계지약침(桂枝藥鍼)이 Streptozotocin 유도 당뇨 흰쥐의 췌장세포 손상에 미치는 보호 효과 (Protective Effects of Cinnamomi Ramulus Herbal Acupuncture on $\beta$-cell Damage of Streptozotocin-induced Diabetic Rat)

  • Objectives : For evaluation of preventive and anti-diabetic activities of Cinnamomi ramulus(CR) herbal acupuncture on pancreatic islet damage in streptozotocin(STZ)-induced diabetic rat. Methods : CR herbal acupuncture was performed at Bisu($BL_{20}$) for 3 weeks subcutaneously starting1 week before STZ i.p. injection. SD rats were divided into four groups(n=10 for each group); 1) NC group, non-treated normal control group, 2) STZ group, STZ administered control group, 3) CR125 group, CR(125mg/kg) + STZ administered group, and 4) CR250 group, CR(250mg/kg) + STZ administered group. Results : Both of CR250 and CR125 groups showed increase in insulin secretion and decrease in the level of serum triglyceride and non-esterified fatty acid in a dose-dependent manner compared to the STZ group. Only CR250 group showed decrease in the levels of glucose and total cholesterol compared to the STZ group. CR herbal acupuncture prevents $\beta$-cell damage of pancreatic islet, showing round figure on the sections of the pancreas. In the pancreatic cells, expressions of iNOS, JNK-2, P-JNK-1/2 and ERK-1/2 were decreased compared to the STZ group. CR herbal acupuncture solution did not show any cytotoxicity by MTS assay and inhibited expressions of iNOS and COX-2 in the STZ-induced diabetic rats. Conclusions : Therefore, we suggest that CR herbal acupuncture may act as a prophylactic as well as a therapeutic modality for diabetes mellitus.

Dietary supplementation with astaxanthin may ameliorate sperm parameters and DNA integrity in streptozotocin-induced diabetic rats

  • Objective: Diabetes mellitus (DM) is known to cause many systemic complications as well as male infertility. Astaxanthin (ASTX) is a powerful antioxidant that is involved in a variety of biologically active processes, including those with anti-diabetes effects. The present study investigates the effect of ASTX on the spermatozoa function in streptozotocin (STZ)-induced diabetic rats. Methods: We divided 30 adult rats into three groups (10 rats per group), with a control group that received corn oil mixed with chow. DM was induced by intra-peritoneal injection of STZ. Eight weeks after the STZ injection, half of the diabetic animals were used as diabetic controls, and the rest were treated with ASTX for 56 days. Then the parameters and chromatin integrity of the epididymal sperm were analyzed using chromomycin A3, toluidine blue (TB), and acridine orange (AO) staining. Results: The count, viability, and motility of the epididymal sperm were decreased significantly in the STZ group in comparison with the control group (count and viability, p<0.001; motility, p<0.01). ASTX increased normal morphology and viable spermatozoa compared to the STZ group (morphology, p=0.001; viability, p<0.05). The percentage of abnormal chromatins in TB and AO staining was higher in the STZ group compared to the control group (p<0.001). The mean percentage of TB and AO positive spermatozoa in STZ rats was significantly lower in the STZ+ASTX group (TB, p=0.001; AO, p<0.05). Conclusion: This study observed that in vivo ASTX treatment partially attenuates some detrimental effect of diabetes. Conversely, ASTX improved sperm viability, normal morphology, and DNA integrity.

