우엉 뿌리 추출물의 최종당화산물 형성 억제 효능 (Inhibition of advanced glycation end product formation by burdock root extract)

  • 항산화, 항암 및 항염증 효능 등의 생리 활성이 보고된 우엉 뿌리의 열수 추출물을 제조하여 advanced glycation end products (AGEs) 형성 저해 효과를 확인하였다. AGEs는 bovine serum albumin (BSA)과 단당류인 glucose, fructose, galactose를 혼합하여 $37^{\circ}C$에서 3주간 배양하여 생성하였고 매주 형광도 측정, fructosamine 분석, ${\alpha}$-dicarbonyl compounds 함량 분석을 통해 AGEs의 생성량을 확인하였다. 우엉 뿌리 추출물의 AGEs 생성 저해능을 AGEs 생성 억제제로 알려져 있는 aminoguanidine (AG)의 저해능과 비교하였다. 우엉 뿌리 추출물은 BSA와 단당류인 glucose, fructose, galactose 각각의 당화반응을 억제하였으며 특히 배양 3주차에서 BSA와 glucose의 당화반응 결과물인 AGEs 생성을 유의적으로 저해하였다. 농도 2 mg/mL 이상의 우엉 뿌리 추출물은 1 mM AG보다 AGEs 생성 저해능이 우수하였으며 농도 4 mg/mL의 우엉 뿌리 추출물은 배양 3주차에서 AGEs 생성을 약 80% 이상 억제하는 효능을 나타냈다. 체내 혈당은 당뇨병과 같은 질환과 식이의 영향을 받으므로 다양한 glucose 농도에서 우엉 뿌리 추출물의 AGEs 생성 억제능을 조사하였다. 그 결과 AGEs 생성은 glucose의 농도에 비례하여 증가하였으며 우엉 뿌리 추출물은 당뇨환자의 식후에 관찰되는 혈당인 25 mM glucose 군에서 1 mM의 AG보다 높은 우수한 저해 효과를 확인하였다. 본 연구 결과는 우엉 뿌리 추출물의 AGEs 생성 억제라는 새로운 기능성을 밝히며 향후 당뇨 합병증 예방 효능을 가진 기능성 식품으로의 개발 가능성을 제시한다.

  • 피부의 주름은 노화과정의 결과로 유도되어지는데, 비효소적 glycation 반응은 주름생성의 원인 중에 하나로 단백질의 비가역적 가교결합에 의해 생성된 최종당화산물(AGEs)이라고 불리는 갈색화합물을 생성한다. 비교적 반감기가 긴 collagen은 쉽게 glycation이 일어나고 collagen의 가교결합(cross-linking)이 일어나면서 여러 단계의 반응을 거쳐 최종적으로 생성된 AGEs가 피부에 축적되어 피부의 탄력감소 및 주름을 일으킨다고 알려져 있다. 피부 내에 glycation된 collagen의 축척은 피부의 탄력성을 유지하는 것을 방해한다. 수종의 식용작물로부터 항주름소재를 개발하기 위하여 메탄올 및 열수추출물을 조제한 후 AGEs 형성을 억제하는 활성을 검색하였다. 먼저 서양향신료 중에서는 월계수잎, 계피, 정향, 오레가노, 로즈마리, 사보리, 팔각과 한약재 중에서는 복분자, 달개비, 삼지구엽초, 가자와 해당화의 메탄올 및 열수추출물에서 최종당화산물 생성 억제활성이 높았다. 그러나 국내 식물 중에서는 단지 참취 및 녹차 추출물의 메탄올과 열수추출물만이 높은 활성을 보였으며 해조류 중에서는 활성이 높은 시료는 없었다.

Carnosine and Anserine in Chicken: Distribution, Age-dependency and their Anti-glycation Activity

  • The imidazole dipeptide carnosine and its methylated anserine analogues are the major histidine containing dipeptides in vertebrate tissue, especially in skeletal muscle, the heart, and the central nervous system. In this study, the carnosine and anserine content in chicken from different parts and of differing ages was determined and their physiological activities were compared. Anserine was more dominant than carnosine in these tissues and both of them significantly decreased with aging in all parts of chicken muscles. Chicken breast muscle showed the highest content of carnosine and anserine than drumstick and wing. Advanced glycated end-product (AGE) formation was inhibited up to 60% by the extract from 20 wk chicken breast and decreased with aging (90 wk). Anti-oxidation activity was also significantly reduced from 61.2% to 52.9% with aging. As results, anti-glycation and anti-oxidation activity of carnosine and anserine extract from chicken muscle increased proportionally to the amount of those peptides in the muscle, while these decreased with the aging process.

