The role of phospholipids in the membrane binding and subsequent insertion of the microsomal protein rabbit cytochrome P450 (P450) lA2 into phospholipid bilayers was investigated. Path of a small hydrophobic molecule through the aqueous cytoplasma is not linear. Partition may favor membrane binding by several orders of magnitude : thus significant membrane association will markedly decrease the cytosolic transport rate. The presence of high concentration of soluble binding proteins for these hydrophobic molecules would compete with membrane association and thereby increase transport rate. For long chain fatty acid molecules, a family of cytosolic binding proteins collectively known as the fatty acid binding proteins(FABP), are thought to act as intracellular transport proteins. This paper examines the mechanism of transfer of fluorescent antyroyloxy-labeled fatty acids(AOFA) from purified FABPs to phosholipid membranes. With the exception of the liver FABP, AOFA is transferred from FABP by collisional interaction of the protein with a acceptor membrane. The rate of transfer increased markedly when membranes contain anionic phospholipids. This suggests that positively charged residues on the surface of the FABP may interact with the membranes. Neutralization of the surface lysine residues of adipocyte FABP decreased fatty acid transfer rate, and transfer was found to proceed via aqueous diffusion rather than collisional interaction. Site specific mutagenesis has further shown that the helix-turn-helix domain of the FABP is critical for interaction with anionic acceptor membranes. Thus cytosolic FABP may function in intracellular transport of fatty acid to decrease their membranes association as well as to target fatty acid to specific subcellular sites of utilization.
Positively charged cytochrome c interacts with the negatively charged mitochondrial inner membrane. This interaction induces conformational changes in bound cytochrome c. In order to estimate the effect of cytochrome c-membrane interaction on the mitochondrial electron transfer, we have investigated oxidation of ferrocytochrom c in the presence of anionic phospholipids.(omitted)
Dipalmitoyl phosphatidylcholine(DPPC)과 dipalmitoyl phosphatidic acid(DPPA) 리포좀내 이중층의 열적 상전이에 미치는 칼슘과 페노치아진 유도체의 영향을 시차 열량 분석계로 연구하였다. 인지질 이중층은 가열시 어느 특정온도에서 급격한 구조적 변화를 가져왔다. 이러한 온도에 의존한 변화는 순수 인지질의 경우 특이할 만큼 예민하였다. 순수 인지질에 페노치아진 유도체를 가하였을 때 순수 인지질의 상전이 온도보다 낮은 온도에서 일어 났으며 상전이 열그림을 넓적하게 만들었다. 칼슘 이온은 DPPC: DPPA(34: 66mol%)의 상전이 온도를 높였다. 그러나 위의 혼합 인지질에 페노치아진 유도체를 가하였을 때 $73^{\circ}C$에서의 작고 넓적한 곡선 부분이 사라졌다. 이러한 현상은 아마도 칼슘과 폐노치아진 유도체의 산성 인지질의 친수부분에서 상호 경쟁적 작용에 기인된 것으로 추정되어진다.
본 연구에서는 인지질계 양쪽성 계면활성제 CDP-W 첨가가 지질 소포체 막과의 상호 작용에 미치는 영향에 관하여 살펴보았다. 이를 위하여 CDP-W 양쪽성 계면활성제와 레시틴 S100-3의 임계 마이셀 농도 및 표면장력 등의 계면 물성을 측정하였다. 또한 pH 변화에 따른 1 wt% 계면활성제 수용액의 제타 전위 측정을 통하여 양쪽성 계면활성제 CDP-W가 양이온 계면활성제에서 음이온 계면활성제로 작용이 전환되는 등전점을 결정하였으며, 이 결과를 바탕으로 pH 변화 및 CDP-W 첨가가 리포좀의 평균 입자 크기, 다분산 지수 및 제타 전위 등과 같은 안정성에 미치는 영향에 살펴보았다. 또한 가장 안정한 상태의 리포좀이 형성되는 pH 6의 조건에서 CDP-W 첨가에 따른 리포좀 막의 형광 이방성, 변형성, 녹는 점 측정 등을 통하여 리포좀 막의 유동성 특성을 측정하고 리포좀 막의 유동성이 포집효율 및 안정성에 미치는 영향에 관하여 살펴보았다.
다가 양이온성 고분자인 polyethylenimine(PEI)를 이용한 plasmid DNA의 세포 전이를 검색했다. 먼저 agarose assay에 의한 2, 10, 25, 및 50KD PEI와 DNA의 중화복합체의 최적비율은 분자량에는 영향을 받지 않았고, 최적의 PEI nitrogen/DNA phosphate 중화 비율은 1.5-2.0(nmol/nmol)로 나타났다. 이 복합체들을 이용한 COS1 세포전이에서는 2KD를 제외하고는 naked DNA에 대비 전이가 증가했고, 이 중에서 특히 25KD PEI는 적정 전이조건에서 DEAE-dextran 혹은 lipofectin 보다 다소 증가된 전이율을 보였다. In vitro 세포전이의 최적 PEI/DNA 비는 7.6-13.3(nmol/nmol)이었고 최적 중화복합체를 이루는 비율보다 높게 나타났다. 용액의 pH에 따른 전이율의 변화는 크게 없었으나 산성일때가 약간 더 증가했다. 세포 표적전이와 독성감소를 위해 인지질분자를 사용한 liposome formulation을 PEI/DNA계에 도입하였다. 그 결과, PC/PE 중성 리포솜이 도입된 경우는 25KD를 제외하고는 PEI 단독일 때 혹은 리포솜 단독일 때 보다 전이율이 2-2.5 배씩 증가했다. 그러나 PEI와 같은 양이온성의 DOTAP/PE 리포솜 도입은 charge repulsion 작용으로 오히려 DOTAP/PE 단독계보다 전이가 감소하는 역효과를 보였다. Liposomal PEI계의 세포독성은 PEI 단독일 때 보다 % cell survival이 10-20% 정도 증가했다. 이 결과들은 PEI가 단독으로도 좋은 전이제로 작용 할 뿐 아니라 세포표적 운반이 가능한 중${\cdot}$음성 리포솜의 효과적인 DNA 응축제로도 이용 될 수 있음을 증명했다.
