Streptozotocin 에 의해 유발된 당뇨병 쥐의 혈당과 당뇨병성 신증에 한약복합처방이 미치는 효과 (Effects of Herb-Combined Remedy(HCR) on Blood Glucose Levels and Diabetic Nephropathy in STZ-induced Diabetic Rats)

  • Diabetes mellitus is a worldwide epidemic. Global projections suggest that most nations will have a doubling of the incidence of Diabetes mellitus(DM) within 20years. The Herb-Combined Remedy(HCR) of DM was known as one of anti-hyperglycaemic agents. But it is unclear whether HCR can be used to treat patients with Diabetic nephropathy or not. The present study was designed to investigate the anti-hyperglycaemic and preventive effects from diabetic nephropathy of HCR in STZ-induced diabetic rats. Treatment with HCR regulated blood glucose levels, but did not affect body weights. In addition, HCR was able to decrease blood glucose levels after 120min. Serum BUN levels were not effective but, serum Creatinine levels were lowered by treatment with HCR or MF. In histopathological observation of renal tissue, both HCR and MF groups showed decreased tendency of inflammation and renal injury. In conclusion, these results demonstrate that HCR is effective to treat patients with DM and also has preventive activity on diabetic nephropathy.

  • The anti-diabetic effects of the methanol extracts of Cortex, Ramulus and Folium part of Eucommia ulmoides Oliver were investigated by the experimental diabetic model in rats. The increased blood glucose level in the diabetic rats induced by alloxan and streptozotocin was significantly lowered and a marked increase in body weight was shown with the treatments of the extracts of Cortex and Folium. The blood insulin level lowered by streptozotocin pretreatment was significantly recovered with the treatment of the extract of Folium. In glucose tolerance test, the extracts of Cortex and Folium displayed a significant inhibitory effect on the glucose level. The extracts of Cortex and Folium also caused a marked inhibition in increased blood pressure in the diabetic rats induced by alloxan.

The Effect of Rutin on Antioxidant and Anti-inflammation in Streptozotocin-induced Diabetic Rats

  • The study examined the antioxidant activities and anti-inflammatory effects of rutin from the streptozotocin (STZ)-induced diabetic rats. Results revealed that the levels of plasma glucose and serum glucose were remarkably higher in the STZ-treated group compared to other groups and were significantly reduced in the STZ+rutin treated group compared to the STZ-treated group. In terms of weight, it significantly increased in all experimental groups during the experiment period except for STZ-induced diabetic group. The weight of the STZ-treated group was remarkably reduced compared to other groups. Regarding the weight of each body organ, the STZ-treated group showed higher organ weight compared to the other groups while STZ+rutin-treated group showed significantly reduced kidney and liver weights compared to those of STZ-treated group. In the pancreas tissue of the STZ-treated group, ${\beta}$-cell destruction and vacuolization were observed. Inflammation in the heart, liver, kidney, and retina tissues were also vividly recorded. In the STZ+rutin administered group, the heart and retina tissues were shown to be preserved normally while the liver and kidney tissues showed reduced histopathology in general compared to the STZ-treated group. Conclusionally, the rutin has the effect on the antioxidant activities and anti-inflammation in the STZ-induced diabetic rats.

당뇨흰쥐의 콜라겐 감소 및 인간 피부 섬유아세포의 MMP-1 증가에 대한 목단피(牧丹皮)의 항피부노화 효과 (Anti-skin-aging effects of Paeonia Suffruticosa Andrews on maintaining skin collagen in STZ-induced diabetic rats and inhibiting MMP-1 systhesis in human skin fibroblasts)

  • Objective : Skin aging is commonly observed in patients with diabetes mellitus, which can be accessed by the amount of skin collagen and matrix metalloproteinase-1 (MMP-1). In the present study, anti-skin-aging effects of Root Cortex of Paeonia Suffruticosa Andrews (PSA), which has been widely used to treat diabetes mellitus, are investigated. Methods : Streptozotocin (STZ) was intraperitoneally injected to rats to induce diabetes. Body weights, feed intake, organ weights, blood glucose, and other biochemical index are determined in both normal and diabetic rats. In order to study the effect of PSA on skin aging, the amount of skin collagen was measured in diabetic rats after PSA treatments. Also, MMP-1 synthesis in UVB-irradiated human skin fibroblasts was investigated. Results : 1. When PSA was administered to STZ-induced diabetic rats, feed intake was significantly increased and blood glucose and total cholesterol were decreased in a dose-dependent manner. However, there are no differences in individual organ weights, GOT, and GPT. 2. A decrease of skin collagen in diabetic rats was significantly suppressed when PSA was treated. 3. PSA also inhibited MMP-1 synthesis in UVB-irradiated normal human skin fibroblasts, similar to retinoid, a well-known effective anti-skin-aging substance. Conclusion: PSA suppressed a collagen decrease in diabetic rats and inhibited MMP-1 synthesis in UVB-irradiated human skin fibroblasts. Therefore, the treatment of PSA is very effective to slow down the skin aging process.