모링가 잎 추출물의 항당뇨, 알코올 대사 및 간 보호 활성 (Anti-Diabetic, Alcohol-Metabolizing, and Hepatoprotective Activities of Moringa (Moringa oleifera Lam.) Leaf Extracts)

  • 본 연구에서는 비타민과 단백질이 풍부하여 높은 영양가를 가지고 있으며, 콜레스테롤 제거 및 간 손상 방지와 염증, 심장병 등의 치료 효과가 있는 것으로 알려진 모링가 잎 추출물의 새로운 소재로서의 가능성을 검토하고자 항산화 활성과 항당뇨, 항염증 및 알코올분해능을 탐색하였다. 모링가 열수 및 에탄올 추출물의 총페놀 함량은 각각 45.49와 63.06 mg TAE/g으로 나타났다. 1,1-Diphenyl-2-picrylhydrazyl radical 소거능은 1 mg/mL 농도의 열수 및 에탄올 추출물에서 각각 60.81%와 71.29%로 에탄올 추출물의 소거능이 높게 나타났다. 모링가 열수 및 에탄올 추출물의 superoxide dismutase 유사 활성은 농도 의존적으로 증가하였으며, 5 mg/mL 농도의 열수 및 에탄올 추출물에서 각각 2.82%와 7.40%로 에탄올 추출물의 활성이 높은 것으로 나타났다. 모링가 추출물의 혈당 강하 효과를 확인하기 위해 실시한 ${\alpha}-glucosidase$ 활성 억제 효과는 5 mg/mL 농도의 열수 및 에탄올 추출물에서 45.19%와 88.14%로 에탄올 추출물의 저해 활성이 유의적으로 높게 나타났다. 숙취 해소능을 알아보기 위해 alcohol dehydrogenase 및 acetaldehyde dehydrogenase 활성을 측정한 결과 두 효소 모두 모링가 열수 및 에탄올 추출물의 농도에 의존적으로 증가하였다. Lipopolysaccharide(LPS)에 의하여 유도된 NO 합성은 열수 및 에탄올 추출물 1 mg/mL 농도에서 28.50%와 7.90%로 LPS 처리군(41.96%)보다 각각 32.1%와 81.2% 현저히 감소하였으며, 모링가 열수 및 에탄올 추출물이 tacrine으로 유도된 Hep3B 간암 세포주에 대하여 유의적인 보호 활성을 나타냈다. 이상의 결과에서와 같이 모링가 잎 열수 및 에탄올 추출물은 우수한 항당뇨, 항염증, 숙취 해소 효과 및 간세포 보호 효과가 있는 것으로 나타났기에 기능성 소재로서의 활용도가 높을 것으로 판단된다.

고지방식이와 STZ 유도 제2형 당뇨 마우스에서 새싹의 항당뇨 효과 (Anti-Diabetic Effects of Sprouts in High-Fat Diet and Streptozotocin-Induced Type II Diabetes Mellitus Mice)

  • 본 연구에서는 고지방식이와 streptozotocin으로 유도된 제2형 당뇨 마우스에서 새싹추출물의 항당뇨 효과를 살펴보았다. 제2형 당뇨 유도 후 4주 동안의 체중 변화를 관찰한 결과 새싹추출물군과 메트포민군에서 유의한 체중 감소 변화가 나타났으며, 식이섭취량에 따른 변화는 모든 실험군 간의 차이가 없었다. 공복혈당은 고지방식이군의 경우 실험종료일까지 지속적으로 상승하였고, 이와 대조적으로 새싹추출물과 메트포민군의 경우 실험 2주째부터 감소하여 실험 종료 4주째에서는 고지방식이군보다 유의적으로 감소하였다. 경구 내당능 검사는 포도당 섭취 후 60분, 90분, 120분에서 새싹추출물과 메트포민군이 고지방식이군보다 유의적으로 감소하였으며, 인슐린 내성 검사는 120분에서 새싹추출물과 메트포민군이 고지방식이군보다 유의성 있게 감소하였다. 혈중 지질 농도 변화에서 총콜레스테롤, 중성지방 그리고 LDL 콜레스테롤 농도는 고지방식이군에 비해 새싹추출물과 메트포민군에서 유의적으로 감소하였고, HDL 콜레스테롤 농도는 고지방식이군에 비해 새싹추출물과 메트포민군에서 증가하였다. 간독성의 지표인 glutamate oxaloacetate transaminase와 glutamate pyruvate transaminase 효소 활성도는 메트포민투여군과 고지방식이군을 제외한 실험군들 간에 유의성이 나타나지 않았다. 혈중 인슐린과 렙틴 농도는 고지방식이군에 비해 새싹추출물과 메트포민군에서 유의적으로 감소하였다. 이상의 결과를 바탕으로 새싹추출물은 제2형 당뇨 마우스에서 체중을 감소시키고 혈중지질 대사 이상을 조절하면서 인슐린 저항성을 감소시켜 혈당을 개선하므로 항당뇨 효과가 있는 우수한 식품 소재로서의 개발 가능성을 시사하고 있다.

