인삼 엑기스의 경구 면역 관용에 관한 연구 (Induction of Immunological Tolerance by Treatment of Ginseng Extract)

  • In order to develop new bioactive functions ginseng extract, it was studies whether the ginseng extracts on the induction of immunological tolerance In mice. Oral immunologic tolerance was induced by the secondary exposure of egg albumin + alum following gastrointestinal exposure nth egg albumin In mice, and the effect on anti EA antibody in blood, 7 cell subset in spleen were Investigated. The results obtained were as follows. EA group and EA + GE group was capable of conferring tolerance, contained a profound for 5 weeks experimental but saline group restricted to induce tolerance. GE group did not show the activity of tolerance by the first immunogens exposure, but induced the tolerance by the secondary exposure. And also spleen T cells, CD 8+ and CD 4+ were decreased. These results suggested that ginseng may affect the induction of immunological tolerance, which may be associated proliferative response of CD 4+ and CD 8+ in splenocyte.

마황의 면역작용에 미치는 효과 (Effect of Ephedrae Herba on the Immune Response in Mice)

  • The effects of Ephedrae herba on cellular and humoral immune responses were investigated in mice. Group A and B of mice received intraperitoneal injection of methanol extracts, ether fraction(E), petroleum ether fraction(PE), ethyl acetate fraction(EA) and water fraction(W) for 5 days or 10 days before sensitization. The other group received i.p. injection of ephedrine (Ep) or pseudoephedrine $({\psi}\;Ep)$ for 1 day prior to sensitization. The change of body and spleen weights showed a tendency of decreasing but that of thymus showed increasing. Ear swelling was maximum at 48 hours after challenge and was significantly decreased in the groups treated with EA, W, Ep, and ${\psi}\;Ep$. HA titer was enhanced in EA, W and EP treated groups, HE titer being significantly decreased.

Inhibitory Effect of Electroacupuncture on Murine Collagen Arthritis and its Possible Mechanisms

  • The influence of electroacupuncture (EA), a traditional Chinese medical treatment, on type Ⅱ collagen-induced arthritis (CIA) was examined in DBA/1J mice in vivo. Mice were immunized intradermally twice at the 3-week interval with bovine type Ⅱ collagen(C Ⅱ). EA stimulation, begun on the 21 simultaneously with the second immunization, was applied at the acupoint equivalent to GV4 three times a week for 3 weeks. The results showed that EA delayed the onset, attenuated the severity of arthritis, and reduced the anti-collagen antibody level. Furthermore, we investigated the impact of EA on the productions of endogenous $interleukin-1{\Beta}$ (IL-1 beta) and prostaglandin E2 (PGE2), and the levels of IL-1 beta mRNA in splenocytes and synovial tissues from C Ⅱ immunized mice on the 45 and cyclooxygenase-2 (COX-2) mRNA in lipopolysaccharide (LPS)-stimulated macrophages of normal mice by using reverse transcriptase-polymerase chain reaction (RT-PCR). EA stimulation significant inhibited the concentrations of splenic endogenous IL-1 beta and serum PGE2. The expression of IL-1 beta mRNA in spleen cells was obviously down-regulated and that in synovial tissues was modestly affected by EA. COX-2 mRNA was highly expressed in cultured peritoneal macrophages when stimulated with LPS. Previous treatment with EA also reduced LPS-stimulated induction of COX-2 mRNA. These data suggest that EA has an inhibitory effect on murine CIA, and the partial mechanism of its therapeutic result may be attributed to inhibiting the productions of IL-1 beta and PGE2 by suppression the IL-1 beta and COX-2 gene activations.
