산초 추출물의 S. mutans 항균활성 및 항산화 효능 연구 (A Study on the Anti-microbial Effect on S. mutans and Anti-oxidant Effect of Zanthoxylum pericarpium Extract)

  • Objectives : The Zanthoxylum pericarpium has been used as oriental spicy seasoning and a medicinal plant from old times. This study was performed to determine the anti-oxidant efficacy of Zanthoxylum pericarpium extract and the anti-microbial effects. Methods : We got Zanthoxylum pericarpium extract using PSE (pressurized solvent extraction) method. The anti-microbial effect of Zanthoxylum pericarpium extract was assessed on Streptococcus mutans(S. mutans) and anti-oxidant effect of the extract was assessed by measuring DPPH radical scavenging activity and SOD like activity. Results : 1. Zanthoxylum pericarpium extract had high anti-microbial activity on S. mutans. 2. DPPH radical scavenging activity significantly increased in the Zanthoxylum pericarpium extract. 3. SOD like activity also significantly increased in the Zanthoxylum pericarpium extract. Conclusions : The PSE extract from Zanthoxylum pericarpium has good anti-microbial and anti-oxidant effects in a concentration-dependent manner.

구강병인균에 대한 마와 꿀풀추출물의 항균.항우식효과 (Anti-microbial and Anticariogenic Activity of Yam and Prunella Extract against Oral Microbes)

  • Yam, Prunella was stepwise extracted with hexane, chloroform, ethyl acetate, butanol, and water. Anti-microbial activity of each extract was investigated. Hexane extract was tested for anti-microbial effect on Streptocaccus mutans, one of causative factor of dental caries. Methanol extracts of 7 plants were investigated to anti-microbial effects on S. mutans KCTC 5316, P. gingivalis KCTC 5352, S. aureus KCTC 1927 by means of agar diffusion method. Methanol extract of Yam and Prunella revealed anti-microbial activity against S. mutans, P. gingivalis, and S. aureus. Also, hexane fraction of Yam revealed anti-microbial activity against S. mutans. In sequence of hexane, chloroform, ethylacetate, butanol fraction by Prunelia acted as potent anti-microbial agent on P. gingivalis. The measured MIC of hexane fraction of Yam and Prunella on S. mutans KCTC 5316 strain was 0.25 mg/ml and 0.5 mg/ml and the MIC of hexane fraction of Prunella on S. aureus was 0.5 mg/ml. The hexane fraction of Yam and Prunella suppressed viable ceil counts(VCC) of S. mutans, especially after 24 hrs. The Prunella hexane fraction suppressed VCC of S. aureus, after 12 and 24 hrs. Tested concentrations were 0.1, 0.25 and 0.5 mg/ml. the results were compared with control (0 mg/ml). The pH of S. mutans media and GTase activity were determined to evaluate the anticariogenic activity of Yam, Prunella hexane fraction. The pH were increased from 5.6 to 7.0-7.2 in concentration of 2.0 mg/ml. Yam hexane extraction revealed 35% inhibition to GTase activity and Punella inhibited 25% of GTase. These results suggest that the hexane extracts of Yam and prunella have Antibacterial activities against S. mutans, P. gingivalis, S. aureus and have preventive effect on dental caries.

Effects of Lactobacillus casei and Aggregatibactor actinomycetemcomitans against Streptococcus mutans according to the Concentration of Sucrose