Biological Activity and Inhibition of Non-Enzymatic Glycation by Methanolic Extract of Rosa davurica Pall. Roots

  • The methanolic extract of Rosa davurica Pall. roots exhibited strong antioxidant activity in a 1,1-diphenyl-2-picryl-hydrazyl (DPPH) radical scavenging assay and was found to be a dose-dependent inhibitor of non-enzymatic formation of advanced glycation end products (AGEs), which are relevant to diabetes complications. HPLC-diode array detector (DAD) analysis of the R. davurica Pall. root extract led to the identification of four compounds: hydrocaffeic acid, catechin, epicatechin, and ellagic acid. Catechin was present in the largest amount and exhibited high antiglycation activity. A CYP3A4 assay was used to investigate potential interactions between drugs and the extract, and results suggest that the R. davurica Pall. root extract had moderate potential for interfering with drug metabolism. The R. davurica Pall. extract did not display anti-inflammatory activity on the level of that for tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) in a lipopolysaccharide (LPS)-stimulated macrophage assay; however, the extract did exhibit low to moderate immunostimulatory activity in a pro-inflammatory macrophage assay. Therefore, we conclude that R. davurica Pall. root is a promising anti-AGE agent with low to moderate risks of associated inflammation or drug interaction.

In-Vitro, Anti-Bacterial Activities of Aqueous Extracts of Acacia catechu (L.F.)Willd, Castanea sativa, Ephedra sinica stapf and shilajita mumiyo Against Gram Positive and Gram Negative Bacteria

  • Objective: Evaluations of the in-vitro anti-bacterial activities of aqueous extracts of Acacia catechu (L.F.)Willd, Castanea sativa, Ephedra sinica stapf and Shilajita mumiyo against gram-positive bacteria (Staphylococcus aureus, Streptococcus pneumonia) and gram-negative bacteria (Escherichia coli, klebsiella pneumoniae, Proteus mirabilis, Pseudomonas aeruginosa) are reasonable since these ethnomedicinal plants have been used in Persian folk medicine for treating skin diseases, venereal diseases, respiratory problems and nervous disorders for ages. Methods: The well diffusion method (KB testing) with a concentration of $250{\mu}g/disc$ was used for evaluating the minimal inhibitory concentrations (MIC). Maximum synergistic effects of different combinations of components were also observed. Results: A particular combination of Acacia catechu (L.F.) Willd, Castanea sativa, Ephedra sinica stapf and shilajita mumiyo extracts possesses an outstanding anti-bacterial activity. It's inhibiting effect on microorganisms is significant when compared to the control group (P<0.05). Staphylococcus aureus was the most sensitive microorganism. The highest anti-bacterial activity against gram-positive bacteria (Staphylococcus aureus, Streptococcus pneumonia) or gram-negative bacteria (Escherichia coli, Klebsiella pneumonia, Proteus mirabilis, and Pseudomonas aeruginosa) was exerted by formula number 2 (table 1). Conclusion: The results reveal the presence of anti-bacterial activities of Acacia catechu, Castanea sativa husk, Ephedra sp. and Mumiyo against gram-positive and gram-negative bacteria. Synergistic effects in a combined formula, especially in formula number 2 (ASLAN$^{(R)}$) can lead to potential sources of new antiseptic agents for treatment of acute or chronic skin ulcers. These results considering the significant anti-bacterial effect of the present formulation, support ethnopharmacological uses against diarrheal and venereal diseases and demonstrate use of these plants to treat infectious diseases.

  • Postmenopausal women aged 50s generally experience gradual changes in body such as decline in antioxidant and estrogen levels as the body ages. To overcome these aging-associated changes, the needs for health functional foods are increasing. Dendropanax morbifera (DM) have antioxidant effects, anti-inflammatory against cancer cells, antidiabetic, and antiatherogenic effect which are associated with postmenopausal symptoms. We analyzed clinical effects of DM on aging-related symptoms by reporting their antioxidant, anticancer and inflammatory activity, etc. and their bioactivity. Data sources EMBASE, SCOPUS, PubMed, Web of Science, and Google Scholar databases were searched up to August 2016 for studies investigating medicinal plants in prevention and treatment of diabetes. The search terms were "Dendropanax morbifera". The reference lists of articles were also reviewed for additional relevant studies. Extracts of DM have various efficacy such as antioxidant, anti-cancer, anti-inflammatory activity and anti-thrombotic effect.

The Role of Intestinal Microflora in Anti-Inflammatory Effect of Baicalin in Mice

  • Baicalin, a main constituent of the rhizome of Scutellaria baicalensis, is metabolized to baicalein and oroxylin A in the intestine before its absorption. To understand the role of intestinal microflora in the pharmacological activities of baicalin, we investigated its anti-inflammatory effect in mice treated with and without antibiotics. Orally administered baicalin showed the anti-inflammatory effect in mice than intraperitoneally treated one, apart from intraperitoneally administered its metabolites, baicalein and oroxylin A, which potently inhibited LPS-induced inflammation. Of these metabolites, oroxylin A showed more potent anti-inflammatory effect. However, treatment with the mixture of cefadroxil, oxytetracycline and erythromycin (COE) significantly attenuated the anti-inflammatory effect of orally administered baicalin in mice. Treatment with COE also reduced intestinal bacterial fecal ${\beta}$-glucuronidase activity. The metabolic activity of human stools is significantly different between individuals, but neither between ages nor between male and female. Baicalin was metabolized to baicalein and oroxylin A, with metabolic activities of $1.427{\pm}0.818$ and $1.025{\pm}0.603$ pmol/min/mg wet weight, respectively. Baicalin and its metabolites also inhibited the expression of pro-inflammatory cytokines, TNF-${\alpha}$ and IL-$1{\beta}$, and the activation of NF-${\kappa}B$B in LPS-stimulated peritoneal macrophages. Of them, oroxylin A showed the most potent inhibition. Based on these findings, baicalin may be metabolized to baicalein and oroxylin A by intestinal microflora, which enhance its anti-inflammatory effect by inhibiting NF-${\kappa}B$ activation.