  • As the average span of human life is continuously increasing, especially the old aged groups, are suffering from various chronic and critical diseases e.g. cardiovascular diseases, coronary heart disease, diabetes, atherosclerosis and alzheimer's etc. However, effective and safe treatment methods have not yet been investigated threathening old aged groups. This research was planned to isolate compounds with the therapeutic potential for the above mentioned chronic diseases from the mulberry leaves. Biological screenings were carried out for the following categories; anti-atherogenic, anti-diabetic and antihypertensive effects. The results were as follows; Mulberry leaves, 20% $\alpha$-treated Gaelyrangppong showed significant 81% of blood glucose lowering effects in alloxan-induced hyperglycemic mice. Particularly, butanol-soluble fractions of mulberry leaves showed the more significant hypoglycemic activity than other fractions in alloxan induced hyperhlycemic mice. Also in the group given 1g/kg doses of extract of mulberry leaves, total cholesterol level was decreased significantly by as much as 49% in hyperlipidemia-induced rats. In Mulberry leaves post-treated group, the atheroscelosis index, HDL-cholesterol/Total-cholesterol, was increased significantly by as much as 91%.

홍국발효 대두의 항산화 및 항당뇨 활성 (Antioxidant and Anti-Diabetic Activities of Soybean Fermented with Monascus)

  • This study was performed to investigate the antioxidant and antidiabetic activities of soybean fermented with Monascus. Also, the changes in the content of isoflavones and Monacolin K were analyzed. It was observed that the glycoside forms of daidzin and genistin were converted to aglycones of daidzein and genistein within 6 days of fermentation. The product can be used as a health functional material that can increase bioavailability. Monacolin K production was found to increase significantly with the progression of fermentation with an increase to 0.04 mg/g and 0.44 mg/g on 6 and 12 days of fermentation, respectively. The DPPH radical scavenging activity of soybean fermented with Monascus was significantly increased compared to that of soybean. The protein expression of inflammation-related genes (TNF-α, IL-6, and COX-2) in the MIN cell was significantly increased in the presence of alloxan compared to the normal group, but a decrease was observed in the presence of soybean fermented with Monascus. In conclusion, soybean fermented with Monascus showed the highest antidiabetic and antioxidant effects. These results suggest that soybean fermented with Monascus has the potential to be used as a beneficial ingredient with antidiabetic and antioxidant effects.

STZ으로 당뇨를 유발한 실험쥐에 대한 Psidium guajava L.과 Lagerstroemia speciosa L. 잎 추출물의 항당뇨 효과 (Antidiabetic Effects of Leaves Extracts of Psidium guajava L. and Lagerstroemia speciosa L. in STZ-induced Rats)

  • 구아바(Psidium guajava L.)와 바나바(Lagerstroemia speciosa L.)는 항당뇨 효과를 갖는 대표적인 약용식물로 알려져 있다. 이들 식물은 다양한 폴리페놀(polyphenol) 성분을 함유하며 당뇨병의 치료에 효과적으로 작용한다. 따라서 본 연구는 이들 추출물을 streptozotocin (STZ)으로 당뇨를 유발한 실험쥐에 섭취시켜 항당뇨 효과를 상호 비교하였다. STZ으로 당뇨를 유발한 실험쥐에 구아바 및 바나바 잎 추출물을 섭취시킨 결과 당뇨쥐에 비해 혈당은 약 19-32%, 총콜레스테롤(total cholesterol)은 약 24-46%, 중성지방(triglyceride)은 22-67% 그리고 유리지방산(free fatty acid) 은 49-71% 감소하였고 또한 인슐린 분비와 췌장에 존재하는 베타세포의 회복도 증가되었다. 한편 구아바 추출물의 섭취가 바나바 추출물의 섭취에 비해 항당뇨 효과가 더 높았는데 그 이유는 바나바 잎에 포함된 친수성 폴리페놀(polyphenol) 성분이 에탄올 추출과정에서 대부분 추출되지 않았고 또한 추출된 코로소릭산(corosolic acid) 은 지금까지 알려진 것과는 달리 항당뇨 활성이 낮았기 때문이다.

