tissue damage in the thigh. Each group was treated under the fellowing conditions. 1. Group I : Control group 2. Group II : Application of direct current 3. Group III : Application of 0.1$\%$ triamcinolone acetonide solution 4. Group IV : Iontophoresis of 0.1$\%$ triamcinolone acetonide solution The degree of anti-inflammation was evaluated by the paw withdrawal latency, the change in volume of foot the change of paw edema, histological change in rats. 1. In paw withdrawal latency, group IV showed the most significant therapeutic effect than the other groups at 0, 3, 6 and 9 hours(p < 0.001). 2. In paw edema experiment in the foot, group IV showed the most significant effect than group I at 0, 3, 6 and 9 hours. It meant that there was effective anti-inflammatory reaction in group I (p < 0.001). 3. In the light microscopic observation, group IV showed the most significant reduction of haemorrhage, hyperemia and infiltrative inflammation. From the results, the iontophoresis with triamcinolone acetonide is more effective than using each groups. It is one of the effective physical agent which delivered large molecular weight drug into the body. The continuous study is needed for many interesting issues of iontophoretic transdermal drug delivery in new future.

Effects of Scolopendra subspinipes multilans Herb-Acupuncture on the Carrageenan induced arthritis in rats (오공약침(蜈蚣藥針)이 Carrageenan으로 유발(誘發)된 흰 쥐의 관절염(關節炎)에 미치는 영향(影響))

  • This study was designed to check of Scolopendra subspinipes multilans Herb-Acupucture on effect of anti-inflammtory function, anti-febrile function and analgestic function in carrageenan-induced arthritic rat. Each of 8 rat were classified into control, sample group. Control group were inject by normal saline and Sample group were injected by Scolopendra subspinipes multilans. Herb-Acupuncture. After elicitating edema and inflammation of Sprgue Dawely(SD) rats by injection of 1% Carrageenan 0.1ml, then the rate of increase of paw edema, CRP, ESR, WBC were checked and rectal-temperature, topical temperature was checked about anti-febrile. writhing syndrome was checked about analgesic. The results were as follows 1. In the sample group, the rate of increase of paw edema was significantly decreased as compared with thatof control group(P<0.05) 2. In the sample group, the rectal-temperature was non-significantly decreased as compared with that of control group (P<0.05) 3. In the sample group, CRP and WBC was significantly decreased as compared with that of control group, but ESR was not.(P<0.05) 4. In the sample group, opical-temperature was significantly decreased as compared with that of control group (P<0.05) 5. In the sample group, analgestic function was not sigiflcntly effected as compared with that of control group(P<0.05) According to the above result, it can be concluded Scolopendra subspinipes multilans Herb-Acupucture showed the treatment effects on the artifical arthritis resulted from carrageenan in rat and it is suggested that more interest and study in the mechanism and clinical use were needed.

  • Angelica dahurica (Umbelliferae) grows in China, Japan, Russia, and Korea. The root of Angelica dahurica has been used as a traditional folk medicine to treat headache and toothache. In this study, the effects of the aqueous extract of Angelica dahurica on acetic acid-induced abdominal pain, carrageenan-induced edema, and thermal hyperalgesia were investigated using mice and rats. The present results showed that the aqueous extract of Angelica dahurica inhibited acetic acidinduced abdominal pain in mice and reduced carrageen-induced edema in rats. The present study showed that the aqueous extract of Angelica dahurica possesses anti-inflammatory and analgesic effects.

The Effect of Hydroalcoholic Gel System on Skin Permeation of Piroxicam and its Anti-inflammatory Activity (피록시캄의 피부투과 및 소염효력에 대한 알코올성 하이드로겔 시스템의 영향)