  • Background: Some studies confirm the reduction of the number of Streptococcus mutans in saliva and dental plaque by Lactobacillus, however, these effects are not always confirmed in in vitro and clinical studies, and only the risk of dental caries has been reported. Our in vitro study aimed to reveal microbial and biochemical changes in the single cultures of S. mutans, Lactobacillus casei and Aggregatibactor actinomycetemcomitans and co-cultures of S. mutans and L. casei or A. actinomycetemcomitans according to sucrose concentration. We also aimed to confirm the anti-oral bacterial and anti-biofilm activities of L. casei and A. actinomycetemcomitans against S. mutans according to sucrose concentration. Methods: S. mutans (KCCM 40105), L. casei (KCCM 12452), and A. actinomycetemcomitans (KCTC 2581) diluted to 5×106 CFU/ml were single cultured, and L. casei or A. actinomycetemcomitans applied at concentrations of 10%, 20%, 30% and 40% to S. mutans were co-cultured with selective medium containing 0%, 1% and 5% sucrose at 36.5℃ for 24 hours. Measurements of bacterial growth value and acid production, disk diffusion and biofilm formation assays were performed. Results: In the medium containing sucrose, the bacterial growth and biofilm formation by S. mutans, L. casei, and A. actinomycetemcomitans were increased. In contrast, 30% and 40% of L. casei in the medium containing 0% sucrose showed both anti-oral bacterial and anti-biofilm activities. This implies that L. casei can be used as probiotic therapy to reduce S. mutans in a 0% sucrose environment. Conclusion: The concentration of sucrose in the oral environment is important for the control of pathogenic bacteria that cause dental caries and periodontitis. To apply probiotic therapy using L. casei for S. mutans reduction, the concentration of sucrose must be considered.

연잎과 민들레 추출물에 의한 S. mutans 성장억제 효과 (The Effect on Growth Inhibition of S. mutans by Lotus Leaf and Dandelion Extracts)

  • 최근에 연잎과 민들레 추출물의 약리작용에 대한 연구가 활발하게 이루어져 항산화작용, 항알레르기효과, 항균작용, 항암활성 등에 관한 연구가 보고되고 있다. 이에 본 연구에서는 colony forming unit와 흡광도를 측정하여 연잎과 민들레 추출물이 S. mutans에 미치는 성장억제효과를 연구하였다. 추출물의 첨가에 따른 S. mutans의 성장억제율을 측정한 결과, 추출물의 농도가 높아질수록 S. mutans의 성장억제율도 높아지는 결과를 얻었다. 이로써 연잎과 민들레 추출물은 S. mutans의 성장을 억제하는 항균효과를 가지고 있음을 본 연구에서 확인 할 수 있었다.

  • In order to demonstrate the effect of specific serum IgG antibody on the adherence of Streptococcus mutans to smooth surface and the mechanism of effective adherence inhibition by IgG antibody, in the present study authors obtained purified IgG from different immunogen preparations of S. mutans NCTC 10449(serotype c) and observed the effect of each IgG preparation on the adherence of each S. mutans strain cultured in different conditions. In addition, the present study was undertaken to observe the cross-reactivity of IgG and the effect of sucrose concentration on the adherence of S. mutans in vitro non-growth condition. The adherence of S. mutans to glass surface was effectively inhibited by serum IgG antibody. At the same IgG concentrations, anti-2% fructose grown/1N NaCl washed S. mutans NCTC 10449 cell showed greater adherence inhibitory effect to S. mutans strains than anti-2% sucrose grown and anti-S. mutans NCTC 10449 cell wall, and the greater inhibitory effects of IgG preparations were observed in assay using 2% fructose grown S. mutans cell preparations than using 0.1% sucrose grown cell preparations. These results suggest that the more effective adherence inhibition by serum IgG antibody is due to the reaction with S. mutans cell surface antigens rather than glucan and cell-associated glucosyltransferase. The greatest adherence inhibitory effect of IgG to S. mutans strains was observed on homologous NCTC 10449 strain and the inhibition cross-reactivities were observed between serotype c, e, and f strains. More pronounced cross-reactivity of adherence inhibition of IgG to S. mutans was observed in assay using anti-2% fructose grown/1N NaCl washed cell than using other IgG preparations, and observed in assay using 2% fructose grown S. mutans cell preparations than 0.1% sucrose grown cell preparations. It was interested that low, but adequate concentration of reactive IgG antibody significantly increased the adherence ability of S. mutans. This result may be due to the formation of small cell aggregates resulted in a increase in the numbers of organisms which adhered to glass surface. The adherence of S. mutans to glass surface was possible in the absence of glucan-synthetic activity. Low level of sucrose significantly increased the adherence ability of S. mutans to glass surface, but excessive amount of sucrose induced large cell aggregates resulted in a decrease in the numbers of organism which adhered.