식용피 밀양3호[Echinochloa esculenta (A. Braun)] 에탄올 추출물의 당뇨모델 마우스에 대 한 항당뇨 활성 (Anti-diabetic Effects of Barnyard Millet Miryang 3 [Echinochloa esculenta (A. Braun)] Grains on Blood Glucose in C57BL/KsJ-db/db Mice)

  • 잡곡의 하나인 식용피는 폴리페놀과 수용성 식이섬유성분이 풍부하고 전분함량이 타 곡류보다 낮으면서 단백질 함양이 높아 영양학적 건강지표가 높은 잡곡이다. 본 연구에서는 항염증 활성이 강하게 확인된 밀양3호를 선발하여 이의 항당뇨 활성에 관해 조사하였다. 식용피 밀양3호의 80% 에탄올(EtOH) 추출물을 당뇨모델인 C57BL/KsJ-db/db mice에 600 mg/kg로 8주간 투여하여 혈당과 혈중 지질에 대한 영향을 조사한 결과, 공복혈당이 당뇨 대조군에 비해 유의적으로 감소하여 개선 되었으며, 경구 내당성도 1주 투여 시부터 개선되는 효과를 보이다가 8주간의 투여 시는 양성대조군인(메트포민 100 mg/kg) 수준으로 경구 내당성이 개선됨을 확인하였다. 이때 체중증가는 메트포민 투여군(10.36±3.15)에는 못 미치나 8.54±2.24로 당뇨 대조군 체중 증가(8.71±1.64)의 수준이었으며, food efficiency rates를 고려하면 식용피가 식이 섭취에는 큰 영향을 주지 않는 것으로 사료되었다. 총 콜레스테롤 농도는 당뇨 대조군의 총콜레스테롤(229.3±47.8)에 비해 식용피 EtOH 추출물 투여군(160.7±7.6)과 메트포민 투여군(176.0±25.6)에서 유의적으로 감소하였다. LDL-콜레스테롤 농도는 식용피 투여군에서 메트포민 투여군 수준으로 당뇨 대조군에 비해 약 41%정도 감소한 것에 반해, 중성지방은 식용피 추출물의 영향을 받지 않았다. 간독성은 식용피 추출물군의 GOT 값이 메트포민 투여군 보다 더 낮았으며, GPT 활성도는 모든 실험군들 간에 유의한 차이가 없었다. 이상의 연구 결과는 식용피 밀양3호가 항 당뇨 관련 우수한 소재 함유 곡류로 관련 질환 예방과 치료에 활용할 수 있음을 시사한다.

Anti-diabetic effect and mechanism of Korean red ginseng extract in C57BL/KsJ db/db mice