  • These studies were designed to determine the effect of hydroalcoholic gel system (lower alkanol concentration: 40-60%) compared to general hydrogel system (lower alkanol concentration: 10-35%) on transdermal delivery of piroxicam and its anti-inflammatory activity. Piroxicam was incorporated into a hydroalcoholic gel and a hydrogel containing polymers, solvents, and cosolvents. The pH of gel was about 6.3-7.3 and the solvent mixtures were composed of water and various concentrations of ethanol (35, 40, 50, and 60%). For the in vitro study, the skin permeation of piroxicam from the gel formulations was investigated using Franz modified diffusion cells fitted with hairless mouse skin. For the in vivo study, the anti-inflammatory activity of hydroalcoholic gel was compared to other commercial products (piroxicam hydrogel and ketoprofen hydrogel) in rat and human. The anti-inflammatory activity was determined using carrageenan induced foot edema model in rat. For the clinical study, it was evaluated from determining efficacy and acceptability with 98 patients suffering from musculoskeletal pain. A novel piroxicam hydroalcoholic gel was successfully formulated in the range of 40-50% of ethanol as solvent, more than 10% of propylene glycol, 5% of $Transcutol^{\circledR}$ and 1 % of benzyl alcohol. The skin permeation of piroxicam using hydroalcoholic gel system was greater than that of general hydrogel system $(flux\;:\;139.1-148.2\;{\mu}g/cm^2/hr\;vs.43.0-84.5 {\mu}g/cm^2/hr)$ in vitro.   • Major symptoms of al1ergic rhinitis are nasal obstruction, sneezing and watery rhinorrhea. Onpyetang has been used to treat for nasal obstruction, which is one of the symptoms of allergic rhinitis. This Experimental study was done to research effects of Onpyetang and transformed Onpyetang(Allii Radix is deducted from Onpyetang) on the anti-allergic effects. We have studied the vascular permeability response induced by serotonin and histamine, the contact dermatitis response induced by picryl chloride, the delayed type hypersensitivity response to SRC, the mice paw edema induced by carrageenin, the writhing syndrome induced by $0.7\%$ acetic acid, and the rectal temperature in febrile rats induced by yeast. The results were as follows : 1. In the vascular permeability response to intradermal serotonin and histamine. Onpyetang proved significant inhibitory effect(P<0.05, p<0.001) But transformed Onpyetang proved significant inhibitory effect only to histamine(P<0.05) 2. In the contact dermatitis response induced by picryl chloride, Onpyetang and transformed Onpyetang proved significant inhibitory effect.(P<0.05, P<0.05) 3. In the delayed type hypersensitivity response induced by SRC, Onpyetang and transformed Onpyetang proved significant inhibitory effect on mice paw edema.. (P<0.05, P<0.05) 4. In the delayed type hypersensitivity response induced by SRC, Onpyetang proved significant inhibitory effect to serum IgE.(P<0.01) But transformed Onpyetang proved insignificant inhibitory effect on serum Ig E. 5. In the mice paw edema induced by carrageenin, Onpyetang and transformed Onpyetang proved significant anti-inflammatory effect. (P<0.01, P<0.01) 6. In the writhing syndrome induced $0.7\%$ acetic acid, Onpyetang and transformed Onpyetang proved significant analgestic effect. (P<0.01, P<0.01) 7. In the rectal temperature in febrile rats induced by yeast, Onpyetang and transformed Onpyetang proved significant anti-pyretic effect.(P<0.001, P<0.01) according to this result Onpyetang was conclude to be effective on anti-allergic, anti-pyretic, anti-inflammatory and analgestic action. but transformed Onpyetang(Allii Radix is deducted from Onpyetang) was not effective on the vascular permeability response to intradermal serotonin and increasing Ig E of delayed type hypersensitivity response induced by SRC. In addition, transformed Onpyetang is not effective as Onpyetang. More study should be done about the role of Allii Radix.

  • To study the effect of Sopoungwhalwheol-tang and Sopoungwhalwheol-tang with cheon chung on the "Tong poung" "yeouk Geul poung" inoriental medicine (Rheumatoid arthrits) measure cure effect on the Analgesic, Anti-inflammatory anti-phlogistic and concentration of albumin's Test. The results are Summarized as follows, 1. Sopoungwhalwheol-tang liquid reduced significantly the pain by acetic acid in accordance with the increase of double dosages, and Sopoungwhalwheol-tang with cheon chung (which was similar with Aspring Group) was more marked effective. 2. Sopoungwhalwheol-tang with cheon chung reduced the edema by carragenine and at 60 minutes, two groups all reduced the edema and at 180 minutes, Sopoungwhalwheol-tang was recognized some effectiveness and Sopoungwhalwheol-tang with cheon chung was recognized excellent effectiveness. 3. Sopoungwhalwheol-tang with cheon chung has more continued Antiphlogistic effect against the fever by yeast which was fallen after 60 minutes of prescription of medicine than Sopoungwhalwheol-tang. 4. Sopoungwhalwheol-tang with cheon chung was more effective against the concentration of albumin by $1\%$ bovine serum albumin than Sopoungwhalwheol-tang. 5. According to the above experiments. Sopoungwhalwheol-tang with cheon chung has greater anti-inflammatory, antiphlogistic Analgestic effect than Sopoungwhalwheol-tang.