Anti-Oral Microbial Effect of Ethanol Extract of Angelica gigas Nakai

  • Background: The Korean name for Angelica gigas Nakai (AGN) is Cham-dang-gui, which grows naturally or is cultivated, and its dried roots are used in traditional herbal medicines. The AGN root exert various pharmacological effects. Despite the various pharmacological effects of the AGN root, there are no reports on its anti-oral microbial effects. The purpose of this study was to reveal the anti-oral microbial effect and the microbial and biochemical changes in oral microorganisms according to the concentration of the ethanol extract of AGN (EAGN) root, and to confirm the possibility of using EAGN as a plant-derived functional substance for controlling oral infectious microorganisms. Methods: Disk diffusion test, growth measurement, biofilm formation assay, and measurements of acid production and buffering capacity were performed to confirm the antibacterial effect of EAGN. Results: EAGN showed anti-oral bacterial effects against Streptococcus mutans and Aggregatibacter actinomycetemcomitans at all concentrations, with S. mutans showing a more susceptible effect at concentrations above 5.0 mg/ml and A. actinomycetemcomitans at 3.75 mg/ml. EAGN treatment significantly reduced A. actinomycetemcomitans growth at all concentrations tested. Biofilm formation was significantly reduced at concentrations above 3.75 mg/ml for S. mutans and 2.5 mg/ml for A. actinomycetemcomitans. Acid production in S. mutans and A. actinomycetemcomitans was significantly increased by treatment with EAGN, and the buffering capacities of S. mutans and A. actinomycetemcomitans increased from an EAGN concentration of 3.75 mg/ml and above. Conclusion: EAGN showed anti-oral bacterial effects against both S. mutans and A. actinomycetemcomitans at concentrations above 3.75 mg/ml, which were thought to be related to the inhibition of their growth and biofilm formation. Therefore, EAGN can be used as a safe functional substance derived from medicinal plants owing to its antibacterial effects against S. mutans and A. actinomycetemcomitans.

천연에센셜오일의 Streptococcus mutans에 의한 치석형성 억제 활성을 이용한 구강관리제품 개발 가능성에 대한 융합연구 (Convergence research on the possibility of development of oral care products using the anti-plaque activity of natural essential oils against Streptococcus mutans)

  • 본 논문은 Lavender, Tea tree, Eucalyptus, Lemongrass 4종의 천연에센셜오일의 Streptpccous mutans 에 의한 치석형성억제 효과에 대하여 연구해 보고자 하였다. Streptpccous mutans 에 의한 치석형성 억제 활성 측정결과인 Lavender 에센셜 오일의 MAC (Minium anti-adhesive concentration, 최소부착저지 농도)가 1.0 % 이고 Tea tree 에센셜 오일, Eucaliptus 에센셜 오일, lemongrass 에센셜 오일의 MAC는 0.5 % 로 나타났다. 에센셜오일의 농도를 달리하여 Streptpccous mutans와 혼합한 배양액을 항온조에서 배양한 후 용액의 pH를 측정하여 최소부착 저지농도 이하에서 산의 생성으로 pH가 낮아진 것을 확인할 수 있었고, 이 결과는 에센셜 오일이 Streptpccous mutans 에 의한 산의 생성을 억제하는 효과가 있다는 것이다. 이러한 결과들로부터 천연에센셜오일 중 특히 Tea tree, Eucalyptus, Lemongrass는 치석생성을 억제하는 천연소재로 구강관리 제품개발에 활용할 수 있는 가능성을 확인할 수 있었다.

지유(地楡) 부탄올 분획물의 Streptococcus mutans 항균활성 및 항산화 효과 (Anti-microbial Effect on Streptococcus mutans and Anti-oxidant Effect of the Butanol Fractions of Sanguisorbae Radix)

