손바닥선인장(Opuntia humifusa) 줄기 추출물의 항당뇨 효과 (Anti-diabetic Effect of Opuntia humifusa Stem Extract)

  • Opuntia humifusa known as the Eastern prickly pear have been used as a treatment of burns, diarrhea, asthma, rheumatism, gonorrhea, and diabetes in alternative medicine. O. humifusa is widely cultivated in the middle and southern provinces of Korea and distributed in North America. The aim of this study is to investigate anti-diabetic effect of O. humifusa stem (OHS) water or 80% MeOH extract using 3T3-L1 adipocytes and db/db mice animal models. OHS 80% MeOH extract at a dose of $250{\mu}g/ml$ significantly increased the glucose uptake and lipid accumulation compared with the control in 3T3-L1 adipocytes. Blood glucose, plasma total cholesterol and triglyceride levels were significantly reduced by oral treatment of OHS 80% MeOH extract (200 mg/kg BW) for 6 weeks in db/db mice. Also, the oral treatment of OHS 80% MeOH extract slightly changed the plasma insulin and insulin resistance levels in db/db mice, but were no significance in comparison to control. Glucose transporter(GLUT)4 expressions of adipose tissue and muscle were significantly increased more than that in the control. Therefore, these results suggest that OHS 80% MeOH extract inhibits the blood glucose level through regulation of lipid profile, insulin resistance, and GLUT4 expression in db/db mice and its diabetic effect is effective more than water extract.

Anti-Cancer Activity of the Flower Bud of Sophora japonica L. through Upregulating Activating Transcription Factor 3 in Human Colorectal Cancer Cells

  • The flower buds of Sophora japonica L (SF), as a well-known traditional Chinese medicinal herb, have been used to treat bleeding-related disorders such as hematochezia, hemorrhoidal bleeding, dysfunctional uterine bleeding, and diarrhea. However, no specific anti-cancer effect and its molecular mechanism of SF have been described. Thus, we performed in vitro study to investigate if treatment of SF affects activating transcription factor 3 (ATF3) expression and ATF3-mediated apoptosis in human colorectal cancer cells. The effects of SF on cell viability and apoptosis were measured by MTT assay and Western blot analysis against cleaved poly (ADP-ribose) polymerase (PARP). ATF3 activation induced by SF was evaluated using Western blot analysis, RT-PCR and ATF3 promoter assay. SF treatment caused decrease of cell viability and increase of apoptosis in a dose-dependent manner in HCT116 and SW480 cells. Exposure of SF activated the levels of ATF3 protein and mRNA via transcriptional regulation in HCT116 and SW480 cells. Inhibition of extracellular signal-regulated kinases (ERK) 1/2 by PD98059 and p38 by SB203580 attenuated SF-induced ATF3 expression and transcriptional activation. Ectopic ATF3 overexpression accelerated SF-induced cleavage of PARP. These findings suggest that SF-mediated apoptosis may be the result of ATF3 expression through ERK1/2 and p38-mediated transcriptional activation.

청황산(靑黃散)의 백혈병(白血病)에 대(對)한 항종양효능(抗腫瘍效能) 연구(硏究) (A literal study of anti-tumor effects of chunghwangsan for leukemia)

  • In the literatual study of anti-tumor effects of chunghwangsan for leukemia, the results were as follows. 1. Chunghwangsan is composed of Indigo naturalis and Realgar. The composing rate is 9 : 1 and it is made into capsule or piece. The basic administration is 0.3g per day and could increase the quantity each day. 2. The effects of Chunghwangsan is expelling toxin and colling, colling blood to detumescence, drying wetness and anticancer are. So it can be used to treat AML, CML and lymphoma. 3. The anticancer component of Indigo naturalis is indirubin which has the effects of suppression the transplanted tumor, activating the phagocyte of macrophage, promoting the maturation of myeloblast to improve cure rate of CML. The anticancer component of Realgar is $As_2O_3$ which has the direct cellular toxicity for leukemia cell. 4. In viewpoints of oriental medicine, leukemia is malignant myeloid neoplasia in which pathogen invade to shaoyin(少陰). So Chunghwangsan which is expelling toxin and colling, colling blood to detumescence, drying wetness and anticancer is effective to leukemia. 5. In clinical reports, Chunghwangsan is often used in CML, and also used in AML, lymphoma and so on. 6. Chunghwangsan is cool-natured, so we must carefully pay attention to pregnant women and hematsdthenic patients. The main side effects are nausea, bone marrow pain, diarrhea, polydefecation, hematokezia and purpora. We sometimes take invigorating stomach medicine to prevent the side effects. 7. If we continuously develop Chunghwangsan and therapy for leukemia with syndrome differentiation. we can improve the response and cure rate for leukemia in the future.