Anti-diabetic Effects of CCCA, CMESS, and Cordycepin from Cordyceps militaris and the Immune Responses in Streptozotocin-induced Diabetic Mice

  • Anti-diabetic effect of various fractions of Cordyceps militaris (CM), CCCA (crude cordycepin containing adenosine), CMESS (ethanol soluble supernatant), and cordycepin were evaluated in streptozotocin (STZ) induced diabetic mice, CMESS showed potent inhibitory activity of 34.7% in starch-loaded mice (2 g/kg) while acarbose as a positive standard exhibited 37.8% of inhibition rate. After 3 days administration (50 mg/kg), cordycepin (0.2 mg/kg), and acarbose (10 mg/kg) dramatically reduced blood glucose level (inhibition ratio: 46.9%, 48.4% and 37.5% respectively). CCCA that has high contents of cordycepin (0.656 mg/4 mg) did not influence on reducing blood glucose level. The proliferation of splenocytes and peritoneal macrophages derived from STZ-induced diabetic mice administered samples were evaluated out by addition of mitogens to see the stability of the usage of these herbal medicines. Proliferation of T-lymphocyte was significantly decreased; while NO production was increased more than two fold to STZ control in the cordycepin-administered group. Changes of serum enzyme levels of glutamic oxaloacetic transaminase (GOT) and glutamic pyruvic transaminase (GPT) were also evaluated. Cordycepin administered group was appeared to acarbose. We conclude that CMESS and cordycepin may be useful tools in the control of blood glucose level in diabetes and promising new drug as an anti-hyperglycemic agent without defects of immune responses and other side effects.

고초균에서 His-Pro 반복서열을 갖는 Oligopeptide의 분비 최적화 및 항당뇨 효과 (The Secretion Optimization of Oligopeptide with His-Pro Repeats in Bacillus subtilis and Its Anti-Diabetic Effects)

  • To verify anti-diabetic effect of oligopeptide with His-Pro repeats (mHP peptide), the oligopeptide was first secreted and optimized using the secretion vector, pRBAS with alkaline protease gene promoter and the signal sequence in Bacillus subtilis and directly the anti-diabetic effect of the mHP peptide was investigated in insulinoma cell, RINm5F cell line. The oligopeptide gene was obtained by annealing oligonucleotides with repeated His-Pro sequence and finally was constructed as 18 dipeptides (108 bp and 4.0 kDa) coding gene, named oligopeptide with His-Pro repeats (mHP peptide) to make cyclo(His-Pro) known to be anti-diabetic effects. The region encoding the oligopeptide gene was subcloned into the pRBAS secretion vector (E.coli-Bacillus shuttle vector) after PCR amplification using the designed primers including initiation and termination codons and His tag, named pRBAS-mHP (6.56 kb). To optimize secretion of the oligopeptide, various culture conditions were investigated in Bacillus subtilis LKS. As a result, the secreted oligopeptide was maximally measured (approximately $59.6{\mu}g/mL$) in 3 L batch culture and the highest secretion was achieved at $30^{\circ}C$, PY medium, and carbon sources (particularly barley and glycerol). In the RINm5F cells treated with 2 mM STZ, the oligopeptide treatment (0.1 mg/mL) restored the cell viability (10%) and reduced the nitric oxide (NO) generation (35%) and DNA fragmentation (90%). And also, insulin secretion level was increased to 17% higher than in STZ-treated RINm5F cells. These results suggest that the oligopeptide with His-Pro repeats could be a candidate material for anti-diabetic agent against STZ-induced diabetes.

Anti-diabetic effects of water extract from the dietary mushroom Neolentinus lepideus in type 2 diabetic db/db mice

  • The objective of this study was to determine the anti-diabetic effect of the water extract of Neolentinus lepideus in a diabetic mouse model. Seven-week-old C57BL/KsJ-db/db mice were fed either a control diet (CD) or diet supplemented with 1% or 5% of N. lepideus water extract (NLWE1 or NLWE5) for 10 weeks. Oral administration of NLWE significantly decreased the body weight gain compared to that of CD-fed group. Mice in the NLWE group had significantly lower levels of fasting serum glucose, fatty acids, and low-density lipoprotein cholesterol compared to those in the control group. These effects were accompanied by reduced fatty liver and improved glucose tolerance in the NLWE group. Taken together, these results suggest that N. lepideus might have potential as a dietary supplement to control diabetes.