  • Objective : Sanguisorbae Radix(SO). that belong to Rosaceae is widely distributed in Asia including Korea, Japan and China. It has been used as traditional medicine from old times as a treatment for anti-inflammatory drugs. This study was designed to investigate the effects of n-BuOH fractions of SO on anti-oxidant effect and anti-microbial effect on $Streptococcus$ $mutans$ ($S.$ $mutans$). Methods : The anti-microbial effect of $n$-BuOH fractions of SO. was assessed by the paper disk diffusion method and anti-oxidant effect was assessed by the DPPH radical scavenging effect, Superoxide anion radical scavenging effect and SOD like ability. Results : DPPH radical scavenging of the $n$-BuOH fraction from SO in 50 ug/mL was shown to be Fr-2 (89.9%), Fr-3 (88.7%), Fr-4 (76.3%), Fr-1 (59.4%), Fr-5 (56.2%). Superoxside anion radical scavenging activity of the n-BuOH fraction from SO. in 50 ug/mL for Fr-3 was shown to be 78%. SOD-like activity of n-BuOH fraction from the SO in 1000 ug/mL for Fr-3 were shown to be 76.1% respectively. The $n$-BuOH fraction from the SO had high anti-microbial effect on $S.$ $mutans$. Conclusion : As a result, the $n$-BuOH fraction from SO. has good anti-microbial and anti-oxidant effects in a concentration-dependent manner.

작약 추출물의 구강병원균에 대한 항균성 및 구강암 세포 증식 억제효과 (Antimicrobial Activities Against Oral Microbes and Growth-inhibitory Effect on Oral Tumor Cell by Extract of Paeonia lactiflora)

  • Paeonia lactiflora was stepwise extracted with hexane, chloroform, ethyl acetate, butanol and water. Anti-microbial activity of each extract was investigated. Methanol extract of P. lactiflora revealed anti-microbial activity against S. mutans, C. albicans, and S. aureus. Also, hexane fraction revealed anti-bacterial activity against S. mutans and ethyl acetate fraction acted as potent anti-microbial agent on C. albicans and S. aureus. The relative growth ratio(RGR) of hexane fraction of P. lactiflora against S. mutans were determined as 77.8% in concentration of 0.125 mg/ml, 98.46% in 0.25 mg/ml and 100% in 0.5 mg/ml. The ethyl acetate fraction of P. lactiflora revealed RGR against C. albicans as 52.5% in concentration of 0.125 mg/ml, 60.83% in 0.25 mg/ml and 78.33% in 0.5 mg/ml. It indicate that increasing concentration increase RGR. The measured minimal inhibitory concentration(MIC) of hexane fraction on S. mutans KCTC 5316 strain was 0.5 mg/ml and MIC of ethyl acetate fraction on C. albicans KCTC 7270 was 2.0 mg/ml. The experiment of inhibition to growth of KB roll(oral squamous cell carcinoma) result 61.9% in butanol, 76.7% in hexane extract of P. lactiflora. The hexane extract exhibit potent inhibition effect to the growth of KB cell. These results suggest that the hexane extract of Paeonia lactiflora has antimicrobial activity against S. mutans and has preventive effect to dental caries in addition to potent inhibition to KB cell growth.

Anti-Biofilm Effect of Egg Yolk Phosvitin by Inhibition of Biomass Production and Adherence Activity against Streptococcus mutans

  • The formation of biofilms on the enamel surface of teeth by Streptococcus mutans is an important step in dental plaque formation, demineralization, and early caries because the biofilm is where other bacteria involved in dental caries attach, grow, and proliferate. The objectives of this study were to determine the effect of phosvitin (PSV) on the biofilm formation, exopolysaccharides (EPS) production, adherence activity of S. mutans, and the expression of genes related to the compounds essential for biofilm formation (quorum-sensing inducers and components of biofilm matrix) by S. mutans. PSV significantly reduced the biofilm-forming activity of S. mutans and increased the degradation of preformed biofilms by S. mutans. PSV inhibited the adherence activity of S. mutans by 31.9%-33.6%, and the production of EPS by 62%-65% depending upon the strains and the amount of PSV added. The expressions of genes regulating the production of EPS and the quorum-sensing-inducers (gtfA, gtfD, ftf, relA, vicR, brpA, and comDE) in all S. mutans strains were down-regulated by PSV, but gtfB was down-regulated only in S. mutans KCTC 5316. Therefore, the anti-biofilm-forming activity of PSV was accomplished through the inhibition of biofilm formation, adherence activity, and the production of quorum-sensing inducers and EPS by S. mutans.