Probiotic Functional Dairy Foods and Health Claims: an Overview

  • The commercial interests in functional foods containing probiotics are gaining significance in view of increasing studies on their role in digestive tract. Probiotic dairy foods containing health-promoting bacteria are important segment of functional food market. Various health benefits have been attributed to specific strains of lactic acid bacteria or food containing these probiotic cultures. Probiotic-containing foods are considered to improve general gut health and natural defenses of body, and lower blood cholesterol level. Specific probiotic microbes can alleviate or prevent diverse intestinal diarrhea-inducing disorders, cause prophylaxis of intestinal and urogenital infections, inhibit mutagenicity of intestinal contents, and reduce incidence of intestinal tumors. Recent increasing evidences on health effects of probiotics have triggered consumer interest in this category of functional foods. Rational approach needs to be applied in selection of strains for probiotic preparation to achieve required functionality. Present article focuses on some prominent probiotic candidates and criteria for their inclusion in functional food sector. Various health claims of probiotics on gastrointestinal disorders, anticarcinogenic effects, and anti-cholestrimic effects, and possible mechanistic explanations for their functionality are highlighted.

Overlapping Guillain-Barr$\acute{e}$ syndrome and Bickerstaff's brainstem encephalitis associated with Epstein Barr virus

  • A flaccid tetraparesis in Bickerstaff's brainstem encephalitis (BBE) is presumed to be a sign of overlapping Guillain-Barr$\acute{e}$ syndrome (GBS). In addition, BBE and Fisher syndrome, which are clinically similar and are both associated with the presence of the immunoglobulin G anti-GQ1b antibody, represent a specific autoimmune disease with a wide spectrum of symptoms that include ophthalmoplegia and ataxia. A 2-year-old boy presented with rapidly progressive ophthalmoplegia, ataxia, hyporeflexia, weakness of the lower extremities, and, subsequently, disturbance of consciousness. He experienced bronchitis with watery diarrhea and had laboratory evidence of recent infection with Epstein-Barr virus (EBV). He was diagnosed as having overlapping GBS and BBE associated with EBV and received treatment with a combination of immunoglobulin and methylprednisolone, as well as acyclovir, and had recovered completely after 3 months. In addition, he has not experienced any relapse over the past year. We suggest that combinations of symptoms and signs of central lesions (disturbance of consciousness) and peripheral lesions (ophthalmoplegia, facial weakness, limb weakness, and areflexia) are supportive of a diagnosis of overlapping GBS and BBE and can be helpful in achieving an early diagnosis, as well as for the administration of appropriate treatments.

한국, 중국, 일본에서 자생하는 '마' 속 식물의 화학 성분과 활성 (Phytochemical and pharmacological profiles of Dioscorea species in Korea, China and Japan)

  • Plants of genus Dioscorea have long been served as important carbohydrate-stuffed foods in the tropical and subtropical regions, and utilized as traditional herb medicines to enhance digestive function, improve anorexia, and treat diarrhea in oriental countries. It is known that around 600 species of Dioscorea are distributed in the world including 107 species in Asia, but actually utilized Dioscorea species are restricted to small numbers. Phytochemical investigations for Dioscorea species have revealed a number of chemical components such as sapogenins, saponins, phenanthrenes, stilbenes, diterpenes and purine derivatives. According to recent pharmacological studies, Dioscorea species possess significant antioxidant, antibacterial and anti-inflammatory activities as well as anticancer, antidiabetic, cholesterol-lowering and hypolipidemic effects. Here, seven Dioscorea species (D. batatas, D. japonica, D. bulbifera, D. opposita, D. tokoro, D. nipponica and D. alata), mainly distributed and used in Korea, China, and Japan, are reviewed to provide their botanical, phytochemical and pharmacological properties were described.