발모 향상을 위한 한약재의 개발 및 효과 확인 (Development and Evaluation of the Herbal Medicine for Hair Growth-promoting Activity)

  • Hair loss affects both men and women of all ages and often significantly affects social and psychologic health. Recent therapeutic approach for hair loss such as finasteride and minoxidil focused on regulation of hormonal system blood flow. However, long-term use of these drugs caused adverse effects. To develop herbal medicine for therapeutic effect on hair growth, here we screened the 10 medicinal herbs (Red ginseng, Licorice, Ulmus, Barberry root, Lycium root, Rehmanniae radix crudus, Sophora root, Sweet flag, Polygala root, Achyranthes) based on oriental medicine literature. We measured cytotoxicity, anti-oxidant activity, and $5-{\alpha}$ reductase inhibitory effect of the herbal medicine on human dermal papilla (DP) cells to investigate therapeutic effect of the herbal medicine. Treatment of the 1% herbal medicine did not show any cytotoxic effects, and cell growth was increased by treatment of the 0.1% herbal medicine. In addition, the herbal medicine showed stronger antioxidant activity than resveratrol and comparable inhibitory activity of $5-{\alpha}$ reductase with finasteride. Furthermore, when applied to in vivo mouse model, we also observed increases in the number and length of hair of the herbal medicine-treated group. These results suggest that the herbal medicine promotes hair growth by its antioxidant activity and inhibitory activity of $5-{\alpha}$ reductase and might therefore be a promising hair growth-promoting agent.

에르고티오네인의 당화 억제 및 항산화 활성에 관한 연구 (Glycation Inhibitory and Antioxidative Activities of Ergothioneine)

  • 에르고티오네인은 생체 내에서 우수한 항산화제로서 산화스트레스로부터 세포 보호제로 알려져 있다. 본 연구에서는 에르고티오네인의 라디칼 소거 활성은 자외선에 조사된 사람 섬유아세포 생존율을 향상 시키는 것을 확인하였으며, 세포노화 지표물질인 senescence-associated ${\beta}$-galactosidase (SA-${\beta}$-gal) 활성을 사람 섬유아세포를 이용하여 확인한 결과, 에르고티오네인 $400{\mu}M$ 처리 농도에서 염색된 세포의 수가 약 45%의 SA-${\beta}$-gal의 레벨이 감소하여 세포 노화(senescence)를 억제하는 것을 확인할 수 있었다. 또한, 에르고티오네인은 글리세르알데하이드에 의해 유도된 최종당화산물(AGEs) 생성을 저해하였으며, 카르복시메칠라이신(CML) 발현을 농도의존적으로 저해하는 것으로 나타내었다. 글리옥살에 의해 최종당화산물의 수용체(RAGE)가 발현된 사람 섬유아세포에 에르고티오네인을 처리하였을 때 RAGE의 발현이 농도의존적으로 감소하는 것을 확인하였다. 따라서, 본 연구결과를 바탕으로 에르고티오네인의 항노화 효과와 최종당화산물의 세포 내 축적을 저해할 수 있는 화장품 소재로서 활용가치가 있음을 확인하였다.

  • With the improvement of economic status and the desire for beauty, the interest in health and skin care is increasing. For these demands, since ages medicinal plants are in vogue. A variety of plants, cosmetics and foods with novel bioactive ingredients for skin care and beauty are under constant research and development. Skin is influenced by various factors such as Ultra-violet rays, stress, hormones and aging which together lead skin to lose elasticity, changes in pigmentation and wrinkle formation. Many medicinal plants have proven effects in skin care and beauty treatment. From this list of medicinal plants, one which is famous for its beauty, flavor and fragrance is Rosa damascene. Rosa damascene has many therapeutic action and postulated pharmacological studies such as anti-arthritic, anti-microbial, cardio protective, anti-inflammatory, antioxidant, analgesic, immune-modulator, gastro-protective, and skin ameliorative effect. Research in the field of Cosmetology has proven the effect of Rosa damascene in rehydrating skin, reducing scars and stretches, acne management, lowering skin pigmentation, delaying wrinkling and is recommended as a skin vitalizing agent. In this review, the morphology, chemical constituents, and some pharmacological activity are discussed.