  • Purpose: Ginseng is a well-known medical plant used in traditional Oriental medicine. Korean red ginseng (KRG) has been known to have potent biological activities such as radical scavenging, vasodilating, anti-tumor and anti-diabetic activities. However, the mechanism of the beneficial effects of KRG on diabetes is yet to be elucidated. The present study was designed to investigate the anti-diabetic effect and mechanism of KRG extract in C57BL/KsJ db/db mice. Methods: The db/db mice were randomly divided into six groups: diabetic control group (DC), red ginseng extract low dose group (RGL, 100 mg/kg), red ginseng extract high dose group (RGH, 200 mg/kg), metformin group (MET, 300 mg/kg), glipizide group (GPZ, 15 mg/kg) and pioglitazone group (PIO, 30 mg/kg), and treated with drugs once per day for 10 weeks. During the experiment, body weight and blood glucose levels were measured once every week. At the end of treatment, we measured Hemoglobin A1c (HbA1c), blood glucose, insulin, triglyceride (TG), adiponectin, leptin, non-esterified fatty acid (NEFA). Morphological analyses of liver, pancreas and white adipose tissue were done by histological observation through hematoxylin-eosin staining. Pancreatic islet insulin and glucagon levels were detected by double-immunofluorescence staining. To elucidate an action of mechanism of KRG, DNA microarray analyses were performed, and western blot and RT-PCR were conducted for validation. Results: Compared to the DC group mice, body weight gain of PIO treated group mice showed 15.2% increase, but the other group mice did not showed significant differences. Compared to the DC group, fasting blood glucose levels were decreased by 19.8% in RGL, 18.3% in RGH, 67.7% in MET, 52.3% in GPZ, 56.9% in PIO-treated group. With decreased plasma glucose levels, the insulin resistance index of the RGL-treated group was reduced by 27.7% compared to the DC group. Insulin resistance values for positive drugs were all markedly decreased by 80.8%, 41.1% and 68.9%, compared to that of DC group. HbA1c levels in RGL, RGH, MET, GPZ and PIO-treated groups were also decreased by 11.0%, 6.4%, 18.9%, 16.1% and 27.9% compared to that of DC group, and these figure revealed a similar trend shown in plasma glucose levels. Plasma TG and NEFA levels were decreased by 18.8% and 16.8%, respectively, and plasma adiponectin and leptin levels were increased by 20.6% and 12.1%, respectively, in the RGL-treated group compared to those in DC group. Histological analysis of the liver of mice treated with KRG revealed a significantly decreased number of lipid droplets compared to the DC group. The control mice exhibited definitive loss and degeneration of islet, whereas mice treated with KRG preserved islet architecture. Compared to the DC group mice, KRG resulted in significant reduction of adipocytes. From the pancreatic islet double-immunofluorescence staining, we observed KRG has increased insulin production, but decreased glucagon production. KRG treatment resulted in stimulation of AMP-activated protein kinase (AMPK) phosphorylation in the db/db mice liver. To elucidate mechanism of action of KRG extract, microarray analysis was conducted in the liver tissue of mice treated with KRG extract, and results suggest that red ginseng affects on hepatic expression of genes responsible for glycolysis, gluconeogenesis and fatty acid oxidation. In summary, multiple administration of KRG showed the hypoglycemic activity and improved glucose tolerance. In addition, KRG increased glucose utilization and improved insulin sensitivity through inhibition of lipogenesis and activation of fatty acid $\beta$-oxidation in the liver tissue. In view of our present data, we may suggest that KRG could provide a solid basis for the development of new anti-diabetic drug.

Sasa borealis extract exerts an antidiabetic effect via activation of the AMP-activated protein kinase

  • Leaf of Sasa borealis, a species of bamboo, has been reported to exhibit anti-hyperglycemic effect. However, its antidiabetic mechanism is not fully understood. In this study, we examined whether an extract of S. borealis activates AMP-activated protein kinase (AMPK) and exerts anti-hyperglycemic effects. Treatment with the S. borealis extract increased insulin signaling and phosphorylation of AMPK and stimulated the expression of its downstream targets, including $PPAR{\alpha}$, ACO, and CPT-1 in C2C12 cells and $PPAR{\alpha}$ in HepG2 cells. However, inhibition of AMPK activation attenuated insulin signaling and prevented the stimulation of AMPK target genes. The S. borealis extract increased glucose uptake in C2C12 cells and suppressed expression of the gluconeogenic gene, PEPCK in HepG2 cells. The extract significantly reduced blood glucose and triglyceride levels in STZ-induced diabetic mice. The extract enhanced AMPK phosphorylation and increased Glut-4 expression in the skeletal muscle of the mice. These findings demonstrated that the S. borealis extract exerts its anti-hyperglycemic effect through activation of AMPK and enhancement of insulin signaling.