오심(惡心), 구토(嘔吐)를 주소(主訴)로 하는 신체형(身體型) 장애(障碍) 환아(患兒)의 치험(治驗) 1예(例) (A clinical case report of somatoform disorder patient complained nausea and vomiting)

  • The somatoform disorder are distinguished by physical symptoms suggesting a medical condition, yet the symptoms are not fully explained by the medical condition, by substance use, or by another mental disorder. This is that an unconscious intrapsychic conflict, wish, or need is converted to a somatic symptom and clinically express various symptoms such as headache, dizziness, nausea, vomiting dyspepsia, diarrhea and constipation, etc. We report a case of somatoform disorder patient, who was 9 years old female and complained of nausea, vomiting and dysdipsia. She had her case diagnosed as somatoform disorder in Yong-dong severance hospital and took anti-depressant (chlomipramine) with counseling for 2 months. After treatment, her emotional instability and depression were improved, yet the somatic symptoms remain same. We diagnosed her case as vomiting induced by deficiency of the stomach(胃虛嘔吐) and administered Bihe-yin(比和飮) to her. After administration of Bihe-yin(比和飮) for one month, her somatic symptoms of nausea, vomiting and dysdipsia were almost disappeared and she got acquired her confidence in school life.

Molecular Modeling of Small Molecules as BVDV RNA-Dependent RNA Polymerase Allosteric Inhibitors

  • Bovine viral diarrhea virus (BVDV), a major pathogen of cattle, is a well-characterized pestivirus which has been used as a good model virus for HCV. The RNA-dependent RNA polymerase (RdRp) plays a key role in the RNA replication process, thus it has been targeted for antivirus drugs. We employed two-dimensional quantitative structure-activity relationship (2D-QSAR) and molecular field analysis (MFA) to identify the molecular substructure requirements, and the particular characteristics resulted in increased inhibitory activity for the known series of compounds to act as effective BVDV inhibitors. The 2D-QSAR study provided the rationale concept for changes in the structure to have more potent analogs focused on the class of arylazoenamines, benzimidazoles, and acridine derivatives with an optimal subset of descriptors, which have significantly contributed to overall anti-BVDV activity. MFA represented the molecular patterns responsible for the actions of antiviral compound at their receptors. We conclude that the polarity and the polarizability of a molecule play a main role in the inhibitory activity of BVDV inhibitors in the QSAR modeling.

Lactobacillus plantarumCLP-1이 돼지바이러스에 미치는 효과 (The Effect of Lactobacillus plantarumCLP-1 on the Swine Viruses)

  • To isolate Lactic acid bacteria for animals, we have screened from Kim-chi, swine intestine, swine feces, and dairy products by random selection and anti-viral, antipathogenic bacteria test. Among them, CLP-1 shown that inhibitory effect against rotavirus, porcine epidemic diarrhea (PED) virus, Salmonella sp, and E.coli. By examining biological property, API-ZYM and identified Lactobacillus plantarum by 16S rDNAgene sequence. CLP-1 determined resistance to low pH and bile salt. Futhermore, the cell body of CLP-1 adhered to the intestinal epithelium tissue of swine and Caco-2 cell. CLP-1 was examined on cell immune system modulating activity in vitro. The whole cell and cell culture supernatant was increasing of interferon-${\beta}$ activity. And then, CLP-1 increased prevention effect by Salmonella enteritidis infection in SPF chickens. And we determined similar result in pigs.

  • The use of practical management factors to maximize pig health improvement cannot guarantee freedom from diseases. Moreover, because of health safety concerns, the use of antibiotics has been restricted in livestock, including pigs. Therefore, the swine industry has been looking for various alternatives to antibiotics to improve pig's health and performance. Clay is a dietary factor generally accepted for improving pig health. It is a naturally occurring material and is primarily composed of fine-grained minerals. It has a specific structure with polar attraction. Because of this structure, clay has the ability to lose or gain water reversibly. In addition, clay has beneficial physiological activities. First, clay has anti-diarrheic and antibacterial effects by penetrating the cell wall of bacteria or inhibiting their metabolism. Second, it can protect the intestinal tract by absorbing toxins, bacteria, or even viruses. When added to the diet, clay has also been known to bind some mycotoxins, which are toxic secondary metabolites produced by fungi, namely in cereal grains. Those beneficial effects of clay can improve pigs' health and performance by reducing pathogenic bacteria, especially pathogenic Escherichia coli, in the intestinal tract. Therefore, it is suggested that clay has a remarkable potential as an antibiotics alternative.