양격산화탕(凉膈散火湯)이 당뇨흰쥐의 전뇌허혈에 미치는 영향 (Effects of Yanggyuksanhwa-tang on Global Cerebral Ischemia of Diabetic Rats Induced by Streptozotocin)

  • This study evaluated neuroprotective effects of Yanggyuksanhwa-tang (YST) on global cerebral ischemia of diabetic rats. On primary experiment, diabetic condition in rats was induced by streptozotocin injection. Secondarily, global cerebral ischemia was induced by bilateral occlusion of the common carotid artery with hypotension (BCAO) under the diabetic condition. Then neuroprotective effect of YST was observed with changes of neuronal c-Fos and Bax expressions, and GFAP expression in the brain tissues by using immunohistochemistry. YST treatment was resulted significant decrease of c-Fos expression in CA1 hippocampus induced by BCAO on diabetic rats. YST treatment was resulted significant decrease of Bax expression in CA1 hippocampus induced by BCAO on diabetic rats. YST treatment was resulted significant decrease of c-Fos expression in cerebral cortex and caudoputamen induced by BCAO on diabetic rats. YST treatment was resulted significant decrease of GFAP expression in cerebral cortex induced by BCAO on diabetic rats. These results suggest that YST has effects on neuroprotection against cerebral ischemic damage under diabetic condition. And it is supposed that neuroprotective effect of YST reveals by anti-apoptosis mechanism.

천년초 선인장 줄기의 항당뇨 및 항산화 효과 (Anti-diabetic and Anti-oxidative Effects of Opuntia humifusa Cladodes)

  • 본 연구에서는 천년초 줄기 분말을 이용한 식품 개발에 활용하고자 천년초 줄기 분말을 용매별로 추출하여 항당뇨 및 항산화 활성을 시험하였다. 천년초 줄기의 ethyl acetate fraction을 $1,000{\mu}g/mL$ 처리했을 때 ${\alpha}$-amylase 억제활성이 $89.94{\pm}1.15%$로 가장 높았으며, 다른 fraction들은 $12.19{\pm}2.39{\sim}28.14{\pm}2.06%$로 낮았다. ${\alpha}$-Glucosidase 억제활성은 ethyl acetate fraction이 $29.01{\pm}0.49%$로 가장 높았으며, 75% ethanol fraction은 $23.48{\pm}9.89%$였고 water fraction이 $12.59{\pm}1.35%$로 가장 낮았다. 용매 분획별 total phenolics 함량과 total flavonoids 함량은 ethyl acetate fraction에서 각각 $196.02{\pm}5.26$$114.00{\pm}10.03{\mu}g/mg$으로 가장 높게 나타났다. Ethyl acetate fraction의 DPPH 및 ABTS radical 소거능은 농도 의존적으로 각각 $88.78{\pm}0.30%$$97.27{\pm}0.94%$로 가장 높게 나타났으며, 대조구인 BHT 및 비타민 C와 비슷하였다. FRAP 활성은 chloroform 및 ethyl acetate fraction에서 각각 $1.119{\pm}0.018$$1.057{\pm}0.014abs(593nm)$로 다른 fraction보다 높게 나타났다. 전체적으로 천년초 줄기의 ethyl acetate fraction의 항당뇨 및 항산화 효과가 가장 높게 나타났으며, 상업적으로 사용되는 BHT 및 비타민 C와 비슷한 항산화 효과를 나타내는 천연소재를 발굴함으로써 천년초 줄기가 항당뇨 및 항산화 활성을 갖는 식품소재로서 활용 가능성이 높은 것으로 판단되었다.

당뇨병 동물모델에서 여정엽(女貞葉) 추출물의 항당뇨 및 항염증 효과 (Anti-diabetic and Anti-Inflammatory Effects of Water Extract of Ligustrum japonicum Leaves in db/db Mouse)

  • Objectives : In this study, we investigated the anti-diabetic and anti-inflammatory effects of water extract from leaves of Ligustrum japonicum (WLJ) in db/db mouse. Methods : The db/db mice were treated orally with WLJ (300 mg/kg/day) for 10 weeks to examine the long-term effects on hyperglycemia and glomerular tissue as well as biochemical and functional abnormalities in the kidney. Results : WLJ treatment markedly reduced plasma levels of glucose, triglyceride, creatinine, and systolic blood pressure in diabetic db/db mouse. Treatment of WLJ significantly increased plasma level of high density lipoprotein (HDL)-cholesterol. We also found that overexpressions of vascular cellular adhesion molecule (VCAM)-1 and endothelin (ET)-1 were observed in aortic tissue of db/db mouse, whereas, WLJ suppressed both expression of VCAM-1 and ET-1 in aorta. In renal tissue, overexpressions of ICAM-1 and TGF-${\beta}1$ were found in untreated db/db mouse, however, significantly decreased those levels by WLJ treatment. The insulin immunoreactivity of the pancreatic islets remarkably increased in WLJ treated db/db mouse compared with untreated db/db mouse. Taken together, WLJ treatment ameliorated hyperglycemia and hyperlipidemia via improvement of insulin secretion and lipid metabolism, respectively. Furthermore, WLJ treatment also ameliorated hypertension via inhibition of inflammatory process in vascular and renal tissues. Conclusions : Ligustrum japonicum has an anti-diabetic and anti-inflammatory effects in db/db mouse. Thus, these results suggested a beneficial effect of Ligustrum japonicum in treatment with diabetes and diabetic vasculopathy.
